Killing of Cryptosporidium sporozoites by Lactoferrin.

Intestinal infection caused by Cryptosporidium is a major contributor to diarrhea morbidity and mortality in young children around the world. Current treatments for children suffering from cryptosporidiosis are suboptimal. Lactoferrin is a glycoprotein found in breast milk. It has showed bacteriostatic and antimicrobial activity in the intestine. However, the effects of lactoferrin on the intestinal parasite Cryptosporidium have not been reported. In this study, we investigated the anticryptosporidial activity of human lactoferrin on different stages of Cryptosporidium. Physiologic concentrati

Development of sporozoite vaccines.

Protective immunity against malaria has been achieved in hosts ranging from birds to man by repeated inoculation of irradiated sporozoites. The main antigens involved in protective immunity to sporozoites are the circumsporozoite (CS) proteins, which are part of a family of proteins, covering the whole surface membrane of the parasite, and which have similar physico-chemical and antigenic properties. Monovalent fragments of monoclonal antibodies to CS proteins neutralize sporozoite infectivity. All monoclonal antibodies recognize a single immunodominant region within the various CS proteins, a

The novel Plasmodium gallinaceum sporozoite protein, Pg93, is preferentially expressed in the nucleus of oocyst sporozoites.

To study gene expression differences between oocyst and salivary gland sporozoites, cDNA libraries previously constructed from the two sporozoite populations of the avian malaria parasite, Plasmodium gallinaceum, were used in a subtractive hybridization protocol to isolate Pg93, a novel oocyst sporozoite gene. Pg93 encodes a putative approximately 76 kDa translated protein that was predicted to localize to the nucleus. Transcriptional analysis indicates that Pg93 is preferentially expressed in oocyst sporozoites versus salivary gland sporozoites. Immunolocalization assays confirm both the nucl

Infectivity and neutralization of Cryptosporidium parvum sporozoites.

Cryptosporidiosis, a diarrheal disease of calves and humans caused by the coccidian parasite Cryptosporidium parvum, is terminated in hosts with normal immune systems. To assess the mechanisms of immunity in cryptosporidiosis, it is necessary to isolate and quantitate sporozoites, the infective stage of Cryptosporidium spp. Here we report the (i) separation of infective C. parvum oocysts from calf feces by ether extraction, sieving, and hypochlorite treatment; (ii) separation of viable C. parvum sporozoites from intact and excysted oocysts by anion-exchange chromatography; and (iii) quantitati

Large-scale production of Plasmodium vivax sporozoites.

Mass-scale production of Plasmodium vivax sporozoites in Anopheles stephensi was achieved using the chimpanzee (Pan troglodytes) as a source of infective blood. Membrane feeding was as successful as feeding mosquitoes directly on the animal so long as the time between drawing the blood and feeding was restricted to 45 min. Longer delays such as 2-3 h resulted in loss of infectivity in terms of oocyst production. The selected strain of A. stephensi was highly susceptible to P. vivax (Chesson strain). A strain of A. stephensi relatively refractory to P. falciparum showed no cross-refractoriness

Flowcytometric and ImageStream Rna-Fish Gene Expression, Quantification and Phenotypic Characterization of Blood Sporozoites and Sporozoites From Human Malaria Species.

We adapted the RNA FISH Stellaris method to specifically detect the expression of Plasmodium genes by flow cytometry and ImageStream (Flow-FISH). This new method accurately quantified the erythrocytic forms of (1) Plasmodium falciparum and Plasmodium vivax and (2) the sexual stages of P vivax from patient isolates. ImageStream analysis of liver stage sporozoites using a combination of surface circumsporozoite protein (CSP), deoxyribonucleic acid, and 18S RNA labeling proved that the new Flow-FISH is suitable for gene expression studies of transmission stages. This powerful multiparametric sing

IMRAS-Immunization with radiation-attenuated Plasmodium falciparum sporozoites by mosquito bite: Cellular immunity to sporozoites, CSP, AMA1, TRAP and CelTOS.

BACKGROUND: Immunization with radiation-attenuated sporozoites (RAS) by mosquito bites provides >90% sterile protection against Plasmodium falciparum malaria in humans. We conducted a clinical trial based on data from previous RAS clinical trials that suggested that 800-1200 infected bites should induce ~50% protective vaccine efficacy (VE) against controlled human malaria infection (CHMI) administered three weeks after the final immunization. Two cohorts were immunized separately. VE was 55% in Cohort 1 but 90% in Cohort 2, the cohort that received a higher first dose and a reduced (fractiona

In vitro production of infectious Plasmodium falciparum sporozoites.

An effective vaccine is needed for the prevention and elimination of malaria. The only immunogens that have been shown to have a protective efficacy of more than 90% against human malaria are Plasmodium falciparum (Pf) sporozoites (PfSPZ) manufactured in mosquitoes (mPfSPZ)1-7. The ability to produce PfSPZ in vitro (iPfSPZ) without mosquitoes would substantially enhance the production of PfSPZ vaccines and mosquito-stage malaria research, but this ability is lacking. Here we report the production of hundreds of millions of iPfSPZ. iPfSPZ invaded human hepatocytes in culture and developed to ma

VIABILITY AND INFECTIVITY OF CRYOPRESERVED PLASMODIUM VIVAX SPOROZOITES.

Plasmodium vivax presents a great challenge to malaria control because of the ability of its dormant form in the liver, the hypnozoite, to cause relapse in otherwise fully recovered patient. Research efforts to better understand P. vivax hypnozoite biology have been hampered by the limited availability of its sporozoite form responsible for liver infection. Thus, the ability to cryopreserve and recover P. vivax sporozoites is an essential procedure. In this study, protective effects of hydroxyethyl starch (HES) alone and in combination with other cryoprotectants on P. vivax sporozoite recovery

Active and passive immunization against Plasmodium yoelii sporozoites.

Three subunit vaccines based on the major repeat, (QGPGAP)n, and flanking regions of the Plasmodium yoelii circumsporozoite protein were designed, produced, and tested. All were immunogenic, but none gave consistent protection against a 40-200 sporozoite challenge. To demonstrate that antibodies to P. yoelii CS protein could provide protection we established a passive transfer model. Passive transfer of NYS1, an IgG3 MAb against the P. yoelii CS protein, protected 100% of mice against challenge with 5000 P. yoelii sporozoites. Binding of NYS1 to sporozoites was inhibited by incubation with (QG

Spit it out: rhoptry organelles in Plasmodium sporozoites.

Plasmodium sporozoites (SPZs), the infective stage of malaria-causing parasites, colonize the salivary glands (SGs) of Anopheles mosquitoes and are transmitted to a vertebrate host during a blood meal. SPZs are injected into the skin, travel to the liver, and invade hepatocytes to replicate to form erythrocyte-invading merozoites. In Plasmodium, as in other apicomplexan parasites, invasion of host cells requires the coordinated secretion of apical secretory organelles, the micronemes and the rhoptries. This review highlights recent advances in unraveling the function of rhoptries in Plasmodium

Vaccination using normal live sporozoites under drug treatment.

Sterile immunity against malaria has been obtained in mammalian hosts exclusively through vaccination with whole parasite preparations. Induction of complete protection against challenge was obtained using sporozoites attenuated by irradiation or genetic manipulations. It has been demonstrated recently that immunization with normal sporozoites under chloroquine cover confers sterile protection in mice and humans, using substantially fewer parasites and injections than with irradiated sporozoite immunization. Subsequently, it was shown that other drugs can substitute for chloroquine. We describ

MAEBL Contributes to Plasmodium Sporozoite Adhesiveness.

The sole currently approved malaria vaccine targets the circumsporozoite protein-the protein that densely coats the surface of sporozoites, the parasite stage deposited in the skin of the mammalian host by infected mosquitoes. However, this vaccine only confers moderate protection against clinical diseases in children, impelling a continuous search for novel candidates. In this work, we studied the importance of the membrane-associated erythrocyte binding-like protein (MAEBL) for infection by Plasmodium sporozoites. Using transgenic parasites and live imaging in mice, we show that the absence

Tissue-dependent protection mechanisms of antibodies targeting Plasmodium sporozoites.

Malaria remains a major global health burden. Antibodies targeting the circumsporozoite protein (CSP), the main surface protein of Plasmodium sporozoites, have emerged as a promising prophylactic tool. Antibodies that bind to the CSP central repetitive region are the foundation of protection against sporozoite infection elicited by CSP-based vaccines. These antibodies have been shown to provide protection in both animal models and humans, although how they confer protection is not fully understood. This review explores how anti-CSP antibodies can neutralize sporozoites and how they mediate pro

Dissection-independent production of Plasmodium sporozoites from whole mosquitoes.

Progress towards a protective vaccine against malaria remains slow. To date, only limited protection has been routinely achieved following immunisation with either whole-parasite (sporozoite) or subunit-based vaccines. One major roadblock to vaccine progress, and to pre-erythrocytic parasite biology in general, is the continued reliance on manual salivary gland dissection for sporozoite isolation from infected mosquitoes. Here, we report development of a multi-step method, based on batch processing of homogenised whole mosquitoes, slurry, and density-gradient filtration, which combined with fr

Retention and survival of Plasmodium sporozoites in the spleen.

A central role of the spleen during the Plasmodium erythrocytic infection has been extensively described and acknowledged. However, whether it also influences the sporozoites in their journey toward the liver it is still unclear. In this paper we report that the intravenously inoculated P. yoelii and P. berghei sporozoites are retained in the spleen. In situ observations of parasite expressing GFP revealed the presence of live sporozoites in the splenic parenchyma for at least three hours after their inoculation. This suggests that the spleen might play an additional role beyond one of simply

Complete attenuation of Plasmodium falciparum sporozoites by atovaquone-proguanil.

To develop a Plasmodium falciparum (Pf) vaccine that precludes replication inside the host for improved vaccine safety, we tested chemo-attenuation (CVac) of sporozoites (SPZ) with atovaquone-proguanil (AP). In mice, P. berghei sporozoites administered with AP invaded hepatocytes, arrested early, and induced robust protection, which correlated with parasite-specific effector-memory CD8+ T cell responses. In a clinical trial of PfSPZ-CVac (AP), in which three doses of 5.12 × 104 or 1.5 × 105 PfSPZ were administered by direct venous inoculation combined with oral single-dose AP (1000/400 mg), bl

Ultrastructure, fractionation and biochemical analysis of Cryptosporidium parvum sporozoites.

Sporozoites of the apicomplexan parasite Cryptosporidium parvum were subjected to cell disruption and subcellular fractionation using a sucrose density step gradient. With this procedure, highly enriched preparations of the parasite membrane, the micronemes, dense granules and amylopectin granules were produced. No separate fraction containing rhoptries was obtained, however this organelle was found in defined fractions of the gradient, still associated with the apical tip of the sporozoites. Using negative staining, the internal structure of the micronemes was revealed by transmission electro

Cryptosporidium parvum merozoites share neutralization-sensitive epitopes with sporozoites.

Sporozoites and merozoites are stages in the life cycle of Cryptosporidium parvum that can cyclically infect intestinal cells, causing persistent infection and severe diarrhea in immunodeficient patients. Infection by sporozoites can be neutralized by surface-reactive mAb. We show that merozoite infectivity can also be neutralized by surface-reactive mAb. To do this, viable C. parvum merozoites were isolated by differential and isopycnic. centrifugation, and distinguished from sporozoites by transmission electron microscopy. Differential reactivity with a panel of seven mAb was used to determi

Plasmodium berghei Radiation-Attenuated Sporozoite-Immunized Mice Require Infectious Sporozoite Challenge to Maintain Protective Immunity.

Plasmodium radiation-attenuated sporozoites (RAS) confer sterile protection in mammalian hosts. The duration of protection is affected by the dose of RAS, the route of immunization, and the timing of primary challenge (PC). Giving PC shortly after the last Plasmodium berghei (Pb) RAS immunization of C75BL/6 mice led to the long-term sterile protection, whereas delaying PC beyond 6 months resulted in parasitemia. The mechanisms responsible for the divergent outcome remain unknown. Because liver effector/memory CD8+T cells are associated with lasting protection, herein we asked if any functions
