Representative strains of Streptococcus mutans and Streptococcus sobrinus showed differences in susceptibility to members of the monobactam group of beta-lactam antibiotics: S. sobrinus was less sensitive than S. mutans. The minimum inhibitory concentrations of aztreonam (AZT) and carumonam, both of which belong to this group, were 2,000 microg/ml for S. sobrinus and 125 microg/ml for S. mutans. Further addition of fosfomycin, bacitracin and sodium chloride to Mitis Salivarius agar (MS) supplemented with AZT resulted in growth inhibition of S. mutans and oral streptococci other than S. sobrinu

Streptococcus sobrinus is a member of the Streptococcus mutans group and is a cariogenic bacterium similar to S. mutans. One of the characteristics of these bacteria is bacitracin resistance, and BceAB (MbrAB), an ABC transporter of S. mutans, has been reported to be involved in bacitracin resistance. However, the factors responsible for bacitracin resistance in S. sobrinus are unknown. In this study, 11 S. sobrinus strains were isolated from the oral cavity, and their bacitracin susceptibility was assessed. The results showed that 9 strains were low-susceptible to bacitracin (MIC = 128-512 µg

The mutans group of streptococci is considered to play a key role in the etiology of dental caries. We have evaluated the ability of different substances to prevent dental plaque formation without affecting Streptococcus sobrinus viability. Viable organisms were detected as CFU/mL in agar plates and bacterial adherence was assessed by dry weight. We studied 23 compounds and we demonstrated that phenyl salicylate, phenylmercuric nitrate and potassium iodate are more effective to inhibit adhesion without showing antibacterial activity.

We previously found that tonsillar application of antigen induces a strong antibody response to Streptococcus sobrinus in saliva and blood plasma. Rabbits immunized against S. sobrinus by tonsillar application were highly resistant to experimental dental caries triggered by oral inoculation of living S. sobrinus organisms with sucrose. In the present study, we examined the reaction of S. sobrinus antigens to the antibodies induced by the tonsillar application of S. sobrinus AHT-k in rabbits and compared them to those antibodies induced by intramuscular injection. In an enzyme-linked immunosorb

Streptococcus mutans is a major pathogenic habitant of oral caries. Owing to its physiological and biochemical features, it prevails in the form of plaque biofilm together with another important mutans streptococci species, Streptococcus sobrinus. Both species are considered as initiators of cavity lesions, and biofilm is essential to the dental caries process. Compared with the planktonic populations, the biofilm form has higher resistance to environmental conditions and antibiotics. Dental plaques also secure the long-term survival of microorganisms and protection from any stress conditions.

BACKGROUND: Mutans streptococci (Streptococcus mutans and S. sobrinus) are considered to be major etiologic agents of dental caries. Using a polymerase chain reaction method, we detected those bacteria from 145 outpatients (6-30 years old) with intellectual disabilities (ID) and their presence was compared with the incidence of dental caries.
METHODS: Plaque samples were collected from all erupted tooth sites in subjects with a sterile toothbrush. A dental examination was performed to determine the number of decayed and filled teeth (DFT score) in permanent dentition using the WHO caries diag

Cariogenic dental plaque may be assimilated to a biofilm resulting from the adhesion of S. mutans, then from the coaggregation of other streptococci, or other genus. We used a static monospecific biofilm model. Supports or bacteria were treated with inhibitors before adhesion in order to clarify the nature of adhesins responsible for the primary adhesion of S. mutans and S. sobrinus on Tygon. To determine the bindings of coaggregation, inhibitors were applied on one-day-old biofilms. Analysis of effects were performed by automatic inoculator Spiral (Interscience) for microbiological methods, a

OBJECTIVES: Dental caries are caused by organic acids produced by cariogenic bacteria through carbohydrate metabolism. Suppression of acid production without disrupting the oral microbiome is a promising preventive strategy against dental caries. Surfactin, a naturally derived biosurfactant, has several biological activities. However, its effects on acid production by cariogenic bacteria remain unclear. In this study, the effects of surfactin on lactate production, growth, biofilm formation, and metabolic activity of Streptococcus sobrinus, were investigated.
METHODS: In vitro assays were per

Nicotinamide adenine dinucleotide (NAD+) serves as a critical cofactor in redox reactions and metabolic transformations catalyzed by NAD-dependent enzymes and is essential for bacterial survival and virulence. The biosynthesis of NAD+ in the cariogenic pathogen Streptococcus Sobrinus (S. sobrinus), a pivotal participant in oral cavities of children and adolescents with a history of caries, has yet to be explored. Bioinformatics, genetics, and biochemical techniques were used to identify NAD+ biosynthesis pathways and corresponding regulator in S. Sobrinus. S. sobrinus lacks de novo NAD+ synthe

We studied the effect of oral infection with cariogenic micro-organisms on alveolar bone loss and root surface caries subsequent to gingivectomy in rats. Thirty-six rats were fed diet MIT 200 (67% sucrose); one-half, the controls, had antibiotics added to the diet. At ages 18, 19, and 21 days, the experimental group was orally infected with streptomycin-resistant Streptococcus sobrinus (mutans) 6715 and Actinomyces viscosus M-100. At age 30 days, all animals were subjected to a gingivectomy on maxillary and mandibular left quadrants. The experimental group was re-inoculated at age 36 days. Ani

To evaluate interleukin 1 (IL-1) as a mucosal adjuvant in the induction of salivary antibodies to Streptococcus sobrinus, S. sobrinus together with IL-1 was applied through the palatine tonsils of rabbits. IL-1 caused approximately 50 and 100% increases in the antibodies reacting against S. sobrinus fragments in the saliva and blood plasma, respectively, compared to the antibodies in those same fluids after tonsillar applications of S. sobrinus alone. In the case of the addition of IL-1, the antibodies reacting to the protein antigens of S. sobrinus increased in each fluid, without affecting t

BACKGROUND: Streptococcus mutans and Streptococcus sobrinus are Gram-positive bacteria involved in the development of dental caries, as they are able to form biofilms on tooth enamel, ferment sugars into acids, and survive under acidic conditions. This ultimately leads to a local lowering of the pH value on the tooth surface, which causes enamel cavities.
HYPOTHESIS: One measure to reduce caries is to limit the growth of cariogenic bacteria by using two anti-bacterial agents with different mechanisms of action. The hypothesis of this study was that the anti-bacterial activity of ω-6 polyunsat

Streptococcus mutans and S. sobrinus are associated with the development of dental caries. These bacteria were detected by PCR and then their presence was compared with the incidence of dental caries in 77 Japanese pre-school children. Plaque samples were collected from all erupted tooth sites in the subjects, aged 3-5 years old and each with primary dentition, with a sterile toothbrush. A dental examination was performed for dmft (decayed, missing, filled, total) with the WHO caries diagnostic criteria. In all subjects, the prevalence of S. mutans and S. sobrinus was 72.8% and 61.1%, respecti

The development of intricate dental biofilms and an imbalance in the oral microbiota are linked to Early Childhood Caries (ECC) as a multifactorial oral disease. Although Streptococcus mutans has been extensively studied, other microorganisms, including Streptococcus sobrinus and Candida albicans, also contribute significantly to the severity of ECC. Probiotics like Limosilactobacillus reuteri, which produce antimicrobial metabolites, such as reuterin, have shown potential as biofilm-modulating agents, but their effects on dual-species biofilms from ECC clinical isolates remain poorly understo

The prevalence of caries on exposed root surfaces in 88-year-old subjects with and without salivary levels of Streptococcus sobrinus was studied. Ninety-two individuals were examined with regard to root caries lesions and fillings. The root caries index (RCI) was calculated and related to salivary flow rate and buffer capacity, plaque score and salivary counts of Streptococcus mutans, S. sobrinus and lactobacilli. In 89 subjects with exposed root surfaces, all but 2 harbored mutans streptococci; 51 subjects carried S. mutans only, 35 both S. sobrinus and S. mutans, and 1 S. sobrinus only. The
