[Orthodontic treatment and mechanical stress].

In orthodontic treatment, the role of periodontal ligament is essential for the teeth to move within the alveolar process against the mechanical stress. Mesenchymal stem cells that differentiate to chondrocytes, adipocytes, and nerve cells is known to exist in the PDL. In both process of physiological and orthodontic tooth movement, bone resorption mainly occurs at the anterior region toward the direction of tooth movement, and bone apposition/formation at the posterior region to the direction of the tooth movement, and bone turnover occurs locally and site specifically, which results in const

[Mechanical stress and tissue engineering].

Recently, people start to pay attention to the effects of physical factors on cell differentiation and function. In this review, we introduce the effect of mechanical stress on chondrocytes for engineerd cartilage by "Tissue engineering" . To reconstruct articular cartilage, it is important to re-differentiate chondrocytes that have de-differentiated along with cell proliferation in vitro . We show here hydrostatic pressure that is a mechanical stress applying articular cartilage, promotes re-differentiation of de-differentited chondrocytes by increasing expression of aggrecan and type II coll

Biosynthetic solutions to mechanical stress: why do mechanical stresses drive altered metabolism?

Cells reside in mechanically stressful microenvironments where protrusive and traction forces from neighboring cells and cell-intrinsic forces derived from adhesions to the extracellular matrix (ECM) establish a dynamical "Mechanoreciprocity". Mechanoreciprocity is the concept that cells respond to the physical properties of the cellular microenvironment by reciprocally exerting proportional forces [1]. However, if feed-forward mechanical stress responses based on energetically demanding cytoskeletal dynamics are the only ways cells reciprocally respond to physical cues, we would not observe t

Mechanical analysis of femoral stress-riser fractures.

BACKGROUND: A stress-riser fracture develops when stress in an object is higher than that in the surrounding material. The aim of this retrospective study was to evaluate all consecutive femoral stress-riser fractures during a period of >10 years and to verify high-risk factors by using mechanical analysis. METHODS: Forty clinical fractures caused by local stress concentration in the femoral cortical bone were included. Risk factors were analyzed, and Sawbones models were used to verify their effect. One hundred thirty-six models were divided into 6 groups to simulate clinical cases of stress

Mechanical stress models of Alzheimer's disease pathology.

INTRODUCTION: Extracellular accumulation of amyloid-β protein and intracellular accumulation of tau in brain tissues have been described in animal models of Alzheimer's disease (AD) and mechanical stress-based diseases of different mechanisms, such as traumatic brain injury (TBI), arterial hypertension (HTN), and normal pressure hydrocephalus (NPH). METHODS: We provide a brief overview of experimental models of TBI, HTN, and NPH showing features of tau-amyloid pathology, neuroinflammation, and neuronal loss. RESULTS: "Alzheimer-like" hallmarks found in these mechanical stress-based models we

Stress Ulcer Prophylaxis during Invasive Mechanical Ventilation.

BACKGROUND: Whether proton-pump inhibitors are beneficial or harmful for stress ulcer prophylaxis in critically ill patients undergoing invasive ventilation is unclear. METHODS: In this international, randomized trial, we assigned critically ill adults who were undergoing invasive ventilation to receive intravenous pantoprazole (at a dose of 40 mg daily) or matching placebo. The primary efficacy outcome was clinically important upper gastrointestinal bleeding in the intensive care unit (ICU) at 90 days, and the primary safety outcome was death from any cause at 90 days. Mechanical stress on canola seed during dehulling.

The mature canola seed comprises an embryo, a group of structures that have a high content of protein and oil, and a fibrous seed coat held together by endosperm tissues. High-quality canola meal production requires removal of fiber-rich seed hull that typically contributes little nutrient value. Canola hull accounts for more than 70.0% of the insoluble fiber within the seed, which limits its uses as feed. Dehulling canola seed, before oil extraction, increases the protein content and decreases fiber content in the meal, to up to 50.0% and to down to 10.0%, respectively. Due to the complexity

Association between mechanical stress and bone remodeling.

Stress patterns and levels were analyzed by use of the three-dimensional finite element method. Three-dimensional models were constructed for the human upper central incisor (1205 nodes and 920 elements) and the sheep metacarpus (240 nodes and 128 elements). Orthodontic and orthopedic forces were applied to the tooth and the bone, simulating orthodontic tooth movement and experimental loading test for the bone. Three principal stresses were determined in the alveolar bone and the sheep long bone. The following results were obtained. 1. Stress distributions in the lateral alveolar bone were sim

Endoplasmic reticulum stress regulates mechanical stress-induced ossification of posterior longitudinal ligament.

PURPOSE: The pathogenesis of ossification of posterior longitudinal ligament (OPLL) is not completely clear. Previous study has confirmed a single-pass type I endoplasmic reticulum (ER) membrane protein kinase (PERK), which is a major transducer of the ER stress, participates in the process of OPLL in vitro. This study aimed to demonstrate the role of ER stress in mechanical stress (MS)-induced OPLL. METHODS: The posterior longitudinal ligaments were collected intraoperatively. The expression of ER stress markers in ligament tissue samples was compared between OPLL and non-OPLL patients in vi

Mechanical stress and mechanosensitive protein in skin tumors.

The skin is highly susceptible to external physical stimuli, and mechanical stresses contribute to certain skin tumors. Focusing on keloid, basal cell carcinoma, cutaneous squamous cell carcinoma and melanoma, we systematically examined the roles and mechanisms between mechanical stresses, such as tensile stress and matrix stiffness, and mechanosensitive proteins, including Piezo, transient receptor potential (TRP), integrin family, Yes-associated protein (YAP) and transcriptional coactivator with PDZ-binding motif (TAZ), in skin tumor progression. Additionally, we analyze existing research ch

Atypical Unilateral Sacroiliitis Secondary to Mechanical Stress Injury.

We report for the first time a case of atypical unilateral sacroiliitis secondary to mechanical stress injury. Unilateral sacroiliitis can be caused by a variety of etiologies. The first diagnosis to rule out is infection since it requires urgent treatment to avoid its serious consequences. Spondyloarthritis can be manifested by unilateral sacroiliitis in its early stage. Sacral fractures should always be looked for on the imaging modalities performed. In our case, no signs of infection or systemic disease were found. No fracture was seen on the imaging examinations and we had negative culture

The Role of Mechanical Stress in Hidradenitis Suppurativa.

Mechanical stress can act as a possible trigger in the development of hidradenitis suppurativa (HS). The mechanical stress has been supported by (1) the special biomechanical conditions in the typically topographic areas of HS; (2) the indirect proof of similar findings in associated follicular occlusion diseases such as acne mechanica and pilonidal sinus disease, and in limb amputees after expression of mechanical forces; (3) pathohistologic, ultrasonography, and immunologic findings; and (4) overweight patients seem to be most susceptible to the effects of mechanical stress.

Aggravation of cardiac myofibroblast arrhythmogeneicity by mechanical stress.

AIMS: Myofibroblasts (MFBs) as appearing in the myocardium during fibrotic remodelling induce slow conduction following heterocellular gap junctional coupling with cardiomyocytes (CMCs) in bioengineered tissue preparations kept under isometric conditions. In this study, we investigated the hypothesis that strain as developed during diastolic filling of the heart chambers may modulate MFB-dependent slow conduction. METHODS AND RESULTS: Effects of defined levels of strain on single-cell electrophysiology (patch clamp) and impulse conduction in patterned growth cell strands (optical mapping) wer

A novel mechanism of mechanical stress-induced hypertrophy.

Angiotensin II (AII) type 1 (AT1) receptor plays a critical role in load-induced cardiac hypertrophy. We have recently found a novel mechanism of mechanical stress-induced activation of the AT1 receptor, which is independent of AII. Mechanical stretch did not activate ERKs in HEK293 cells and COS7 cells which had no AT1 receptor, but when AT1 receptor was overexpressed in these cells, stretch activated ERKs, Galphaq and JAK2. An AT1 receptor blocker, candesartan, inhibited stretch-induced activation of ERKs in these cells. Stretch also activated ERKs in COS7 cells expressing AT1 mutant which d

Mechanical stress in the urbanized Roman Phoenician coast.

OBJECTIVE: Skeletal populations from Byblos, Beirut, and Tyre were studied to assess mechanical stress along the Roman Phoenician coast. MATERIALS: The sample included 153 adult skeletons. METHODS: Skeletal remains were macroscopically assessed for osteoarthritis, intervertebral disc disease (IDD), and Schmorl's nodes. RESULTS: The Byblos population experienced higher levels of mechanical stress than the Beirut and Tyre ones. Sex-based differences were also found in all skeletal assemblages with males likely engaging in physically more demanding tasks. CONCLUSIONS: The variation in mechani

Melaninization Reduces Cryptococcus neoformans Susceptibility to Mechanical Stress.

Melanin is a complex pigment that is found in various fungal species and is associated with a multitude of protective functions against environmental stresses. In Cryptococcus neoformans, melanin is synthesized from exogenous substrate and deposited in the cell wall. Although melanin is often cited as a protector against mechanical stress, there is a paucity of direct experimental data supporting this claim. To probe whether melanin enhances cellular strength, we used ultrasonic cavitation and French cell press pressure to stress cryptococcal cells and then measured changes in cellular morphol

[Mechanical stress promotes cartilage repair in inflammatory environment].

OBJECTIVE: To investigate the effect and mechanism of mechanical stress on cartilage repair in inflammatory environment. METHODS: The chondrogenic progenitor cells (CPCs) were isolated from the knee joint cartilage of patients with osteoarthritis (OA) undergoing total knee arthroplasty. The CPCs were cultured and expanded in a 3-D scaffold constructed with alginate. Intermittent hydrostatic pressure (IHP) was applied in a inflammatory environment induced by IL-1β, and Western blot was used to detect the expression of MAPK signaling pathway proteins. Cell proliferation was detected by CCK-8 me

Microtubules self-repair in response to mechanical stress.

Microtubules--which define the shape of axons, cilia and flagella, and provide tracks for intracellular transport--can be highly bent by intracellular forces, and microtubule structure and stiffness are thought to be affected by physical constraints. Yet how microtubules tolerate the vast forces exerted on them remains unknown. Here, by using a microfluidic device, we show that microtubule stiffness decreases incrementally with each cycle of bending and release. Similar to other cases of material fatigue, the concentration of mechanical stresses on pre-existing defects in the microtubule latti

Chronic intermittent mechanical stress increases MUC5AC protein expression.

Increased abundance of mucin secretory cells is a characteristic feature of the epithelium in asthma and other chronic airway diseases. We showed previously that the mechanical stresses of airway constriction, both in the intact mouse lung and a cell culture model, activate the epidermal growth factor receptor (EGFR), a known modulator of mucin expression in airway epithelial cells. Here we tested whether chronic, intermittent, short-duration compressive stress (30 cm H(2)O) is sufficient to increase the abundance of MUC5AC-positive cells and intracellular mucin levels in human bronchial epith
