Conundrums of Localized Surface Plasmon Resonance Biosensors.

Localized surface plasmon resonance (LSPR) biosensing offers label-free, real-time detection of biomolecular interactions with high sensitivity and compact instrumentation. However, despite widespread use, several persistent and poorly understood phenomena limit the reproducibility, quantitation, and interpretability of LSPR signals. This perspective examines six recurring "conundrums" in LSPR biosensing, with a focus on their physical origins, experimental manifestations, and current approaches to mitigation. I review puzzling cases where analyte binding induces either red or blueshifts of th

Engineering Antiviral Agents via Surface Plasmon Resonance.

Surface plasmon resonance (SPR) is used to measure hemagglutinin (HA) binding to domain-swapped Cyanovirin-N (CV-N) dimer and to monitor interactions between mannosylated peptides and CV-N's high-affinity binding site. Virus envelope spikes gp120, HA, and Ebola glycoprotein (GP) 1,2 have been reported to bind both high- and low-affinity binding sites on dimeric CVN2. Dimannosylated HA peptide is also bound at the two low-affinity binding sites to an engineered molecule of CVN2, which is bearing a high-affinity site for the respective ligand and mutated to replace a stabilizing disulfide bond i

Localized Surface Plasmon Resonance in Semiconductor Nanocrystals.

Localized surface plasmon resonance (LSPR) in semiconductor nanocrystals (NCs) that results in resonant absorption, scattering, and near field enhancement around the NC can be tuned across a wide optical spectral range from visible to far-infrared by synthetically varying doping level, and post synthetically via chemical oxidation and reduction, photochemical control, and electrochemical control. In this review, we will discuss the fundamental electromagnetic dynamics governing light matter interaction in plasmonic semiconductor NCs and the realization of various distinctive physical propertie

Analysis of Protein Interactions by Surface Plasmon Resonance.

Surface plasmon resonance is an optical technique that is utilized for detecting molecular interactions, such as interactions that occur between proteins or other classes of molecules. Binding of a mobile molecule (analyte) to a molecule immobilized on a thin metal film (ligand) changes the refractive index of the film. The angle of extinction of light that is completely reflected after polarized light impinges upon the film, is altered and monitored as a change in detector position for a dip in reflected intensity (the surface plasmon resonance phenomenon). Because the method strictly detects

Fragment Screening of MIF by Surface Plasmon Resonance.

The cytokine MIF is implicated in several autoimmune and inflammatory conditions and is elevated in several types of cancer, making it a promising therapeutic target. Small-molecule MIF inhibitors provide tools to elucidate MIFs' immune regulatory roles, and their further optimization could lead to potential therapeutics. Surface plasmon resonance (SPR) is a relatively high-throughput and sensitive biophysical technique used in drug discovery for screening small organic compounds (termed fragments) for their interactions with macromolecules. SPR analysis can provide the association and dissoci

Surface Plasmon Resonance Clinical Biosensors for Medical Diagnostics.

The design and application of sensors for monitoring biomolecules in clinical samples is a common goal of the sensing research community. Surface plasmon resonance (SPR) and other plasmonic techniques such as localized surface plasmon resonance (LSPR) and imaging SPR are reaching a maturity level sufficient for their application in monitoring biomolecules in clinical samples. In recent years, the first examples for monitoring antibodies, proteins, enzymes, drugs, small molecules, peptides, and nucleic acids in biofluids collected from patients afflicted with a series of medical conditions (Alz

Electrochemistry on a localized surface plasmon resonance sensor.

The optical signal of a localized surface plasmon resonance (LSPR)-based sensor combined with electrochemistry was investigated. Gold nanoparticles were immobilized on an indium tin oxide (ITO) substrate, which functioned as working electrode. Using cyclic voltammetry synchronized with LSPR sensing, surface reactions on gold were detected both electrically and optically. In the capacitive charging regime, optical signals linear to the applied potential were detected. Gold was found to be dissolved above the oxidation potential and partially redeposited during the reduction, which changed size

Surface-plasmon-resonance-enhanced cavity ring-down detection.

The cavity ring-down technique is used to probe the absolute optical response of the localized surface plasmon resonance (SPR) of a gold nanoparticle distribution to adsorption of trichloroethylene (TCE) and perchloroethylene (PCE) from the gas phase. Extended Mie theory for a coated sphere with a particle-size-dependent dielectric function is used to elucidate size-dispersion effects, the size-dependence of the SPR sensitivity to adsorption, and the kinetics of adsorption. An approximate Gaussian distribution of nanospheres with a mean diameter of 4.5 nm and a standard deviation of 1.1 nm, as

Determination of a localized surface plasmon resonance mode of Cu7S4 nanodisks by plasmon coupling.

Plasmon properties such as peak position, extinction cross-section and local electric field intensity are strongly dependent on excited, localized surface plasmon resonance (LSPR) modes. In non-spherical copper chalcogenide nanoparticles, assignment of the LSPR peaks to the corresponding oscillation modes has been controversial and requires experimental verification. We determined the in-plane LSPR mode of roxbyite Cu7S4 nanodisks from the plasmon coupling effect of nanodisks in solution. Compared with individual Cu7S4 nanodisks, self-assembled Cu7S4 nanodisk arrays in chloroform exhibited a b

Surface plasmon resonance aptasensor for Brucella detection in milk.

A Surface Plasmon Resonance (SPR) aptasensor was developed for the detection of Brucella melitensis (B. melitensis) in milk samples. Brucellosis is a bacterial zoonotic disease with global distribution caused mostly by contaminated milk or their products. Aptamers recognizing B. melitensis were selected following a whole bacteria-SELEX procedure. Two aptamers were chosen for high affinity and high specificity. The high affinity aptamer (B70 aptamer) was immobilized on the surface of magnetic silica core-shell nanoparticles for initial purification of the target bacteria cells from milk matrix.

Surface plasmon resonance-based immunoassay for human fetuin A.

This article describes a highly-sensitive surface plasmon resonance (SPR)-based immunoassay (IA) for human fetuin A (HFA), a specific biomarker for atherosclerosis and hepatocellular carcinoma. The assay is based on a novel immobilization procedure that simply involves the dilution of an anti-HFA capture antibody (Ab) in 1% (v/v) 3-aminopropyltriethoxysilane (APTES), followed by its dispensing on a KOH-treated gold (Au)-coated SPR chip and incubation for 30 min. The developed SPR IA detected 0.3-20 ng mL(-1) of HFA with a limit of detection and sensitivity of 0.7 ng mL(-1) and 1 ng mL(-1), res

Serodiagnosis of Lyme borreliosis infection using surface plasmon resonance.

BACKGROUND: Label-free biosensors are ideally suited for the direct monitoring of binding events without the need for additional labeling substances; however, their application in the field of serodiagnosis is not trivial. The major problem is the unspecific adsorption of blood serum components to the sensor surface. METHODS: A surface plasmon resonance (SPR) sensor has been used for the direct detection of Lyme borreliosis specific antibodies in blood serum. The combination of an optimal dilution factor with the addition of suitable detergents minimizes the unspecific adsorption. Serum sampl

Characterization of polysaccharide conformational epitopes by surface plasmon resonance.

SPR techniques can provide a wealth of insight into the nature of protein-carbohydrate interactions. Information not obtained readily by other methodologies can be gathered relatively quickly in a label-free manner with low sample consumption. This chapter focused on two applications in which SPR has been used to map conformational epitopes on bacterial polysaccharides recognized by protective antibodies. In one example, methods for demonstrating the conformational nature of the epitope recognized by an anti-GBS antibody were described. Dramatic epitopic stabilization at 2 repeating units with

Super-resolution imaging using surface plasmon resonance cavity lithography.

Surface plasmonic lithography (SPL) utilizes the lateral propagation of light on the surface of metals, which generates transverse excitation, resulting in evanescent waves that participate in imaging to break the diffraction limit. However, a challenge is the difficulty in achieving stable evanescent wave imaging. This paper proposes a surface plasmon resonance cavity lithography (SPRCL) technique, which achieves super-resolution imaging and explores the effects of various factors on this technique. The general dispersion relation for surface plasmon polaritons in a double-resonator cavity st

Portable Surface Plasmon Resonance Detector for COVID-19 Infection.

Methods based on nucleic acid detection are currently the most commonly used technique in COVID-19 diagnostics. Although generally considered adequate, these methods are characterised by quite a long time-to-result and the necessity to prepare the material taken from the examined person-RNA isolation. For this reason, new detection methods are being sought, especially those characterised by the high speed of the analysis process from the moment of sampling to the result. Currently, serological methods of detecting antibodies against the virus in the patient's blood plasma have attracted much a

Surface Plasmon Resonance Analysis of PfEMP1 Interaction with Receptors.

A detailed understanding of the interaction between the highly variant Plasmodium falciparum erythrocyte membrane proteins 1 (PfEMP1) and their human binding partners is essential to explain their roles in disease development in malaria, as well as to understand how antibodies can inhibit these interactions and how the parasite manages to evade such an immune response. This chapter focuses on using surface plasmon resonance (SPR) as a reproducible, high-throughput method to quantitatively characterize these interactions. We describe how to utilize protein A or A/G and streptavidin for protein

Surface Plasmon Resonance Imaging Detection of Sub-femtomolar MicroRNA.

MicroRNA (miRNA) is a promising new type of biomarkers but at a low fM level and hard to be analyzed. Herein proposed is an innovated surface plasmon resonance imaging (SPRi) method merged with a novel in-plane and vertical signal amplification strategy, that is, orthogonal signal amplification to enable a direct determination of sub-fM miRNA-15a (a multiple tumor diagnostic biomarker). The core idea is to add more mass on a target sample spot first along the surficial direction, then upward from the surface. In detection of miRNA, this was realized by coupling a miRNA-initiated surficial cycl

Surface plasmon resonance of two-segmented Au-Cu nanorods.

Two-segmented gold-copper nanorods were electrodeposited inside the pores of polycarbonate track-etched membranes from two separate solutions. The PCT membranes were dissolved in dichloromethane (CH(2)Cl(2)) and the solvent was replaced by methanol solution. Optical absorption spectra of two-segmented Au-Cu nanorods dispersed in methanol showed two peaks which were related to the transverse mode of copper and the longitudinal mode of gold. By increasing the length of the gold segment, when the total length of both metals was fixed at 1 µm, the copper and gold peaks shifted to the blue and red
