Diversity and antibiotic resistance patterns of Sphingomonadaceae isolates from drinking water.

Sphingomonadaceae (n = 86) were isolated from a drinking water treatment plant (n = 6), tap water (n = 55), cup fillers for dental chairs (n = 21), and a water demineralization filter (n = 4). The bacterial isolates were identified based on analysis of the 16S rRNA gene sequence, and intraspecies variation was assessed on the basis of atpD gene sequence analysis. The isolates were identified as members of the genera Sphingomonas (n = 27), Sphingobium (n = 28), Novosphingobium (n = 12), Sphingopyxis (n = 7), and Blastomonas (n = 12). The patterns of susceptibility to five classes of antibiotics

Ileal mucosa-associated microbiota overgrowth associated with pathogenesis of primary biliary cholangitis.

The small intestinal mucosa-associated microbiota (MAM) can potentially impact the etiology of primary biliary cholangitis (PBC). Herein, we investigate the MAM profile to determine its association with liver pathology in patients with PBC. Thirty-four patients with PBC and 21 healthy controls who underwent colonoscopy at our hospital were enrolled in our study. We performed 16S ribosomal RNA gene sequencing of MAM samples obtained from the mucosa of the terminal ileum and examined the relationship between the abundance of ileal MAM and chronic nonsuppurative destructive cholangitis using live

Altered blood microbiome in patients with HCV-related Child-Pugh class B cirrhosis.

BACKGROUND: Altered bacterial translocation is associated with changes in hepatic function and the progression from compensated to decompensated cirrhosis. Child-Turcotte-Pugh (CTP) score is an essential indicator of liver severity. Thus, we aimed to study differences in the blood microbiome together with metabolome profile between HCV-infected patients with CTP class B (CTP-B, significant functional compromise) and patients with CTP class A (CTP-A, well-compensated cirrhosis). METHODS: We conducted a cross-sectional study in patients with advanced HCV-related cirrhosis (n = 88) stratified by

Microbial aerobic degradation of 4-isopropylnitrobenzene by Sphingobium yanoikuyae strain SG1.

4-Isopropylnitrobenzene (4-IPNB) is a nitroaromatic compound commonly employed as an intermediate in pesticide synthesis and chemical manufacturing. Despite its potential environmental persistence and ecological risks, the microbial degradation pathway of 4-IPNB remains largely unknown. In this study, a Gram-negative bacterium, designated Sphingobium yanoikuyae strain SG1, was isolated from a pesticide manufacturing site in Brazil for its ability to utilize 4-IPNB as the sole added source of carbon, nitrogen, and energy. Aerobic degradation of 4-IPNB by strain SG1 was accompanied by nitrite re

Haloalkane dehalogenases other than LinB can contribute to the γ-hexachlorocyclohexane utilization.

Haloalkane dehalogenases (HLDs) convert halogenated compounds to corresponding alcohols by a simple hydrolytic mechanism. Although many bacterial strains possess HLDs or HLD homologues, LinB is the only HLD known to be involved in the utilization of γ-hexachlorocyclohexane (γ-HCH), a man-made chlorinated pesticide. In this study, to gain insight into the functional evolution of HLDs toward γ-HCH utilization, the linB gene in γ-HCH-degrading Sphingobium japonicum strain UT26 was replaced by 7 other HLD or HLD homologous genes, including 3 putative ancestral enzymes. Interestingly, strains carry

Biodegradation of Triphenyl Phosphate by Gordonia polyisoprenivorans YC-XJ4: Environmental Adaptability and Bioremediation Potential.

Triphenyl phosphate (TPHP) is a widely used organophosphate flame retardant (OPFR) with concerning environmental persistence and toxicity. Microbial degradation is a promising removal strategy, but highly efficient degraders outside the Sphingomonadaceae family remain rarely reported. In this study, a highly efficient TPHP-degrading strain, designated YC-XJ4, was isolated from plastic-waste-contaminated soil and identified as Gordonia polyisoprenivorans through genomic analysis. The strain exhibited robust degradation activity over broad ranges of temperature (15-40 °C), pH (6.0-10.0), and sal

Aromatic Dimer Dehydrogenases from Novosphingobium aromaticivorans Reduce Monoaromatic Diketones.

Lignin is a potential source of valuable chemicals, but its chemical depolymerization results in a heterogeneous mixture of aromatics and other products. Microbes could valorize depolymerized lignin by converting multiple substrates into one or a small number of products. In this study, we describe the ability of Novosphingobium aromaticivorans to metabolize 1-(4-hydroxy-3-methoxyphenyl)propane-1,2-dione (G-diketone), an aromatic Hibbert diketone that is produced during formic acid-catalyzed lignin depolymerization. By assaying genome-wide transcript levels from N. aromaticivorans during growt

Biodegradation of a phthalate plasticizer, di-isononyl phthalate (DINP), by Sphingobium chungbukense.

In this study, di-isononyl phthalate (DINP) was efficiently degraded by Sphingobium chungbukense KCTC 2955. The optimal conditions for DINP (100 mg L(-1)) degradation by S. chungbukense in a mineral salts medium were found to be pH 7.0, 30 degrees C, and stirring at 200 rpm. The maximum specific rate of DINP degradation was found to be concentration dependent, with a maximum of 4.12 mg DINP L(-1) h(-1). DINP was transformed rapidly by S. chungbukense, with the formation of monoisononyl phthalate (MIP) and phthalic acid, which subsequently degraded further. These results highlight the potential

Catabolism of β-5 linked aromatics by Novosphingobium aromaticivorans.

UNLABELLED: Aromatic compounds are an important source of commodity chemicals traditionally produced from fossil fuels. Aromatics derived from plant lignin can potentially be converted into commodity chemicals through depolymerization followed by microbial funneling of monomers and low molecular weight oligomers. This study investigates the catabolism of the β-5 linked aromatic dimer dehydrodiconiferyl alcohol (DC-A) by the bacterium Novosphingobium aromaticivorans. We used genome-wide screens to identify candidate genes involved in DC-A catabolism. Subsequent in vivo and in vitro analyses of

Bacterial Catabolism of β-Hydroxypropiovanillone and β-Hydroxypropiosyringone Produced in the Reductive Cleavage of Arylglycerol-β-Aryl Ether in Lignin.

Sphingobium sp. strain SYK-6 converts four stereoisomers of arylglycerol-β-guaiacyl ether into achiral β-hydroxypropiovanillone (HPV) via three stereospecific reaction steps. Here, we determined the HPV catabolic pathway and characterized the HPV catabolic genes involved in the first two steps of the pathway. In SYK-6 cells, HPV was oxidized to vanilloyl acetic acid (VAA) via vanilloyl acetaldehyde (VAL). The resulting VAA was further converted into vanillate through the activation of VAA by coenzyme A. A syringyl-type HPV analog, β-hydroxypropiosyringone (HPS), was also catabolized via the sa

PhcK mediates transport of a β-5-type lignin-derived dimer in Sphingobium lignivorans SYK-6.

Lignin is a major component of plant cell walls; however, its heterogeneous and complex structure has hindered its efficient utilization. Recently, strategies that combine chemical pretreatment with microbial conversion to produce valuable chemicals have attracted attention. To develop an ideal microbial platform for this purpose, it is essential to elucidate the complete bacterial catabolic system for lignin-derived aromatic compounds. Here, we identified an inner membrane transporter gene involved in the uptake of a metabolite of dehydrodiconiferyl alcohol (DCA), a lignin-derived β-5 dimer,

Occurrences of 2-methylisoborneol and geosmin -degrading bacteria in a eutrophic reservoir and the role of cell-bound versus dissolved fractions.

As taste-and-odor outbreaks are common in surface waters worldwide, extensive studies have focused on the identification of microorganisms involved in the production of 2-methylisoborneol (MIB) and geosmin (GSM). However, fewer studies have tried to identify potential degraders in natural environments. Eagle Creek Reservoir, a temperate and eutrophic water body, experienced two major seasonal odorous outbreaks in 2013 with maximal concentrations of 99.1 (MIB) and 77.3 ng L-1 (GSM). Fractionation analyses of the odorous compounds showed that MIB was found more frequently in the dissolved fracti

Functional and phylogenetic evidence of a bacterial origin for the first enzyme in sphingolipid biosynthesis in a phylum of eukaryotic protozoan parasites.

Toxoplasma gondii is an obligate, intracellular eukaryotic apicomplexan protozoan parasite that can cause fetal damage and abortion in both animals and humans. Sphingolipids are essential and ubiquitous components of eukaryotic membranes that are both synthesized and scavenged by the Apicomplexa. Here we report the identification, isolation, and analyses of the Toxoplasma serine palmitoyltransferase, an enzyme catalyzing the first and rate-limiting step in sphingolipid biosynthesis: the condensation of serine and palmitoyl-CoA. In all eukaryotes analyzed to date, serine palmitoyltransferase is

Identification of key degraders for controlling toxicity risks of disguised toxic pollutants with division of labor mechanisms in activated sludge microbiomes: Using nonylphenol ethoxylate as an example.

Efficient management of disguised toxic pollutants (DTPs), which can undergo microbial degradation and convert into more toxic substances, necessitates the collaboration of diverse microbial populations in wastewater treatment plants. However, the identification of key bacterial degraders capable of controlling the toxicity risks of DTPs through division of labor mechanisms in activated sludge microbiomes has received limited attention. In this study, we investigated the key degraders capable of controlling the risk of estrogenicity associated with nonylphenol ethoxylate (NPEO), a representati

Degradation of 17β-estradiol by Novosphingobium sp. ES2-1 in aqueous solution contaminated with tetracyclines.

17β-estradiol (E2) often coexists with tetracyclines (TCs) in wastewater lagoons at intensive breeding farms, threatening the quality of surrounding water bodies. Microbial degradation is vital in E2 removal, but it is unclear how TCs affect E2 biodegradation. This primary study investigated the mechanisms of E2 degradation by Novosphingobium sp. ES2-1 in the presence of TCs and assessed the removal efficiency of E2 by strain ES2-1 in natural waters containing TCs. E2 biodegradation was unaffected at TCs concentrations below 0.1 mg L-1 yet significantly inhibited at TCs above 10 mg L-1. As ele
