TLC-image analysis of non-chromophoric tuberostemonine alkaloid derivatives in Stemona species.

A simple, selective, precise, and accurate thin-layer chromatographic (TLC) image analytical method was developed and validated for simultaneous quantification of the major components in the root extracts of Stemona tuberosa (tuberostemonine, tuberostemonine N and neotuberostemonine)), and S. phyllantha (tuberostemonine and tuberostemonine A). The analysis was performed by TLC on silica gel 60 F254 aluminum plates using a mixture of dichloromethane: ethyl acetate: methanol: ammonium hydroxide (50:45:4:1) as mobile phase. Post-derivatization was employed by dipping the TLC plate into Dragendorf

Anticoccidial activity and its potential Mechanisms of Stemona tuberosa against Eimeria tenella: In Vivo and In Vitro Investigations and Host Intestinal Protection.

Coccidiosis causes substantial economic losses in poultry, and current control relies largely on chemotherapeutics. Plant-derived extracts offer low toxicity, environmental safety, and reduced resistance risks. This study evaluated the anticoccidial activity of Stemona tuberosa ethanol extract (STEE) and explored its underlying mechanisms. Anticoccidial efficacy was assessed through oocyst sporulation, sporozoite invasion, anticoccidial index (ACI), hematological parameters, inflammatory markers, and intestinal protective proteins. Mechanistic analyses included 16S rRNA sequencing and western

New alkaloids from Stemona tuberosa and structural revision of tuberostemonols P and R.

Three Stemona alkaloids named stemotuberines A-C (1-3) with unique C17N frameworks, presumably formed by elimination of the C-11-C-15 lactone ring of the stichoneurine skeleton, were isolated from the roots of Stemona tuberosa. Their structures were elucidated by spectroscopic analysis, X-ray diffraction, and computational methods. Compounds 2 and 3 showed inhibition (IC50 values of 37.1 and 23.2 μM, respectively) against LPS-induced nitric oxide production in RAW 264.7 cells. In addition, concern was expressed about the reported plant origin (S. sessilifolia) of the recently described alkaloi

Synthesis of the Common Core Structure of the Stemofoline Alkaloids.

A novel synthetic route to the common core structural motif of the stemofoline alkaloids has been developed. The key transformations include (1) an intramolecular 1,3-dipolar cycloaddition reaction of a highly functionalized nitrone, (2) the subsequent formation of a caged structure via lithiated allylic sulfoxide, and (3) the concomitant sila-Pummerer reaction of α-silylalkenyl sulfoxide to prepare a thioester precursor. A series of stereochemistries on the highly caged core structure characteristic of the stemofoline alkaloids was successfully assembled.

Two pectic polysaccharides from Stemona tuberosa Lour. roots with detailed structural features display potent immunoenhancing activity via TLR signaling pathway.

Stemona tuberosa Lour., a noted traditional Chinese medicine, exerts potent pharmacological effects, yet its key pectic polysaccharides, especially their chemical structures, remain poorly understood, impeding further investigation and development. In this study, two pectin polysaccharides (STP-3-1 and STP-3-2) were isolated from the roots for the first time. Using HPLC, methylation-GC-MS, and 1D/2D NMR, their structures were elucidated. Both contain a rhamnogalacturonan I (RG-I) backbone with 1,5-α-arabinan and 1,4-β-galactan side chains; STP-3-1 is exclusively RG-I with higher galactose cont

Toxicity of Stemona root against onion cutworm by using different contacting methods and solvents.

Roots of Stemona tuberoso have been extracted by using fixed-bed contacting method with serial and parallel technique. In order to extract secondary metabolites from roots of Stemona sp., various organic solvents with different polarities were employed. Optimal solvents for the extraction of effective ingredients from root were determined. The results showed that in most cases the yields of crude extract were highest by using parallel technique rather than serial technique. The highest yields of crude extract by using both techniques were obtained with acetone, followed by water, ethanol, meth
