Three root-parasitic witchweed (Striga) species can cause up to total loss of grain and legume crops for millions of farmers in Africa. The damage is initiated before Striga emerges from the soil. Four decades of resistance breeding, especially maize, have at best conferred localized moderate resistance that has not greatly increased yields. Recent innovative basic research has led to major advances that are herein evaluated in biocontrol, formulating suicidal germination stimulants and gene-edited crops. A mycoherbicide was able to become a commercial success because the innovators rendered t

The root hemiparasite witchweed (Striga spp.) is a devastating agricultural pest that causes losses of up to $1 billion US annually in sub-Saharan Africa. Development of resistant crops is one of the cost-effective ways to address this problem. However, the molecular mechanisms underlying resistance are not well understood. To understand molecular events upon Striga spp. infection, we conducted genome-scale RNA sequencing expression analysis using Striga hermonthica-infected rice (Oryza sativa) roots. We found that transcripts grouped under the Gene Ontology term defense response were signific

The high-level view of global food systems identifies three all-encompassing barriers to the adoption of food systems solutions: knowledge, policy, and finance. These barriers, and the siloed characteristics of each of these, have hindered the development and adoption of microbial herbicides. How knowledge, policy, and finance are related to the Toothpick Project's path of commercializing a new bioherbicide, early in the scope of the industry, is discussed here. The Toothpick Project's innovation, developed over four decades and commercialized in 2021, uses strains of Fusarium oxysporum f.sp.

Parasitic plants of the Orobanchaceae family, particularly Striga hermonthica, rank among the world's most devastating agricultural weeds (Jamil et al., 2021). They inflict massive yield losses on staple cereal crops across Africa, including sorghum, millet, and rice, as well as globally on other essential crops such as maize, rapeseed, tomato, and legumes (Parker et al., 2009; Bouwmeester, 2018). These root parasites have evolved a specialized organ called the 'haustorium' that connects to the vascular system of the host to siphon water, minerals, and nutrients (Yang et al., 2015). A critical

The present study investigated the antiglycation activities of extracts and fractions of Phyllanthus amarus, Chrysanthellum americanum, Striga hermonthica and, based on cytotoxicity results, evaluated the anti-inflammatory potential of the ethyl acetate fraction of Striga hermonthica (EtOAc-Sh) in AGE-challenged THP-1 monocytes. The in vitro methylglyoxal (MGO)-bovine serum albumin (BSA) assay revealed notable inhibition of AGE formation by EtOAc-Sh (IC50 = 100.1 ± 0.001 μg/mL; quercetin 1.23 μM, gallic acid 0.131 μM, rutin 0.0021 μM), with rutin used as the standard (IC50 = 402 ± 0.30 μM). Witchweed (Striga hermonthica), also called striga, is a parasitic weed that causes high yield losses in maize on more than 200 000 ha in Kenya alone. A new commercial, biological herbicide developed in Kenya is able to control striga effectively. The product was approved for use by the Pest Control Products Board in Kenya in September, 2021. It is self-produced in villages using a secondary inoculum provided by a commercial company. The formulated product has some disadvantages, which are a complicated production process, a very short shelf life and high application rate. Additionally, the pr

Low soil nitrogen (low-N), Striga hermonthica infestation and vitamin A deficiency in normal endosperm maize are major challenges confronting maize production and nutrition of the people of sub-Saharan Africa (SSA). Development of pro-vitamin A (PVA) maize hybrids with combined resistance/tolerance to the two stress factors is crucial in mitigating the food insecurity and nutrition challenges resulting from low-N deficiency and Striga infestation. One hundred and fifty hybrids plus six hybrid checks were evaluated under low-N, Striga-infested and optimal conditions in Nigeria for two years. Th

The parasitic weed Striga has led to billions of dollars' worth of agricultural productivity loss worldwide. Striga detects host plants using compounds of the strigolactone class of phytohormones. Early steps in the strigolactone signaling pathway involve substrate binding and hydrolysis followed by a conformational change to an "active" or "closed" state, after which it associates with a MAX2-family downstream signaling partner. The structures of the inactive and active states of strigolactone receptors are known through X-ray crystallography, and the transition pathway from the inactive to a

Vitamin A deficiency, drought, low soil nitrogen (low-N), and Striga hermonthica parasitism of maize (Zea mays L.) cause malnutrition and food insecurity in sub-Saharan Africa. The objectives of this study were to determine combining abilities of extra-early provitamin A (PVA) lines, classify them into heterotic groups (HGs), identify testers, and determine yield stability of hybrids under contrasting environments in two trials. In Trial 1, 190 F1 hybrids plus six checks were tested under Striga-infested, drought, and stress-free environments in Nigeria from 2015-2017. In Trial 2, 35 extra-ear

Obligate parasitic plants, particularly members of the Orobanchaceae family, including Striga and Orobanche, greatly devastate crop production. Here, we synthesize recent advances in understanding the molecular and ecological dynamics underlying parasitic plant-host interactions, focusing on critical stages of parasitism: germination, host detection, haustorium formation, and resource extraction. Orobanchaceous parasites exploit host-derived strigolactones (SLs) to break seed dormancy, whereas Cuscuta species do not rely on SLs for germination. Instead, chemotropic responses to host-exuded com

The plant growth regulator zaxinone is essential for proper rice growth and development. Additionally, zaxinone and its two synthetic mimics, MiZax3 and MiZax5, have been shown to significantly promote crop growth and reduce infestation by the root parasitic plant Striga by suppressing strigolactone (SL) production, highlighting their potential for field application. Here, we developed 4 new MiZax through structural modifications of the methoxybenzene ring in MiZax3 and evaluated their effects on plant growth and SL exudation. These newly developed mimics enhanced rice growth and reduced SL re

Although strigolactones play a critical role as rhizospheric signaling molecules for the establishment of arbuscular mycorrhizal (AM) symbiosis and for seed germination of parasitic weeds, scarce data are available about interactions between AM fungi and strigolactones. In the present work, we present background data on strigolactones from studies on their seed germination activity on the parasitic weeds Orobanche and Striga, the importance of nitrogen and phosphorus for this seed germination activity, and what this could mean for AM fungi. We also present results on the susceptibility of plan

Parasitic plants are a special group deriving their nutrients from another plant, some of which such as witchweeds (Striga spp.) and broomrapes (Orobanche and Phelipanche spp.) are referred as weeds responsible for severe crop losses in agriculture. The parasite attaches to and feeds off its host using a haustorium, which also facilitates the transport of various molecules between the parasite and its host. These translocation molecules have received extensive attention from researchers. In this review, we summarize the existing knowledge on the transfer of molecules such as pathogens, herbici

Reductive metabolism of strigolactones (SLs) in several plants was investigated. Analysis of aquaculture filtrates of cowpea and sorghum each fed with four stereoisomers of GR24, the most widely used synthetic SL, revealed stereospecific reduction of the double bond at C-3' and C-4' in the butenolide D-ring with preference for an unnatural 2'S configuration. The cowpea metabolite converted from 2'-epi-GR24 and the sorghum metabolite converted from ent-GR24 had the methyl group at C-4' in the trans configuration with the substituent at C-2', different from the cis configuration of the synthetic

Strigolactones (SLs) are important phytohormones that regulate plant architecture, stress response, and adaptation. SLs exuded by roots also act as signals that allow symbiotic fungi and root-parasitic Striga plants to detect their host. Carlactone, the precursor of SLs, is derived from all-trans-β-carotene through the sequential action of the enzymes DWARF27 (D27), carotenoid cleavage dioxygenase 7 (CCD7), and CCD8. D27 catalyses the isomerisation between all-trans and 9-cis-β-carotene, enriching 9-cis-β-carotene, which is the substrate for CCD7. D27 paralogues (D27likes) have also been repor

Polyphenols are one of the largest plant-derived natural product and they play an important role in plants' defense as well as in human health and disease. A number of them are pleiotropic molecules and have been shown to regulate signaling pathways, immune response and cell growth and proliferation which all play a role in cancer development. Hematological malignancies on the other hand, are cancers of the blood. While current therapies are efficacious, they are usually expensive and with unwanted side effects. Thus, the search for newer less toxic agents. Polyphenols have been reported to po
