Senotherapeutics for Knee Osteoarthritis.

Osteoarthritis (OA) is a chronic disease that imposes a significant economic burden and deteriorates quality of life. Nevertheless, current therapeutic options for OA are limited to symptomatic remedies. As such, there is a high interest in novel methods for treating or preventing OA. One of the most promising medication modalities is through the clearance of cells that are cell cycle arrested but resistant to apoptosis, termed senescent cells. Additionally, these cells are also resistant to alternative programmed cell death modes, such as ferroptosis and pyroptosis. Senescent cells tend to ac

Wound Healing, Senescence, and Senotherapeutics.

SIGNIFICANCE: Dermal repair is a tightly regulated process that becomes progressively impaired with aging, chronic diseases, and persistent tissue injury. Senescent cells (SnCs) accrue in the skin following damage and with advancing age, contributing to alterations in tissue homeostasis and repair. However, the role of SnCs in normal and impaired wound repair is pleiotropic and context-dependent, particularly in the context of age and chronic disease. Furthermore, the use of senotherapeutics that target SnCs can slow aging, mitigate chronic disease, and promote rejuvenation of aged tissues, im

Senotherapeutics in Cancer and HIV.

Cellular senescence is a stress-response mechanism that contributes to homeostasis maintenance, playing a beneficial role during embryogenesis and in normal adult organisms. In contrast, chronic senescence activation may be responsible for other events such as age-related disorders, HIV and cancer development. Cellular senescence activation can be triggered by different insults. Regardless of the inducer, there are several phenotypes generally shared among senescent cells: cell division arrest, an aberrant shape, increased size, high granularity because of increased numbers of lysosomes and va

Cellular senescence and senotherapeutics in cardiovascular diseases.

Cellular senescence (CS) is characterized by stable cell cycle arrest and is resistant to growth-promoting stimuli allied with aging. Cardiac senescent cells (SCs) are highly heterogeneous cells that can regulate the pathophysiology of cardiovascular diseases (CVDs). SCs accumulate in the cardiovascular system, leading to typical age-related cardiovascular conditions. Such conditions advance in cardiovascular pathologies, including heart failure, coronary artery disease, cardiac fibrosis, etc., by evocating the production of proinflammatory mediators and profibrotic senescence-associated secre

Mitochondria-targeted senotherapeutic interventions.

Healthy aging is the art of balancing a delicate scale. On one side of the scale, there are the factors that make life difficult with aging, and on the other side are the products of human effort against these factors. The most important factors that make the life difficult with aging are age-related disorders. Developing senotherapeutic strategies may bring effective solutions for the sufferers of age-related disorders. Mitochondrial dysfunction comes first in elucidating the pathogenesis of age-related disorders and presenting appropriate treatment options. Although it has been widely accept

Revolutionizing cancer treatment with senotherapeutics: a current perspective.

Cellular senescence is a double-edged sword in cancer biology, initially acting as a tumor-suppressive mechanism but later contributing to cancer progression and therapy resistance. Senescent cells, characterized by stable cell cycle arrest, secrete a complex array of bioactive molecules known as the senescence-associated secretory phenotype (SASP), which fosters chronic inflammation, disrupts tissue architecture, and promotes tumorigenesis through paracrine signaling. Accumulation of these cells in the tumor microenvironment can enhance malignancy, drive metastasis, and impair treatment outco

Cellular Senescence in Human Skin Aging: Leveraging Senotherapeutics.

BACKGROUND: As the largest organ in the human body, the skin is continuously exposed to intrinsic and extrinsic stimuli that impact its functionality and morphology with aging. Skin aging entails dysregulation of skin cells and loss, fragmentation, or fragility of extracellular matrix fibers that are manifested macroscopically by wrinkling, laxity, and pigmentary abnormalities. Age-related skin changes are the focus of many surgical and nonsurgical treatments aimed at improving overall skin appearance and health. SUMMARY: As a hallmark of aging, cellular senescence, an essentially irreversibl

Senotherapeutic potential against xeroderma pigmentosum.

BACKGROUND: Xeroderma pigmentosum (XP) is an inherited photoaging syndrome caused by mutations in genes involved in the nucleotide excision repair (NER) pathway. XP patients exhibit hypersensitivity to ultraviolet (UV) radiation, leading to accelerated skin aging and requiring lifelong sun avoidance. In this study, we aimed to investigate UV-induced cellular senescence in XP melanocytes and explore potential senotherapeutic strategies. METHODS: Melanocytes differentiated from iPSCs derived from an XP-A patient and a healthy individual, respectively, were used to induce senescence. Senescence

Fisetin is a senotherapeutic that extends health and lifespan.

BACKGROUND: Senescence is a tumor suppressor mechanism activated in stressed cells to prevent replication of damaged DNA. Senescent cells have been demonstrated to play a causal role in driving aging and age-related diseases using genetic and pharmacologic approaches. We previously demonstrated that the combination of dasatinib and the flavonoid quercetin is a potent senolytic improving numerous age-related conditions including frailty, osteoporosis and cardiovascular disease. The goal of this study was to identify flavonoids with more potent senolytic activity. METHODS: A panel of flavonoid

Senotherapeutic approach to age-related endocrine diseases.

Senescent cells progressively accumulate in the endocrine glands and their target tissue during the biological aging process. Senescence leads to hormonal imbalances contributing to various age-related endocrine diseases (AREDs). Cellular senescence, characterized by irreversible cell-cycle arrest, becomes more prevalent in advanced age, and the senescent cells release pro-inflammatory and pro-fibrotic factors, exacerbating endocrine dysregulation. Senescence-associated secretory phenotype (SASP) contributes to the pathogenesis of AREDs such as metabolic syndrome, sarcopenia, osteoporosis, and

Is there a role for senotherapeutics in the treatment of giant cell arteritis?

Giant cell arteritis (GCA) is a large-vessel vasculitis predominantly affecting individuals over 50 years, with a peak incidence between 70 and 80 years. Its pathogenesis involves complex immunological cascades triggered by vascular dendritic cell activation, recruitment of CD4⁺ T cells and monocytes, T helper lymphocyte polarization, and vascular remodeling driven by myofibroblasts and fibroblasts. Growing evidence implicates cellular senescence (particularly in vascular smooth muscle cells, fibroblasts, endothelial cells, and immune cells) in the pathophysiology of GCA. Senescent cells (SC)

A novel senotherapeutic strategy with azithromycin for preventing endometriosis progression.

BACKGROUND: Endometriosis is an estrogen-dependent chronic inflammatory disease, however the mechanisms underlying inflammation remain unclear. Non-hormonal drugs that can prevent endometriosis progression and resolve endometriotic infertility are urgently required. We thus focused on cellular senescence as a novel feature of endometriosis. Senescent cells cause chronic inflammation via the senescence-associated secretory phenotype (SASP) factor. It has been reported the effects of senolysis for various diseases in recent years. The aim of this study was to validate the involvement of cellular

Endothelial TLR4 at the Crossroads of Vascular Senescence and Targeted Senotherapeutics.

Traditionally viewed as a classic sentinel of innate immunity, Toll-like receptor 4 (TLR4) has emerged as a crucial pathological driver within the vascular endothelium, with its identity dictated by cell-type specificity, microenvironmental context, and fluctuating expression levels. This review critically examines the dual functional nature of endothelial TLR4, delineating how it transitions into a primary driver of vascular aging and organ-specific vascular pathologies. The profound spatial, temporal, and cellular heterogeneities of TLR4 signaling expose the fundamental inadequacies of conve

Emerging strategies in senotherapeutics: from broad-spectrum senolysis to precision reprogramming.

Cellular senescence, originally described as a finite proliferative arrest in cultured somatic cells, has since been recognized as a central mechanism underlying aging and the development of age-associated disorders. The progressive accumulation of senescent cells (SnCs) promotes chronic inflammation through the senescence-associated secretory phenotype (SASP) and circumvents immune-mediated clearance by upregulating pro-survival and immune checkpoint pathways. Early "first-generation" senolytics, including navitoclax (ABT-263) and the dasatinib-quercetin (D + Q) combination, provided proof-of

Cellular senescence and inflammageing: from mechanisms to senotherapeutic interventions.

Cellular senescence is a context-dependent cellular state characterised by persistent cell-cycle arrest, epigenetic remodelling, metabolic reprogramming and acquisition of a senescence-associated secretory phenotype. Transient senescence contributes to embryogenesis, tissue repair and tumour suppression, whereas persistent senescent cell populations accumulate with advancing age across multiple tissues, in part owing to declining immune-mediated clearance and intrinsic resistance to apoptosis, thereby promoting chronic systemic inflammation, tissue fibrosis, stem-cell dysfunction and propagati

Senotherapeutic drug treatment ameliorates chemotherapy-induced cachexia.

Cachexia is a debilitating skeletal muscle wasting condition for which we currently lack effective treatments. In the context of cancer, certain chemotherapeutics cause DNA damage and cellular senescence. Senescent cells exhibit chronic activation of the transcription factor NF-κB, a known mediator of the proinflammatory senescence-associated secretory phenotype (SASP) and skeletal muscle atrophy. Thus, targeting NF-κB represents a logical therapeutic strategy to alleviate unintended consequences of genotoxic drugs. Herein, we show that treatment with the IKK/NF-κB inhibitor SR12343 during a c

Targeting cellular senescence through nutritional senotherapeutics in brain aging and neurodegenerative diseases.

Cellular senescence has emerged as a key contributor to brain aging and neurodegenerative diseases, which are increasing in prevalence as life expectancy rises. Senescent cells accumulate in multiple brain cell populations and secrete the senescence-associated secretory phenotype (SASP), promoting chronic neuroinflammation, mitochondrial dysfunction, blood-brain barrier disruption, and progressive neuronal damage. This review examines the role of cellular senescence in brain aging and neurodegenerative diseases and discusses senotherapeutic strategies aimed at either eliminating senescent cell

Senotherapeutic strategies for ovarian aging: Mechanistic insights and translational perspectives.

Ovarian aging manifests as progressive depletion of the primordial follicle reserve and declining oocyte quality, culminating in menopause and associated systemic comorbidities. Cellular senescence emerges as a central driver of this process, exerting effects through cell-cycle arrest pathways and a proinflammatory senescence-associated secretory phenotype (SASP) that disrupts the follicular niche and stromal architecture. However, current senolytic and senomorphic interventions for ovarian aging remain largely at the mechanistic and preclinical stages, and their translational potential in hum
