[The efficiency of photometric determination of skatole in slaughtered swine].

In a group of 264 boars, including both cryptorchides and hermaphrodites, skatole was measured using a photometric method developed in Denmark. It resulted in a concentration of skatole equivalents (SE) in the fat of 0.14 ppm in belly tissue and 0.18 ppm in flomen tissue on average, respectively. 10.6% of belly and 16.3% of flomen tissue exceeded the limiting value of 0.25 ppm skatole for boar taint. The photometric determination of skatole is based on a relatively unspecific reaction according to Ehrlich. Therefore, other derivatives of indole concerning the decomposition of tryptophan are re

Mechanistic insight into the role of immunocastration on eliminating skatole in boars.

The accumulation of skatole in fat tissue is one of the predominant factors, causing boar taint. The present study was aimed to understand the mechanism whereby active immunization against GnRH (immunocastration) eliminates skatole in boars. Thirty-six boars were assigned within litter into three groups (n = 12): control, surgically castrated, or immunized against GnRH at 10 wk of age (with a booster 8 wk later). Faecal and blood samples (for skatole and skatole-regulatory hormone profiles) were collected at 4-wk intervals until boars were slaughtered (26 weeks). Immunocastration reduced (P <

Skatole and indole concentrations in Longissimus dorsi and fat samples of pigs.

A HPLC-NP (normal phase-high performance liquid chromatography) method for determining the concentration of skatole and indole in Longissimus dorsi samples is described. Lipids containing skatole and indole were extracted in chloroform:methanol (2:1) at room temperature and dehydrated by liquid-liquid extraction with an aqueous solution saturated with 10% of sodium chloride. The organic phase was evaporated to dryness and redissolved in 10 ml of hexane:2-propanol (92:8). Indolic compounds were separated on a Hypersil aminopropylsilica column (5 μm) (250×4.6 mm i.d.). The mobile phase was hexan

Elucidation of a novel skatole degradation pathway and key genes in Rhodococcus rhodochrous S3.

Skatole is a major component of the objectionable smell of manure and has an extremely low odor threshold, leading to severe odor problems and potential threats to animal and human health. Yet the molecular mechanisms underlying microbial skatole degradation remain poorly understood. To date, only a limited number of skatole monooxygenases have been reported; these enzymes belong to the Group E flavin-dependent monooxygenases and catalyze the initial oxidation of skatole, but the subsequent degradation pathways and associated genes remain unclear. In this study, we isolated a skatole-degrading

The structural basis of malodorant skatole formation by the glycyl radical enzyme indoleacetate decarboxylase.

Glycyl radical enzymes (GREs) catalyze challenging chemical reactions using a posttranslationally installed glycyl radical cofactor. One such enzyme, indoleacetate decarboxylase (IAD), performs the radical-based decarboxylation of indole-3-acetate (I3A) to form the malodorant molecule skatole. In addition to being an odor nuisance, skatole is a human and livestock lung toxin, a suspected carcinogen, and a mosquito attractant, all of which impact human health, agriculture, food production, and wastewater treatment. Here, we use cryogenic electron microscopy to solve a 2.45-Å resolution structur

Rapid determination of skatole and indole in pig back fat by normal-phase liquid chromatography.

A rapid normal-phase high-performance liquid chromatographic method for the quantitative determination of indole and skatole in pig back fat samples has been developed. The compounds were extracted by shaking the samples at ambient temperature in hexane-2-propanol (92:8, v/v). The sample preparation procedure was simple because it was not necessary to remove the fat from the samples. The compounds were separated on a 250 x 4.6 mm I.D., 5 micron Hypersil aminopropylsilica column. Fluorescence (excitation at 280 nm and emission at 360 nm) was used for selective detection. Recoveries for skatole

Fecal skatole and indole and breath methane and hydrogen in patients with large bowel polyps or cancer.

The object of this study was to explore the use of fecal skatole and indole and breath methane and hydrogen as metabolic markers of the anaerobic colonic flora in patients with unresected large bowel cancer or polyps. Patients with descending or sigmoid colon cancer were more likely to be breath methane excretors than control subjects, patients with proximal colon cancer, and patients with rectal cancer. Control subjects excreting breath methane excreted less fecal skatole than breath methane excretors in the following groups: patients with adenomatous polyps, all patients with colorectal canc

Natural Product Skatole Ameliorates Lipotoxicity-Induced Multiple Hepatic Damage under Hyperlipidemic Conditions in Hepatocytes.

Skatole (3-methylindole, 3MI) is a natural-origin compound derived from plants, insects, and microbial metabolites in human intestines. Skatole has an anti-lipid peroxidation effect and is a biomarker for several diseases. However, its effect on hepatocyte lipid metabolism and lipotoxicity has not been elucidated. Hepatic lipotoxicity is induced by excess saturated free fatty acids in hyperlipidemia, which directly damages the hepatocytes. Lipotoxicity is involved in several metabolic diseases and hepatocytes, particularly affecting nonalcoholic fatty liver disease (NAFLD) progression. NAFLD i

Elevated fat skatole levels in immunocastrated, surgically castrated and entire male pigs with acute dysentery.

Boar taint is due to androstenone and skatole (3-methyl-indole) accumulation in fat tissues. During a study to investigate the effect of immunocastration on fattening pigs, an outbreak of acute dysentery occurred caused by Lawsonia intracellularis and Brachyspira hyodysenteriae and resulted in cachexia and high mortality. Low androstenone levels in the immunocastrates (0.25 ± 0.04 μg/g liquid fat) suggested that the immunocastration had been effective, but unusually high skatole concentrations in fat tissues were found not only in entire males, but also in surgical castrates and immunocastrate

Single-Enzyme Conversion of Tryptophan to Skatole and Cyanide Expands the Mechanistic Competence of Diiron Oxidases.

Skatole is a pungent heterocyclic compound derived from the essential amino acid l-tryptophan by bacteria in the mammalian digestive tract. The four-step anaerobic conversion of tryptophan to skatole is well-established; though, to date, no aerobic counterpart has been reported. Herein, we report the discovery of the oxygen-dependent skatole synthase SktA that single-handedly converts 5-bromo-l-tryptophan to 5-bromoskatole, obviating the need for a multienzyme process. SktA is part of a three-gene biosynthetic gene cluster (BGC) in the cyanobacterium Nostoc punctiforme NIES-2108 and functions

Breed-dependent differences in gut microbiota linked to indole and skatole levels in Hezuo and DLY pigs.

BACKGROUD: Indole and skatole are major odorous compounds generated in pig farms and their emissions affect the sustainable development of pig industry seriously. At present, microbial interventions represent an efficient and environmentally sustainable strategy for reducing odor emissions. Research on functional microorganisms specifically targeting indole and skatole emission reduction in pigs remains limited. To compare the concentrations of indole and skatole in Hezuo and DLY pigs, and analysis differences in microbial community characteristics between the two pig breeds, aimed to screen t

Regulation of CYP2A6 protein expression by skatole, indole, and testicular steroids in primary cultured pig hepatocytes.

CYP2A6 is one of the enzymes involved in the hepatic metabolism of a naturally produced compound, skatole, in the pig. Low CYP2A6 activity has been linked to excessive accumulation of skatole in pig adipose tissue and development of the phenomenon "boar taint." CYP2A6 activity varies between male and female animals, suggesting the involvement of sex hormones in regulation of the enzyme activity and/or expression. The present study investigated whether pig hepatic CYP2A6 protein expression is regulated by the testicular steroids testosterone, androstenone, or estrone sulfate using primary cultu

Hepatic Gene Expression and Metabolite Profiles of Androstenone and Skatole Relative to Plasma Estrone Sulfate Levels in Boars.

Testicular steroids can alter the activity and expression of enzymes within the liver and may influence the metabolism of skatole and androstenone, which are responsible for boar taint. Plasma levels of estrone sulfate (E1S) are indicative of the steroidogenic capacity of the boar and are variable between animals of similar live weights at slaughter. This study aimed to characterize the relationship between steroidogenic capacity and the metabolism of boar taint compounds by relating plasma E1S levels at slaughter weight to the expression levels of genes regulating the metabolism of androsteno

Preliminary Test of the Reduction Capacity for the Intestinal Adsorption of Skatole and Indole in Weaning Piglets by Pure and Coated Charcoal.

To reduce the risk of boar taint, intact male piglets are immuno- or surgically castrated. One alternative is reducing skatole by adding skatole reducing or adsorbing substances to the boars' diet. Charcoal with a high capacity for adsorbing skatole and indole in vitro (tested before, data not shown) was fed to the boars to test the hypothesis that a fat coating prevents the unspecific adsorption of charcoal before entry into the large intestine while increasing skatole adsorption. Twelve male and six female weaning piglets with initial body weights of 7.74 ± 0.75 kg were fed for 18 (or 19) da

Genomic Characterization and Biotechnological Potential of a Skatole-Degrading Bacillus pumilus Strain hhy901 Isolated from Bovine Manure.

Skatole (3-methylindole) is a primary malodorous compound emitted from livestock manure, posing significant challenges for odor control in intensive animal production. In this study, a skatole-degrading bacterial strain, designated hhy901, was isolated from fresh cattle manure and identified as Bacillus pumilus through polyphasic taxonomy, including 16S rRNA and gyrB gene phylogeny, morphological characterization, biochemical profiling, and whole-cell fatty acid analysis. Under an initial skatole concentration of 100 mg/L in Bacillus liquid medium, strain hhy901 removed 87.2% of skatole within

Harnessing a marine-derived Rhodococcus strain for cross-habitat bioaugmentation of malodorant skatole: Performance and ecological response.

Skatole, primarily generated via the anaerobic metabolism of tryptophan, is a pervasive nitrogenous heterocyclic malodorant. While ubiquitous in various environments, its microbial remediation in saline ecosystems remains poorly documented. In this study, a skatole-degrading microbial consortium was enriched from marine sediments. High-throughput sequencing revealed distinct successional dynamics during domestication, with Pseudomonas becoming predominant (89.23%) and Rhodococcus persisting as a potential functional taxon (1.98%).
