Intraflagellar Transport (IFT) and Sperm Formation.

Intraflagellar transport (IFT) is a conserved mechanism for cilia formation. Twenty-two IFT components form the IFT-A complex (six components) and IFT-B complex (sixteen components). Driven by kinesin and dynein motor proteins, these IFT complexes are involved in the trafficking of proteins needed for cilia assembly by anterograde transport and retrograde transport. IFT core components also associate with other proteins for cilia formation. Mutations in IFT core components result in ciliogenesis defects and human diseases, including male infertility. Sperm flagella are specialized motile cilia

Importance of sperm morphology during sperm transport and fertilization in mammals.

After natural or artificial insemination, the spermatozoon starts a journey from the site of deposition to the place of fertilization. However, only a small subset of the spermatozoa deposited achieves their goal: to reach and fertilize the egg. Factors involved in controlling sperm transport and fertilization include the female reproductive tract environment, cell-cell interactions, gene expression, and phenotypic sperm traits. Some of the significant determinants of fertilization are known (i.e., motility or DNA status), but many sperm traits are still indecipherable. One example is the infl

Sperm transport and survival in the mare.

Following the deposition of semen in the mares uterus, spermatozoa must be transported to the site of fertilization, be maintained in the female tract until ovulation occurs, and be prepared to fertilize the released ovum. Sperm motility, myometrial contractions, and a spontaneous post-mating uterine inflammation are important factors for the transport and survival of spermatozoa in the mares reproductive tract. Fertilizable sperm are present in the oviduct within 4 hours after insemination. At this time, the uterus is the site of a hostile inflammatory environment. Our data suggest that sperm

Sperm transport in the female reproductive tract.

At coitus, human sperm are deposited into the anterior vagina, where, to avoid vaginal acid and immune responses, they quickly contact cervical mucus and enter the cervix. Cervical mucus filters out sperm with poor morphology and motility and as such only a minority of ejaculated sperm actually enter the cervix. In the uterus, muscular contractions may enhance passage of sperm through the uterine cavity. A few thousand sperm swim through the uterotubal junctions to reach the Fallopian tubes (uterine tubes, oviducts) where sperm are stored in a reservoir, or at least maintained in a fertile sta

Effect of sperm dosage transportation in stallions: Effect on sperm DNA fragmentation.

Artificial insemination programs for horses usually involve ex vivo handling and transporting of sperm. The present experiment was designed to: (i) assess the effect of transportation on sperm DNA integrity at different time post semen collection, and (ii) evaluate if sperm DNA quality deteriorates rapidly beyond 24 h of cooled storage. After collection, the ejaculates were extended using INRA 96 and semen was prepared for prompt analysis (A0) or 24 h/48 h cooled-shipping (B24 and C48 respectively). Each sample was assessed for sperm DNA fragmentation index (SDFI) at time 0 and after incubatio

ATP activation of peritubular cells drives testicular sperm transport.

Spermatogenesis, the complex process of male germ cell proliferation, differentiation, and maturation, is the basis of male fertility. In the seminiferous tubules of the testes, spermatozoa are constantly generated from spermatogonial stem cells through a stereotyped sequence of mitotic and meiotic divisions. The basic physiological principles, however, that control both maturation and luminal transport of the still immotile spermatozoa within the seminiferous tubules remain poorly, if at all, defined. Here, we show that coordinated contractions of smooth muscle-like testicular peritubular cel

Oxytocin--a stimulator of directed sperm transport in humans.

Rhythmic peristaltic contractions of the muscular wall of the non-pregnant uterus, as well as rapid sperm transport from the vagina to the Fallopian tubes, have long been documented by means of vaginal sonography and hysterosalpingoscintigraphy. Uterine peristaltic activity reaches a maximum before ovulation and is controlled via oestradiol secretion from the dominant follicle systemically and into the utero-ovarian countercurrent system; it is also enhanced by oxytocin. In this study, the effect of oxytocin and its receptor antagonist atosiban on uterine peristalsis and thus directed sperm tr

Transport intracytoplasmic sperm injection (ICSI): a cost-effective alternative.

PURPOSE: Transport in vitro fertilization (IVF) programs are operational in a lot of countries and especially popular in The Netherlands, where IVF activities are strictly regulated. Since the introduction of intracytoplasmic sperm injection (ICSI) in the IVF laboratory, many laboratories are now setting up this new technique, which necessitates major investments in terms of infrastructure and specialized personnel. METHODS: We present a cost effective alternative, consisting of patient selection, preparation, and oocyte retrieval at one center and transport of oocytes to a second center, whe

Effect of sperm numbers and concentration on sperm transport and uterine inflammatory response in the mare.

Our objective was to determine whether the concentration of cooled sperm inseminated influenced sperm transport and intensity of the uterine inflammatory reaction 2, 4 and 24h after insemination. Experimental subjects were 189 estrous mares with a dominant follicle > or =35 mm in diameter and no bacterial growth or neutrophils detected in uterine smears. Each mare was randomly assigned to receive one of the following intrauterine treatments (volume, 20 mL): insemination with 5x10(6) mL(-1) or 25x10(6) mL(-1) or 50x10(6) mL(-1) sperm diluted in 3 mL seminal plasma (SP) and 17 mL skim milk; semi

Oxidative stress and epididymal sperm transport, motility and morphological defects.

OBJECTIVES: Radical oxidative species (ROS) have an important effect on sperm quality and quantity. Oxidative stress (OS) occurs when production of potentially destructive reactive oxygen species (ROS) exceeds the body's own natural antioxidant defenses, resulting in cellular damage. OS is a common pathology seen in approximately half of all infertile men. Increased ROS generation and reduced antioxidant capacity is negatively correlated with sperm concentration and motility in infertile men. For the first time, we used a more stable and reliable sensitive carbonyl protein (CP) detection metho

Sperm storage and transport following natural mating and artificial insemination.

Recent observations in turkey and chicken hens show that sperm storage in both species is a highly inefficient process. After artificial insemination (AI), less than 1% of spermatozoa inseminated are selected for transport to and enter the sperm storage tubules (SST). It has been shown that the sperm selection process is orchestrated within the vagina and not at the level of the SST. At least two mechanisms are involved in the selection of spermatozoa fit for sperm storage, one being mechanical (motility) and the other biochemical in nature (sperm-vaginal mucosa interactions). Furthermore, it

[Regulation of sperm hyperactivation by transporters involved in Na+ homeostasis].

Mammalian sperm have to undergo several physiological and biochemical changes to be fertilization-competent. These changes are collectively called "capacitation". Capacitated sperm show frantic whiplash-like flagellar movement called "hyperactivation". Recently, it has been reported that treatments of sperm to enhance hyperactivation improve in vitro fertilization (IVF) and development of the fertilized embryos. This fact indicates that hyperactivation is an attractive target to improve assisted reproductive technologies. However, the detailed mechanisms which regulate hyperactivation have not

Utero-tubal sperm transport and its impairment in endometriosis and adenomyosis.

The uterus is composed of different smooth muscle layers that serve various functions. First, menstrual debris is expulsed at the time of the menses. Second, sperm is transported in the preovulatory phase to maximize fertility, and third, the human embryo is placed in an adequate setting during implantation. Endometriosis is a gynecologic disorder leading to severe pain symptoms such as severe pain during menstruation (dysmenorrhea), chronic pelvic pain, pain during sexual intercourse (dyspareunia), and abnormal uterine bleeding. Besides, endometriosis is often associated with female infertili

PCM1 orchestrates centrosomal and flagellar protein transport to promote sperm maturation.

Centriolar satellites facilitate the delivery of proteins required for centrosome assembly and function, and rely on the nucleation of the scaffold protein pericentriolar material 1 (PCM1). During sperm maturation, centrosomes undergo remodeling to facilitate neckpiece formation and flagellogenesis. However, the role and mechanism of PCM1 in centrosome remodeling and sperm maturation remain to be elucidated. Herein we show that PCM1 is downregulated in patients with non-obstructive azoospermia. Pcm1 knockout mice exhibit disrupted spermiogenesis, characterized by a disorganized manchette and h

Fate of fluorescent stained sperm following insemination: new light on oviducal sperm transport and storage in the turkey.

A novel approach was used to evaluate the distribution of sperm in the oviduct of turkey hens inseminated before or after the onset of egg production. Prior to insemination, sperm were stained with the nuclear fluorescent stain bisbenzimide. Sperm distribution in the sperm storage tubules (SST) of the uterovaginal junction and the infundibular tubular glands was determined by use of simultaneous differential interference contrast and fluorescence microscopy. In hens inseminated and examined prior to the onset of egg production, 94% of the SST contained sperm (21% were filled). In contrast, in

Two modifiers of sperm transport within the fallopian tube of the rat.

In several mammals studied, ovulation appears to stimulate coordinated translocation of a few potential fertilizing spermatozoa to the ampulla from the caudal isthmus of the Fallopian tube. The present experiments demonstrate that, in the rat, this movement is regulated to a considerable degree within each duct by the ipsilateral ovary. Unilateral ovariectomy had no effect on ipsilateral sperm transport into the uterus, but this brought a unilateral quenching of sperm transport to the ampulla of the oviduct, from which spermatozoa were often totally or almost absent. This suppression was alway

Methylxanthines stimulate calcium transport and inhibit cyclic nucleotide phosphodiesterases in abalone sperm.

Experiments were designed to determine the mechanism by which methylxanthines elevate abalone sperm cAMP concentrations and induce the acrosome reaction (AR). Theophylline or, more effectively, 1-methyl-3-isobutylxanthine (MIX) inhibit the cyclic nucleotide phosphodiesterase activities of abalone sperm homogenates. 45Ca2+ uptake by sperm is also stimulated by theophylline, and more effectively by MIX, and this stimulatory effect is blocked by KCN. Verapamil, a compound known to antagonize Ca2+ conductance, has no effect on the Ca2+ or MIX-induced cAMP elevation at concentrations up to 200 micr

The KIF6-RBP Complex Orchestrates mRNA Transport Required for Sperm Flagellar Assembly.

The precise assembly of the sperm flagellum is essential for male fertility and has long been ascribed to kinesin-2-driven intraflagellar transport (IFT) of protein cargoes. However, during late spermiogenesis, when transcription activity is largely silenced, how the spatiotemporally regulated delivery of flagellar components remains poorly understood. Here, we systematically screened kinesin genes in asthenozoospermic patients and identified two homozygous deleterious KIF6 variants in unrelated men characterized by complete sperm immotility. Mouse models carrying the corresponding mutations r

Transport, Distribution and Elimination of Mammalian Sperm Following Natural Mating and Insemination.

The integrity of transport, distribution and elimination of sperm in the female genital tract plays a pivotal role for successful reproduction in mammals. At coitus, millions or billions of sperm are deposited either into the anterior vagina (human, primates), the cervix (most mammalian species) or the uterus (pig). In most species, the first anatomical barrier is the cervix, where spermatozoa with poor morphology and motility are filtered out by sticking to the cervical mucus. The second anatomical barrier is the uterotubal junction (UTJ) with its tortuous and narrow lumen. Finally, only a fe

Establishment of a transport system for mouse epididymal sperm at refrigerated temperatures.

The exchange of genetically engineered mouse strains between research facilities requires transporting fresh mouse sperm under refrigerated temperatures. Although sperm generally maintains fertility for 48 h at cold temperatures, in vitro fertilization rates of C57BL/6 mouse sperm are low after 48-h cold storage. Furthermore, 48 h is often not sufficient for the specimens to reach their destinations. To increase the availability of this technology, we aimed to extend the cold storage period while maintaining sperm fertility. In this study, we determined the optimal medium for sperm preservatio
