Antigens of Streptococcus mutans. I. Characterization of a serotype-specific determinant from Streptococcus mutans.

A membrane-associated glycerol teichoic acid antigen has been isolated from Streptococcus mutans AHT and a similar antigen has been demonstrated to be present in each of the other Bratthall serotype a organisms studied. Trichloroacetic acid-extracted material was resolved into two phosphorus-containing antigenic fractions (B and C) by agarose chromatography. Fraction B was preliminarily identified as a phospholipid moiety with a glycerol-to-phosphorus ratio of 2:1, and fraction C showed a ratio of 1:1 indicative of a glycerol teichoic acid. This latter fraction also was associated with glucose

Pulp response to Streptococcus mutans.

The maxillary molar pulps of germ-free rats were mechanically exposed, and suspensions of a strain of freshly grown Streptococcus mutans were applied to the pulp wounds. The pulps were left open to the oral environment, and the animals were maintained in the isolator until they were killed in groups after 2, 7, and 28 days. After 2 days there was little evidence of any pulp response. In the 7-day group early pulp necrosis was present. No evidence of inflammatory infiltration was detected in either the 2- or 7-day animals. After 28 days there was extensive pulp necrosis in many specimens. Denti

Characterization of MreCD in Streptococcus mutans.

BACKGROUND: Activities that control cell shape and division are critical for the survival of bacteria. However, little is known about the circuitry controlling these processes in the dental caries pathogen Streptococcus mutans. METHODOLOGY: We designed experiments to characterize two genes, mreC and mreD, in S. mutans. Assays included cell morphology imaging, protein interaction analysis, transcriptomics, proteomics, and biofilm studies to generate a comprehensive understanding of the role of MreCD in S. mutans. RESULTS: Consistent with mreCD participating in cell elongation, cells lacking t

Streptococcus mutans Extracellular Vesicles Regulate Early Streptococcus sanguinis Nanoadhesion.

Dental caries remains a globally prevalent disease, driven by microbial dysbiosis and competitive ecological shifts within the oral biofilm. A key interaction in this context is the antagonism between Streptococcus mutans and Streptococcus sanguinis, two early colonizers of the dentin surfaces. While bacterial extracellular vesicles (bEVs) have emerged as important mediators of microbial communication, their role in interspecies adhesion and early biofilm dynamics remains poorly understood. This study explored how S. mutans-derived bEVs, isolated from planktonic cultures and biofilms grown on

Fluoride uptake by Streptococcus mutans 6715.

The short-term kinetics of fluoride uptake by cells from 20- to 22-h cultures of Streptococcus mutans strain 6715 were studied using rapid filtration and centrifugation techniques. Saline-suspended organisms were diluted with fluoride-containing solutions buffered at four different pH values (2.0, 4.0, 5.5, and 8.2). Fluoride disappearance from the medium was inversely related to pH and to the duration of the exposure at any given pH. The uptake was rapid and extensive at the lower pH values and decreased as the pH increased. Media fluoride concentrations subsequently increased; i.e., fluoride

Early acquisition of Streptococcus mutans for children.

Existing evidence reveals that in Early Oral Infection the main route of transmission of Streptococcus mutans is mother-child saliva contacts and that initial acquisition takes place during a specific period of time denominated "Window of Infectivity" that lapses between 6 and 30 months of the child's life, with a higher risk between 18 and 30 months of age. The aim of the present study was to analyze Early Oral Infection. The levels of Streptococcus mutans in saliva and bacterial plaque in the binomial mother-child 6 to 18 months after childbirth were evaluated. Twenty-four mother-child binom

Role of sucrose in the fitness of Streptococcus mutans.

INTRODUCTION: Dental caries has been closely linked to fermentable carbohydrates as key environmental factors. Sucrose has been identified as the most cariogenic carbohydrate. Streptococcus mutans, considered to be the primary pathogen causing dental caries, is able to utilize sucrose as a nutrient source, partially for the production of intracellular storage components and for the production of extracellular glucans via the glucosyltransferases GtfB, GtfC, and GtfD. The following study explores the competitiveness and fitness of S. mutans when grown with different concentrations of sucrose.

[The role of Streptococcus mutans in human caries].

Definite proof has been found that S. mutans, and probably S. sobrinus and lactobacilli, are responsible for caries in man. The aciduria of S. mutans encourages the selection inside the plaque and is directly responsible for its cariogenicity. Other aciduric species, as S. sobrinus, cause caries on the smooth surface of the tooth, where the principal causes of the rampant caries reside. If during the eruption of the tooth the fissures become colonized in depth by S. mutans, the development of caries becomes a highly probable event. Instead, if the colonization of the tooth by S. mutans takes p

Asymmetrical dental caries and Streptococcus mutans infection.

A 27-year-old woman chewed unilaterally and had restorations and caries only on the right side of the dentition. The left side had a greater amount of soft and hard deposits and more severe periodontal disease. Plaque samples from occlusal and interproximal tooth surfaces were collected and cultured. The proportion of Streptococcus mutans of the total anaerobic count in the plaque samples from the right side was 1,000 to 10,000 times as high as from the left side. The proportion of S mutans of the total streptococcus was 6% to 20% on the right and less than 1% on the left side.

Trans-Cinnamaldehyde-Fighting Streptococcus mutans Using Nature.

Streptococcus mutans (S. mutans) is the main cariogenic bacterium with acidophilic properties, in part due to its acid-producing and -resistant properties. As a result of this activity, hard tooth structures may demineralize and form caries. Trans-cinnamaldehyde (TC) is a phytochemical from the cinnamon plant that has established antibacterial properties for Gram-positive and -negative bacteria. This research sought to assess the antibacterial and antibiofilm effects of trans-cinnamaldehyde on S. mutans. TC was diluted to a concentration range of 156.25-5000 μg/mL in dimethyl sulfoxide (DMSO)

Biological roles of extracellular proteins in Streptococcus mutans.

Streptococcus mutans remains a key etiologic agent of dental caries by thriving in dysbiotic plaque biofilms that sustain low pH and abundant extracellular matrix. Although acidogenesis and extracellular polysaccharide (EPS) synthesis are long recognized virulence traits, accumulating evidence indicates that a coordinated network of extracellular proteins is the operational interface that links carbohydrate metabolism to adhesion, matrix architecture, quorum-guided behaviors, and ecological competition. Here we review major surface-anchored and secreted proteins and peptides that drive carioge

Systematic Analysis of Lysine Succinylation in Streptococcus mutans.

Lysine succinylation (Ksuc) is a novel post-translational modification (PTM), which regulates biological functions in bacteria. Streptococcus mutans has been identified as a predominant cariogenic pathogen responsible for the initiation and progression of dental caries. However, lysine succinylation in S. mutans has not yet been investigated. In this study, a global lysine succinylome was analyzed to examine Ksuc in S. mutans. Overall, 2250 succinylated sites in 580 proteins were identified. Quantitative analysis demonstrated that Ksuc substrates were substantially altered in the biofilm growt

YkuR functions as a protein deacetylase in Streptococcus mutans.

Protein acetylation is a common and reversible posttranslational modification tightly governed by protein acetyltransferases and deacetylases crucial for various biological processes in both eukaryotes and prokaryotes. Although recent studies have characterized many acetyltransferases in diverse bacterial species, only a few protein deacetylases have been identified in prokaryotes, perhaps in part due to their limited sequence homology. In this study, we identified YkuR, encoded by smu_318, as a unique protein deacetylase in Streptococcus mutans. Through protein acetylome analysis, we demonstr

MecA in Streptococcus mutans is a multi-functional protein.

UNLABELLED: Our recent studies have shown that deficiency of MecA in Streptococcus mutans significantly affects cell division, growth, and biofilm formation. In this study, an in vitro mixed-species model, proteomics, and affinity pull-down assays were used to further characterize the MecA-mediated regulation in S. mutans. The results showed that compared with the wild type, UA159, the mecA mutant significantly reduced its production of glucans and weakened its ability to facilitate mixed-species biofilm formation. Relative to the wild type, the mecA mutant also displayed unique characteristic

The virulence of Streptococcus mutans and the ability to form biofilms.

In some diseases, a very important role is played by the ability of bacteria to form multi-dimensional complex structure known as biofilm. The most common disease of the oral cavity, known as dental caries, is a top leader. Streptococcus mutans, one of the many etiological factors of dental caries, is a microorganism which is able to acquire new properties allowing for the expression of pathogenicity determinants determining its virulence in specific environmental conditions. Through the mechanism of adhesion to a solid surface, S. mutans is capable of colonizing the oral cavity and also of fo

Streptococcus mutans: a rare cause of retroperitoneal abscess.

Retroperitoneal abscesses are uncommonly encountered clinical entities and they represent serious surgical infections associated with significant mortality rates because of their insidious clinical manifestations and diagnostic difficulty. The source of retroperitoneal infections is usually an organ contained within or abutting the retroperitoneum, usually the kidney and the microorganisms most commonly isolated are gram-negative bacilli. Gram-positive cocci, mainly staphylococcal species and rarely streptococcal species, are a less common cause of retroperitoneal abscess and are usually isola

Cytokine induction by Streptococcus mutans and pulpal pathogenesis.

Chronic pulpal inflammation under caries appears to be elicited by bacterial antigens that diffuse into the pulp through dentinal tubules. This prompted the hypothesis that cytokines elicited by antigens from Streptococcus mutans, which frequently dominates shallow lesions, could play a major role in eliciting the initial T-cell response in the pulp. To test this, we examined the ability of S. mutans to stimulate T cells and elicit cytokines and used Lactobacillus casei, which often predominates in deep carious lesions where B cells and plasma cells predominate, as a control. In addition, the
