Deposition of Gadolinium in the Central and Peripheral Nervous Systems and Its Effects on Sensory, Cognitive, and Athletic Implications after Multiple Injections of Gadolinium-Based Contrast Agents in Rats.

BACKGROUND AND PURPOSE: After repeat administration of gadolinium-based contrast agents (GBCAs), the association between gadolinium retention in the central and peripheral nervous systems and the main manifestations of myelopathy and progressive neurologic symptoms remains unclear. We investigated the effects of the repeat administration of GBCAs on gadolinium retention in the central and peripheral nervous systems and the sensory, cognitive, and athletic implications. MATERIALS AND METHODS: Forty-eight male Wistar rats (6 weeks of age) were randomly divided into 4 experimental groups (12 rat

Paclitaxel causes degeneration of both central and peripheral axon branches of dorsal root ganglia in mice.

BackgroundPeripheral neuropathy is a common and dose-limiting side effect of many cancer chemotherapies. The taxane agents, including paclitaxel (Taxol(®)), are effective chemotherapeutic drugs but cause degeneration of predominantly large myelinated afferent sensory fibers of the peripheral nervous system in humans and animal models. Dorsal root ganglia (DRG) neurons are sensory neurons that have unipolar axons each with two branches: peripheral and central. While taxane agents induce degeneration of peripheral axons, whether they also cause degeneration of central nervous system axons is not

The Use of Capsaicin in the Treatment of Swallowing Disorders.

Swallowing disorders are a common clinical issue that significantly impacts patients' quality of life and nutritional intake. Recent years have seen a growing interest in capsaicin, a natural compound, due to its potential neuroprotective effects and its ability to stimulate the swallowing reflex. This review summarizes the research progress on the application of capsaicin in the treatment of swallowing disorders, detailing its mechanisms of action, clinical applications, and future research directions. By doing so, this paper aims to provide insights into the potential of capsaicin as a thera

The influence of turbulence on the sensory basis of rheotaxis.

Rheotaxis is a widespread behavior with many potential benefits for fish and other aquatic animals, yet the sensory basis of rheotaxis under different fluvial conditions is still poorly understood. Here, we examine the role that vision and the lateral line play in the rheotactic behavior of a stream-dwelling species (Mexican tetra, Astyanax mexicanus) under both rectilinear and turbulent flow conditions. Turbulence lowered the flow speed at which threshold levels of rheotactic performance were elicited, an effect that was independent of sensory condition. Compared to fish with access to visual

The olfactory system and its disorders.

The sense of smell is greatly underappreciated, despite the fact that it monitors the intake of airborne agents into the human respiratory system and determines to a large degree the flavor and palatability of foods and beverages. In addition to enhancing quality of life, this primary sensory system warns of spoiled foods, leaking natural gas, polluted air and smoke, and mediates basic elements of communication (e.g., mother-infant interactions). It is now apparent that smell dysfunction is among the first clinical signs of such neurodegenerative diseases as Alzheimer's disease and sporadic Pa

[Cholinergic systems in bronchospasm].

A neuronal cholinergic system is widely distributed in larger airways, particularly around bifurcations. These neurons form efferent pathways from the brainstem ending in the motor end-plate on smooth muscle, and ciliated and mucus-producing cells. Impulses originating in various sensory endings in the airways, from the central nervous system and from many sensory receptors throughout the body, influence the activity of the cholinergic system thereby controlling the patency of the airways and modulating the rate and depth of breathing. A non-neuronal cholinergic system present on mast cells fa

Experimental animal models of drug-induced sensorineural hearing loss: a narrative review.

OBJECTIVE: This narrative review describes experimental animal models of sensorineural hearing loss (SNHL) caused by ototoxic agents. BACKGROUND: SNHL primarily results from damage to the sensory organ within the inner ear or the vestibulocochlear nerve (cranial nerve VIII). The main etiology of SNHL includes genetic diseases, presbycusis, ototoxic agents, infection, and noise exposure. Animal models with functional and anatomic damage to the sensory organ within the inner ear or the vestibulocochlear nerve mimicking the damage seen in humans are employed to explore the mechanism and potentia

Cough: The Emerging Role of the TRPA1 Channel.

Sneezing, cough, mucus secretion, and bronchoconstriction represent the main components of a coordinated and efficient reaction direct to expel or neutralize irritant agents from the respiratory system. A dense network of sensory nerves localized from the nose to the lower airways beneath the epithelium subserves this function. A variety of receptors and channels present in sensory nerve terminals by sensing irritant stimuli activate the system in emergence and initiate protective reflex responses, including cough. Previous and recent literature highlights the prominent role of some transient

Capsaicin and the stomach. A review of experimental and clinical data.

Capsaicin, the pungent principle of hot pepper, because of its ability to excite and later defunctionalize a subset of primary afferent neurons, has been extensively used as a probe to elucidate the function of these sensory neurons in a number of physiological processes. In the rat stomach, experimental data provided clear evidence that capsaicin-sensitive (CS) sensory nerves are involved in a local defense mechanism against gastric ulcer. Stimulation of CS sensory nerves with low intragastric concentrations of capsaicin protected the rat gastric mucosa against injury produced by different ul

Capsaicin in the Management of Peripheral Neuropathy: A Review.

PURPOSE OF REVIEW: The purpose of this review is to provide an update on the efficacy and safety of capsaicin in the treatment of neuropathic pain. RECENT FINDINGS: Capsaicin can provide prolonged relief from pain in various neuropathies. One proposed mechanism is the ability to cause degeneration of hypersensitized nerve tissue and incite regeneration of healthy nerve fibers. Capsaicin provides more significant relief than placebo and is non-inferior to pregabalin in the treatment of peripheral neuropathy. It is emerging as an alternative but effective and well-tolerated treatment for neurop

Local therapeutic strategies for neurocutaneous dysesthesia: from capsaicin to cannabinoids.

BACKGROUND: Neurocutaneous dysesthesia (ND) is characterized by abnormal skin sensations such as pruritus or burning without an underlying skin condition. It is caused by nerve damage or compression. Location and extent of symptoms vary greatly depending on the affected somatosensory site(s). Treatment is challenging and step-by-step guidelines are lacking. Local therapy offers a valuable first step, especially in mild cases or when systemic agents are contraindicated. Although numerous studies have described a variety of local treatment options for ND, no clear overview of these options exist

A behavior-environment information loop drives sensory navigation.

As organisms navigate the environment to locate critical resources, their behavioral actions must be tightly coupled to their sensory inputs. Here, we introduce an information-theoretic framework that quantifies this coupling using transfer entropy, which measures information flow between sensory inputs and behavioral outputs. Information flow from sensory inputs to behavior defines a "reactive" component of a navigational strategy, whereas information flow from behavior to sensory inputs defines an "active" component, whereby actions shape subsequent sensory experiences. Analyzing these bidir

Axonal PPARγ promotes neuronal regeneration after injury.

PPARγ is a ligand-activated nuclear receptor best known for its involvement in adipogenesis and glucose homeostasis. PPARγ activity has also been associated with neuroprotection in different neurological disorders, but the mechanisms involved in PPARγ effects in the nervous system are still unknown. Here we describe a new functional role for PPARγ in neuronal responses to injury. We found both PPAR transcripts and protein within sensory axons and observed an increase in PPARγ protein levels after sciatic nerve crush. This was correlated with increased retrograde transport of PPARγ after injury

Sensory, parasympathetic, and sympathetic neural influences in the nasal mucosa.

Neural mechanisms contribute to many nasal symptoms and syndromes. Sensory nerve stimulation by irritants, mast cell products, and inflammatory mediators leads to sneezing and other systemic reflexes. Parasympathetic reflexes and sensory axon responses combine to increase nasal blood flow, fill venous sinusoids (which thickens the mucosa and reduces nasal patency), induce plasma extravasation, and stimulate glandular secretion of mucous and serous cell products. These putative roles for nerves and neuropeptides in pathologic events open new therapeutic avenues. Anticholinergic agents, peptide

Capsaicin-sensitive intestinal mucosal afferent mechanism and body fat distribution.

This report summarizes clinical and experimental data in support of the hypothesis that capsaicin-sensitive intestinal mucosal afferent mechanism plays a role in regulating body fat distribution. Epidemiological data have revealed that the consumption of foods containing capsaicin is associated with a lower prevalence of obesity. Rural Thai people consume diets containing 0.014% capsaicin. Rodents fed a diet containing 0.014% capsaicin showed no change in caloric intake but a significant 24% and 29% reduction in the visceral (peri-renal) fat weight. Increase in intestinal blood flow facilitate

Sensory innervation shapes cancer immunotherapy efficacy and adverse events.

Sensory neurons innervate the tumor microenvironment (TME) and are active contributors to tumor progression and immune evasion. Across multiple cancer types, higher intratumoral nerve density is associated with worse clinical outcomes, including increased lymph node metastasis, local recurrence, and reduced survival. This relationship is driven, at least in part, by local immunosuppression arising from neuropeptide and neurotransmitter paracrine signaling within the TME. Nociceptive sensory neurotransmission impacts tumor-infiltrating immune cells to promote T-cell exhaustion, mast cell degran

Nanowired drug delivery to enhance neuroprotection in spinal cord injury.

Spinal cord injury (SCI) is a serious clinical situation for which no suitable drug therapy exists. SCI often results in paraplegia or quadriplegia and, apart from the personal trauma leads to huge costs to society for rehabilitation or day-to-day life support. Sensory motor dysfunction following SCI is mainly a consequence of the slowly progressing cord pathology after primary injury that worsens over tine. Thus, almost all sensory and motor nerve control and pathways passing through spinal cord and reflexes are compromised in SCI patients. As a result their peripheral nervous system, autonom

Bortezomib alters microtubule polymerization and axonal transport in rat dorsal root ganglion neurons.

Bortezomib is part of a newer class of chemotherapeutic agents whose mechanism of action is inhibition of the proteasome-ubiquitination system. Primarily used in multiple myeloma, bortezomib causes a sensory-predominant axonal peripheral neuropathy in approximately 30% of patients. There are no established useful preventative agents for bortezomib-induced peripheral neuropathy (BIPN), and the molecular mechanisms of BIPN are unknown. We have developed an in vitro model of BIPN using rat dorsal root ganglia neuronal cultures. At clinically-relevant dosages, bortezomib produces a sensory axonopa

Current understanding of the molecular mechanisms of chemotherapy-induced peripheral neuropathy.

Chemotherapy-induced peripheral neuropathy (CIPN) is the most common off-target adverse effects caused by various chemotherapeutic agents, such as cisplatin, oxaliplatin, paclitaxel, vincristine and bortezomib. CIPN is characterized by a substantial loss of primary afferent sensory axonal fibers leading to sensory disturbances in patients. An estimated of 19-85% of patients developed CIPN during the course of chemotherapy. The lack of preventive measures and limited treatment options often require a dose reduction or even early termination of life-saving chemotherapy, impacting treatment effic
