An acute social defeat stressor in early puberty increases susceptibility to social defeat in adulthood.

Syrian hamsters readily display territorial aggression. If they lose even a single agonistic encounter, however, hamsters show striking reductions in aggressive behavior and increases in submissive behavior, a distinct behavioral change that we have previously termed conditioned defeat. This acute social defeat stressor is primarily psychological and is effective in both males and females. Therefore, we maintain that this procedure presents an ideal model for studying behavioral and physiological responses to social stress. Here, we demonstrate that social avoidance following social defeat is

The effect of escapable versus inescapable social defeat on conditioned defeat and social recognition in Syrian hamsters.

Male Syrian hamsters are naturally aggressive animals that reliably defend their home territory against intruding conspecifics. Hamsters that lose agonistic encounters subsequently exhibit a striking change in their agonistic behavior, however, expressing no aggression and instead becoming highly submissive, a behavioral change that we have termed conditioned defeat. We have generally employed an inescapable defeat training protocol when studying conditioned defeat. The purpose of the present study was to determine if conditioned defeat is an epiphenomenon of the inescapable defeat experience

The social defeat hypothesis of schizophrenia: an update.

According to the social defeat (SD) hypothesis, published in 2005, long-term exposure to the experience of SD may lead to sensitization of the mesolimbic dopamine (DA) system and thereby increase the risk for schizophrenia. The hypothesis posits that SD (ie, the negative experience of being excluded from the majority group) is the common denominator of 5 major schizophrenia risk factors: urban upbringing, migration, childhood trauma, low intelligence, and drug abuse. The purpose of this update of the literature since 2005 is to answer 2 questions: (1) What is the evidence that SD explains the

Juvenile social defeat stress exposure persistently impairs social behaviors and neurogenesis.

Adverse juvenile experiences, including physical abuse, often have negative health consequences later in life. We investigated the influence of social defeat stress exposure as juveniles on neuropsychological behaviors, and the causal role of glucocorticoids in abnormal behaviors and impairment of neurogenesis in mice exposed to the stress. The juvenile (24-day-old) and adult (70-day-old) male C57BL/6J mice were exposed to social defeat stress induced by an aggressive ICR mouse. Social defeat stress exposure as juveniles, even for 1 day, induced persistent social avoidance to the unfamiliar IC

Chronic social defeat stress causes retinal vascular dysfunction.

PURPOSE: The roles of vascular dysfunction and chronic stress have been extensively discussed in the pathophysiology of glaucoma. Our aim was to test whether chronic stress causes retinal vascular dysfunction and therewith induces retinal ganglion cells (RGCs) loss. METHODS: Twelve mice underwent chronic social defeat (CSD) stress, while 12 mice received control treatment only. Intraocular pressure (IOP) was measured with a rebound tonometer. Blood plasma corticosterone concentration and adrenal gland weight were used to assess stress levels. Brn-3a staining in retinas and PPD staining in opt

Epigenetic Signatures of Social Defeat Stress Varying Duration.

Stress-induced mental disorders, including depression and anxiety disorders, constitute a global issue in contemporary society due to treatment complexity and the diversity of manifestations. Understanding the molecular mechanisms of these disorders presents a significant challenge for neurobiology. We investigated the effects of social defeat stress (SDS) of varying durations (10 and 30 days) on behavioral patterns and the H3K4me3 (trimethylation at the 4th lysine residue of histone H3) landscape in the prefrontal cortex of C57BL/6 mice. Furthermore, we compared these data with previously pub

Social Defeat Stress, Sex, and Addiction-Like Behaviors.

Social confrontation is a form of social interaction in animals where two conspecific individuals confront each other in dispute over territory, during the formation of hierarchies, and during breeding seasons. Typically, a social confrontation involves a prevailing individual and a yielding individual. The prevailing individual often exhibits aggressive postures and launches attacks, whereas the yielding individual often adopts postures of defeat. The yielding or defeated animals experience a phenomenon known as social defeat stress, in which they show exaggerated stress as well as autonomic

Modelling social fear in mice: advancements in social fear conditioning and social defeat paradigms.

Social anxiety disorder (SAD) is a common and chronic mental illness whose pathogenesis is not yet fully understood, and treatment responses are often suboptimal. Animal models play a crucial role in elucidating its neurobiological mechanisms and developing novel therapeutic approaches. This article systematically reviews two primary mouse modelling paradigms: social fear conditioning (SFC) and social defeat. The SFC model induces specific social fear behaviors by pairing a social stimulus with an aversive event, whereas the social defeat model, by simulating social conflict and psychological

Chronic social defeat stress gives rise to social avoidance through fear learning.

Chronic social defeat stress (CSDS), a widely used rodent model of stress, reliably leads to decreased social interaction in stress susceptible animals. Here, we investigate a role for fear learning in this response using male 129 Sv/Ev mice, a strain that is more vulnerable to CSDS than the commonly used C57BL/6 strain. We first demonstrate that defeated 129 Sv/Ev mice avoid a CD-1 mouse, but not a conspecific, indicating that motivation to socialize is intact in this strain. CD-1 avoidance is characterized by approach behavior that results in running in the opposite direction, activity that

Social fear memory as a framework for understanding social defeat stress and psychopathology.

Social defeat is a potent stressor in both rodents and humans that, under conditions of chronic or severe stress, can lead to persistent social avoidance. This review synthesizes evidence supporting the view that social defeat promotes the formation of social fear memory through associative learning mechanisms that parallel classical fear conditioning. Drawing on preclinical models, we describe how the context of social defeat functions as a conditioned stimulus, while the experience of defeat serves as an unconditioned stimulus, giving rise to fear-related behaviors such as freezing and socia

Network adaptability governs resilience and susceptibility to social defeat.

Stress resilience-the ability to maintain mental health after adversity-manifests in humans and rodents as efficient threat-safety discrimination and fear extinction. Although behavioral differences between resilient and susceptible individuals are well documented, the brain-wide structural signatures that distinguish resilience, susceptibility, and impaired threat learning remain poorly defined. Using a translational mouse model of chronic social defeat stress in male C57BL/6 J mice (n = 55), combined with preregistered multimodal magnetic resonance imaging with graph-theoretical analyses, we

Social defeat stress aggravates asthma airway inflammation through neutrophils.

BACKGROUNDS: Psychological stress is a recognized contributor to asthma progression and is associated with increased neutrophils. OBJECTIVE: To investigate the role of neutrophils and NETosis in psychosocial stress aggravates asthma airway inflammation. METHODS: We constructed murine asthma models under social defeat stress (SDS) intervention. Wild-type,Csf3-deficient (Csf3-/-, neutropenic), andPadi4-deficient (Padi4-/-, impaired NETosis) mice were used to explore neutrophil-driven mechanisms. RESULTS: In murine asthma models, SDS exacerbated airway hyperresponsiveness, inflammation, and re

Repeated social defeat causes bone loss from mouse femur.

INTRODUCTION: Clinical studies have demonstrated that depression increases the risk of bone fracture and impaired bone growth. However, the mechanisms by which psychosocial stress aggravates bone properties remain to be fully elucidated. Repeated social defeat (RSD) is a murine model system that recapitulates key physiological, immunological, and behavioral alterations observed in humans exposed to repeated psychosocial stress. The objective of this study was to examine whether RSD alters bone characteristics and to explore the mechanisms underlying these effects. METHODS: Twenty-four wild-ty

Chronic Social Defeat Stress in Early Adolescent Male Mice.

Physical abuse and trauma in childhood is reported in as many as 1 in 7 children and is a major risk factor for the development of psychiatric diseases, such as Post-Traumatic Stress Disorder (PTSD), in adolescence and adulthood. Hallmark behavioral symptoms of PTSD include avoidance of cues and contexts associated with trauma. The ability to model long-lasting changes in the brain due to early life trauma is critical to determine potential circuit and molecular targets for the modulation of resultant symptoms. This manuscript describes a protocol for modeling early life physical and psycholog

Ramelteon protects against social defeat stress-associated abnormal behaviors.

Psychological stress affects the neuroendocrine regulation, which modulates mental status and behaviors. Melatonin, a hormone synthesized primarily by the pineal gland, regulates many brain functions, including circadian rhythms, pain, sleep, and mood. Selective pharmacological melatonin agonist ramelteon has been clinically used to treat mood and sleep disorders. Posttraumatic stress disorder (PTSD) is a psychiatric condition associated with severe trauma; it is generally triggered by traumatic events, which lead to severe anxiety and uncontrollable trauma recall. We recently reported that re

Resilience to social defeat stress in adolescent male mice.

Adverse social experiences during adolescence are associated with the appearance of mental illness in adulthood. Social defeat (SD) is an ethologically valid murine model to study the consequences of social stress. In adolescent mice, SD induces depressive-like behaviors, increased anxiety and potentiates the reinforcing effects of cocaine and alcohol. However, not all mice exposed to SD will be susceptible to these effects. Adult mice resilient to the effects of SD show a consistent phenotype being resilient to depressive-like behaviors and to the increase in cocaine and alcohol consumption.

Neuroinflammatory Dynamics Underlie Memory Impairments after Repeated Social Defeat.

UNLABELLED: Repeated social defeat (RSD) is a murine stressor that recapitulates key physiological, immunological, and behavioral alterations observed in humans exposed to chronic psychosocial stress. Psychosocial stress promotes prolonged behavioral adaptations that are associated with neuroinflammatory signaling and impaired neuroplasticity. Here, we show that RSD promoted hippocampal neuroinflammatory activation that was characterized by proinflammatory gene expression and by microglia activation and monocyte trafficking that was particularly pronounced within the caudal extent of the hippo

A re-evaluation of social defeat as an animal model of depression.

Social defeat by aggressive Tryon Maze Dull (TMD) rats, resulting in loss of rank of a previously dominant rat, has recently been advanced as a model of loss of self-esteem in humans. Since low self-esteem is a major symptom of depression, a further claim has been made that loss of rank can be used as a model of depression. In support of this claim, it has been suggested that loss of rank can be reversed by the antidepressant imipramine. However, antidepressant treatment has not yet been shown to reverse the effects of defeat for more than a single test session. Consequently, the present study

Effects of environmental enrichment during adolescence on social defeat effects.

Social defeat (SD) is a well established model that increases addiction vulnerability accompanied by depressive- and anxiety-like behaviors. Environmental enrichment (EE) has been shown to enhance resilience and mitigate stress-induced behavioral alterations. Here, we investigated the protective effects of EE during adolescence, both before and during SD encounters, on stress-induced anxiety, depression and the increased conditioned rewarding effects of a subthreshold cocaine dose in adulthood. We employed the social interaction test (SIT) to categorize mice into resilient and susceptible phen
