An outbreak of suspected solanine poisoning in schoolboys: Examinations of criteria of solanine poisoning.

Seventy eight schoolboys became ill after eating potato at lunch on the second day of the autumn term. Seventeen of the boys required admission to hospital. The gastrointestinal, circulatory, neurological and dermatological findings and the results of laboratory investigations were in keeping with solanine poisoning. The illness affected the junior boys and all the monitors but no other senior boys or staff. This pattern was compatible with the consumption of a relatively small number of toxic potatoes believed to have come from a bag (A) left in the stores since the summer term. The amount of

α-Solanine attenuates chondrocyte pyroptosis to improve osteoarthritis via suppressing NF-κB pathway.

α-Solanine has been shown to exhibit anti-inflammatory and anti-tumour properties; however, its efficacy in treating osteoarthritis (OA) remains ambiguous. The study aimed to evaluate the therapeutic effects of α-solanine on OA development in a mouse OA model. The OA mice were subjected to varying concentrations of α-solanine, and various assessments were implemented to assess OA progression. We found that α-solanine significantly reduced osteophyte formation, subchondral sclerosis and OARSI score. And it decreased proteoglycan loss and calcification in articular cartilage. Specifically, α-sol

Study on the apoptosis of human melanoma cells induced by solanine in vitro and its mechanisms.

Melanoma (malignant melanoma, MM) is a highly aggressive tumor, ranking as the third most common cutaneous malignancy and characterized by high metastatic potential, high mortality rates, and poor prognosis. Solanine, a major steroidal alkaloid found in potatoes, has widely reported anticancer benefits, though its inhibitory effects on melanoma cells are less studied. This study aimed to observe the effects of solanine on the proliferation, apoptosis, and related apoptotic proteins in melanoma A375 and A2058 cells and to investigate its possible anti-tumor mechanisms. The morphological changes

Fast aqueous synthesis of hydrogel-immobilized papain-CuSO4@SiO2 for on-site solanine detection.

Solanine is a toxic glycoalkaloid naturally present in potatoes and other Solanaceae crops, and its accumulation during storage poses a serious risk to food safety. However, current detection methods such as high-performance liquid chromatography, and mass spectrometry, are costly, labor-intensive, and unsuitable for rapid on-site analysis. In this study, we present a fast, green, and cost-effective colorimetric sensing platform for solanine detection based on a biomimetic papain-CuSO₄@SiO₂ nanozyme. The nanozyme was synthesized in water via a one-step reaction at room temperature, completing

α-Solanine induces ROS-mediated autophagy through activation of endoplasmic reticulum stress and inhibition of Akt/mTOR pathway.

α-Solanine is a glycoalkaloid found in species of the nightshade family including potato. It was primarily reported to have toxic effects in humans. However, there is a growing body of literature demonstrating in vitro and in vivo anticancer activity of α-solanine. Most of these studies have shown activation of apoptosis as the underlying mechanism in antitumor activity of α-solanine. In this study, we report α-solanine as a potential inducer of autophagy, which may act synergistically or in parallel with apoptosis to exert its cytotoxic effect. Induction of autophagy was demonstrated by sever

Molecular encapsulation of the aminosteroid phytotoxins solanidine and α-solanine by sugammadex: new insights into atomic-level interactions.

Using cyclodextrins (CDs) as antidotes is of great interest in various fields of research due to their characteristic feature to strongly capture certain small apolar bioactive molecules. The application of a γ-CD derivative, sugammadex (SGM), as an antidote is exploited in the clinical practice to specifically reverse the effect of neuromuscular blocking agents. In the present work, we investigated the capability of SGM to capture naturally occurring aminosteroid phytotoxins, that have no known antidote to date. Using NMR spectroscopy, we identified exceptionally stable inclusion complexes be

Antimicrobial Activity Against Phytopathogens and Inhibitory Activity on Solanine in Potatoes of the Endophytic Bacteria Isolated From Potato Tubers.

As an important global crop, the potato (Solanum tuberosum L.) contains the endotoxin solanine that leads to human poisoning and major economic losses. Poisoning symptoms and even acute poisoning may occur when the content of solanine in potatoes exceeds 200 mg/kg. In addition, potatoes are susceptible to some pathogenic bacteria, including Streptomyces scabies and Erwinia carotovora subsp. atroseptica (Van Hall) dye, which can cause potato scab and potato blackleg disease, respectively. In this study, 37 culturable endophytic bacteria strains were obtained from potato tubers based on the cult

Long-Term Selection with α-Solanine Enhances Tolerance and Drives Multilevel Physiological Remodeling in Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae).

The fall armyworm, Spodoptera frugiperda (J.E. Smith) (Lepidoptera: Noctuidae), is a polyphagous pest exposed to plant defensive chemicals. We tested whether sustained selection by the potato steroidal glycoalkaloid α-solanine induces changes in tolerance, life-history performance, and physiology. A potato-sprout-derived preparation was standardized for α-solanine content by targeted LC-ESI-MS/MS, and a selected line was maintained for 12 generations on a diet containing 1.0 mg α-solanine g-1. Selected and unselected lines were compared across normal artificial diet, α-solanine diet, and potat

Effects of α‑solanine on human head and neck squamous cell carcinoma cells and human umbilical vein endothelial cells in vitro.

α-solanine is a glycoalkaloid that is commonly found in nightshades (Solanum) and has a toxic effect on the human organism. Among other things, it is already known to inhibit tumor cell proliferation and induce apoptosis in tumor cell lines. Due to its potential as a tumor therapeutic, the current study investigated the effect of α-solanine on head and neck squamous cell carcinoma (HNSCC). In addition, genotoxic and antiangiogenic effects on human umbilical vein endothelial cells (HUVECs) were evaluated at subtoxic α-solanine concentrations. Cytotoxicity and apoptosis rates were measured in tw

Anti-Cancer Role of Dendrosomal Nano Solanine in Chronic Myelogenous Leukemia Cell Line through Attenuation of PI3K/AKT/mTOR Signaling Pathway and Inhibition of hTERT Expression.

BACKGROUND: Solanine was primarily known as a toxic compound. Nonetheless, recently the apoptotic role of solanine through suppression of PI3K/AKT/mTOR signaling pathway has been shown against many malignancies except chronic myelogenous leukemia (CML). Sustaining the aforementioned pro-survival pathway, BCR-ABL fused oncoprotein in CML activates NF-kB and c- MYC for apparent immortalizing factor hTERT. Since solanine is a poor water-soluble molecule, herein, a nanocarrier was employed to intensify its pernicious effect on cancerous cells. OBJECTIVE: The current research aimed at evaluating t

Is there any association between blood glycoalkaloid levels and anencephaly in human?

AIM: In various experimental animal studies, it has been proven that solanine, a subtype of glycoalkaloids, is responsible for neural tube defects. However, there have not been any human studies yet in this area. Our aim is to investigate whether there are any connections between blood glycoalkaloid levels and anencephaly in humans. METHODS: Blood and amniotic fluid samples were taken from patients diagnosed with fetal anencephaly during pregnancy. The samples from patients with normal pregnancies were taken as well and was compared to the patients with fetal anencephaly during pregnancy. We

Pharmacology and toxicology of chaconine and tomatine.

The pharmacological responses produced by alpha chaconine and tomatine on guinea pig ileum, on the isolated electrically stimulated frog ventricle, and recordings of EEG, ECG, respiration and blood pressure in the rabbit showed no essential differences from those produced by alpha solanine. The LD50 values of chaconine and solanine in the mouse and rabbit are also similar and suggest that compounds other than these are probably responsible for the predominant toxic effects of certain hybrid potatoes in man and animals.
