[Reparative and neoplastic spheroid cellular structures and their mathematical model].

Spheroid cell structures in the cell cultures have been described and are used for studying cell-cell and cell- matrix interactions. At the same time, spheroid cell structure participation in the repair and development of cancer in vivo remains unexplored. The aim of this study was to investigate the cellular composition of spherical structures and their functional significance in the repair of squamous epithelium in human papilloma virus-associated cervical pathology--chronic cervicitis and cervical intraepithelial neoplasia 1-3 degree, and also construct a mathematical model to explain the d

Sonoprinting nanoparticles on cellular spheroids via surface acoustic waves for enhanced nanotherapeutics delivery.

Nanotherapeutics, on their path to the target tissues, face numerous physicochemical hindrances that affect their therapeutic efficacy. Physical barriers become more pronounced in pathological tissues, such as solid tumors, where they limit the penetration of nanocarriers into deeper regions, thereby preventing the efficient delivery of drug cargo. To address this challenge, we introduce a novel approach that employs surface acoustic wave (SAW) technology to sonoprint and enhance the delivery of nanoparticles onto and into cell spheroids. Our SAW platform is designed to generate focused and un

3D cellular spheroids as tools for understanding carboxylated quantum dot behavior in tumors.

BACKGROUND: Monolayer cell cultures have been considered the most suitable technique for in vivo cellular experiments. However, a lot of cellular functions and responses that are present in natural tissues are lost in two-dimensional cell cultures. In this context, nanoparticle accumulation data presented in literature are often not accurate enough to predict behavior of nanoparticles in vivo. Cellular spheroids show a higher degree of morphological and functional similarity to the tissues. METHODS: Accumulation and distribution of carboxylated CdSe/ZnS quantum dots (QDs), chosen as model nan

Mineralization-driven cellular dynamics and tissue remodeling in scaffold-free MC3T3-E1 spheroids.

Bone mineralization is a critical process in bone tissue formation and remodeling, yet the underlying mechanisms in three-dimensional (3D) cellular environments remain unclear. In this study, we fabricated scaffold-free spheroids using mouse osteoblast-like MC3T3-E1 cells and evaluated their long-term morphological and mechanical changes to elucidate the mineralization process. The spheroids were cultured for up to 35 days in osteogenic medium, and their size reduction (53.2 % compared to 2-day spheroid), cellular viability, collagen accumulation, and mineral deposition were systematically ana

Resolving femtogram-scale cellular nanomagnetism via high-gradient magnetophoresis of spheroids in microfluidic chips.

Quantifying trace amounts of magnetic iron in living cells remains a major challenge in cellular biology and nanomedicine. Existing approaches such as magnetometry or elemental analysis either require large cell populations or cannot discriminate endogenous iron from magnetic phases, preventing measurements at the femtogram-per-cell scale. Here, we introduce a high-gradient magnetophoresis approach combined with multicellular spheroids to achieve femtogram-level sensitivity in intact 3D cellular assemblies. By exploiting the scaling of magnetic force with spheroid volume and hydrodynamic drag

Generation and Quantification of Cytotoxic Lymphocytes Following Oncolytic Virus Infection of Multi-cellular Tumor Spheroids.

Oncolytic viruses (OVs) rapidly and specifically replicate in and kill tumor cells. OV-targeted infection of malignant cells has the potential to create an "inflammatory storm" that stimulates both innate and adaptive anti-tumor immune responses. The generation of anti-tumor immunity following OV treatment has been shown to be crucial for effective therapy. Therefore, establishing methodologies to measure the generation of anti-tumor T cell responses following OV infection in in vitro assays, which better mimic the complexity of the human tumor microenvironment (TME), will be critical to harne

Investigations of cardiac fibrosis rheology by in vitro cardiac tissue modeling with 3D cellular spheroids.

Cardiac fibrosis refers to the abnormal accumulation of extracellular matrix within the cardiac muscle, leading to increased stiffness and impaired heart function. From a rheological standpoint, knowledge about myocardial behavior is still lacking, partially due to a lack of appropriate techniques to investigate the rheology of in vitro cardiac tissue models. 3D multicellular cardiac spheroids are powerful and versatile platforms for modeling healthy and fibrotic cardiac tissue in vitro and studying how their mechanical properties are modulated. In this study, cardiac spheroids were created by

Uncovering the Potential of 3D Spheroids for Modeling the Cellular Crosstalk Within the Tumor Microenvironment in HNSCC.

Head and neck squamous cell carcinoma (HNSCC) progression is profoundly influenced by dynamic cellular crosstalk within the tumor microenvironment (TME), where interactions between cancer cells, stromal components, and immune populations orchestrate therapeutic resistance, metastasis, and immune evasion. This review examines the utility of three-dimensional (3D) spheroid models as physiologically relevant platforms for investigating these cellular interactions compared with conventional two-dimensional (2D) cultures. By incorporating cancer-associated fibroblasts (CAFs), tumor-associated macro

Impaired expansion of trophoblast spheroids cocultured with endometrial cells overexpressing cellular retinoic acid-binding protein 2.

We previously showed that the cellular retinoic acid-binding protein 2 (CRABP2) gene was up-regulated during the implantation window period in the endometrium of patients with unexplained recurrent spontaneous abortion. Here, we report that trophoblast spheroids cocultured with a human endometrial cell line stably overexpressing CRABP2 are defective in expansion and exhibit apoptosis, suggesting that the altered expression level of endometrial CRABP2 is involved in abnormal endometrium-trophoblast interaction, which leads to implantation failure.

Fiber-Optic Quantum Dots Sensor for Dynamic and Quantitative Thermal Monitoring of Spheroids toward Single-Cellular Resolution.

Accurate monitoring of thermal dynamics at the single-cellular scale is crucial for elucidating metabolism, signaling pathways and disease mechanisms, yet remains hampered by the limited stability and spatiotemporal resolution of current sensors. Here, we report a fiber-optic quantum dots sensor (FOQDS), which is based on a tapered fiber optic tip functionalized with CdSe/CdS/ZnS QDs and encapsulated by a dense Al2O3 layer to ensure exceptional long-term stability. FOQDS features a linear thermal response with high sensitivity (145 pm/°C), a temporal resolution of <10 μs, with the potential to

Creation of Formalin-Fixed, Paraffin-Embedded 3D Lung Cancer Cellular Spheroids for the Optimization of Immunohistochemistry Staining Procedures.

In an era of precision medicine important treatment decisions are dictated by expression of clinically informative tumor protein biomarkers. These biomarkers can be detected by immunohistochemistry (IHC) performed in tumor tissue sections obtained from biopsies or resections. Like all experimental procedures, IHC needs optimization for several of its steps. However, the investigator must avoid optimizing the IHC procedure using valuable human biopsy samples which may be difficult to obtain. Ideally, valuable biopsy samples should only be subjected to IHC once the IHC protocol has been optimize

Altered Magnesium Environments Restrict Colorectal HT-29 Spheroid Growth by Disturbing Cellular Mg2+ Homeostasis.

Dysregulated magnesium (Mg2+) homeostasis contributes to colorectal cancer (CRC), yet its context-dependent function within the tumor microenvironment remains unresolved. This study aimed to determine how sustained low and high extracellular Mg2+ environments affect CRC spheroid (SP) growth and Mg2+ homeostasis using HT-29 SPs. We analyzed Mg2+ flux, the expression of Mg2+ transporters (e.g., Transient Receptor Potential Melastatin (TRPM) 6), viability, apoptotic and autophagic markers, and phospho-/oxidoproteomic alterations. Both Mg2+ extremes destabilized SP architecture, reduced viability,

Soft microfingers with flexible tactile sensor using liquid metal for in situ evaluation of cellular spheroid stiffness.

Inflatable soft microfingers integrated with liquid metal strain gauge sensors have been developed for the in situ evaluation of cellular spheroid stiffness. Cellular spheroids ranging from several hundred micrometers to a few millimeters in diameter, typical three-dimensional cellular aggregates, have been used as biological models. Cellular spheroids are evaluated using various methods, of which microscopic imaging is the most popular. This study focuses on mechanical characteristics, such as stiffness, related to the internal structure of cellular spheroids and reports an evaluation device

Physical Properties and Cellular Metabolic Characteristics of 3D Spheroids Are Possible Definitive Indices for the Biological Nature of Cancer-Associated Fibroblasts.

The current study's objective was to elucidate some currently unknown biological indicators to evaluate the biological nature of cancer-associated fibroblasts (CAFs). For this purpose, four different CAFs, CAFS1, CAFS2, SCC17F and MO-1000, were established using surgical specimens from oral squamous cell carcinomas (OSCC) with different clinical malignant stages (CAFS1 and CAFS2, T2N0M0, stage II; SCC17F and MO-1000, T4aN2bM0, stage IVA). Fibroblasts unrelated to cancer (non-CAFs) were also prepared and used as controls. Initially, confirmation that these four fibroblasts were indeed CAFs was

A Novel Cellular Spheroid-Based Autophagy Screen Applying Live Fluorescence Microscopy Identifies Nonactin as a Strong Inducer of Autophagosomal Turnover.

Dysregulation of the basal autophagic flux has been linked to several pathological conditions, including neurodegenerative diseases and cancer. In addition, autophagy has profound effects on the response of tumor cells to therapy. Hence, the search for pharmacological modulators of autophagy is of great clinical relevance. We established a drug screening assay in which the autophagic flux is measured by recording the fluorescence emission of the tandem fusion protein mRFP-GFP-LC3 by dynamic live-cell imaging. We optimized the assay for the identification of autophagy modulators in three dimens

Standard chemotherapy impacts on in vitro cellular heterogeneity in spheroids enriched with cancer stem cells (CSCs) derived from triple-negative breast cancer cell line.

Triple-negative breast cancer is a heterogeneous disease with high recurrence and mortality, linked to cancer stem cells (CSCs). Our study characterized distinct cell subpopulations and signaling pathways to explore chemoresistance. We observed cellular heterogeneity among and within the cells regarding phenotyping and drug response. In untreated BT-549 cells, we noted plasticity properties in both CD44+/CD24+/CD146+ hybrid cells and CD44-/CD24+/CD146+ epithelial cells, enabling phenotypic conversion into CD44+/CD24-/CD146- epithelial-mesenchymal transition (EMT)-like like breast CSCs (BCSCs).

Proteomic analysis reveals a metabolic switch and cellular remodelling in pancreatic-like cell spheroids cocultured with hepatocyte-like cells using an interconnected liver-pancreas-on-chip system.

Systemic interactions between the liver and pancreas are critical for regulating lipid and carbohydrate homeostasis. These organs are also deeply interdependent in pathological conditions such as insulin resistance, where the liver fails to respond appropriately to insulin stimulation, leading to dysfunction in both organs. To investigate the crosstalk between these tissues using human models, we developed an advanced organ-on-chip (OoC) device hosting human induced pluripotent stem cell-derived liver and pancreatic tissues. The proteomic response of pancreatic-like cell spheroids cocultured w

Cellular alterations identified in pluripotent stem cell-derived midbrain spheroids generated from a female patient with progressive external ophthalmoplegia and parkinsonism who carries a novel variation (p.Q811R) in the POLG1 gene.

Variations in the POLG1 gene encoding the catalytic subunit of the mitochondrial DNA polymerase gamma, have recently been associated with Parkinson's disease (PD), especially in patients diagnosed with progressive external ophthalmoplegia (PEO). However, the majority of the studies reporting this association mainly focused on the genetic identification of the variation in POLG1 in PD patient primary cells, and determination of mitochondrial DNA copy number, providing little information about the cellular alterations existing in patient brain cells, in particular dopaminergic neurons. Therefore
