Multifaceted Effects of Starch Branching Enzyme and Soluble Starch Synthase Gene Editing in Rice with Different Wx Genotypes.

Rice endosperm, the major edible portion of the grain, plays an important role in regulating blood glucose and preventing intestinal diseases by increasing its resistant starch (RS) content. Previous studies have shown that suppressing amylopectin biosynthesis via genome editing can increase RS content. However, the influence of different Waxy (Wx) allelic backgrounds on RS accumulation in edited lines has not been systematically evaluated. In this study, we used glutinous rice Yunan Heixiangnuo (HXN) with a nonfunctional wx allele and indica rice Yixiang 1B (YX1B) with a weak Wxb allele as ba

Finding granule-bound starch synthase enzymes with high activity using free-energy calculations.

Starch is a branched glucose polymer comprising amylopectin, of high molecular weight with many short branches, and amylose, of lower molecular weight and only a few long-chain branches. Granule-bound starch synthase I (GBSSI) is one of the main enzymes controlling amylose synthesis and structure. Production of different GBSSI mutants is laborious, this study retrospectively examined the relationship between mutation-induced changes in glucan binding and experimentally reported GBSSI activity. Atomistic molecular dynamics simulations and free-energy calculations were performed for GBSSI comple

Identification of an abundant 56 kDa protein implicated in food allergy as granule-bound starch synthase.

Rice, the staple food of south and east Asian counties, is considered to be hypoallergenic. However, several clinical studies have documented rice-induced allergy in sensitive patients. Rice proteins with molecular weights of 14-16, 26, 33, and 56 kDa have been identified as allergens. Recently, it was documented that the 56 kDa rice allergen was responsible for rice-induced anaphylaxis. The 14-16 kDa allergens have been identified as α-amylase inhibitors; the 26 kDa protein has been identified as α-globulin; and the 33 kDa protein has been identified as glyoxalase I. However, the identity of

Soluble starch synthase IVa determines cooked rice elongation in waxy and non-waxy rices (Oryza sativa L.).

SIGNIFICANCE: Cooked rice elongation (CRE) is an attractive grain quality reported only in Basmati from India and Pakistan and in Paw San Hmwe from Myanmar. Several reports on the discovery of genes controlling CRE were unequivocal in rice. Here, we design new experiments to uncover genes that significantly affect CRE in waxy and non-waxy rice varieties. MATERIALS AND METHODS: We created 105 BC1F2 lines derived from a cross between a high-CRE waxy rice, NHN, and Pink+4 (#20A09), a low-CRE, low-glycemic-index, non-waxy rice, to generate two distinct phenotypic pools for QTL-seq analysis: high-

Starch synthase IIIa and starch branching enzyme IIb-deficient mutant rice line ameliorates pancreatic insulin secretion in rats: screening and evaluating mutant rice lines with antidiabetic functionalities.

Diabetes mellitus is a metabolic disease spreading worldwide that has been reported to worsen the development and progression of other diseases (cancer, vascular diseases and dementia). To establish functional rice lines with anti-postprandial hyperglycaemic effects, we developed mutant rice lines, which lack one or two gene(s) related to starch synthesis, and evaluated their effects. Powder of mutant rice lines or other grains was loaded to rats fasted overnight (oral grain powder loading test). Incremental area under time-concentration curves (iAUC) were calculated with monitored blood gluco

Age-dependent starch-sucrose metabolic transition during bulb development of Fritillaria unibracteata.

BACKGROUND: The bulb of Fritillaria unibracteata is a valuable traditional Chinese medicinal material whose bioactive compound accumulation is closely linked to carbon metabolism. As a perennial herb, its bulbs undergo multi-year developmental cycles, with starch and sucrose serving as key storage and mobilizable carbohydrates, respectively. However, the age-dependent dynamics of starch-sucrose interconversion during bulb development and its expression patterns remain poorly understood, limiting our comprehension of the carbon allocation dynamics that support biomass accumulation and medicinal

HvPHO1 Controls a Glucose 1-Phosphate-SnRK1 Signalling Module That Coordinates Starch Accumulation in Barley Endosperm.

Developing barley grains must balance the use of imported sucrose between soluble sugars and storage starch, but how this balance is controlled remains unclear. Here we show that loss of the plastidial α-glucan phosphorylase HvPHO1 causes starch and soluble sugars to accumulate concurrently in barley grains. Developing pho1 grains showed sustained increases in glucose 1-phosphate (G1P) and adenosine diphosphate glucose (ADP-glucose), and exogenous G1P also promoted starch accumulation, supporting a stimulatory effect of elevated G1P on starch synthesis. G1P directly bound sucrose non-fermentin

Production of high-amylose starch with low digestibility in a green marine microalga Tetraselmis subcordiformis by delaying high-bicarbonate induction.

A photoautotrophic green marine microalga Tetraselmis subcordiformis was applied to sustainably produce high-amylose starch (HAS) from CO2 via high-bicarbonate (HB) induction. Delaying the addition of HB from Day 0 to Day 3 overcome the HB stress and significantly improved photosynthetic activity, resulting in 7.2-times higher starch productivity and the production of HAS with the total starch content of 21.81 % and amylose/amylopectin ratio of 1.51 obtained on Day 6, which represented the highest HAS levels among the photoautotrophic microalgae reported hitherto. The HB treatment improved the

Physiological and Transcriptome Analyses Provide Insights into the Response of Grain Filling to High Temperature in Male-Sterile Wheat (Triticum aestivum L.) Lines.

High-temperature (HT) stress frequently affects the early and middle stages of grain filling in hybrid seed production regions. Photo-thermo-sensitive male-sterile (PTMS) wheat lines, which play a critical role as female parents in hybrid seed production, face challenges under HT conditions. However, the mechanisms governing grain filling in PTMS lines under HT stress remain poorly understood. This study used the BS253 line to investigate the effects of HT on grain filling, primarily focusing on the transition from sucrose unloading to starch synthesis. The findings indicated that HT significa

Loss-of-function of SSIIa and SSIIIa confers high resistant starch content in rice endosperm.

Rice (Oryza sativa L.) endosperm accumulates huge amounts of starch. Rice starch is highly digestible, potentially enhancing the occurrence of blood sugar- and intestine-related diseases such as type 2 diabetes. Resistant starch (RS) is hardly digestible in small intestine but can be converted into beneficial short-chain fatty acids in large intestine, potentially reducing the incidence of these diseases. However, it is still difficult to produce a high RS rice variety. Here, we report that simultaneous deficiency of soluble starch synthases IIa and IIIa confers high RS content in rice endospe

Pilot-scale duckweed-CO2 wastewater bioremediation system: a synergistic approach for nutrient removal, carbon capture, and biomass production.

This study presents a pilot-scale duckweed-based wastewater treatment system enhanced with CO2 fertilization to improve nutrient removal, CO2 capture, and biomass productivity. Three strains of Landoltia punctata were cultivated continuously for five months. In spring, CO2 supplementation significantly increased the biomass yield and starch and protein productivity (by more than 12.2, 2.4 and 3.5 g/m2/d, respectively), representing increases of 189-259 %, 1,108-2,330 % and 143-166 %, respectively, compared with those of the control group. Integrated metabolomic and transcriptomic analyses reveal that starch and sucrose metabolism regulate maize kernel hardness.

Maize kernel hardness, largely determined by the structural and compositional characteristics of the endosperm, is a key trait affecting grain quality, milling performance, and storage stability. Although vitreous and starchy endosperms exhibit markedly different physical properties, the underlying metabolic and transcriptional mechanisms-particularly the crosstalk between primary and specialized metabolic pathways-remain insufficiently understood. In this study, we conducted integrated metabolomic and transcriptomic analyses, along with cytological observations, to investigate these mechanism

The ss3a/be2b double mutant yields high resistant starch through cooperative regulation of starch fine structure in indica rice.

Rice starch biosynthesis is governed by multiple enzymes, among which soluble starch synthase IIIa (SSIIIa) and starch branching enzyme IIb (BEIIb) play critical but distinct roles in determining starch fine structure. However, their combined effects on starch fine structure and physicochemical properties are not fully understood. Herein, we generated a double mutant ss3a/be2b in the indica variety GLA4 by crossing a radiation-induced SSIIIa mutant with a CRISPR/Cas9-generated BEIIb knockout mutant. The double mutant displayed a floury endosperm with irregular starch granules, an amylose conte

Integrating GWAS and Transcriptome Analysis Identifies Candidate Genes for Kernel Starch Quality Traits in Maize.

Maize (Zea mays L.) starch quality is a complex trait with significant implications for grain processing and industrial applications. However, the genetic basis underlying starch quality, particularly for gelatinization and thermodynamic properties, remains poorly understood. In this study, we evaluated 12 starch quality traits, including seven gelatinization characteristics, four thermodynamic traits, and kernel starch content (KSC) in a diverse panel of 335 maize inbred lines. Considerable phenotypic variation was observed for all traits. A total of 228 quantitative trait loci (QTLs) were si

Hydrogen sulfide and ethylene regulate photosynthesis, sugar metabolism, and tolerance to heat stress in the presence of sulfur in rice.

Heat stress impacts photosynthesis and carbohydrate metabolism, challenging food security. To comprehend the mechanisms of thermotolerance, we examined the role of ethylene (ET) and hydrogen sulfide (H2S) with or without sulfur (S) in rice (Oryza sativa L.). Both ET and H2S promoted heat stress tolerance more conspicuously in the presence of S, restoring the balance between carbon assimilation and utilization. The enhanced photosynthesis in ET and H2S-treated plants under heat stress was linked with increased relative expression of Rubisco subunits rbcS and rbcL and carbohydrate metabolizing,

Comparative analysis of ultraviolet-B radiation and abscisic acid revealing distinct mechanistic approaches to drought tolerance in highland barley.

Drought tolerance in highland barley (qingke) is modulated by distinct responses to ultraviolet-B radiation (UVB) and abscisic acid (ABA). Plants were exposed to two drought levels (moderate: 50 % soil water content and severe: 30 % soil water content), alongside two UVB intensities (low: 5-6 kJ m-2 d-1 and high: 14-15 kJ m-2 d-1), and two ABA concentrations (low: 70 μM and high: 150 μM). Physiological analyses revealed that UVB exposure significantly reduced the photosynthetic rate by 46.5 % under severe drought conditions. In contrast, ABA treatment helped to maintain the rate and improved w
