Optimization of degradation conditions for sulfachlorpyridazine by Bacillus sp. DLY-11 and analysis of biodegradation mechanisms.

Sulfachloropyridazine (SCP) is a common sulfonamide antibiotic pollutant found in animal excreta. Finding highly efficient degrading bacterial strains is an important measure to reduce SCP antibiotic pollution. Although some strains with degradation capabilities have been screened, the degradation pathways and biotransformation mechanisms of SCP during bacterial growth are still unclear. In this study, a strain capable of efficiently degrading SCP, named Bacillus sp. DLY-11, was isolated from pig manure aerobic compost. Under optimized conditions (5 % Vaccination dose, 51.5 ℃ reaction temperat

Inhibition of recombinant Pneumocystis carinii dihydropteroate synthetase by sulfa drugs.

Forty-four sulfa drugs were screened against crude preparations of recombinant Pneumocystis carinii dihydropteroate synthetase. The apparent Michaelis-Menten constants (Km) for p-aminobenzoic acid and 7,8-dihydro-6-hydroxymethylpterin pyrophosphate were 0.34 +/- 0.02 and 2.50 +/- 0.71 microM, respectively. Several sulfa drugs, including sulfathiazole, sulfachlorpyridazine, sulfamethoxypyridazine, and sulfathiourea, inhibited dihydropteroate synthetase approximately as well as sulfamethoxazole, as determined by the concentrations which cause 50% inhibition and/or by Ki. Activity of Sulfa Drugs and Their Combinations against Stationary Phase B. burgdorferi In Vitro.

Lyme disease is a most common vector-borne disease in the US. Although the majority of Lyme patients can be cured with the standard two- to four-week antibiotic treatment, at least 10%-20% of patients continue to suffer from prolonged post-treatment Lyme disease syndrome (PTLDS). While the cause for this is unclear, one possibility is that persisting organisms are not killed by current Lyme antibiotics. In our previous studies, we screened an FDA drug library and an NCI compound library on B. burgdorferi and found some drug hits including sulfa drugs as having good activity against B. burgdorf

Sulfadiazine, sulfamethazine and sulfachloropyridazine removal using three different porous materials: Pine bark, "oak ash" and mussel shell.

This work focuses on studying the efficacy of three different by-products to adsorb three antibiotics (sulfadiazine, SDZ; sulfamethazine, SMT; sulfachloropyridazine, SCP). These antibiotics can be considered pollutants of the environment when they reach water, as well as in cases where they are spread on soils through irrigation or contained in sewage sludge or livestock manure. In this study, batch-type adsorption/desorption experiments were performed for each of the three sulfonamides, adding 7 different concentrations of the antibiotics, going from 1 to 50 μmol L-1, and with contact time of

New Models for Estimating the Sorption of Sulfonamide and Tetracycline Antibiotics in Soils.

Sulfonamides (SAs) and tetracyclines (TCs) are two classes of widely used antibiotics. There is a lack of easy models for estimating the parameters of antibiotic sorption in soils. In this work, a dataset of affinity coefficients (Kf and Kd) of seven SA/TC antibiotics (i.e., sulfachlorpyridazine, sulfamethazine, sulfadiazine, sulfamethoxazole, oxytetracycline, tetracycline, and chlortetracycline) and associated soil properties was generated. Correlation analysis of these data showed that the affinity coefficients of the SAs were predominantly affected by soil organic matter and cation exchange

Monodrug efficacies of sulfonamides in prophylaxis for Pneumocystis carinii pneumonia.

A remarkably high rate of adverse events is associated with the use of trimethoprim-sulfamethoxazole in patients with human immunodeficiency virus type 1 infection. We examined the efficacies of sulfonamides alone in the prevention of Pneumocystis carinii pneumonitis, with the assumption that at least some of the adverse events with the drug combination might be due to trimethoprim. With the immunosuppressed rat model, eight sulfonamides were studied at 100, 10, and 1.0 mg/kg/day (10 rats per dosage and drug). P. carinii infection was prevented in all animals (100%) receiving dosages of as lit

Effects of pine bark amendment on the transport of sulfonamide antibiotics in soils.

In the present work, laboratory column experiments were carried out to study the effect of pine bark amendment (at doses of 0, 12, 48 and 96 Mg ha-1) on the transport of three sulfonamide antibiotics (sulfadiazine -SDZ-, sulfamethazine -SMT-, and sulfachloropyridazine -SCP-) through two crop soils. All three sulfonamides showed high mobility in the unamend soils, with absence of retention in most cases. However, some differences were detected regarding the degree of interactions between sulfonamides and soils, being higher for soil 1, which was attributed to its higher organic carbon content.

Sulfate radical-based oxidation of antibiotics sulfamethazine, sulfapyridine, sulfadiazine, sulfadimethoxine, and sulfachloropyridazine: Formation of SO2 extrusion products and effects of natural organic matter.

The widespread occurrence of sulfonamide antibiotics in the environment has raised great concerns about their potential to proliferate antibacterial resistance. Sulfate radical (SO4•-) based advanced oxidation processes (SR-AOPs) are promising in-situ chemical oxidation (ISCO) technologies for remediation of soil and groundwater contaminated by antibiotics. The present study reported that thermally activated persulfate oxidation of sulfonamides (SAs) bearing six-membered heterocyclic rings, e.g., sulfamethazine (SMZ), sulfapyridine (SPD), sulfadiazine (SDZ), sulfadimethoxine (SDM), and sulfach

Assessing the spread of sulfachloropyridazine in poultry environment and its impact on Escherichia coli resistance.

Sulfachloropyridazine (SCP) is an antimicrobial (AM) commonly used in the poultry industry. This drug is excreted as the original compound, which may accumulate in litter. This work was done to assess whether SCP residues from droppings of broiler chickens that were treated with therapeutic doses of this drug spread into the production environment and to determine if these events were associated with the selection of resistant bacteria. To this end, broiler chickens were raised under controlled conditions, and their droppings and litter were processed to detect and identify SCP residues using

WS2 significantly enhances the degradation of sulfachloropyridazine by Fe(III)/persulfate.

The use of antibiotics has become an indispensable part of the production and life of human society. Among them, sulfonamide antibiotics widely used in humans and animals are considered to be one of the most crucial antibiotics. However, antibiotics are difficult to degrade naturally, leading to an accumulation in the environment and a potential hazard to human health. In this paper, WS2 as a co-catalyst could reduce trace Fe(III) to Fe(II) which exhibited a great activating ability to PS through the exposed W(IV) active sites, and formed the Fe(III)/Fe(II) cycle to degrade sulfachloropyridazi

Broad-Specificity Aptamer of Sulfonamides: Isolation and Its Application in Simultaneous Detection of Multiple Sulfonamides in Fish Sample.

Sulfonamide antibiotics (SAs) are widely used in animal husbandry and aquaculture, and the excess residues of SAs in animal-derived foods will harm the health of consumers. In reality, various SAs were alternately used in animal husbandry and aquaculture, and thus, it is urgent need to develop simple and high-throughput methods for simultaneously detecting multiple SAs or groups of SAs in order to realize rapid screening of total SAs residues in animal-derived foods. We herein isolated a broad-specificity aptamer for SAs by using a multi-SAs systematic evolution of ligands by exponential enric

Quantum chemical study of the photolysis mechanisms of sulfachloropyridazine and the influence of selected divalent metal ions.

Sulfonamides have been found in aquatic environments. Degradation of sulfachloropyridazine (SCP) mainly proceeds through direct and indirect photolysis in the aquatic environment. However, the mechanisms underlying the triplet photolysis of SCP and the influence of metal ions on the photolysis mechanism have not yet been fully explained. In this study, we elucidated the triplet photolysis mechanisms of SCP and the effects of three selected metal ions (Zn(2+), Ca(2+), and Cu(2+)) on the SCP photolysis mechanisms using quantum chemical calculation. Peroxymonosulfate activation using a composite of copper and nickel oxide coated on SBA-15 for the removal of sulfonamide antibiotics.

The sluggish Ni(II)/Ni(III) redox cycle does not benefit perxymonosulfate (PMS) activation for recalcitrant pollutant degradation. To solve this problem, a heterogeneous catalyst, Cu0.2Ni0.8O/SBA-15 (CNS), was constructed to activate PMS for decomposing two sulfonamide antibiotics, sulfachlorpyridazine (SACP) and sulfapyridine (SAP). SACP and SAP were completely degraded over Cu0.2Ni0.8O/SBA-15/PMS (CNSP) after 90 min. O2.- was the dominant active species involved in the degradation of SACP and SAP. Structural analysis and elemental valence state observations indicated that Cu(Ⅰ) provided elec

Residues of selected sulfonamides, non-steroidal anti-inflammatory drugs and analgesics-antipyretics in surface water of the Elbe river basin (Czech Republic).

The occurrence of human as well as veterinary drug residues in surface water is caused by their insufficient removal ability from wastewater. Drug residues disturb the natural balance of water ecosystem, have a negative effect on non-target organisms and pose a significant risk for human health. The main aim of this study was to determine the concentration of residues of eight drugs from the group of sulfonamides (sulfathiazole, sulfadiazine, sulfamethazine, sulfamethoxazole, sulfadimethoxine, sulfadoxine, sulfamerazine, sulfachlorpyridazine), four drugs from the non-steroidal anti-inflammator

Zr-based multivariate metal-organic framework for rapid extraction of sulfonamide antibiotics from water and food samples.

Based on multiple ligands strategy, a series of multivariate metal organic frameworks (MTV-MOFs) named as PCN-224-DCDPSx were prepared using one-pot solvothermal method to extract and remove sulfonamide antibiotics (SAs). The pore structure and adsorption performance can be further regulated by modulating the doping ratios of medium-tetra(4-carboxylphenyl) porphyrin and 4,4'-dicarboxydiphenyl sulfones. The MTV-MOFs of PCN-224-DCDPS1.0 possesses very large specific surface area (1625 m2/g). Using PCN-224-DCDPS1.0 as sorbent, a dispersive solid-phase extraction method was developed to extract an

In vitro and in vivo binding of trimethoprim and sulphachlorpyridazine to equine food and digesta and their stability in caecal contents.

Binding of antibiotics to food has received little attention in equine medicine, although such binding could potentially reduce the bioavailability and clinical efficacy. In the present study, binding of trimethoprim (TMP) and sulphachlorpyridazine (SCP) to hay, grass silage and concentrate was investigated in vitro in buffer at pH 6.8 at different concentrations. The binding of TMP and SCP to caecal contents was also studied. In addition, the degradation of TMP and SCP by the caecal microflora was investigated by incubating sterilized and non-sterilized caecal contents for 3 h at 37 degrees C

An LC-ESI-MS/MS method for residues of fluoroquinolones, sulfonamides, tetracyclines and trimethoprim in feedingstuffs: validation and surveillance.

An increasing concern about food safety has been observed over the years. The presence of drugs residues in food is one of the major subjects of research in food safety. Feedingstuffs can be responsible for carryover into the food chain of residues of several drugs. This paper describes the development, validation and application of a fast and simple method for analysis of 24 antibiotic residues in feedingstuffs for cattle, pigs and poultry. Analytes include compounds from different antimicrobials classes, such as sulfonamides (sulfadiazine, sulfamethazine, sulfamethoxazole, sulfaquinoxaline,

Diagnosis and Management of Diabetes Mellitus in a Bali Mynah (Leucopsar rothschildi).

An 18-year-old female Bali mynah (Leucopsar rothschildi) was presented for polyphagia, weight loss, and incoordination. Diabetes mellitus was diagnosed based on the history and clinical findings, including persistent hyperglycemia with concurrent hypoinsulinemia and glucosuria. A treatment protocol was developed that led to improvement of clinical signs and management of hyperglycemia over several months. Because of the advanced age of the animal, difficulty in maintaining euglycemia, and the stress of handling and treatment, euthanasia was elected 167 days after initial presentation. At postm
