The rate of transmammary transmission of Stronglyloides ratti was examined in albino rats in terms of the route of subcutaneous (s.c.) migration from the infection site (the skin) to the cranium. Inoculation sites nearer the cranium resulted in less frequent transmammary infection. The maximum number of adult worms was recovered from the sucklings when the mother was inoculated in her hindquarter and sucklings were allowed to feed for 30-36 h after inoculation (AI). Few worms were recovered from sucklings when they were allowed to nurse during periods of less than 24 h AI or greater than 42 h

Experimental infections of C57Bl/6 mice with strongyloides ratti produced marked resistance to challenge. Resistance to reinfection was induced by as few as six filariform larvae. The degree of resistance produced by soluble somatic antigens prepared from filariform larvae of S. ratti was comparable to that seen after a primary infection. The addition of Freund's adjuvants to the antigen did not increase resistance significantly. Intraperitoneal implantation of micropore chambers containing filariform larvae failed to induce resistance to reinfection. Considerable resistance to a challenge inf

Strongyloides spp. are common parasites of vertebrates and two species, S. ratti and S. venezuelensis, parasitize rats; there are no known species that naturally infect mice. Strongyloides ratti and S. venezuelensis overlap in their geographical range and in these regions co-infections appear to be common. These species have been widely used as tractable laboratory systems in rats as well as mice. The core biology of these two species is similar, but there are clear differences in aspects of their within-host biology as well as in their free-living generation. Phylogenetic evidence suggests th

Strongyloides ratti has a complex life-cycle with two adult generations, one free-living and dioecious and one parasitic and female only. The parasitic females reproduce by parthenogenesis, but it is unclear whether this is mitotic or meiotic in nature. This question has been addressed genetically by analysing the progeny of parasitic females that were heterozygous at an actin locus for evidence of allelic segregation. Such progeny were similarly heterozygous showing that segregation had not occurred. It was therefore concluded that reproduction in the parasitic female of S. ratti is functiona

Eight days after mother rats were injected with 4000 infective larvae of Strongyloides ratti at different stages of lactation the numbers of adult worms in their intestines were uniformly low (less than 3% of the dose) compared with unmated controls (mean = 25%). Those in their litters varied from 12% on day 5 to a maximum of 47% on day 17 post partum. These data, which do not correlate with lactational performance, imply that parasite movements in lactating rats are controlled by qualitative, not quantitative, consequences of humoral events. The numbers of worms in litters are concluded to be

The elimination of adult parasites from the intestines of rats after a first and second infection of Strongyloides ratti was studied. Expulsion after a second infection was anamnestic, indication that the response is immunologic. Intestinal mast cell responses accompanied damage and expulsion of worms, the secondary mast cell response being more rapid but less intense that the first. Antithymocyte serum suppressed expulsion as well as both the intestinal mast cell and circulating eosinophil responses to primary infection.

BACKGROUND: Strongyloidiasis is a truly neglected tropical disease, but its public health significance is far from being negligible. At present, only a few drugs are available for the treatment and control of strongyloidiasis.
METHODOLOGY/PRINCIPAL FINDINGS: We investigated the activity of tribendimidine against third-stage larvae (L(3)) of Strongyloides ratti in vitro and against juvenile and adult stages of the parasite in vivo. S. ratti larvae incubated in PBS buffer containing 10-100 microg/ml tribendimidine died within 24 hours. A single 50 mg/kg oral dose of tribendimidine administered

Strongyloides ratti is a nematode parasite of rats. It is able to undergo two types of development outside the host: heterogonic (free-living adults and sexual reproduction) and homogonic (direct larval development). Homogonic development has a number of similarities with the development of the dauer stage of free-living nematodes, including Caenorhabditis elegans. Using isofemale lines of the parasite, factors that control this developmental choice have been investigated. Isofemale lines can be selected for both heterogonic and homogonic development, but are still able to respond to environme

Most recent authors suggest that larvae of the threadworm, Strongyloides ratti, migrate from a cutaneous infection site to the small intestine via the naso-frontal region of the head. However, the proportion of larvae that successfully reach the intestine having followed this pathway had not been determined. Using compartmental analysis we have obtained a comprehensive quantitative description of larval migration during a primary infection of rats with S. ratti. Mean residence times in organs through which larvae migrate were calculated and the proportion of the larvae arriving in the small in

Infective larvae of homogonic Strongyloides ratti grown in faecal culture with 32P or 75Se acquired a significant amount of radioactivity which was firmly attached to them. Heating removed most of the 32P but left 75Se in place. Subcutaneous injection of virgin and nursing mother rats with living and heat-killed radioactive larvae resulted in a pattern of labelling in the small intestine of injected animals and, in the case of 75Se, those of suckling pups, which can only be explained if labelled worms follow the natural migratory routes. The use of this tool in migratory studies is discussed,

In order to explain the hypocorticosteronemia which establishes itself in rats with Strongyloides ratti parasites, we are proceeding with a functional exploration of the hypothalamo-hypophyso-corticosurrenal axis in experimentally infested rodents. The adreno-cortico-tropic-hormone test (A.C.T.H.), stress to ether and the Lysin-8-Vasopressin and Metopirone tests lead to concordant results: -- the hypothalamo-pituitary-corticosurrenal present a functional integrity which allows it to react normally to direct stimulation or to feed-back; -- the deficiency seems to relate to the hypothalamic secr

The course of infection with Strongyloides ratti was examined in congenitally hypothymic CBA/H, C57Bl/6 and BALB/c nude mice. The intensity of infection and the duration of faecal larval excretion were both increased in nude mice when compared with intact mice. Adult worms persisted in the small intestine of nude mice for at least 6 weeks. Greater worm burdens were found in such mice after subcutaneous injection as compared with percutaneous infection. Hypothymic mice did not acquire resistance to re-infection. It is concluded that both the spontaneous expulsion of worms in primary infection a

Parasitic nematodes show levels of genetic diversity comparable to other taxa, but the functional consequences of this are not understood. Thus, a large body of theoretical work highlights the potential consequences of parasite genetic diversity for the epidemiology of parasite infections and its possible implications for the evolution of host and parasite populations. However, few relevant empirical data are available from parasites in general and none from parasitic nematodes in particular. Here, we test two hypotheses. First, that different parasitic nematode genotypes vary in life-history

The possibility of coproantigen detection in the diagnosis of strongyloidiasis was investigated in rats experimentally infected with Strongyloides ratti. A capture ELISA was developed using affinity-purified polyclonal antibodies raised in rabbits against adult and filariform larval somatic antigens. The first detection of coproantigen coincided with patency and the coproantigen level remained elevated even when the parasite count was low. Following challenge infection, when examination of stool for larvae or ova was negative, coproantigen was detected by anti-adult somatic capture ELISA. Desp

Strongyloides ratti is a parasitic nematode that naturally infects wild rats. However, most laboratory rat and mouse strains are fully susceptible to infection. Immunocompetent BALB/c and C57BL/6 mice terminate S. ratti infections within a month in the context of a canonical type 2 immune response and remain semi-resistant to a re-infection. The course of infection can be divided into three phases: (a) the tissue migration phase of the infective third-stage larvae during the first two days; (b) the early intestinal phase, including the molting to the adult parasites and embedding in the mucosa

The immunological environment experienced by parasitic nematodes varies greatly between hosts and is particularly influenced by whether or not a host has been previously infected. How a parasitic nematode responds to these different environments is poorly understood, but may allow a parasite to ameliorate the adverse effects of host immunity on parasite fitness. Here we use a microarray approach to identify genes in the parasitic nematode Strongyloides ratti that exhibit differential transcription between different rat host immunological environments, and between replicate lines of S. ratti se
