Neurotoxic shellfish poisoning.

Neurotoxic shellfish poisoning (NSP) is caused by consumption of molluscan shellfish contaminated with brevetoxins primarily produced by the dinoflagellate, Karenia brevis. Blooms of K. brevis, called Florida red tide, occur frequently along the Gulf of Mexico. Many shellfish beds in the US (and other nations) are routinely monitored for presence of K. brevis and other brevetoxin-producing organisms. As a result, few NSP cases are reported annually from the US. However, infrequent larger outbreaks do occur. Cases are usually associated with recreationally-harvested shellfish collected during o

Paralytic shellfish poisoning and palytoxin poisoning in dogs.

BACKGROUND: Fatal cases of exposure to paralytic shellfish toxins and palytoxins have occurred in companion animals but are poorly described. METHODS: We describe one case of paralytic shellfish poisoning (PSP) and three cases of palytoxin poisoning in dogs. RESULTS: Mild PSP occurred following ingestion of crab while walking on a beach. Analysis confirmed the presence of paralytic shellfish toxins, particularly decarbamoyl saxitoxin, in clinical samples and marine organisms. This case occurred shortly after an outbreak of PSP in dogs on the eastern coast of England. Palytoxin poisoning occu

Paralytic shellfish poisoning: a review.

Paralytic shellfish poisoning (PSP) in man results from the consumption of mussels, clams, and oysters that have fed on toxic dinoflagellates. Motile, marine protozoa of the dinoflagellate group often produce "blooms," i.e., red tides, which color the sea. Not all genera or species are toxic to fish and mammals, nor are the toxic principles the same in all poisonous protozoa. At least 5 of the group are known to cause poisonings in man. Shellfish poisonings other than PSP are also recognized. The PSP toxin, saxitoxin, is concentrated in the viscera and occasionally in the mantle and syphon of

Paralytic shellfish poisoning in southern China.

The rapidly expanding mariculture and commercial region along the southern coast of China has experienced sporadic outbreaks of paralytic shellfish poisoning for nearly 30 years, yet virtually nothing is known of the nature of that toxicity or of the causative organisms. This study presents the first direct comparisons of the high performance liquid chromatography toxin composition profiles of shellfish implicated in paralytic shellfish poisoning outbreaks in Daya Bay with Alexandrium tamarense cultures established from those waters. The three cultures that were analyzed produced an unusually

Paralytic shellfish poisoning detection by surface plasmon resonance-based biosensors in shellfish matrixes.

The detection of paralytic shellfish poisoning (PSP) toxins in contaminated shellfish is essential for human health preservation. Ethical and technical reasons have prompted the search for new detection procedures as an alternative to the mouse bioassay. On the basis of the detection of molecular interactions by surface plasmon resonance (SPR) biosensors, an inhibition assay was developed using an anti-GTX2/3 antibody (GT13-A) and a saxitoxin-CM5 chip. This assay allowed for quantification of saxitoxin (STX), decarbamoyl saxitoxin (dcSTX), gonyautoxin 2,3 (GTX2/3), decarbamoyl gonyautoxin 2,3

Paralytic Shellfish Poisoning Outbreak-Oregon, United States, 2024.

In May 2024, the Oregon Poison Center notified the Oregon Health Authority of multiple people with suspected paralytic shellfish poisoning (PSP) who had consumed self-harvested mussels. Two days before this report, part of the Oregon Coast was closed to mussel harvesting after detection of paralytic shellfish toxins (PST) above the closure threshold (≥80 µg PST/100 g shellfish tissue) in mussels at one site. Following these illness reports, the closure was expanded northward to the Oregon-Washington border to include harvest areas reported by ill persons. An outbreak investigation was initiate

Phycotoxins: chemistry, mechanisms of action and shellfish poisoning.

Phycotoxins are natural metabolites produced by micro-algae. Through accumulation in the food chain, these toxins may concentrate in different marine organisms, including filter-feeding bivalves, burrowing and grazing organisms, herbivorous and predatory fish. Human poisoning due to ingestion of seafood contaminated by phycotoxins has occurred in the past, and harmful algal blooms (HABs) are naturally occurring events. Still, we are witnessing a global increase in HABs and seafood contaminations, whose causative factors are only partially understood. Phycotoxins are small to medium-sized natur

Domoic Acid and Amnesic Shellfish Poisoning - A Review.

A new type of seafood toxicity, called amnesic shellfish poisoning, was described from 107 human cases after individuals consumed mussels containing domoic acid harvested from Prince Edward Island, Canada, in 1987. Most of these cases experienced gastroenteritis, and many older persons or others with underlying chronic illnesses developed neurologic symptoms including memory loss. Standard treatment procedures for the neurologic condition were not effective and three patients died. Domoic acid is a known neurototoxin, and it is believed that in these cases enough toxin was absorbed through the

Shellfish and fish poisoning related to the toxic dinoflagellates.

At least four different species of the toxic dinoflagellates cause shellfish and fish poisoning in the United States: Ptychodiscus brevis, neurotoxic shellfish poisoning; Protogonyaulax catenella and P tamarensis, paralytic shellfish poisoning; and Gambierdiscus toxicus, ciguatera fish poisoning. These three disorders have similar clinical manifestations, primarily neurologic and alimentary. A complete history is essential; confirmation, while dependent on specific laboratory analysis, is usually based on a history of ingestion of fish or shellfish previously associated with these types of poi

TRPV1 as a key determinant in ciguatera and neurotoxic shellfish poisoning.

Ciguatera fish poisoning and neurotoxic shellfish poisoning are distinct clinical entities characterized by gastrointestinal and neurological disturbances, following the consumption of certain reef fish and shellfish containing toxic polyether compounds sporadically present in certain toxic marine dinoflagellates. The biotransformation and bioaccumulation of gambierol and brevetoxin, and their congeners, are believed to be involved in the pathogenesis of these "food-chain diseases", for which no effective treatments are available. Here, we describe for the first time the potent effect of gambi

Azaspiracid poisoning (AZP) toxins in shellfish: toxicological and health considerations.

It has been almost a decade since a previously unknown human toxic syndrome, azaspiracid poisoning (AZP), emerged as the cause of severe gastrointestinal illness in humans after the consumption of mussels (Mytilus edulis). Structural studies indicated that these toxins, azaspiracids, were of a new unprecedented class containing novel structural features. It is now known that the prevalent azaspiracids in mussels are AZA1, AZA2 and AZA3, which differ from each other in their degree of methylation. Several hydroxylated and carboxylated analogues of the main azaspiracids have also been identified

[Clinical analysis of paralytic shellfish poisoning following ingestion of oysters].

We describe five patients of paralytic shellfish poisoning (PSP) following ingestion of oysters in January 1991, in Nagato, Yamaguchi. The five patients (four men and one woman, age range:37-80 years) developed symptoms three-nine hours after consuming oysters. Symptoms included paresthesia of the mouth and tongue (three), paresthesia of the extremities (five), and quadriparesis (five). The median duration of neurological symptoms was 16 hours. All five patients completely recovered. Although PSP is rare as compared with globe fish poisoning in Japan, the fatality rate of PSP is 8%-9%, with de

Application of an anti-idiotypic antibody as a mimetic competing antigen in a direct competitive ELISA for the detection of paralytic shellfish poisoning toxins in shellfish.

The previously developed direct competitive ELISA to detect PSP toxins (PSP-ELISA) was modified by employing an enzyme-conjugated anti-idiotypic antibody as a competing antigen. The detection limit of the modified PSP-ELISA in shellfish tissue was estimated to be 0.07 MU/g. When the modified PSP-ELISA was evaluated using the 192 toxin-contaminated shellfish samples, the modified PSP-ELISA results showed a sensitivity of 100% (88 of 88) and a specificity of 92.3% (96 of 104) compared to the MBA results.

A receptor binding assay for paralytic shellfish poisoning toxins: recent advances and applications.

We recently described a high throughput receptor binding assay for paralytic shellfish poisoning (PSP) toxins, the use of the assay for detecting toxic activity in shellfish and algal extracts, and the validation of 11-[3H]-tetrodotoxin as an alternative radioligand to the [3H]-saxitoxin conventionally employed in the assay. Here, we report a dramatic increase in assay efficiency through application of microplate scintillation technology, resulting in an assay turn around time of 4 h. Efforts are now focused on demonstrating the range of applications for which this receptor assay can provide d

Isodomoic acid C, an unusual amnesic shellfish poisoning toxin from Pseudo-nitzschia australis.

An unusual isomer of domoic acid (1), isodomoic acid C (2), has been found in New Zealand shellfish contaminated by amnesic shellfish poisoning (ASP) toxins and was shown to be produced by a local strain of the pennate diatom Pseudo-nitzschia australis. A bulk culture of this strain was used to isolate 2. The structure was determined from spectroscopic data and was shown to correspond to that of 2 from a Japanese red seaweed, the only other reported occurrence of this compound. The affinity of 2 for GluR6 glutamate receptors was 240-fold lower than for 1, indicating low neurotoxic potential.

Paralytic shellfish poisoning toxins induce xenobiotic metabolising enzymes in Atlantic salmon (Salmo salar).

Paralytic shellfish poisoning (PSP) toxins have been implicated as the causative agent of a number of fish kills. Exposure experiments indicate that fish are susceptible to PSPs by intraperitoneal (i.p.) and oral administration, while sampling of fish affected by toxic blooms reveals that these toxins can be accumulated. In spite of the potential impact to marine fisheries, little research has been conducted on the potential metabolism and detoxification of PSPs in marine fishes. Previous work by this group has shown that the xenobiotic metabolising enzyme (XME) cytochrome P-450 (CYP1A) is ind
