The chemical neuroanatomy of vagus nerve stimulation.

In this short overview a reappraisal of the anatomical connections of vagal afferents is reported. The manuscript moves from classic neuroanatomy to review details of vagus nerve anatomy which are now becoming more and more relevant for clinical outcomes (i.e. the therapeutic use of vagus nerve stimulation). In drawing such an updated odology of central vagal connections the anatomical basis subserving the neurochemical effects of vagal stimulation are addressed. In detail, apart from the thalamic projection of central vagal afferents, the monoaminergic systems appear to play a pivotal role. S

Chemical stimulation sustains bioluminescence of living light materials.

Bioluminescence offers a powerful tool for real-time, label-free sensing for living materials. However, conventional approaches often rely on mechanical stimulation, which is difficult to standardize, localize, and sustain. Here, we introduce a chemical strategy to stimulate and sustain bioluminescence in the marine dinoflagellate Pyrocystis lunula, enabling durable, adaptive, light-emitting living materials. By embedding P. lunula into 3D-printed, alginate scaffolds, we engineered architecturally stable constructs with long-term cellular retention, viability, and light-emitting capacity. Expo

Acetazolamide attenuates chemical-stimulated but not thermal-stimulated acute pain in mice.

AIM: Acetazolamide (AZA), a carbonic anhydrase (CA) inhibitor, has been found to alleviate inflammatory and neuropathic pain in rats. In the present study, we investigated the effects of AZA on thermal- and chemical-stimulated acute pain in mice and the possible mechanisms underlying the effects. METHODS: Five acute pain models based on thermal and chemical stimuli were established to investigate the effects of AZA on different types of nociception in mice. The antinociceptive effects of methazolamide (another CA inhibitor) and diazepam (a positive allosteric modulator of GABAA receptor) were

Nervous outflow from skeletal muscle following chemical noxious stimulation.

1. In order to determine the nervous outflow from skeletal muscle during chemically induced muscle pain, the impulse activity of various types of muscle afferents in response to close intra-arterial injections of pain-producing substances (bradykinin, 5-hydroxytryptamine, histamine and potassium) was studied in anaesthetized cats using a single fibre recording technique.2. By administration of algesic agents in doses which produce pain in man and pain reactions in animals, about half of the group IV and two thirds of the group III muscle afferents could be activated. In contrast, group II and

Puberty in monkeys is triggered by chemical stimulation of the hypothalamus.

Gonadal quiescence prior to puberty in primates results from a diminished secretion of the pituitary gonadotropic hormones, follicle-stimulating hormone and luteinizing hormone, which, in turn, is occasioned by an interruption of pulsatile release of gonadotropin-releasing hormone (GnRH) from the hypothalamus during this phase of development. A discharge of GnRH may be provoked from the hypothalamus of prepubertal monkeys, however, by an i.v. injection of N-methyl-D-aspartate (NMDA), an analog of the putative excitatory neurotransmitter, aspartate. Since this action of NMDA is blocked by the s

An endoscopic method for thermal and chemical stimulation of the human oesophagus.

Previous methods for visceral thermal stimulation have lacked control of the temperature rate and visual inspection of the organ. The aims of this study was to develop a method for linear control of heat stimulation in the human oesophagus combined with endoscopy, to assess the reproducibility of this method and to investigate sensitivity to thermal stimulation of the distal oesophagus before and after acid perfusion. A probe with a 2.8 mm endoscope inside was constructed permitting heat and chemical stimulation. Three different temperature ramps were applied in the distal oesophagus in 12 hea

Chemical and immunological analysis of prey-derived vomeronasal stimulants.

We have isolated seven proteins from earthworm preparations that are chemoattractive to garter snakes. Three of these proteins have been purified to homogeneity: two from aqueous earthworm wash (EWW) and one from electric shock-induced earthworm secretion (ESS). One of the two highly purified proteins from EWW has a relative molecular mass of 20 kDa and contains free sulfhydryl groups that appear to play a functional role in its chemoattractivity. The other purified protein from EWW has a molecular mass of 3 kDa (low molecular weight protein, LMW). The highly purified chemoattractive protein (

Chemical sympathectomy and clorgyline-induced stimulation of rat pineal melatonin synthesis.

The response to administration of the specific monoamine oxidase A (MAO-A) blocker clorgyline was investigated in adult male Sprague-Dawley rats which were sympathectomized by injection of the false neurotransmitter 6-hydroxydopamine as newborns. In intact animals which served as controls, the contents of pineal indoles melatonin, serotonin, 5-hydroxytryptophan were augmented, and the content of 5-hydroxyindoleacetic acid decreased 90 min following clorgyline injections when compared to saline receiving rats. Sympathectomized animals exhibited similar responses but these were less pronounced.

Chemical Stimulation of Rodent and Human Cortical Synaptosomes: Implications in Neurodegeneration.

Synaptic plasticity events, including long-term potentiation (LTP), are often regarded as correlates of brain functions of memory and cognition. One of the central players in these plasticity-related phenomena is the α-amino-3-hydroxy-5-methylisoxazole-4-propionate receptor (AMPAR). Increased levels of AMPARs on postsynaptic membranes thus constitute a biochemical measure of LTP. Isolated synaptic terminals (synaptosomes) are an excellent ex vivo tool to monitor synaptic physiology in healthy and diseased brains, particularly in human research. We herein describe three protocols for chemically

Respiratory responses to chemical stimulation in patients with obstructive sleep apnoea.

Chemical control of breathing in obstructive sleep apnoea (OSA) patients has been studied by many authors. The results of previous studies, especially those dealing with hypoxic drive, are discordant. Respiratory responses were studied during hypercapnic and hypoxic stimulation in a group of 37 normocapnic patients with OSA during wakefulness. The diagnosis of OSA was established by standard polysomnography. These patients had increased apnoea/hypopnoea index (AHI; 51 +/- 22 (mean +/- SD)), obesity (body mass index (BMI) 32.4 +/- 5.6 kg.m-2) and normal lung function tests. The control group co

Sex differences in inflammation evoked by noxious chemical, heat and electrical stimulation.

Neurogenic inflammation (NI) is a feature of several inflammatory pain conditions in which females are overrepresented. Therefore, we asked if there are sex differences in the inflammatory response evoked by well known neurogenic stimuli. We compared the amount of plasma extravasation (PE), a measure of inflammation, in the hindpaw skin of male and female rats caused by subcutaneous injection of capsaicin, application of noxious heat (51 degrees C water bath) or electrical stimulation of the saphenous nerve. We also compared the amount of PE in males and females evoked by substance P (SP), the

Chemical stimulation of the area postrema induces cardiorespiratory changes in the cat.

The purpose of our study was to determine the cardiorespiratory effects of exciting cell bodies of the area postrema of the cat. This was accomplished by local application of L-glutamic acid (bilateral application of 5 microliter of a 250-1000 mM solution) and kainic acid (bilateral application of 5 microliter of a 40 mM solution) to the area postrema of chloralose-anesthetized cats while monitoring arterial pressure, heart rate, tidal volume and respiratory rate. These excitatory amino acids activate neuronal cell bodies but not axons of passage. L-Glutamic acid produced a dose-dependent incr

Effects of age on the axon reflex response to noxious chemical stimulation.

The axon reflex flare response to noxious stimulation of the skin is mediated by polymodal nociceptors of "C" fibre primary afferent nerves. Topical application of capsaicin initiates such a flare. The mediator of the response is presumed to be substance P. In this study we examined the flare response to topical capsaicin (1g/L in 70% alcohol for 30 minutes; n = 220) and the substance P content of autopsied skin (3mm punch biopsy; n = 14). The area of the flare response was measured by tracing a Leitz ASM probe onto traced flare outlines. Skin was extracted in acetic acid and acetone and subst

Gastric and pyloric motor response to sympathetic nerve stimulation after chemical sympathectomy.

This study evaluated the effect of electrical stimulation of the greater splanchnic nerve on gastric and gastroduodenal motility in 6-hydroxydopamine (6-OHDA)-pretreated anesthetized dogs. Biopsies from the stomach, pylorus and duodenum were obtained and analyzed by catecholamine fluorescence microscopy. Degeneration of adrenergic terminals in Auerbach's plexus was complete at 1 week. The majority of stimulations in control dogs or dogs treated with 6--OHDA produced contractions in the gastric body, antrum, and pylorus and relaxation of inhibition in the duodenum. The excitatory motor response

Stimulation of chemically induced rectal carcinogenesis by chronic ethanol ingestion.

The effect of chronic ethanol administration on 1, 2-dimethylhydrazine-induced rectal carcinogenesis was investigated in 32 paired male Sprague-Dawley rats fed a nutritionally-adequate liquid diet containing 36% of the total calories as either ethanol or isocaloric carbohydrates. Chronic ethanol ingestion increased the total number of rectal tumors significantly (17 vs 6; P less than 0.02), whereas no cocarcinogenic effect of ethanol was observed in other parts of the intestine. Alcohol did not influence tumor size or histopathology. A 47% increase in the activity of mucosal alcohol dehydrogen

NMDA sensitization and stimulation by peroxynitrite, nitric oxide, and organic solvents as the mechanism of chemical sensitivity in multiple chemical sensitivity.

Multiple chemical sensitivity (MCS) is a condition where previous exposure to hydrophobic organic solvents or pesticides appears to render people hypersensitive to a wide range of chemicals, including organic solvents. The hypersensitivity is often exquisite, with MCS individuals showing sensitivity that appears to be at least two orders of magnitude greater than that of normal individuals. This paper presents a plausible set of interacting mechanisms to explain such heightened sensitivity. It is based on two earlier theories of MCS: the elevated nitric oxide/peroxynitrite theory and the neura
