New alkaloids from Stemona tuberosa and structural revision of tuberostemonols P and R.

Three Stemona alkaloids named stemotuberines A-C (1-3) with unique C17N frameworks, presumably formed by elimination of the C-11-C-15 lactone ring of the stichoneurine skeleton, were isolated from the roots of Stemona tuberosa. Their structures were elucidated by spectroscopic analysis, X-ray diffraction, and computational methods. Compounds 2 and 3 showed inhibition (IC50 values of 37.1 and 23.2 μM, respectively) against LPS-induced nitric oxide production in RAW 264.7 cells. In addition, concern was expressed about the reported plant origin (S. sessilifolia) of the recently described alkaloi

Total synthesis of the putative structure of stemonidine: the definitive proof of misassignment.

[structure: see text] The total synthesis of the putative structure of the Stemona alkaloid stemonidine has been completed. The key transformations include a 1,3-dipolar cycloaddition of a chiral nitrone derived from (S)-prolinol and a spirolactonization process involving the generation of the critical stereocenter. The NMR data of the synthetic material do not match those reported for the natural product. It is concluded that the structure assigned to stemonidine is incorrect, and it must be reassigned as stemospironine.

Suppression of IL-2 production and proliferation of CD4(+) T cells by tuberostemonine O.

Tuberostemonine stereoisomers are natural alkaloids found in Stemona tuberosa, that are known to have anti-inflammatory and anti-infective properties. Tuberostemonine alkaloids inhibit inflammation by suppressing the expression of inflammatory mediators such as cyclooxygenase and nitric oxide synthase. However, the direct immunomodulatory properties of tuberostemonine alkaloids in T cells have not been elucidated so far. In this study, the activities in T cells of tuberostemonine N (TbN) and a novel alkaloid, tuberostemonine O (TbO), isolated from S. tuberosa, were investigated. Although TbN d

LC-MS/MS-Based Lipidomic Profiling Reveals the Regulatory Effect of Stemoninoamide on TGF-β1-Induced Myofibroblast Transition in HFL1 Cells.

The dried roots of Stemona tuberosa, known as "Bai-Bu" in traditional Chinese medicine, have been employed as a therapeutic agent for the treatment of pertussis and tuberculosis. The chemical composition of S. tuberosa is dominated by alkaloids which have been reported to possess antifibrosis effect. Herein, ELISA kit and immunofluorescence analyses showed that the alkaloid of stemoninoamide (Stem) isolated from S. tuberosa exhibited inhibitory effect on the production of α-SMA and Col-1 of HFL1 cells stimulated by TGF-β1. Further lipidomic analysis based on LC-MS/MS revealed differential meta

Vinylogous Mannich reaction in total synthesis of alkaloids.

Covering: up to July 2026The vinylogous Mannich reaction (VMR) enables γ-selective carbon-carbon bond formation between vinylogous nucleophiles and imines or iminium ions, yielding δ-aminocarbonyl frameworks. Compared with the classical Mannich reaction, VMR shows distinct advantages in remote functionalization and convergent bond formation, facilitating the rapid assembly of nitrogen-containing heterocycles, multiple stereogenic centers, and densely functionalized intermediates. As a result, this reaction has become an increasingly valuable strategy for generating complexity in the total synt

Tuberostemonine alleviates high-fat diet-induced obesity and hepatic steatosis by increasing energy consumption.

Obesity is of public concern worldwide, and it increases the probability of developing a number of comorbid diseases, including NAFLD. Recent research on obesity drugs and health demands have shown the potential of natural plant extracts for preventing and treating obesity and their lack of toxicity and treatment-related side effects. We have demonstrated that tuberostemonine (TS), an alkaloid extracted from the traditional Chinese medicine Stemona tuberosa Lour can inhibit intracellular fat deposition, reduce oxidative stress, increase cellular adenosine triphosphate (ATP), and increase mitoc

Oral toxicity of various Stemona collinsiae crude extracts against nymph and adult stages of American cockroach, Periplaneta americana (Dictyoptera: Blattodea).

Stemona collinsiae exhibits insecticidal resistance against various pests and insect vectors. However, insecticidal activity of S. collinsiae roots has not been tested for some insect vectors, including the American cockroach, Periplaneta americana. The synanthropic insect P. americana is a reservoir of pathogenic and non-pathogenic microorganisms and a cause of infectious diseases and cockroach allergy. This important vector transmits microorganisms to animals and humans to cause vector-borne diseases. This research involved detection of the nymphicidal and adulticidal activities of S. collin
