A rare case of human subconjunctival setariasis due to Setaria equina infection is reported herein. A 15-years old girl was referred with a 24h history of edema and redness in her left eye. On slit lamp examination, a thread-like cylindrical worm was moving in the subconjunctival area. The worm was extracted, stained and measured 110mm in length 510μm in width. The isolated worm was identified as adult female S. equina based on morphometric criteria. Identification of the species of the worm was confirmed using molecular methods. For this purpose, the 12S rRNA gene was PCR-amplified and the pu

The authors describe a trap utilized in the field to catch possible intermediate hosts of Setaria labiatopapillosa attracted by naturally infected bovine. The trap was bulky and stable but also decomposable and removable. It was tested in an area of Friuli Venezia Giulia (Udine province, NE Italy) during 21 nights in the summer 1994 (29/6-29/9) and allowed to catch 16159 mosquitoes and 218 other Diptera, showing a very good efficiency.

BACKGROUND: Ocular setariasis is an ectopic infection caused by a parasite under the genus Setaria. Adult worms belong to the Setariidae family and typically reside in the peritoneal cavity of ungulates. However, immature forms of these species may aberrantly migrate to the eyes of cattle, buffalo, goats, horses and several other hosts, leading to corneal opacity and blindness. Here, we have distinguished the Setaria digitata collected from both equine and buffalo hosts based on the morphology, molecular profiling of mitochondrial cytochrome c oxidase subunit 1 (Cox1), cytochrome c oxidase sub

Equine ocular setariasis is mainly caused by Setaria digitata, and the identification of this filarial nematode is based on morphology. However, morphological characterization alone is insufficient for the detection and differentiation of S. digitata from its congeners. In Thailand, the molecular detection of S. digitata is lacking and its genetic diversity is still unknown. This study aimed to phylogenetically characterize equine S. digitata from Thailand based on sequences derived from the mitochondrial cytochrome c oxidase subunit 1 (COI), the mitochondrial small subunit ribosomal DNA (12S

An adult Indian buffalo (Bubalus bubalis) presented with corneal opacity, irritation, and excessive lacrimation from the left eye in the Referral Veterinary Polyclinic-Teaching Veterinary Clinical Complex (RVC-TVCC), Indian Veterinary Research Institute, Izatnagar. Clinical examination revealed a whitish thread-like worm in the left eye's anterior chamber. The worm was surgically removed from the eye with supportive nerve blocks. Light microscopy was used for parasite morphological identification, which provided insight into the worm as female Setaria sp. Genomic DNA was isolated, and polymera

Three Shiba goats aged 1 to 7 years kept in Ibaraki prefecture in Japan were presented with chief complaint of lumbar paralysis or gait abnormalities. As cerebrospinal setariasis were suspected in all cases at the first stage, ivermectin was administered to treat, but the response was insufficient. Necropsy revealed abscess formation on the ventral side of the spine at T5 in Case 1, T5-6 in Case 2, and C7-T1 in Case 3, causing compression of the spinal cord in all three cases. In addition to cerebrospinal setariasis, vertebral abscess should be considered as a cause of paresis or gait abnormal

The occurrence of Setaria digitata in a horse is reported for the first time in Malaysia. An 8-year-old Thoroughbred cross mare was referred to the University Veterinary Clinic with the primary complaint of corneal opacity and excessive eye discharge. After initial treatment with Terramycin eye ointment, corneal opacity cleared partially to reveal a moving thread-like cylindrical worm in the anterior chamber of the eye. The parasite was successfully removed surgically, and examination under the light microscope revealed that the isolated worm (length = 45 mm) was a 5th stage larva of S. digita

An 18-month-old Arabian-English filly resident in southwest France was referred for evaluation of a conjunctival mass in the right eye (OD). A pink, solid, and mobile nodular formation, measuring approximately 1.2 × 0.8 cm was found under the superior nasal bulbar conjunctiva during an ophthalmic examination that was otherwise normal. The mass was surgically removed using a standing procedure. Cytological examination of fine-needle aspirates from the mass revealed a mixed eosinophilic-lymphocytic inflammation. Histological examination confirmed the dense and diffuse eosinophilic-lymphocytic in

A fairly clean antigenic cuticle was isolated from the S. digitata by dissection. Crossed immunoelectrophoresis of cuticular antigens against rabbit antiserum to cuticular antigens gave 30 anodic and 5 cathodic precipitin arcs. The cuticle antiserum cross reacted with muscle, uterus and pseudocoelomic fluid. When the antiserum was absorbed individually with these cross reacting somatic preparations, analysis against cuticle antigens gave only a limited number of precipitin arcs. But the results are clear enough to indicate the presence of cuticle specific antigens. As the cuticle of the parasi

The main problem regarding the chemotherapy of filariasis is that no safe and effective drug is available yet to combat the adult human filarial worms. Setaria cervi, the causal organism of setariasis and lumbar paralysis in cattle, is routinely employed as a model organism for conducting biochemical and enzymatic studies on filarial parasites. In view of the practical difficulties in procuring human strains of Wuchereria bancrofti and Brugia malayi for drug screening, the bovine filarial parasite S. cervi, resembling the human species in having microfilarial periodicity and chemotherapeutic r

Setaria digitata is a filarial parasite that causes fatal cerebrospinal nematodiasis in goats, sheep and horses, resulting in substantial economic losses in animal husbandry in the tropics. Due to its close resemblance to Wuchereria bancrofti, this nematode is also frequently used as a model organism to study human lymphatic filariasis. This review highlights numerous insights into the morphological, histological, biochemical, immunological and genetic aspects of S. digitata that have broadened our understanding towards the control and eradication of filarial diseases.

BACKGROUND: The aim of these studies was to determine the level of infection of European bison by nematodes belonging to the genus Setaria.
MATERIAL AND METHODS: From February 2006 to February 2007 21 bisons from 6 months to 20 years old shot in Białowieza Forest have been examined. During the necropsy pleural and peritoneal cavities were macroscopically examined for the presence of nematodes. The parasites were removed from tissues and fixed in 70% ethanol with 5% addition of glycerol and then identified to the species. Samples of blood were taken from shot bison and after centrifugation in

Surface antigens of adult filarial parasite S. digitata was isolated by employing techniques from manual dissection to treatment with detergents. Among the surface antigen preparations (SAPs), the activities of marker enzymes such as alkaline phosphatase, adenosine triphosphatase and 5' nucleotidase were higher with that isolated by triton X-100 technique (SAP2). On SDS-PAGE, the SAP2 has three major proteins with molecular weights 17, 29 and 36 KD which were consistent with the PBS soluble cuticular proteins (SAP1). Besides these, few other minor protein bands were also observed with the othe

Excretory Secretory (ES) material isolated from the culture fluid of S. digitata was highly antigenic. Neither oesophagus nor excretory cells and excretory pore of the parasite showed reasonable fluorescence with ES antisera. However, the uterine tissue and the egg showed strong fluorescence. The egg showed fluorescence mainly in the space between embryo and egg membrane (amniotic fluid). The amniotic fluid was highly antigenic and appears to be the most important source of ES material released by the filarial parasites.

Release of macromolecules by S. digitata, in 9 different media under in vitro condition have been studied. A direct relationship between microfilariae (mf) release and associated folin positive materials was seen in majority of the cases. High activities of hydrolytic enzymes such as protease, collagenase, alkaline phosphatase and lipase were detected in the excretary-secretary products and worm preparations. Activity of collagenase could not be detected in the male worm under experimental conditions.

Infective larvae of Wuchereria, Brugia, Breinlia, Dirofilaria and Setaria species from an experimental vector, Aedes togoi, are compared. The distinctive bubble-like caudal papillae of Wuchereria bancrofti are readily distinguishable from the protuberant ones of Brugia spp; the 'ear-like' papillae of Breinlia are distinct from the 'knob-like' ones of Dirofilaria or the 'thorn-like' terminal papilla of Setaria.

Localization of different enzymes of PEP-succinate pathway has been done in Setaria cervi, a bovine filarial worm. Succinate dehydrogenase and fumarate reductase were localized in mitochondria rich particulate fraction while all other enzymes were cytosolic. The in vitro effect of certain antifilarial/anthelmintic agents on these enzymes was also investigated. Sumarmin, at low concentration, could cause a marked inhibition of most of the enzymes of this pathway. Centperazine, an antifilarial drug being developed by CDRI showed significant inhibitory action on pyruvate kinase, lactate dehydroge

Filarial worms, Setaria yehi, were found in 286 of 1,045 white-tailed deer (Odocoileus virginianus) from 13 of 16 southeastern states. Infections were widespread in the piedmont and coastal plain physiographic provinces, but rarely occurred in deer of the mountains or in southern Florida. Infections were significantly more prevalent among fawn and yearling deer. Infections with S yehi largely accounted for a mild fibrinous peritonitis commonly observed among southeastern deer.
