Canine Staphylococcaceae circulating in a Kenyan animal shelter.

Animal shelters, especially in resource-poor countries, bring together pets from different regions and with different backgrounds. The crowding of such animals often results in infectious diseases, such as respiratory infections. This study characterized Staphylococcaceae from diseased and apparently healthy dogs housed in an animal shelter in Kenya, to determine their antibiotic resistance profiles, their genetic relatedness, and the presence of dominant clones. Therefore, bacteria were collected from all 167 dogs present in the shelter in June 2015 and screened for Staphylococcaceae using st

Bats Are Carriers of Antimicrobial-Resistant Staphylococcaceae in Their Skin.

Bats have emerged as potential carriers of zoonotic viruses and bacteria, including antimicrobial-resistant bacteria. Staphylococcaceae has been isolated from their gut and nasopharynx, but there is little information about Staphylococcaceae on bat skin. Therefore, this study aimed to decipher the Staphylococci species in bat skin and their antimicrobial susceptibility profile. One hundred and forty-seven skin swabs were collected from bats during the spring and summer of 2021 and 2022. Bats were captured in different areas of the Metropolitan Region of São Paulo, Brazil, according to the degr

Clonal dissemination of methicillin-resistant Staphylococcaceae between Algerian sheep farms.

OBJECTIVES: Sheep farming represents an important economic sector in Algeria, and the potential dissemination of methicillin-resistant Staphylococcaceae (MRS) is a critical veterinary and public health concern. This study aimed to determine the prevalence and types of MRS in ovine in Algeria and characterize them using whole-genome sequencing (WGS) analysis. METHODS: Two hundred sheep from 20 different Algerian farms across 3 regions were screened for nasal colonization with MRS. The isolates were identified by matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALD

Birds as Sentinels of Methicillin-Resistant Staphylococcaceae in Brazilian Protected Areas.

Antimicrobial resistance (AMR) poses a growing threat to One Health, with wild birds serving as sentinels and reservoirs for resistant pathogens. Despite Brazil's rich avian biodiversity, studies on AMR in wild bird populations are limited. This study investigated methicillin-resistant Staphylococcaceae in wild birds from Southern Brazil, analyzing molecular characteristics of isolates. Oropharyngeal swabs were collected from 248 birds in two protected areas and two wildlife rehabilitation centers over one year. The sample included resident, partially migratory, and migratory species from terr

Flies from meat processing facilities are carriers of multidrug-resistant Escherichia coli and diverse Staphylococcaceae species.

Flies are important vectors of antimicrobial resistance (AMR) in food production, but their role in meat processing environments remains poorly characterized. This study isolated Enterobacteriaceae and Staphylococcaceae (including Mammaliicoccus sciuri) from flies collected in bovine and swine slaughterhouses and meat processing industries. Phenotypic AMR was assessed, and the presence of selected antimicrobial resistance and virulence genes (mecA, sea, see, sec, stx1, stx2, and eae) was verified. Out of 60 flies collected, a total of 34 isolates were identified using MALDI-TOF with 13 differe

Impact of the Stringent Stress Response on the Expression of Methicillin Resistance in Staphylococcaceae Strains Carrying mecA, mecA1 and mecC.

The acquisition of the resistance determinant mecA by Staphylococcus aureus is of major clinical importance, since it confers a resistant phenotype to virtually the entire large family of structurally diverse β-lactam antibiotics. While the common resistance determinant mecA is essential, the optimal expression of the resistance phenotype also requires additional factors. Previous studies showed that the great majority of clinical isolates of methicillin-resistant S. aureus (MRSA) have a heterogeneous resistant phenotype, and we observed that strains carrying methicillin genetic determinants o

Environmental spread of mecA-carrying Staphylococcaceae in a Brazilian veterinary teaching hospital: A phenotypic and molecular epidemiological investigation.

Antimicrobial resistance significantly threatens human, animal, and environmental health, necessitating coordinated surveillance and intervention strategies. This study aimed to assess the presence of antimicrobial-resistant Staphylococcaceae on high-contact surfaces within the dermatology sector of a Brazilian veterinary teaching hospital. Thirty-eight high-touched surfaces were swabbed. Staphylococcus spp. was isolated from two sampling moments - after cleaning (M1) and after attendance activities (M2) using mannitol salt agar. Isolates were identified using MALDI-TOF, and an antimicrobial s

pTJK, a rare Mammaliicoccus lentus phage with broad-host-range, antibiofilm, and synergistic interactions with antimicrobials against resistant Staphylococcaceae.

Antimicrobial resistance in Staphylococcaceae poses a serious threat to human and animal health, driving the search for alternative strategies such as bacteriophage therapy. Here, we describe the isolation and characterization of pTJK, one of the first bacteriophages reported to infect Mammaliicoccus lentus, recovered together with its host strain TJK24 from sewage-contaminated stream water. pTJK is a novel lytic phage with a broad host range, capable of infecting multiple Mammaliicoccus and Staphylococcus species. It significantly reduced biofilm formation and disrupted preformed biofilms, in

Detectable effects of bacterial phylogeny on virulence but not phage efficacy in an in vivo Galleria mellonella model.

Predicting the outcomes of bacteria-phage interactions is a central challenge in microbial ecology, particularly across heterogeneous host environments. Evolutionary relatedness among bacteria can explain variation in phage susceptibility in vitro, but it remains unclear whether this predictive relationship persists in complex host environments and to what extent relatedness also explains variation in other bacterial traits, such as virulence. Here, we used an in vivo infection model to quantify how bacterial phylogeny and host environmental context mediate bacterial virulence and bacteria-pha

From Farm to Community: Dispersal of Potentially Pathogenic Staphylococcus and Mammaliicoccus Species and Antimicrobial Resistance Across Shared Environments.

Staphylococcaceae are a diverse bacterial family with important implications for human and animal health. This study highlights the One Health relevance of their environmental dispersal, particularly, by identifying closely related or genetically identical strains circulating between farm and community environments. Environmental Staphylococcaceae strains were isolated from animal farms and interconnected areas within a university setting, both influenced by anthropogenic activities. Genetic diversity analysis via GTG5-PCR revealed that specific strains circulated across these environments, li

Technological Applications of Macrococcus caseolyticus and its Impact on Food Safety.

Macrococcus spp. are Gram-positive cocci that belong to the Staphylococcaceae family; they are closely related to staphylococci, but, unlike staphylococci, they are not considered as human pathogens. Macrococcus spp. are recognized as relevant veterinary pathogens, and their presence has been reported in food products of animal origin. Macrococcus caseolyticus, the most studied species of the Macrococcus genus, is associated with the development of aroma and flavor in fermented foods and is, thus, used as starter cultures in fermentations. However, certain important issues regarding food safet

The nasal cavity microbiota of healthy adults.

BACKGROUND: The microbiota of the nares has been widely studied. However, relatively few studies have investigated the microbiota of the nasal cavity posterior to the nares. This distinct environment has the potential to contain a distinct microbiota and play an important role in health. RESULTS: We obtained 35,142 high-quality bacterial 16S rRNA-encoding gene sequence reads from the nasal cavity and oral cavity (the dorsum of the tongue and the buccal mucosa) of 12 healthy adult humans and deposited these data in the Sequence Read Archive (SRA) of the National Center for Biotechnology Inform

Diversification of the Staphylococcal Cassette Chromosome Through Distinct Mechanisms of Horizontal Transfer.

The staphylococcal cassette chromosome mec (SCCmec) is a mobile genetic element that carries the mecA gene conferring resistance to beta-lactam antibiotics. While SCCmec is widely disseminated in Staphylococcus aureus, its diversity and evolutionary history across different taxonomic scales have not been investigated in detail. To elucidate the mechanisms governing the diversification of SCCmec, we carried out the largest systematic analysis of SCCmec to date. We focused on the Staphylococcaceae family, which is the primary cellular host of SCCmec. We scanned 2,556 complete genomes, representi
