Methods in molecular biology (Clifton, N.J.)2026eng
After an injury, adult mammals do not regenerate or repair their central nervous system in general, and their spinal cord in particular, leading to life-long disabilities. However, many aquatic species such as zebrafish, salamanders and frogs, as well as some land reptiles (turtles and lizards) have the remarkable ability to regenerate their spinal cords. In the case of anurans such as Xenopus, this is a transient ability that disappears after metamorphosis, making it an interesting model with accessible regenerative and nonregenerative stages within the same species. Here, we will summarize o

Vertebrates exhibit a range of regenerative capacities following spinal cord injury. At one end of the spectrum are chief regenerators, including teleost fish and urodele amphibians. At the other end, most mammalian species exhibit limited repair and multicellular complications following spinal cord injury. Pro-regenerative immune, glial and neuronal injury responses underlie innate spinal cord repair in highly regenerative vertebrates. In many instances, fundamental mechanisms of spinal cord repair represent ancestral neuroprotection mechanisms that are conserved but become overwhelmed by ant

Spinal cord injury remains difficult to treat because of the intrinsically limited regenerative capacity of neurons. Although neural progenitor cell (NPC) therapies are promising, inadequate graft survival, uncontrolled differentiation and weak functional integration continue to restrict outcomes. Here we report biohybrid microrobots called NPCbots, fabricated by integrating human-induced pluripotent-stem-cell-derived NPCs with magnetoelectric nanoparticles, enabling wireless magnetic navigation and non-invasive neuronal stimulation. A lab-on-a-chip platform allows scalable fabrication and mai

Spinal cord injury in humans is a life-changing condition with no effective treatment. However, many non-mammalian vertebrates can fully regenerate their spinal cord after injury. Frogs such as Xenopus can regenerate the spinal cord at larval stages, but lose this capacity at metamorphosis. This makes them ideal models to elucidate molecular pathways underlying regenerative capacity by comparing responses to spinal cord injury in regenerative (R) and non-regenerative (NR) stages of the same species. Here we use quantitative proteomics with Isobaric Tags for Relative and Absolute Quantification

This hypothetical study explores a potential therapeutic strategy for patients with recent complete spinal cord injuries (≤6 weeks since injury approximately). We review current literature and identify five key determinants of regenerative failure: inflammation, glial scar formation, deficit in molecular guidance, loss of structural guidance, and persistence of Wallerian debris. Although each of these barriers can be addressed by existing interventions, their efficacy depends partially on a constrained sequence of application. Based on this, a temporally orchestrated strategy is proposed, comp

BACKGROUND: Nearly 11,000 cases of spinal cord injury (SCI) are reported in the United States annually. Current management options give a median survival time of 38 years; however, no rehabilitative measures are available. Stem cells have been under constant research given their ability to differentiate into neural cell lines replacing non functional tissue. Efforts have been made to establish new synapses and provide a conducive environment, by grafting cells from autologous and fetal sources; including embryonic or adult stem cells, Schwann cells, genetically modified fibroblasts, bone strom

Adult urodele amphibians possess extensive regenerative abilities, including lens, jaws, limbs, and tails. In this study, we examined the cellular events and time course of spinal cord regeneration in a species, Plethodon cinereus, that has the ability to autotomize its tail as an antipredator strategy. We propose that this species may have enhanced regenerative abilities as further coadaptations with this antipredator strategy. We examined the expression of nestin, vimentin, and glial fibrillary acidic protein (GFAP) after autotomy as markers of neural precursor cells and astroglia; we also t

Extensive regeneration of sensory axons into the spinal cord can be achieved experimentally after dorsal root injury, but no effort has been made to target regenerating axons and restore a normal lamina-specific projection pattern. Ectopic axon growth is potentially associated with functional disorders such as chronic pain and autonomic dysreflexia. This study was designed to target regenerating axons to normal synaptic locations in the spinal cord by combining positive and negative guidance molecules. Previously, we observed that, after dorsal rhizotomy, overexpression of NGF leads to robust

Stem cell therapy has been widely studied as a promising treatment for spinal cord injury (SCI). However, a lack of functional scaffolds for stem cell therapy to address SCI leads to low therapeutic efficacy due to poor survival of transplanted cells. To address these challenges, this study aims to enhance regenerative potential of spinal cord organoids (SCOs) by employing extracellular matrix (ECM) recapitulating spinal cord-specific microenvironment. Decellularized spinal cord-derived ECM (ScEM) supports 3D culture for development, maturation, and functionality of human induced pluripotent s

INTRODUCTION: Spinal cord injury (SCI) disrupts crucial neural pathways and causes severe neurological deficits that impact the quality of life. Although preliminary research has shown some promise of Spinal Cord Stimulation (SCS) in promoting axonal regeneration and remyelination, a clear understanding of the pertinent literature remains lacking. This scoping review consolidates evidence on the effects of SCS in promoting axonal growth and myelin development, preserving spared tissue integrity, and facilitating functional recovery following SCI.
METHODS: Pre-clinical studies from 1946 to 202

Spinal cord injury (SCI) causes significant sensory and motor deficits and paralysis due to the limited regenerative capacity of the mammalian central nervous system (CNS). The inflammatory response triggered by the injury is critical for successful spinal cord regeneration. Galanin is a neuropeptide acting primarily as an inhibitory neuromodulator or neurotransmitter in the central and peripheral nervous systems. Galanin is also expressed in various non-neuronal cell types, including immune cells. Galanin has been implicated in neuroprotection and inflammation; however, its contribution to sp

The spinal cord is one of the most important part of the human nervous system and great importance is placed on developing the best treatment for its damage. 3D bio-printing technology, and the fabrication of special scaffolds using it, is a potential solution for regenerating damage in spinal cord injuries (SCIs). Bio-printing can be divided into indirect and direct bio-printing, while among the bio-printing methods, inkjet bio-printing, fused deposition modeling (FDM), extrusion bio-printing, or light-assisted bio-printing can be distinguished. The last group can be in turn divided into seve

Directing the organization of cells into a tissue with defined architectures is one use of biomaterials for regenerative medicine. To this end, hydrogels are widely investigated as they have mechanical properties similar to native soft tissues and can be formed in situ to conform to a defect. Herein, we describe the development of porous hydrogel tubes fabricated through a two-step polymerization process with an intermediate microsphere phase that provides macroscale porosity (66.5%) for cell infiltration. These tubes were investigated in a spinal cord injury model, with the tubes assembled to

Segmentectomy of the spinal cord (SC) was performed in 28 rats at the level of the tenth thoracic vertebra. Scar formation at the operation site was studied, along with the completeness of the anatomical integrity of the SC, in control animals (group 1) and after filling of the defect with the neural matrix Spherogel (group 2) and with Spherogel containing embryonic nerve cells (group 3). The defect in the SC in animals of the control group was filled with fibrin masses by 1-2 weeks, while connective tissues scars were already formed by this time in rats of experimental groups 2 and 3. By 10-1

Spinal cord (SC) segmentectomy was performed at ThX level in 28 rats. The scar formation in place of surgery and the degree of restoration of anatomical integrity were studied in control rats (group I) and in rats, in which SC defect was replaced by either Spherogel neuromatrix (group II) or Spherogel containing dispersed embryonic nerve cells (group II). While in group I rats by weeks 1-2 SC defect was filled with fibrin masses, in rats of experimental groups II and III the formation of connective tissue scar was found by the same time. By weeks 10-11 these animals demonstrated partial restor

Spinal cord injuries often occur at the cervical level above the phrenic motor pools, which innervate the diaphragm. The effects of impaired breathing are a leading cause of death from spinal cord injuries, underscoring the importance of developing strategies to restore respiratory activity. Here we show that, after cervical spinal cord injury, the expression of chondroitin sulphate proteoglycans (CSPGs) associated with the perineuronal net (PNN) is upregulated around the phrenic motor neurons. Digestion of these potently inhibitory extracellular matrix molecules with chondroitinase ABC (denot

In mammals, spinal cord injury often results in permanent impairment of motor function owing to ineffective tissue regeneration. Unlike mammals, zebrafish have the remarkable ability to regenerate many tissues, including the spinal cord. Cross-species comparison is an attractive approach for revealing regeneration-specific mechanisms, but the large evolutionary distance between species sometimes hinders direct comparison. Recent studies have revealed that another model fish species, medaka, has a low regenerative ability in some tissues, making comparisons with them advantageous to revealing r

Nerve regeneration in hemisectioned spinal cords with and without supporting materials was examined in rats. Cell migration and newly formed capillaries surrounded by laminin-positive materials appeared at the lesioned site 7 days postoperatively. Reactive astrocytes increased at the lesioned site and extended into the spinal cord within 3 weeks. However, cell reactions decreased by 4 weeks and a cavity formed at the lesioned site in all animals. Regenerated axons were found only proximal to the lesioned site at 4 to 8 weeks. In the presence of supporting material (sciatic nerve containing pro

Spinal cord injury is a severe clinical problem, and research searching activity molecular that can promote spinal cord injury repairing is very prevalent. Mst3b can promote repair of damaged peripheral nerves and the optic nerve, but has been rarely reported in spinal cord injury research. Through detecting its expression in different periods of injured spinal cord, we found that the expression of Mst3b was significantly upregulated in injured spinal cord neurons. Increasing Mst3b expression using adenovirus in vivo and in vitro promoted axonal regeneration of spinal cord neurons, which led t

While previous research has identified key cell types involved in this process, the underlying molecular and cellular mechanisms remain largely unexplored. In this study, we used single-cell RNA sequencing to profile distinct cell populations at different stages of spinal cord injury in zebrafish. Our analysis revealed that multiple subpopulation
