Silene latifolia and S. dioica are model systems in studies of plant reproduction, chromosome evolution and sexual dimorphism, but sexing of plants based on morphology is only possible from flowering stage onwards. Both species show homogametic females (XX) and heterogametic males (XY).•Here we developed two assays (primer pairs ss816 and ss441) for molecular sexing of S. latifolia and S. dioica, targeting length polymorphisms between the X and Y-linked copies of the spermidine synthase gene. The two assays were successful in identifying known (flowering-stage) males and females from UK and Spa

Patterns of codon usage bias (CUB) convey useful information about the selection on synonymous codons induced by gene expression and contribute to an understanding of substitution patterns observed at synonymous sites. They can also be informative about the distinctive evolutionary properties of sex chromosomes such as genetic degeneration of the Y chromosome, dosage compensation, and hemizygosity of the X chromosome in males, which can affect the selection on codon usage. Here, we study CUB in Silene latifolia, a species of interest for studying the early stages of sex chromosome evolution. W

This work aims to investigate the nature and the specific mechanisms by which polycarboxylic compounds participate in the tolerance of Silene vulgaris to Cr with special attention given to the rhizosphere system. This knowledge is important to use this species in the implementation of phytoremediation technologies in Cr-polluted soils. According to the results, chromium is chelated and mobilized by the citric and malic acids in plant tissues, while oxalic acid might participate in the reduction and chelation of Cr in the rhizosphere. At the applied doses, the response of both exudation rate an

Interactions, antagonistic or mutualistic, can exert selection on plant traits. We explored the role of Hadena bicruris, a pollinating seed predator, as a selective agent on its host, the dioecious plant Silene latifolia. We exposed females from artificial-selection lines (many, small flowers (SF) vs. few, large flowers (LF)) to this moth. Infestation did not differ significantly between lines, but the odds of attacked fruits aborting were higher in SF females. We partitioned selection between that caused by moth attack and that resulting from all other factors. In both lines, selection via mo

Some plants have massive sex-linked regions. To test hypotheses about their evolution, we sequenced the genome of Silene latifolia, in which giant heteromorphic sex chromosomes were first discovered in 1923. It has long been known that the Y chromosome consists mainly of a male-specific region that does not recombine with the X chromosome and carries the sex-determining genes and genes with other male functions. However, only with a whole Y chromosome assembly can candidate genes be validated experimentally and their locations determined and related to the suppression of recombination. We desc

We carried out a global survey of all major types of transposable elements in Silene latifolia, a model species with sex chromosomes that are in the early stages of their evolution. A shotgun genomic library was screened with genomic DNA to isolate and characterize the most abundant elements. We found that the most common types of elements were the subtelomeric tandem repeat X-43.1 and Gypsy retrotransposons, followed by Copia retrotransposons and LINE non-LTR elements. SINE elements and DNA transposons were less abundant. We also amplified transposable elements with degenerate primers and use

A new anther smut fungus, Microbotryum crypticum is described based on specimens collected from the Japanese endemic plant, Silene miqueliana (Caryophyllaceae) in Osaka Prefecture, as well as from herbarium specimens of S. miqueliana preserved at the National Museum of Nature and Science (TNS) in Japan. Microbotryum crypticum is phylogenetically distinct from other anther smut fungi reported on Silene species and has relatively small spores compared to previously described Microbotryum species. Host preference further supports this new species. The presence of infected S. miqueliana plants in

Introduction: The incidence of acquired laryngotracheal stenosis is on rise, being caused usually due to prolonged intubation or trauma. Its management has thus become a common clinical challenge for Otolaryngologists and Head and Neck surgeons. Further, with advances in Critical care medicine, Cardiopulmonary surgery and Neurosurgery, the number of intubation-related laryngotracheal injuries is increasing. Objectives: To assess the outcome of patients who underwent Partial Cricotracheal Resection (PCTR) for subglottic stenosis on basis of pre-defined parameters. Design of Study: This is a pro

Silene compacta Fischer ex Hornem., a member of Caryophyllaceae family, is a wild plant with significant ornamental and ecological value, now threatened by habitat loss and climate change. This study assessed the genetic diversity of 40 S. compacta genotypes collected from four—geographically distinct regions of Türkiye (Bolu, İzmir, Isparta, and Nevşehir) using 26 morphological traits and 10 inter-simple sequence repeats (ISSR) primers. Principal Component Analysis of morphological data grouped the genotypes into five clusters, with the first two components explaining 61.5% of the total varia

The reactivity of halogen-substituted silenes (silicon-carbon doubly bonded compounds) is not well understood relative to alkenyl halides. Herein, we report the reductive dimerization and complexation of a stable bromosilene (1). The reaction of 1 with an equimolar amount of KC8 furnished a 1,3-disilabicyclo[1.1.0]butane (trans-3) featuring a remarkably short bridgehead Si-Si bond [2.204(1) Å] and a very wide Si-Si-Cexocyclic angle of 173.0(1)°. Bulky alkyl groups on the bridgehead silicon atoms and less steric hindrance at the bridging carbon atoms lead to the short bridgehead bond. Bromosile

Sex chromosomes undergo various modifications that affect their synapsis during meiosis. While most of the genome achieves full synapsis by the end of pachytene, the nonrecombining regions of XY (or ZW) chromosomes often remain asynaptic and fail to form physical associations at metaphase I. Despite significant progress in animal models, the meiotic behavior dynamics of plant sex chromosomes remain largely unexplored. In this study, we employed super-resolution microscopy to analyze 3D chromosome organization and the localization of key meiotic proteins. Namely, we studied the dynamics of Asyn

The dioecious plant Silene latifolia has heteromorphic sex chromosomes, and comparison of the positions of sex-linked genes indicates that at least three large inversions have occurred during the evolution of the Y chromosome. In this article, we describe the isolation of a new sex-linked gene from S. latifolia, which provides new information on the evolution of this plant's young sex chromosomes. By using reverse-transcription polymerase chain reaction degenerate primers based on the Arabidopsis thaliana sequence of WUSCHEL, a flower-development gene, we found two copies in S. latifolia, whic

The effect of oral administration of a 50% ethanol extract of Silene aprica (SA) on acute liver injury was examined in rats intoxicated with carbon tetrachloride (CCl4) and acetaminophen. The results indicated that SA protected the liver from CCl4- and acetaminophen-induced injury as judged by morphological and biochemical observations. An increase in both lipid peroxidation (LPO) and triglyceride concentrations occurred in the liver with CCl4 injection, SA administration significantly reduced these changes.

Sexual dimorphism is widespread among species with separate sexes, and its extent is thought to be governed by the differential expression of thousands of genes between males and females known as sex-biased genes (SBGs). SBGs have been studied in numerous species, but rarely through comparative approaches, which limits our understanding of their evolution, especially during multiple, independent transitions to separate sexes. We sequenced the transcriptomes of nine dioecious species (with separate males and females), two gynodioecious species (with separate females and hermaphrodites), and two

The genus Silene includes a plethora of dioecious and gynodioecious species. Two species, Silene latifolia (white campion) and Silene dioica (red campion), are dioecious plants, having heteromorphic sex chromosomes with an XX/XY sex determination system. The X and Y chromosomes differ mainly in size, DNA content and posttranslational histone modifications. Although it is generally assumed that the sex chromosomes evolved from a single pair of autosomes, it is difficult to distinguish the ancestral pair of chromosomes in related gynodioecious and hermaphroditic plants. We designed an oligo pain

Understanding the origin of new species is a central goal in evolutionary biology. Diverging lineages often evolve highly heterogeneous patterns of genetic differentiation; however, the underlying mechanisms are not well understood. We investigated evolutionary processes governing genetic differentiation between the hybridizing campions Silene dioica (L.) Clairv. and S. latifolia Poiret. Demographic modelling indicated that the two species diverged with gene flow. The best-supported scenario with heterogeneity in both migration rate and effective population size suggested that a small proporti

The overuse of antibiotics has led to the emergence of multidrug-resistant bacteria, which are resistant to various antibiotics. Combination therapies using natural compounds with antibiotics have been found to have synergistic effects against several pathogens. Synergistic natural compounds can potentiate the effects of polymyxins for the treatment of Acinetobacter baumannii infection. Out of 120 types of plant extracts, only Silene armeria extract (SAE) showed a synergistic effect with polymyxin B (PMB) in our fractional inhibitory concentration and time-kill analyses. The survival rate of G

The current study aimed to determine the metabolites present in the crude methanol extract of Silene kuschakewiczii Regel & Schmalh. by UHPLC-MS. Total phenol and flavonoid contents were examined in the methanol extract. UHPLC-MS analysis allowed the characterisation of 20 metabolites in S. kuschakewiczii, mainly flavonoids and oleanane-type triterpenes. Among them, the major compounds were schaftoside-2-O-glucoside and saponarin. The crude methanol extract also showed the ability to scavenge free radicals effectively (DPPH: 13.12 mg TE/g; ABTS: 53.42 mg TE/g). In addition, it exhibited remark

Dioecy is a breeding system with separate females and males, where sex can be determined by sex chromosomes (for instance, XY males and XX females). Dioecy is generally associated with the evolution of sexual dimorphism. In this study, we tested whether small RNAs (sRNAs) play a role in sexual dimorphism and sex chromosome evolution in Silene latifolia. We generated new data for female and male sRNAs and DNA methylation. We showed that sRNAs are most of the time female-biased in S. latifolia, suggesting that the presence of the Y chromosome globally impacts the epigenome by diluting sRNAs geno
