[Hypocholesteremic action of trepang (Stichopus japonicus) lipids].

The hypocholesteremic effect displated by the Japan Sea trepang is shown. The experiments were carried out on rabbits with hypocholesteremia caused by fasting. Total cholesterol, beta-lipoproteids, lipoid phosphorus, total phospholipids and the cholesterol/phospholipids ratio were determined in the blood of hypercholesteremic animals. The introduction of total trepang lipids to the hypercholesteremic fasting rabbits resulted in normalization of the lipids metabolism, this being indicative of the favourable effect produced by such lipids.

Immunomodulatory effect of Stichopus japonicus acid mucopolysaccharide on experimental hepatocellular carcinoma in rats.

Stichopus japonicus acid mucopolysaccharide (SJAMP) is an important biologically active compound that can be extracted from the body wall of the sea cucumber. The present study investigated the anti-tumor and immunomodulatory effects of SJAMP in an experimental hepatocellular carcinoma (HCC) model in rats. Three doses of SJAMP (17.5 mg/kg, 35 mg/kg, and 70 mg/kg administered 5 days/week via oral gavage) were given to rats with diethylnitrosamine (DEN)-induced HCC. SJAMP treatment significantly inhibited DEN-induced HCC by reducing both the number and mean volume of nodules, decreasing serum a-

OVULATION AND THE FINE STRUCTURE OF THE STICHOPUS CALIFORNICUS (ECHINODERMATA: HOLOTHUROIDEA) FECUND OVARIAN TUBULES.

The ovary of Stichopus californicus consists of several size classes of tubules, which insert into a central gonad basis. The largest tubules contain the oocytes that will be spawned in the current season. All tubules are composed of three layers. Outermost is a complex peritoneum composed of epithelial cells, axons and muscle cells. The fine structure of the peritoneal neurons suggests their involvement in neurosecretory activity. Between the basal laminae of the peritoneum and the inner epithelium is the ovarian connective tissue compartment, including the genital hemal sinus. This sinus pro

Identification of novel anti-obesity saponins from the ovary of sea cucumber (Stichopus japonicus).

The potential anti-obesity effects of sea cucumber extract have been reported. However, the individual saponins responsible for these effects are yet to be isolated and characterized. This study aimed to identify the most effective sea cucumber body part for inhibiting lipid accumulation in adipocytes and to elucidate the compounds responsible for this effect using nuclear magnetic resonance (NMR) techniques. Sea cucumber ovary 80 % ethanol extract (SCOE) demonstrated remarkable efficacy in inhibiting adipocyte differentiation compared to other sea cucumber body parts with 50 % or 80 % ethanol

Production, optimisation and characterisation of angiotensin converting enzyme inhibitory peptides from sea cucumber (Stichopus japonicus) gonad.

In this study, production of bioactive peptides with angiotensin converting enzyme (ACE) inhibitory activity from sea cucumber (Stichopus japonicus) gonad using commercial protamex was optimised by response surface methodology (RSM). As a result, the optimal condition to achieve the highest ACE inhibitory activity in sea cucumber gonad hydrolysate (SCGH) was hydrolysis for 1.95 h and E/S of 0.75%. For further characterisation, three individual peptides (EIYR, LF and NAPHMR) were purified and identified. The peptide NAPHMR showed the highest ACE inhibitory activity with IC50 of 260.22 ± 3.71 μM

Fucoidan from Stichopus chloronotus relieved DSS induced ulcerative colitis through inhibiting intestinal barrier disruption and oxidative stress.

Intestinal barrier disruption and oxidative stress are the major pathological features during the ulcerative colitis (UC). The present research aimed to explore the amelioration property of fucoidan of Stichopus chloronotus (FucSc) against dextran sulfate sodium (DSS) resulted UC. The findings from our study suggest that treating with Fuc-Sc improved the integrity of the Caco-2 monolayer through raising its TEER value, reducing LDH release, promoting the tight junction proteins (TJs) in Caco-2 cells, and shielding the cells from decreases of these proteins caused by H2O2. Besides, Fuc-Sc activ

Transcriptomic characterization of the intestine in Stichopus monotuberculatus under gradient temperature stress and HSP gene family-mediated molecular adaptation.

The increasing frequency of extreme temperature events under climate change poses a growing threat to the stability of tropical sea cucumber aquaculture. To characterize the molecular responses of the tropical sea cucumber Stichopus monotuberculatus to acute temperature stress, juveniles were exposed for 96 h to 15 °C, 20 °C, 25 °C, 30 °C, and 35 °C, followed by transcriptomic profiling of the intestine. By transcriptomic analysis, 2258, 634, 1618, and 2980 differentially expressed genes (DEGs) were identified at 15, 20, 30, and 35 °C compared to control, respectively. More DEGs were generally

Structural, biochemical, transcriptomic, and metabolomic changes in the body wall of sea cucumber (Stichopus monotuberculatus) across different stages of autolysis.

Stichopus monotuberculatus is a highly prized marine delicacy that undergoes post-harvest autolysis at a rate exceptionally faster than common species like Apostichopus japonicus. Driven by this pronounced phenotypic difference, we hypothesized that the autolysis of S. monotuberculatus is governed by an exceptionally accelerated cascade of oxidative and lipid disruptions, which rapidly overwhelms the classical protease and apoptosis mechanisms. To test this hypothesis, a trauma induced degradation model was established by incision and air exposure, and body-wall samples across seven time point

Purification and Structural Analyses of Sulfated Polysaccharides from Low-Value Sea Cucumber Stichopus naso and Anticoagulant Activities of Its Oligosaccharides.

Three polysaccharides (SnNG, SnFS and SnFG) were purified from the body wall of Stichopus naso. The physicochemical properties, including monosaccharide composition, molecular weight, sulfate content, and optical rotation, were analyzed, confirming that SnFS and SnFG are sulfated polysaccharides commonly found in sea cucumbers. The highly regular structure {3)-L-Fuc2S-(α1,}n of SnFS was determined via a detailed NMR analysis of its oxidative degradation product. By employing β-elimination depolymerization of SnFG, tri-, penta-, octa-, hendeca-, tetradeca-, and heptadeca-saccharides were obtain

Genome-wide identification of the HSP70 superfamily in tropical sea cucumber Stichopus monotuberculatus and their expression analysis under low-salinity stress.

Heat shock proteins (HSPs) are a group of evolutionarily conserved molecular chaperones that serve as indispensable core regulators in preserving cellular homeostasis and orchestrating organismal stress responses. The tropical sea cucumber Stichopus monotuberculatus, a high-value aquaculture species, is sensitive to fluctuations in environmental salinity-a challenge that has emerged as a critical bottleneck limiting its large-scale commercial cultivation. However, no systematic investigation has been conducted to characterize the HSP70 superfamily in S. monotuberculatus and elucidate its funct

Role of Calpain in Autolysis of Sea Cucumber Apostichopus japonicus.

Calpain is a family of cysteine proteases whose role in sea cucumber autolysis was previously unreported. In this study, specific inhibition of calpain notably alleviated autolysis in the respiratory tree and intestine, significantly reduced body wall hardness loss. During an 8 h autolysis period, calpain activity in the body wall continuously declined, whereas activity in the respiratory tree and intestine peaked at 2 h. The contents of calpain 5 & 9 were highest in the respiratory tree and remained relatively stable, whereas these two isoforms showed distinct temporal patterns in the intesti

The Sea Cucumber Holobiont and Probiotics: Recent Progress on Apostichopus japonicus.

After the first definition of the term "Holobiont" by Margulis in the introduction of symbiosis as "Association throughout a significant portion of the life history" in 1991 [1], the understanding of holobiont has become an important goal in modern biology today [2]. Recent advances in microbial collection, genome/metagenome/transcriptome sequencings, and bioassays for host-microbes interactions push us towards a fuller understanding of holobiont in various aspects of life on Earth. Historically, holobiont and related hologenome concepts have been tested and expanded through research on marine

CircRNA254 functions as the miR-375 sponge to inhibit coelomocyte apoptosis via targeting BAG2 in V. splendidus-challenged Apostichopus japonicus.

Circular RNAs (circRNAs) function as immune regulators in many biological processes in mammals, while their function and underlying mechanisms in invertebrates are largely unexplored. In this study, the competing endogenous RNA (ceRNA) mechanism of circRNA that sponges miR-375 and thus regulates AjBAG2-mediated coelomocyte apoptosis was evaluated in Apostichopus japonicus. The results showed that circRNA254 (circ254) was significantly down-regulated in the intestines and coelomocytes after Vibrio splendidus challenge or Lipopolysaccharide exposure, which matched the RNA-seq results in A. japon

Alterations in mitochondrial structure and function in response to environmental temperature changes in Apostichopus japonicus.

Global temperatures have risen as a result of climate change, and the resulting warmer seawater will exert physiological stresses on many aquatic animals, including Apostichopus japonicus. It has been suggested that the sensitivity of aquatic poikilothermal animals to climate change is closely related to mitochondrial function. Therefore, understanding the interaction between elevated temperature and mitochondrial functioning is key to characterizing organisms' responses to heat stress. However, little is known about the mitochondrial response to heat stress in A. japonicus. In this work, we i

Effects of temperature and salinity on CO2 fluxes dynamics and respiration metabolism in the sea cucumber Apostichopus japonicus (Selenka).

Global warming has led to higher temperatures and extreme precipitation events, significantly affecting the physiological status of sea cucumbers and causing substantial economic losses in the aquaculture industry. This study explored the effects of high temperature (HT), low salinity (LS), and their combination (TS) on CO2 fluxes at the water-air interface, energy allocation, and the expression of respiratory metabolism-related genes in sea cucumbers. The results showed that HT and/or LS treatments reduced growth performance by 8.37-21.64 % and increased respiratory metabolic carbon content b
