Genistein mediates the anti-adipogenic actions of Sophora japonica L. extracts.

Previous studies showed that feeding diets containing the mature fruits of Sophora japonica L. prevented body weight gain and reduced fat mass in high-fat diet-induced obese mice. This observation has led to the hypothesis that extracts from S. japonica L. may inhibit adipocyte differentiation of preadipocytes. To elucidate the possible mechanisms for the anti-obesity action of S. japonica L., its effects on adipocyte differentiation were investigated in C3H10T1/2 mesenchymal stem cells and 3T3-L1 preadipocyte cells. The mature fruit of S. japonica L. was partitioned with ethanol, hexane, dich

Characterization and Identification of Potential Allergenic Proteins in Sophora japonica L. Pollen.

Sophora japonica L. pollen (SJP) is an emerging cause of urban seasonal allergies such as asthma and rhinitis. This study systematically evaluated the allergenicity of SJP using both in vivo and in vitro experiments. In vivo, a mouse model was employed in which SJP induced typical allergic symptoms, including sneezing and rubbing behavior, elevated serum IgE and IL-4 levels, and reduced IFN-γ levels. In vitro, IgE immunoblotting and ELISA using sera from allergic patients identified five potential allergenic protein bands separated by SDS-PAGE. LC-MS analysis further revealed a novel potential

Dichloromethane extracts of Sophora japonica L. stimulate osteoblast differentiation in mesenchymal stem cells.

Sophora japonica L. fruit prevents bone loss by inhibiting osteoclast activity. We hypothesized that S japonica L. extracts could promote osteoblast differentiation. To test this hypothesis, we investigated the effect of S japonica L. on osteoblast differentiation and identified the bioactive compound(s) from S japonica L. The mature fruit of S japonica L. was partitioned with ethanol, hexane, dichloromethane (DCM), ethyl acetate, and butanol, and their effects were tested on osteoblast differentiation of C3H10T1/2 cells. DCM fractionated extracts were identified as the most osteogenic fractio

Effects of Roasting on Antibacterial and Antioxidant Properties of Sophora japonica Buds-The Involvements of Rutin and Quercetin Constituents.

Roasting is the most common thermal processing method established for Sophora japonica (SJ) buds applied as traditional medicines, and it has also been reported to alter several of their therapeutic functions. However, there have been no studies investigating the influences of roasting on the effects of these materials against bacteria. Therefore our study was performed to examine the alterations that this process would induce in SJ buds' antibacterial properties. Fresh buds were subjected to hot air drying or different roasting methods, as described in Materia Medica, including yellow-, dark

Sophora japonica Extract Supplementation Facilitates Piglets' Early Weaning Transition by Alleviating Intestinal Oxidative Stress, Inflammation, and Microbiota Dysbiosis.

The early weaning of piglets is still widely practiced in modern swine production, although it is frequently associated with intestinal dysfunction, microbiota dysbiosis, and even diarrhea. A total of 126 piglets were randomly allotted to three groups (seven pens per group) and fed either a basal diet, a basal diet supplemented with 0.05% SJE, or a basal diet supplemented with 0.1% SJE. Supplementation with 0.1% SJE effectively decreased fecal scores on days 7 and 14 post-weaning and improved growth performance on day 14 post-weaning in early-weaned piglets. In addition, SJE supplementation im

Hand-Held Electrostatic Spray Ionization Mass Spectrometry Enables On-Site Metabolomic Analysis of Sophora japonica and Robinia pseudoacacia.

Mass spectrometry plays a key role in agricultural and food chemistry, yet direct ionization of plant samples is complicated by endogenous salts and other matrix components that destabilize conventional direct-current nanoelectrospray ionization (DC-nanoESI). To address this limitation in point-of-need analysis, we developed hand-held electrostatic spray ionization (HESSI), which produces stable ionization from nanoliter sample volumes without direct electrical contact with the solution. A compact pulsed-discharge module and dielectric barrier limit electrochemical reactions and reduce the ris

A TCM formula comprising Sophorae Flos and Lonicerae Japonicae Flos alters compositions of immune cells and molecules of the STAT3 pathway in melanoma microenvironment.

A traditional Chinese medicine (TCM) formula (SL) comprising Sophorae Flos and Lonicerae Japonicae Flos was used for treating melanoma in ancient China. We have previously shown that an ethanolic extract of SL (SLE) possesses anti-melanoma effects and suppresses STAT3 signaling in vitro and in vivo. STAT3 has been linked to the development of melanoma immunosuppressive microenvironment. In this work, we investigated whether SLE inhibits melanoma growth by reprogramming the tumor microenvironment in mouse and co-culture cell models. In B16F10 melanoma-bearing mice, we found that intragastric ad

Optimization of Enzyme-Assisted Ultrasonic Extraction of a Quercetin-Containing Crude Extract from Sophora japonica Flower Buds and Evaluation of Its In Vitro Antioxidant and Antibacterial Activities.

Flower buds of Sophora japonica are flavonoid-rich edible botanical materials. This study developed an enzyme-assisted ultrasonic protocol for producing a quercetin-containing crude extract. A Box-Behnken design evaluated β-glucosidase concentration, digestion time, and the cellulase-to-pectinase mass ratio. The quadratic model was significant (p < 0.0001; R2 = 0.9848; adjusted R2 = 0.9652). At 14 U/mL β-glucosidase, 72 min of digestion, and a mass ratio of 1.25:1 (w/w), the UV-estimated quercetin-equivalent yield reached 7.34%, compared with 4.17% for ultrasound alone (p < 0.001).
