All animals use two different strategies to navigate: idiothetic or movement-based navigation, and allothetic or landmark-based navigation. A new study reveals that compromised idiothetic navigation underlies disrupted grid cell coding in an early stage Alzheimer's disease mouse model.

Although several previous studies have demonstrated navigational deficits in early-stage symptomatic Alzheimer's disease (AD), navigational abilities in preclinical AD have not been examined. The present investigation examined the effects of preclinical AD and early-stage symptomatic AD on spatial navigation performance. Performance on tasks of wayfinding and route learning in a virtual reality environment were examined. Comparisons were made across the following three groups: Clinically normal without preclinical AD (n = 42), clinically normal with preclinical AD (n = 13), and early-stage sym

Older age is associated with changes in the brain, including the medial temporal lobe, which may result in mild spatial navigation deficits, especially in allocentric navigation. The aim of the study was to characterize the profile of real-space allocentric (world-centered, hippocampus-dependent) and egocentric (body-centered, parietal lobe dependent) navigation and learning in young vs. older adults, and to assess a possible influence of gender. We recruited healthy participants without cognitive deficits on standard neuropsychological testing, white matter lesions or pronounced hippocampal a

Embodied navigation and manipulation are fundamental capabilities for embodied agents operating in physical environments. A key challenge in this process is understanding the spatial context and the affordances of the environment, which involves recognizing how objects can be interacted with (object affordance) and identifying suitable locations for movement and object placement (free space affordance). While Vision-Language Models (VLMs) have shown promise in high-level task planning, their ability to translate reasoning into precise executable actions remains limited, particularly in image-b

Objective. Virtual reality (VR) has become a key tool for researching spatial memory. Virtual environments offer many advantages for research in terms of logistics, neuroimaging compatibility etc. However, it is well established in animal models that the lack of physical movement in VR impairs some neural representations of space, and this is considered likely to be true in humans as well. Furthermore, it is unclear how big the disruptive effect stationary navigation is-how much does physical movement during encoding and recall affect human spatial memory and representations of space? What eff

Spatial memory is core to wayfinding and everyday memory. Interestingly, individuals with schizophrenia using spatial navigation strategies (cognitive mapping) are impaired, whereas those using response-based (e.g., single-landmark) strategies show relatively intact memory performance. We observed abnormal brain communication in schizophrenia participants who used a spatial strategy during a virtual-reality navigation task, particularly between temporal and frontal brain regions. In contrast, schizophrenia participants using a response strategy recruited similar brain systems to healthy partic

Navigation in the real world may rely on a multitude of strategies with distinct advantages and disadvantages; however, the characterization of spontaneous strategy use is scarce. We explore data across three groups engaged in virtual navigation: patients with epilepsy undergoing invasive electrophysiological recordings, two groups undergoing fMRI scanning: young adults at genetic risk for Alzheimer's disease (APOE-ε4 carriers), and healthy controls. We establish metrics to quantitatively characterize participants' trajectories, reflecting putative strategies: straightness, deviation to bounda

INTRODUCTION: Persistent postural-perceptual dizziness (PPPD) is a common chronic dizziness disorder with an unclear pathophysiology. It is hypothesized that PPPD may involve functional dysfunction of the construction of inner cognitive maps, leading to disrupted spatial cognition processes as a core feature. The present studies attempt to unravel the neural mechanisms that underlie spatial navigation in PPPD.
METHODS: Fifty-two participants completed the study: 19 PPPD patients, 20 control subjects with vestibular disorders but without PPPD (with comparable peripheral vestibular function to

Aims of the present article are: 1) assessing vestibular contribution to spatial navigation, 2) exploring how age, global positioning systems (GPS) use, and vestibular navigation contribute to subjective sense of direction (SOD), 3) evaluating vestibular navigation in patients with lesions of the vestibular-cerebellum (patients with downbeat nystagmus, DBN) that could inform on the signals carried by vestibulo-cerebellar-cortical pathways. We applied two navigation tasks on a rotating chair in the dark: return-to-start (RTS), where subjects drive the chair back to the origin after discrete ang

Spatial learning, including encoding and retrieval of spatial memories as well as holding spatial information in working memory generally serving navigation under a broad range of circumstances, relies on a network of structures. While central to this network are medial temporal lobe structures with a widely appreciated crucial function of the hippocampus, neocortical areas such as the posterior parietal cortex and the retrosplenial cortex also play essential roles. Since the hippocampus receives its main subcortical input from the medial septum of the basal forebrain (BF) cholinergic system,

Planning is a fundamental function of both spatial navigation and visuo-spatial tasks, supported by overlapping and distinct neural substrates that remain partially unclear. We conducted an Activation Likelihood Estimation (ALE) meta-analysis on fMRI studies examining active, goal-directed navigation and visuo-spatial planning tasks (i.e., the Tower of London paradigm). Conjunction and contrast analyses revealed a shared network encompassing bilateral frontal regions, including the superior frontal gyrus (SFG), middle frontal gyrus (MFG), anterior insula (INS), and clusters in the left supplem

Astronauts often face orientation challenges while on orbit, which can lead to operator errors in demanding spatial tasks. In this study, we investigated the impact of long-duration spaceflight on the neural processes supporting astronauts' spatial orientation skills. Using functional magnetic resonance imaging (fMRI), we collected data from 16 astronauts six months before and two weeks after their International Space Station (ISS) missions while performing a spatial orientation task that requires generating a mental representation of one's surroundings. During this task, astronauts exhibited

Active navigation seems to yield better spatial knowledge than passive navigation, but it is unclear how active decision-making influences learning and memory. Here, we examined the contributions of theta oscillations to memory-related exploration while testing theories about how they contribute to active learning. Using electroencephalography (EEG), we tested individuals on a maze-learning task in which they made discrete decisions about where to explore at each choice point in the maze. Half the participants were free to make active decisions at each choice point, and the other half passivel

The 'cognitive map' hypothesis proposes that brain builds a unified representation of the spatial environment to support memory and guide future action. Forty years of electrophysiological research in rodents suggest that cognitive maps are neurally instantiated by place, grid, border and head direction cells in the hippocampal formation and related structures. Here we review recent work that suggests a similar functional organization in the human brain and yields insights into how cognitive maps are used during spatial navigation. Specifically, these studies indicate that (i) the human hippoc

This paper uses a Yoked experimental design to investigate the selective advantage of active spatial navigation in 4- to 10-year-old children. Participants were presented with a map game in which they were asked to find the shortest way for a monster to get across town while collecting its five monster friends on the way. Active control was associated with better recall for path length and the area between paths, whereas no differences were observed for the number of corners. Further, active navigation did not enhance episodic memory. Our results indicate that the benefits of active control du

In a familiar city, people can recall scene views (e.g., a particular street corner scene) they could encounter again in the future. Complex objects with multiple features are represented by multiple neural units (channels) in the brain, but when anticipating a scene view, the kind of feature that is assigned to a specific channel is unknown. Here, we studied neural encoding of scene view anticipation during spatial navigation, using a novel data-driven analysis to evaluate encoding channels. Our encoding models, based on functional magnetic resonance imaging (fMRI) activity, provided channel

Cognitive deficits in older adults attributable to Alzheimer's disease (AD) pathology are featured early on by hippocampal impairment. Among these individuals, deterioration in spatial navigation, manifested by poor hippocampus-dependent allocentric navigation, may occur well before the clinical onset of dementia. Our aim was to determine whether allocentric spatial navigation impairment would be proportional to right hippocampal volume loss irrespective of general brain atrophy. We also contrasted the respective spatial navigation scores of the real-space human Morris water maze with its corr

We know little about the ability to explore and navigate large-scale space for people with intellectual disability (ID). In this cross-syndrome study, individuals with Down syndrome (DS), individuals with Williams syndrome (WS) and typically developing children (TD; aged 5-11 years) explored virtual environments with the goal of learning where everything was within the environment (Experiment 1) or to find six stars (Experiment 2). There was little difference between the WS and DS groups when the goal was simply to learn about the environment with no specific destination to be reached (Experim

Individuals with Down Syndrome (DS) and individuals with Williams syndrome (WS) present with poor navigation and elevated anxiety. The aim of this study was to determine the relationship between these two characteristics. Parent report questionnaires measured navigation abilities and anxiety in WS (N = 55) and DS (N = 42) as follows. Anxiety: Spence Children's Anxiety Scale and a novel measure of navigation anxiety. Navigation: Santa Barbara Sense of Direction Scale (SBSOD) and a novel measure of navigation competence. Most individuals were not permitted to travel independently. A relationship
