Crocodilian perivitelline membrane-bound sperm detection.

Advanced reproductive technologies (ART's) are often employed with various taxa to enhance captive breeding programs and maintain genetic diversity. Perivitelline membrane-bound (PVM-bound) sperm detection has previously been demonstrated in avian and chelonian species as a useful technique for breeding management. In the absence of embryotic development within an egg, this technique can detect the presence of sperm trapped on the oocyte membrane confirming breeding, male reproductive status, and pair compatibility. PVM-bound sperm were successfully detected in three clutches of Cuban crocodil

Enigmas of mammalian gamete form and function.

The gametes of man and some other Eutheria have been manipulated successfully for practical reasons, but many gaps remain in our basic understanding of the way that they function. This situation stems not least from a failure to recognize the extent to which eutherian spermatozoa and eggs, and elements related to their operation, have come to differ from those of other groups. Novel features in the male that reflect this include a radical design of the sperm head with the acrosome seeming to function primarily in egg-coat binding rather than its lysis, a multifaceted post-testicular sperm matu

Multifunctional 4D-Printed Sperm-Hybrid Microcarriers for Assisted Reproduction.

Remotely controllable microrobots are appealing for various biomedical in vivo applications. In particular, in recent years, our group has focused on developing sperm-microcarriers to assist sperm cells with motion deficiencies or low sperm count (two of the most prominent male infertility problems) to reach the oocyte toward in-vivo-assisted fertilization. Different sperm carriers, considering their motion in realistic media and confined environments, have been optimized. However, the already-reported sperm carriers have been mainly designed to transport single sperm cell, with limited functi

Two egg-derived molecules in sperm motility initiation and fertilization in the Pacific herring (Clupea pallasi).

Sperm of the Pacific herring are immotile at spawning. Two egg-derived molecules are capable of initiating sperm motility. One is herring sperm activating protein(s) (HSAPs) and the other is sperm motility initiation factor (SMIF). These two motility initiators differ in their location and association with the chorion, and in their isoelectric points and molecular weights. In this study we have investigated the roles of these two inducers with respect to motility and fertilization. Using computer analysis of sperm motility, we found that HSAPs, as well as the C-terminal HSAPs peptide, elicit a

Cellular mechanisms regulating sperm-zona pellucida interaction.

For mammalian spermatozoa to exhibit the ability to bind the zona pellucida (ZP) they must undergo three distinct phases of maturation, namely, spermatogenesis (testis), epididymal maturation (epididymis) and capacitation (female reproductive tract). An impressive array of spermatozoa surface remodeling events accompany these phases of maturation and appear critical for recognition and adhesion of the outer vestments of the oocyte, a structure known as the ZP. It is becoming increasingly apparent that species-specific zona adhesion is not mediated by a single receptor. Instead, compelling evid

Methods for enhancement of sperm function.

From a review of recent advances in human reproduction, it is apparent that therapeutic approaches to male infertility have been revolutionized. While our understanding of sperm function at the molecular level is steadily increasing, the realization of consistent oocyte fertilization by mechanically bypassing natural barriers has given new perspective for future investigation. This chapter reviews current knowledge of adjuvants that enhance sperm function and lend themselves to clinical application. Each compound is presented with recent publications supporting proposed mechanisms of action. S

Estimating the binding ability of collared peccary (Pecari tajacu Linnaeus, 1758) sperm using heterologous substrates.

In collared peccaries, the development of artificial insemination (AI) is scarce, requiring search for alternative methods for the evaluation of sperm fertilizing ability. Thus, the aims of this study were to estimate the binding capability of collared peccaries sperm, using swine oocytes and the egg perivitelline membrane, and to evaluate the prognostic value of sperm parameters on the in vitro interactions among sperm and heterologous substrates. Eleven ejaculates were collected by eletroejaculation and evaluated for viability and morphology by light microscopy, for functionality by hypo-osm

Mechanisms of filtration of morphologically abnormal human sperm by cervical mucus.

It is well known that cervical mucus restricts penetration of morphologically abnormal human sperm, both in vitro and in vivo. However, the mechanisms of such restriction are not well understood. Using videomicrography to simultaneously analyze the motions and morphology of individual human sperm, we analyzed differential penetration of normal and abnormal sperm into fresh human cervical mucus. Abnormal sperm swam slower in mucus than the normal sperm, but their flagellar beat parameters were not commensurately different. Multivariate statistical analysis of the relationship between individual

Rat DCST1 and DCST2 are essential for sperm-egg fusion†.

Physiological functions of fertilization-related genes in male fertility differ among species. The sperm membrane proteins, DC-STAMP domain containing (DCST) 1 and 2, are essential for male fertility in mice and zebrafish. Although dcst2 knockout (KO) zebrafish sperm showed impaired binding to the oolemma, mouse Dcst2 KO sperm showed impaired fusion after binding to the oolemma. Thus, the physiological functions of the target genes must be analyzed in various genetically modified animals. In this study, the physiological function of DCST1/2 in male fertility was examined in Dcst1/2 double-KO (

The functional integrity and fate of cryopreserved ram spermatozoa in the female tract.

Cryopreservation advances capacitation-like changes in ram spermatozoa. These changes are reflected in an increased fertilizing ability compared with fresh spermatozoa, followed by an accelerated decline in fertilizing ability after incubation in vitro or in vivo. Furthermore, frozen-thawed spermatozoa are released earlier than fresh spermatozoa after binding to oviduct cells in vitro, confirming their physiological readiness to participate in fertilization despite their short lifespan. After insemination large numbers of spermatozoa are lost from the female reproductive tract of the ewe via t

The fallopian tube and reproductive health.

The fallopian tube is the essential link between the ovary and the uterus. Its transport mechanisms are now reasonably well understood and include orderly ovum transfer by the fimbria, and ovum/preembryo retention, with transport to the uterus on the third postovulatory day. Sperm/tube interaction ensures a reservoir and storage/activation system at the tubal isthmus. Ovum/tube interaction is driven by the HCO3 ion in the tubal secretions, which also supply pyruvate and other essential substances to the preembryo. Tubal function may be impaired/destroyed by salpingitis, a peritubal disease res

The impact of epididymal proteins on sperm function.

The epididymis is necessary for post-testicular sperm maturation as it provides the milieu required for spermatozoa to gain the ability for progressive movement and fertilization. In the epididymis the sperm protein, lipid and small RNA content are heavily modified due to interaction with luminal proteins secreted by the epididymal epithelium and extracellular vesicles, epididymosomes. This review focuses on epididymal proteins demonstrated to have an effect on sperm functions, such as motility, capacitation, acrosome reaction, sperm-zona pellucida binding and sperm-egg binding, as well as on

Correlation between semen adenosine triphosphate and sperm fertilizing capacity.

The concentration of ATP was determined in a large number of semen samples from fertile men and infertility clinic patients. Results were correlated with routine semen characteristics, in vitro sperm fertilizing capacity, and functional integrity of the sperm membrane. The results confirm the findings of Comhaire et al. and Irvine and Aitken that there are positive correlations between semen ATP concentration and sperm count and sperm motility. In routine laboratory examination of semen specimen, the results agree with those of Irvine and Aitken that semen ATP determination does not appear to

[CatSper in sperm hyperactivation and male infertility: Advances in studies].

The CatSper channel is known as one of the most important Ca²⁺ channels on the cell membrane of mammalian sperm and plays a key role in the motility, hyperactivation and fertilization function of sperm. The CatSper protein, expressed exclusively in the principal piece of the sperm tail, is composed of CatSper1-4 and 5 auxiliary unitsβ,γ,δ and ε, and has an essential part in the functional and structural domains of Ca²⁺as well as in the spatiotemporal regulation of the P-Tyr protein, sperm hyperactivation, efficient sperm migration in the oviduct, egg penetration, and normal fertility. Influence of the quality of the cervical mucus on sperm penetration: comparison of the morphologic features of spermatozoa in 101 postcoital tests with those in the semen of the husband.

The purpose of this study was to determine the frequency of abnormal spermatozoa in mucus by a postcoital test and to compare the findings with the teratospermia in a control analysis of the husband's semen. These data obtained from 101 hypofertile couples were correlated with the quality of mucus, which was classified into three categories according to rigorous scoring procedures. The morphologic features of spermatozoa were evaluated by the same investigator and classified on the basis of a system that allowed for multiple entries. This method provided an exact description when several anoma

Epididymosomes: transfer of fertility-modulating proteins to the sperm surface.

A variety of glycosylphosphatidylinositol (GPI)-linked proteins are acquired on spermatozoa from epididymal luminal fluids (ELF) during sperm maturation. These proteins serve roles in immunoprotection and in key steps of fertilization such as capacitation, acrosomal exocytosis and sperm-egg interactions. Their acquisition on sperm cells is mediated both by membrane vesicles (epididymosomes, EP) which were first reported to dock on the sperm surface, and by lipid carriers which facilitate the transfer of proteins associated with the membrane-free fraction of ELF. While the nonvesicular fraction

Can spermatozoa with abnormal heads gain access to the ovum in artificially inseminated super- and single-ovulating cattle?

The collective efficiency of barriers in the female tract against spermatozoa with abnormal heads was studied. In Experiment 1, Day 6 ova/embryos were recovered nonsurgically from superovulated (n = 24) and single-ovulating (n = 44) cows following artificial insemination with semen of bulls selected for normal spermatozoal motility (> or = 50%) and high content (> 30%) of spermatozoa with misshapen heads, random nuclear vacuoles or the diadem defect. To assess characteristics of spermatozoa capable of traversing barriers in the female tract, accessory spermatozoa were classified morphologicall

Turkey sperm mobility influences paternity in the context of competitive fertilization.

We have devised a novel means of investigating competitive fertilization in turkeys, using microsatellite genotyping to identify male parentage. Our results demonstrate that sperm mobility is a mechanism responsible in part for paternity efficiency in turkeys. Sperm mobility is composed of several parameters in which sperm motility is a component. Differences between ejaculates in the number of sperm penetrating into a dense, insert, nontoxic solution were measured and used to classify males into high, average, or low sperm mobility phenotypes. Microsatellite genotyping was used to determine p

Sperm guidance in mammals - an unpaved road to the egg.

Contrary to the prevalent view, there seems to be no competition in the mammalian female genital tract among large numbers of sperm cells that are racing towards the egg. Instead, small numbers of the ejaculated sperm cells enter the Fallopian tube, and these few must be guided to make the remaining long, obstructed way to the egg. Here, we review the mechanisms by which mammalian sperm cells are guided to the egg.

Quantitative determination of spermatozoa penetration of the perivitelline layer of the hen's ovum as assessed on oviposited eggs.

A technique was developed to assess the number of cock spermatozoa penetrating the perivitelline layer (PL) in oviposited eggs in vivo. Two trials were conducted to test this technique and to establish correlation values between fertility and sperm penetration (SP). First, three Athens Canadian Randombred males, previously tested as having high fertility (100%), were each housed with seven hens. Sperm penetration was determined from eggs laid over a 3-d period (n = 41) with the mean number of spermatozoa penetrating the PL overlying the germinal disc (GD; 1.35 mm2 area) and nongerminal disc (N
