Sialic Acids as Receptors for Pathogens.

Carbohydrates have long been known to mediate intracellular interactions, whether within one organism or between different organisms. Sialic acids (Sias) are carbohydrates that usually occupy the terminal positions in longer carbohydrate chains, which makes them common recognition targets mediating these interactions. In this review, we summarize the knowledge about animal disease-causing agents such as viruses, bacteria and protozoa (including the malaria parasite Plasmodium falciparum) in which Sias play a role in infection biology. While Sias may promote binding of, e.g., influenza viruses

Sialic acids: biomarkers in endocrinal cancers.

Sialylations are post translational modification of proteins and lipids that play important role in recognition, signaling, immunological response and cell-cell interaction. Improper sialylations due to altered sialyl transferases, sialidases, gene structure and expression, sialic acid metabolism however lead to diseases and thus sialic acids form an important biomarker in disease. In the endocrinal biology such improper sialylations including altered expression of sialylated moieties have been shown to be associated with disorders. Cancer still remains to be the major cause of global death an

Expression of sialic acids and other nonulosonic acids in Leptospira.

BACKGROUND: Sialic acids are negatively charged nine carbon backbone sugars expressed on mammalian cell surfaces. Sialic acids are part of a larger family of nonulosonic acid (NulO) molecules that includes pseudaminic and legionaminic acids. Microbial expression of sialic acids and other nonulosonic acids has been shown to contribute to host-microbe interactions in a variety of contexts, including participation in colonization, immune subversion, and behaviors such as biofilm formation, autoagglutination and motility. Previous research has suggested that some spirochetes may also express these

Identification and Evaluation of CMP-Sialic Acids.

Cytidine 5'-monophosphate-sialic acid (CMP-Sia) is a donor substrate of all sialyltransferases in both prokaryotes and eukaryotes, and biosynthesized by the CMP-Sia synthetase (CSS or CMAS) using Sia and cytidine 5'-triphosphate (CTP) as the substrates. Since CSSs have unique substrate specificity for Sia species depending on organisms, cells, and organs, and developmental stages, various kinds of CMP-Sia exist in nature. This chapter describes how to quantitatively detect CMP-Sia in the CSS reaction mixture as well as in cells and organs.

Identification of sialic acids on Leishmania donovani amastigotes.

The presence of Neu5Ac on promastigotes of Leishmania donovani, the causative organism of Indian visceral leishmaniasis, has been reported recently. Here we report the occurrence of Neu5Ac as a major component on amastigotes, as well as Neu5Gc, Neu5,9Ac2 and Neu9Ac5Gc as indicated by fluorimetric high performance liquid chromatography and gas liquid chromatography/electron impact mass spectrometry. Furthermore, binding studies with Sambucus nigra agglutinin (SNA), Maackia amurensis agglutinin (MAA), and various Siglecs, showed the presence of both (alpha2 --> 6)- and (alpha2 --> 3)-linked sial

Malaria parasites do not contain or synthesize sialic acids.

The capacity of Plasmodia to synthesize sialic acids was investigated by adding radioactive acetate to short-term in vitro cultures of the intraerythrocytic asexual forms of three malaria parasites (the human malaria Plasmodium falciparum in Aotus trivirgatus erythrocytes; the simian malaria P. knowlesi in rhesus monkey erythrocytes; the rodent malaria P. berghei in mouse erythrocytes) and to cultures of extracellular zygotes of the avian malaria P. gallinaceum. Radioactive acetate was added to normal rhesus monkey erythrocytes and to cells of the murine myeloma NS-1 for comparison. Although [

Sialic Acid Receptors of Viruses.

Sialic acid linked to glycoproteins and gangliosides is used by many viruses as a receptor for cell entry. These viruses include important human and animal pathogens, such as influenza, parainfluenza, mumps, corona, noro, rota, and DNA tumor viruses. Attachment to sialic acid is mediated by receptor binding proteins that are constituents of viral envelopes or exposed at the surface of non-enveloped viruses. Some of these viruses are also equipped with a neuraminidase or a sialyl-O-acetyl-esterase. These receptor-destroying enzymes promote virus release from infected cells and neutralize sialic

Investigating the Utility of Leukocyte Sialic Acid Measurements in Lysosomal Free Sialic Acid Storage Disorder.

Lysosomal free sialic acid storage disorder (FSASD) is a rare, multisystem neurodegenerative disease caused by biallelic pathogenic variants in SLC17A5, encoding sialin. FSASD is characterized by aberrant accumulation of unconjugated "free" sialic acid (Neu5Ac) within lysosomes. Depending on the specific genetic variants, affected individuals may present with either a rapidly fatal disease or progressive neurodegeneration. While skin fibroblasts have traditionally been used for diagnosis and research, the use of leukocytes in FSASD remains underexplored. This study examined Neu5Ac levels in le

Versatile biosynthetic engineering of sialic acid in living cells using synthetic sialic acid analogues.

Sialic acids are critical components of many glycoconjugates involved in biologically important ligand-receptor interactions. Quantitative and structural variations of sialic acid residues can profoundly affect specific cell-cell, pathogen-cell, or drug-cell interactions, but manipulation of sialic acids in mammalian cells has been technically limited. We describe the finding of a previously unrecognized and efficient uptake and incorporation of sialic acid analogues in mammalian cells. We added 16 synthetic sialic acid analogues carrying distinct C-1, C-5, or C-9 substitutions individually to

Simple fluorimetric method for quantification of sialic acids in glycoproteins.

A simple and rapid fluorimetric method was developed for detection and quantitative analysis of sialic acids in glycoproteins. Sialic acid residues in glycoproteins were specifically oxidized with periodate at 0 degrees C for 45 min. Formaldehyde generated from carbon 9 (C-9) of sialic acid was converted specifically to fluorescent dihydropyridine derivative with acetoacetanilide and ammonia at room temperature for 10 min. The reaction products indicate intense fluorescence with excitation and emission maxima at 388 and 471 nm, respectively. When the reaction was conducted in approximately a 1

Serum concentration of sialic acids in naturally occurring ovine babesiosis.

This study is designated to assess the effect of the severity of Babesia ovis infection on sialic acid concentration in blood sera in naturally infected sheep. Infected animals (diseased group) comprised 38 Iranian fat-tailed sheep, about 1-3 years old, naturally infected with B. ovis, divided into four subgroups with respect to parasitemia rates (low 0.1-0.3 %, moderate 0.4-0.9 %, high 1-2.5 %, and very high >2.5 %). The parasitological diagnosis was confirmed using PCR analysis. As a control group, ten clinically healthy sheep reared under the same management and environmental conditions wer

Histoenzymological detection of sialic acids in the rodent salivary glands.

Sections from the major salivary glands of rats and mice were used to locate, characterize and compare sialoglycoconjugates by means of lectin histochemistry, sialidase digestion, periodate oxidation and potassium hydroxide deacetylation. The gland sialylated macromolecules contained the terminal dimers sialic acid-beta-galactose and sialic acid-alpha-N-acetyl-galactosamine but differed in the varieties of sialic acids and the linkages of sialic acids to penultimate sugars. Indeed, the submandibular and parotid glands exhibited a notable occurrence of periodate labile sialic acids with C7 and/

Variety of sialic acids occurring in the bovine sublingual gland.

Sialoglycoconjugates were investigated in the bovine sublingual gland by direct visualization of sialic acid with specific lectins (LPA, SNA) and by histochemical procedures combined with sialidase digestion and lectins. The most reactive histological structures were found to be acini which contained glycoconjugates with terminal disaccharides consisting of sialic acid linked to galactose or N-acetylgalactosamine. Resistance to periodate oxidation was interpreted as demonstrating a relevant presence of C7, C8 and C9 acetylated sialic acids. KOH-Sialidase-DBA and KOH-Alcian blue sequences allow

Identification of the glycosidically bound sialic acid in mucin glycoproteins that reacts as "free sialic acid" in the Warren assay.

A widely employed colorimetric assay for sialic acids based on periodate oxidation followed by reaction with thiobarbituric acid depends on the formation of a hexos-5-uluronic acid product, the pre-chromogen, by the periodate cleavage of the C6-C7, C7-C8, and C8-C9 bonds in free sialic acid. Glycosidically bound sialic acids are not expected to react in the assay since cleavage cannot occur between C6-C7 to yield the pre-chromogen. However, several investigators have reported the detection of a positive reaction by certain sialoglycoconjugates. In this study, it was found that various mucins b

Sialic Acid metabolism and systemic pasteurellosis.

Pasteurella multocida subsp. multocida is a commensal and opportunistic pathogen of food animals, wildlife, and pets and a zoonotic cause of human infection arising from contacts with these animals. Here, an investigation of multiple serotype A strains demonstrated the occurrence of membrane sialyltransferase. Although P. multocida lacks the genes for the two earliest steps in de novo sialic acid synthesis, adding sialic acid to the growth medium resulted in uptake, activation, and subsequent transfer of sialic acid to a membrane acceptor resembling lipooligosaccharide.
