Enterotoxigenic potential of Staphylococcus intermedius.

Staphylococcal food poisoning (SFP) caused by enterotoxigenic staphylococci is one of the main food-borne diseases. In contrast to Staphylococcus aureus, a systematic screening for the enterotoxins has not yet been performed on the genomic level for the coagulase-positive species S. intermedius. Therefore, the enterotoxigenic potential of 281 different veterinary (canine, n = 247; equine, n = 23; feline, n = 9; other, n = 2) and 11 human isolates of S. intermedius was tested by using a multiplex PCR DNA-enzyme immunoassay system targeting the staphylococcal enterotoxin genes sea, seb, sec, sed

Enterotoxigenicity of Staphylococcus intermedius of canine origin.

Seventy-three staphylococcal strains isolated from pyrodermatitis in dogs were classified as Staphylococcus intermedius (52 strains) or Staphylococcus aureus (21 strains) on the basis of acetoin formation, anaerobic mannitol fermentation, aerobic maltose fermentation, pigmentation, coagulation of human plasma, and reaction on crystal violet agar. Enterotoxin was produced by 13 of the 52 S. intermedius strains and 6 of the S. aureus strains. The highest percentage of enterotoxigenic strains produced enterotoxins C (6 strains), D (7 strains), and E (6 strains). Four strains produced the toxic sh

Antimicrobial susceptibility of Staphylococcus intermedius and Staphylococcus schleiferi isolated from dogs.

The susceptibility to 23 antimicrobial agents was determined in 114 isolates of Staphylococcus intermedius and eight isolates of Staphylococcus schleiferi of canine origin. Overall, 73% of S. intermedius isolates and 37.5% of S. schleiferi isolates were susceptible to all the 23 antimicrobials tested. The large majority of S. intermedius strains retained susceptibility to antimicrobials currently employed in treatment of pyoderma (cephalosporins, cotrimoxazole and association amoxicillin-clavulanic acid) as well as to those effective against staphylococci (fusidic acid, rifampicin and fluoroqu

Enterotoxigenicity of Staphylococcus intermedius strains isolated from dogs.

Dogs (n = 100) were examined for the presence of enterotoxigenic Staphylococcus intermedius. Enterotoxigenic strains were isolated from the nasal cavities of 43 (43.0%) out of 100 dogs. Of the 50 enterotoxigenic strains isolated, 23 (46.0%) produced enterotoxin A, 17 (34.0%) enterotoxins A and C, 8 (16.0%) enterotoxin C, and only two (4.0%) enterotoxins A and B. The amount of enterotoxin produced on semisolid brain heart infusion medium by the 17 enterotoxin A producing strains of S. intermedius was 10-160 ng per ml. Enterotoxin A production by S. intermedius strains was ony one tenth to one h

In vitro indices of tissue adherence in Staphylococcus intermedius.

In vitro indices of adherence showed that strains of Staphylococcus intermedius from lesions of canine pyoderma differed from strains isolated from normal carrier sites in that a significantly greater proportion of pyoderma strains adhered to extracellular matrix proteins whilst fewer adhered to polystyrene. Slime production and a hydrophobicity index did not differ between the groups. This suggests that exposure of extracellular matrix proteins due to underlying disease may result in the selection of a narrower spectrum of strains from amongst those at carrier sites.

Staphylococcus cornubiensis sp. nov., a member of the Staphylococcus intermedius Group (SIG).

We here describe a novel species in the Staphylococcus intermedius Group (SIG) which is phenotypically similar to Staphylococcus pseudintermedius but is genomically distinct from it and other SIG members, with an average nucleotide identity of 90.2 % with its closest relative S. intermedius. The description of Staphylococcus cornubiensis sp. nov. is based on strain NW1T (=NCTC 13950T=DSM 105366T) isolated from a human skin infection in Cornwall, UK. Although pathogenic, NW1T carries no known virulence genes or mobilizable antibiotic resistance genes and further studies are required to assess t

Catheter-related bacteremia associated with coagulase-positive Staphylococcus intermedius.

We report a case of catheter-related bacteremia in a 63-year-old patient caused by Staphylococcus intermedius. Clinical resolution of the infection was obtained after removal of the intravenous device and antibiotic treatment. This observation emphasizes the risk of confusion between S. intermedius and Staphylococcus aureus if only a coagulase test is done.

Staphylococcus intermedius: current knowledge on a pathogen of veterinary importance.

It has been 16 years since the identification of S. intermedius as a new species. Numerous investigations using cell wall and DNA analytic methods have now clearly demonstrated significant differences to warrant the creation of this new species. However, studies investigating virulence factors associated with S. intermedius have not uncovered evidence that differentiates virulent from non-virulent isolates. Therefore, at the present time, it is difficult for veterinary clinicians and microbiologists to determine the clinical significance of many S. intermedius isolates. Host-bacterial interact

Beware of the pet dog: a case of Staphylococcus intermedius infection.

Staphylococcus intermedius is a common commensal and pathogen in dogs and cats and only rarely has been identified as causing human infection. We report a human case of postoperative sinus infection caused by methicillin-resistant S. intermedius. A 28-year-old woman with a history of endoscopic pituitary adenoma resection presented with 3 weeks of foul smelling nasal discharge. Nasal endoscopy revealed purulent sinus drainage. Cultures, initially misidentified as coagulase-negative Staphylococcus and then as Staphylococcus aureus, revealed the presence of S. intermedius. Cultures from the pati

Adherence by Staphylococcus intermedius to canine keratinocytes in atopic dermatitis.

The adherence of Staphylococcus intermedius to canine keratinocytes in normal dogs was compared to that in dogs suffering from atopic dermatitis, primary seborrhoea and bacterial pyoderma. Statistically significant greater adherence by S. intermedius to keratinocytes occurred in atopic dogs and dogs suffering from pyoderma when compared with the normal group (P < 0.01) and dogs suffering from primary seborrhoea (P < 0.05). This is similar to the results of a study of human atopic dermatitis by Cole and Silverberg (1986) who demonstrated increased adherence by S. aureus to keratinocytes from at

Enterotoxin production by strains of Staphylococcus intermedius and Staphylococcus aureus isolated from dog infections.

Sixty-six strains of S. intermedius and 10 of S. aureus isolated from infected dogs were examined for enterotoxin production. 39.5% of the strain (37.9% of S. intermedius and 50% of S. aureus) produced one or more enterotoxins. The predominant types produced by S. intermedius were C1 and C2, and only two of the strains synthesized enterotoxin A. One of the S. aureus strains produced the toxic shock syndrome toxin 1.

Characteristics of Staphylococcus intermedius isolates from diseased and healthy dogs.

Staphylococcus intermedius isolates from diseased and healthy dogs were examined for production of extracellular enzymes and toxins, and phage patterns. There were no significant differences between the two groups of isolates in the production rates of DNase, protease, lipase, gelatinase, hyaluronidase, hemolysins, protein A, and TSST-1, or in phage patterns. But the production rate of enterotoxins in isolates from diseased dogs was significantly higher than that in isolates from healthy dogs. PFGE analysis was performed with isolates from different body sites in individual dogs. In 3 of 6 hea

Efficacy of tylosin tartrate on canine Staphylococcus intermedius isolates in vitro.

In the past 5 years, the incidence of canine skin infections caused by resistant strains of Staphylococcus (pseud)intermedius has increased. Many older antibiotics are used to treat these infections because the sensitivity can be demonstrated in vitro. Additionally, many of these older drugs are efficacious and unlikely to induce multidrug resistance. More than a decade ago, the antibiotic tylosin tartrate was reported to be efficacious in vitro and in vivo against Staphylococcus intermedius. The purpose of this study was to determine whether S. (pseud)intermedius isolated from untreated pyode

Characterization of a variant of the SCCmec element in a bloodstream isolate of Staphylococcus intermedius.

Here we report for the first time, a detailed characterization of a variant of the SCCmec element, in a methicillin-resistant Staphylococcus intermedius human isolate. S. intermedius is a coagulase-positive zoonotic microrganism, recently classified as a separate species. In routine clinical laboratory practice, the coagulase production is used as criterion of pathogenicity related to S. aureus, but S. intermedius is frequently misidentified-being mistaken for S. aureus-and consequently its real incidence underestimated. S. intermedius have been found only occasionally in human beings, and met

Antimicrobial drug susceptibility of Staphylococcus intermedius clinical isolates from canine pyoderma.

A total of 50 Staphylococcus intermedius strains isolated in France from canine pyodermas in 2002 were investigated for their susceptibility to various antimicrobial drugs. Antimicrobial susceptibility was assessed using a 2-fold serial dilution method in Mueller-Hinton agar, and the minimal inhibitory concentrations (MICs) were determined. About 62% of the 50 strains tested were producers of beta-lactamase and categorized as penicillin-resistant. About 26% demonstrated resistance to sulphonamides, 46% to oxytetracycline, 30% to chloramphenicol, 28% to streptomycin, kanamycin, neomycin or eryt

Comparative ribotyping of Staphylococcus intermedius from dogs, pigeons, horses and mink.

Strains of Staphylococcus (S.) intermedius from dogs, pigeons, horses and mink were typed by comparison of rRNA gene restriction fragment length polymorphisms (ribotyping) and the resulting ribotypes examined by cluster-analysis. Digestion of whole-cell DNA with HindIII resulted in 9 ribotypes with 3 to 4 bands. Separation of isolates from different host animal species was not possible. EcoRI yielded 11 different patterns with 4 to 9 fragments. The EcoRI-ribotypes of all canine strains grouped in one cluster encompassing four closely related ribotypes. Isolates were indistinguishable with resp

Staphylococcus intermedius: clinical presentation of a new human dog bite pathogen.

Staphylococcus intermedius is a Gram-positive, coagulase-positive coccus that can be distinguished from Staphylococcus aureus by routine microbiological testing. Whereas S intermedius is recognized as flora and pathogen of dogs, it has never been isolated from human infections. We hypothesized that S intermedius may cause human dog bite wound infections and that it has been previously misidentified as S aureus. Fourteen isolates from clinically infected dog bite wounds that were originally identified as S aureus were subjected to further testing; three (22%) were found to be S intermedius. The

Identification of protein A from Staphylococcus intermedius isolated from canine skin.

Protein A was identified in cell wall-bound and secreted forms from Staphylococcus intermedius isolated from canine skin. A direct binding radioimmunoassay for the detection of bacterial surface Fc receptors identified 48 of 50 S intermedius isolates that contained cell wall-bound protein A. Using a competitive binding radioimmunoassay for the detection of Fc-reactive proteins in bacterial culture supernatants, we identified 9 of 50 clinical isolates of S intermedius that secreted measurable quantities of an Fc receptor into the culture medium. Concentrated culture supernatants from these isol
