UNLABELLED: Polygraphic sleep recording was performed in 20 narcoleptics with one or more of the auxiliary symptoms, 4 narcoleptics with only sleep attacks and 10 normals during one night and into the following day. Total sleep time in the narcoleptics did not differ significantly from that in the normals. Sleep of the narcoleptics with auxiliary symptoms was unstable with frequent awakening. The temporal organization of the REM--NREM sleep cycle was irregular in the narcoleptics with auxiliary symptoms, compared with those in the other two groups. Percentage of Stage 1 was significantly large

This article summarizes the key literature describing the effects of the pancreatic beta-cell hormone amylin on eating. One of the first described and best investigated effects of amylin on eating is its physiological effect to control meal size by inducing satiation. This effect is very rapid, short-lasting and probably directly reflects the meal-induced increase in circulating amylin levels. Evidence provided by many groups suggests that the effect of amylin on eating is directly mediated by humoral action in the central nervous system rather than by peripheral receptors. It is also clear th

Bulimia nervosa (BN) is characterized by the recurrent consumption of excessive amounts of food (binge eating) followed by inappropriate compensatory behaviors. A leading hypothesis is that the persistence of BN may be due, in part, to a disturbance in the development of satiation. Because patients with BN consume larger meals than controls, previous studies have not been able to directly compare the development of satiation. In order to address this problem, subjects consumed large meals of predetermined size without knowing when they would be stopped. Twenty-one women with BN and 13 control

Leptin is a hormone secreted into the systemic blood primarily by white adipose tissue. However, leptin also is synthesized and stored by cells in the gastric mucosa. Because gastric mucosal leptin is secreted in response to ingestion of a meal, we hypothesized that it might contribute to satiation (meal termination) by acting on gastrointestinal vagal afferent neurons. To test whether leptin is capable of acutely reducing short-term food intake, we measured consumption of a liquid meal (15% sucrose) following low-dose leptin administration via the celiac artery, which perfuses the upper gastr

The brain is the seat of body weight homeostasis. However, our inability to control the increasing prevalence of obesity highlights a need to look beyond canonical feeding pathways to broaden our understanding of body weight control1-3. Here we used a reverse-translational approach to identify and anatomically, molecularly and functionally characterize a neural ensemble that promotes satiation. Unbiased, task-based functional magnetic resonance imaging revealed marked differences in cerebellar responses to food in people with a genetic disorder characterized by insatiable appetite. Amylin plays an important role in the control of nutrient fluxes. It is cosecreted with insulin and reduces eating by promoting meal-ending satiation. This effect seems to depend on a stimulation of amylin receptors in the area postrema. Subsequent to area postrema activation, the neural signal is conveyed to the forebrain via distinct relays in the nucleus of the solitary tract and the lateral parabrachial nucleus to the lateral hypothalamic area and other hypothalamic nuclei; the functional roles of these relays in amylin's eating inhibitory effect have not been fully investigated. This review summarizes the present knowledge concerning the anorectic action of the pancreatic hormone amylin. It focuses mainly on the role of amylin as a short term satiating peptide. Since there is some evidence however that basal amylin levels might play a role in the long term control of food intake and/or body weight, this aspect will be discussed briefly towards the end of this review. Concerning amylin as a satiating hormone, it is well established that amylin is released during meals, and that exogenous amylin leads to a dose-related reduction in meal size. Amylin has a rapid onset an

OBJECTIVE: The objective of this study was to validate the use of impedance for measurement of antral contractions and to determine the relationship between food-induced changes in gastric motility and satiation.
RESEARCH METHODS AND PROCEDURES: In Experiment 1, three dogs were implanted with an antral strain gauge and bipolar electrodes for measurement of local tissue impedance. Impedance and strain gauge recordings were obtained simultaneously during antral contractions to correlate impedance changes with contractile events. In Experiment 2, seven dogs were implanted with two pairs of gastr

Two single-case experiments demonstrated the efficacy of satiation therapy with adult males who had long-standing deviant sexual interests. The procedure involves the pairing of prolonged masturbation (1 hour) with the verbalization by the patient of his deviant sexual fantasies and in both cases the designs permitted the attribution of control over aberrant responding to the satiation therapy. The results are discussed in terms of the possible active ingredients of the procedure.

Hunger is evolutionarily hardwired to ensure that an animal has sufficient energy to survive and reproduce. Just as important as knowing when to start eating is knowing when to stop eating. Here, using spatially resolved single-cell phenotyping, we characterize a population of neuropeptidergic neurons in the brainstem's dorsal raphe nucleus (DRN) and describe how they regulate satiation. These neurons track food from sensory presentation through ingestion, integrate these signals with slower-acting humoral cues, and express cholecystokinin (CCK). These CCK neurons bidirectionally regulate meal

Satiation procedures involving increased food intake recently have been used to reduce rates of ruminative vomiting in mentally retarded children. Use of these nonaversive intervention procedures has been limited almost exclusively to clients who are seriously underweight since excessive weight gain is a common side effect. The purpose of the present study was to determine whether modified satiation, provided by unlimited quantities of low calorie foods, could decelerate ruminative vomiting to clinically acceptable rates without producing undesirable weight gain. The two subjects were a severe

Neuropeptides have important roles in neural plasticity, spiking and behaviour1. Yet, many fundamental questions remain regarding their spatiotemporal transmission, integration and functions in the awake brain. Here we examined how MC4R-expressing neurons in the paraventricular nucleus of the hypothalamus (PVHMC4R) integrate neuropeptide signals to modulate feeding-related fast synaptic transmission and titrate the transition to satiety2-6. We show that hunger-promoting AgRP axons release the neuropeptide NPY to decrease the second messenger cAMP in PVHMC4R neurons, while satiety-promoting POM

BACKGROUND: Asimadoline, a kappa-opioid agonist, reduces visceral sensitivity in experimental animal models and may decrease satiation and postprandial fullness in healthy individuals. However, its effect on satiation in functional dyspepsia is unclear, and any symptom benefit has not been explored.
AIM: To evaluate the effects of asimadoline on satiation volume and postchallenge symptoms in functional dyspepsia.
METHODS: A randomized, double-blind trial evaluated gastric satiation and symptoms before and after 8 weeks of asimadoline 0.5 mg (n = 13) or 1.0 mg (n = 13) or placebo (n = 14) b.d

The effects of food satiation on rumination and a collateral self-stimulatory behavior were examined in three profoundly retarded individuals. For all three individuals, the provision of unlimited quantities of cereal and milk during mealtime resulted in reductions in rumination. Decreases were also seen in collateral behaviors for all three subjects, although these responses were not specifically treated. Rumination and self-stimulation increased during a withdrawal condition for the three individuals, with experimental control being regained once the satiation procedure was re-instituted. Fa

Studying communication between the gut and the brain is as relevant and exciting as it has been since Pavlov's discoveries a century ago. Although the efferent limb of this communication has witnessed significant advances, it is the afferent, or sensory, limb that has recently made for exciting news. It is now clear that signals from the gut are crucial for the control of appetite and the regulation of energy balance, glucose homeostasis, and more. Ghrelin, discovered just a few years ago, is the first gut hormone that increases appetite, and it may be involved in eating disorders. The stable

The factors influencing appetite in humans are poorly understood. There is a weak relation between appetite and gastric emptying in normal subjects. Recent studies have shown that fasting and postprandial antral areas increase in patients with functional dyspepsia compared with normal subjects. We evaluated the hypothesis that antral area, and hence antral distention, is a significant determinant of postprandial fullness. Fourteen normal subjects had simultaneous measurements of gastric emptying by scintigraphy and antral area by ultrasound after ingestion of 350 mL 20% glucose. Fullness and h

Satiety signals arising from the gastrointestinal (GI) tract and related digestive organs during food ingestion and digestion are conveyed by vagal sensory afferents to the hindbrain nucleus of the solitary tract (NST). Two intermingled but chemically distinct NST neuronal populations have been implicated in meal size control: noradrenergic (NA) neurons that comprise the A2 cell group, and glucagon-like peptide-1 (GLP-1)-positive neurons. Previous results indicate that A2 neurons are activated in a meal size-dependent manner in rats that have been acclimated/entrained to a feeding schedule in

Despite advances in elucidating obesity pathophysiology, predicting individual responses to weight loss interventions remains challenging. Cifuentes et al.1 developed a predictive model integrating genetic risk scores and anthropometric parameters to estimate caloric intake to satiation, demonstrating potential in forecasting weight loss trajectories with phentermine-topiramate and liraglutide therapies.
