General pathology of experimental swine vesicular disease.

Twenty-four minimal disease pigs were inoculated intracerebrally, intravenously or intradermally with an English strain of swine vesicular disease virus. In the skin, snout, tongue and tonsil the main lesion was a full-thickness coagulative necrosis of the stratified squamous epithelium. In the renal pelvis, bladder, tonsillar crypts and the collecting ducts of salivary glands and pancreas, epithelial degeneration with the formation of periodic acid-Schiff-positive material were consistent features of this disease. Histopathological examination alone could not be relied upon to differentiate b

Experimental swine vesicular disease, pathology and immunofluorescence studies.

Two day old piglets were inoculated intravenously with 1 ml of swine vesicular disease virus UK-G 27-72 isolate. Using infectivity tests, immunofluorescent staining and gross and histopathological examination, pathogenesis of the infection was studied in tissue specimens collected daily from one through seven days postinoculation. Swine vesicular disease virus had a strong affinity for the epithelia of the tongue, snout, coronary band and lips, the myocardium and the lymphoid elements of the tonsil and the brain stem. The virus had the greatest affinity for the epithelium of the tongue. Howeve

High dose of vesicular stomatitis virus-vectored Ebola virus vaccine causes vesicular disease in swine without horizontal transmission.

ABSTRACTThe recent impact of Ebola virus disease (EVD) on public health in Africa clearly demonstrates the need for a safe and efficacious vaccine to control outbreaks and mitigate its threat to global health. ERVEBO® is an effective recombinant Vesicular Stomatitis Virus (VSV)-vectored Ebola virus vaccine (VSV-EBOV) that was approved by the FDA and EMA in late 2019 for use in prevention of EVD. Since the parental virus VSV, which was used to construct VSV-EBOV, is pathogenic for livestock and the vaccine virus may be shed at low levels by vaccinated humans, widespread deployment of the vaccin

Crystal structure of Swine vesicular disease virus and implications for host adaptation.

Swine vesicular disease virus (SVDV) is an Enterovirus of the family Picornaviridae that causes symptoms indistinguishable from those of foot-and-mouth disease virus. Phylogenetic studies suggest that it is a recently evolved genetic sublineage of the important human pathogen coxsackievirus B5 (CBV5), and in agreement with this, it has been shown to utilize the coxsackie and adenovirus receptor (CAR) for cell entry. The 3.0-A crystal structure of strain UK/27/72 SVDV (highly virulent) reveals the expected similarity in core structure to those of other picornaviruses, showing most similarity to

Identification of neutralizing epitopes on a European strain of swine vesicular disease virus.

Six neutralizing monoclonal antibodies (MAbs) were used to isolate MAb neutralization-resistant (MAR) mutants from a recent European strain of swine vesicular disease virus (SVDV), ITL/9/93. Sequencing of MAR mutants identified two epitopes located at positions analogous to sites 2A (VP2) and 3B (VP3) on poliovirus (PV) which have been previously identified on a Japanese strain of SVDV. A third epitope near to the C terminus of VP1, not previously recognized on SVDV, was tentatively identified in a region analogous to site 1 of PV. A fourth epitope, located in the C-terminal region of VP3, has

Effects of quaternary ammonium compounds with 0.1% sodium hydroxide on swine vesicular disease virus.

The effects of quaternary ammonium compounds (QACs) with sodium hydroxide on swine vesicular disease virus (SVDV), an enterovirus were studied. Didecyldimethylammonium chloride (DDAC) with 0.1% NaOH showed a stronger effect against SVDV than other QACs with 0.1% NaOH. The effect of DDAC with 0.1% NaOH was strong at 40 degrees C. DDAC was effective against SVDV at pH values around 11.0, but not in the distilled water control. The effect of DDAC with 0.1% NaOH was already observed at 1 min after mixing of the DDAC with SVDV. Observation under an electron microscopy revealed that the probable mec

Swine vesicular disease: virological studies of experimental infections produced by the England-72 virus.

Pigs exposed to relatively small amounts of virus by intradermal inoculation of the feet or by skin sacrification developed clinical disease. Large amounts of virus were recovered from samples taken from the nose, mouth, pharynx, rectum and the prepuce or vagina during the first week of infection and smaller amounts during the second week. Virus was recovered from the faeces of most animals 16 days after infection and from one animal for 23 days. Pigs in contact with inoculated animals were killed at intervals before the appearance of clinical disease. The distribution and amounts of virus in

[Laboratory diagnosis of foot-and-mouth disease and swine vesicular disease in the years 1962-1988 in Greece].

During 26 years (1962-1988) 499 samples from FMD-suspicious cases were examined in Greece. These materials came from 348 (70%) cattle, 95 (19%) pigs and 56 (11%) sheep and goats. The cattle with 197 (72.4%) positive cases seems to play the most important roll in FMD. The different isolated virus types belonged in 60 cases to type A, in 187 to type O, in 14 to type C, in 6 to type SAT 1 and in 2 cases to type ASIA 1, respectively. SVD was isolated in 3 cases from the same area and at the same time. Most samples have been examined by means of CF, cell culture, unweaned mice or by a combined way

Swine vesicular disease viruses isolated from healthy pigs in non-epizootic period. II. Vesicular formation and virus multiplication in experimentally inoculated pigs.

An infection experiment was carried out on pigs with swine vesicular disease virus isolated from healthy pigs (SVDV-H). Inoculation was done by two routes, intradermal in the coronary band of the foot and oral. Observation was made on the formation of vesicles and their spread, the virus contents of serum, swab of the oral cavity, and feces, the vicissitude of neutralizing antibody titers, and the distribution of virus in the body. From its results the pathogenicity of virus was judged. In the pigs inoculated intradermally there was a difference in the extension of the area involved in vesicul

Persistent infection is a rare sequel following infection of pigs with swine vesicular disease virus.

Nine isolates from pigs persistently infected with a recent Italian isolate of swine vesicular disease (SVD) virus, ITL/9/93, were collected sequentially over 121 days and were characterized antigenically and biochemically. There was an accumulation of amino acid (aa) substitutions in the capsid proteins throughout the carrier state that could be correlated with alterations in antigenicity in virus isolates collected late stage in infection. The aa substitutions detected mainly occurred in VPI and antigenic changes were detected in late isolates both at antigenic site 1, resulting in loss of b

[Serological examinations for swine vesicular disease (SVD) in a closed pig breeding herd using ELISA].

In a closed pig establishment housing about 18,000 pigs, 2895 gilts were tested pre-export for SVD (swine vesicular disease) antibodies using Ceditest/PrioCHECK SVDV-AB ELISA. 130 gilts (4.5%) tested positive. In addition, 561 animals of this farm were sampled per random for SVD serology. One in 241 weaners (0.4%), eight in 150 gilts (5.3%) and 18 in 170 (10.6%) pregnant sows tested ELISA SVD-antibody positive. Of the ELISA positive samples, 23 tested positive in VNT (virus neutralization test). Of these, 20 VNT-positive animals were re-sampled two weeks later and re-tested via ELISA and VNT i

Viral meningitis epidemics and a single, recent, recombinant and anthroponotic origin of swine vesicular disease virus.

BACKGROUND AND OBJECTIVES: Swine vesicular disease virus (SVDV) is a close relative of the human Enterovirus B serotype, coxsackievirus B5. As the etiological agent of a significant emergent veterinary disease, several studies have attempted to explain its origin. However, several key questions remain, including the full biological ancestry of the virus, and its geographical and temporal origin. METHODOLOGY: We sequenced near-complete genomes of 27 SVDV and 13 coxsackievirus B5 samples, all originally isolated between 1966 and 2006, and analysed these in conjunction with existing sequences an

Complete nucleotide sequence of a coxsackie B5 virus and its relationship to swine vesicular disease virus.

We report the first complete nucleotide sequence of the picornavirus coxsackievirus B5 (CB5), strain 1954/UK/85, an isolate from a case of hand-foot-and-mouth disease. We have compared the sequence with those of other coxsackie B viruses, coxsackievirus A9, poliovirus and swine vesicular disease virus (SVDV). The genes encoding the three major capsid proteins are most closely related to those of SVDV but the 5' and 3' noncoding regions and the P3 gene are more similar to the corresponding regions in the other coxsackie B viruses than to those of SVDV. These observations are considered in the l

Serological and Molecular Detection of Senecavirus A Associated with an Outbreak of Swine Idiopathic Vesicular Disease and Neonatal Mortality.

We performed a longitudinal field study in a swine breeding herd that presented with an outbreak of vesicular disease (VD) that was associated with an increase in neonatal mortality. Initially, a USDA Foreign Animal Disease (FAD) investigation confirmed the presence of Senecavirus A (SVA) and ruled out the presence of exotic agents that produce vesicular lesions, e.g., foot-and-mouth disease virus and others. Subsequently, serum samples, tonsil swabs, and feces were collected from sows (n = 22) and their piglets (n = 33) beginning 1 week after the onset of the clinical outbreak and weekly for

Brain and spinal cord lesions in pigs inoculated with swine vesicular disease (UKG strain) virus and coxsackievirus B5.

Pigs inoculated intravenously with swine vesicular disease virus (UKG strain), those inoculated with coxsackievirus B5, and other pigs exposed by pen contact to the same viruses developed diffuse encephalomyelitis. Perivascular cuffing, with lymphocytes and formation of neuroglia cell foci, were most prominent in telencephalon, diencephalon, and mesencephalon. Encephalitis was of mild to severe intensity. Severity of lesions was more extensive and severe in the pigs exposed to swine vesicular disease virus. Pen contact exposure to either of the 2 viruses caused a more severe central nervous sy

Emergence of Two Porcine Variants of Human Coxsackievirus B5 and B4 in the 20th Century That Caused Swine Vesicular Disease: A Retrospective Review.

In this review, we examine the occurrence of two independent, single recombination events which occurred between human enteroviruses (Picornaviridae, Enterovirus, Enterovirus betacoxsackie). These recombination events contributed to the emergence of two viruses which adapted to pigs. These viruses have caused epizootics of swine vesicular disease (SVD) for many years. As was shown previously, the classical SVD virus (SVDV-1) originated from human coxsackievirus B5. The strain T75 (SVDV-2) emerged from human coxsackievirus B4 in the Tambov region of Russia, where it circulated from 1975 to 1977

The N-terminal region of the VP1 protein of swine vesicular disease virus contains a neutralization site that arises upon cell attachment and is involved in viral entry.

The N-terminal region of VP1 of swine vesicular disease virus (SVDV) is highly antigenic in swine, despite its internal location in the capsid. Here we show that antibodies to this region can block infection and that allowing the virus to attach to cells increases this blockage significantly. The results indicate that upon binding to the cell, SVDV capsid undergoes a conformational change that is temperature independent and that exposes the N terminus of VP1. This process makes this region accessible to antibodies which block virus entry.

First report and genetic characterization of Seneca Valley virus (SVV) in Chile.

Seneca Valley virus (SVV) is a non-enveloped RNA virus and the only member of the Senecavirus A (SVA) species, in the Senecavirus genus, Picornaviridae family. SVV infection causes vesicular lesions in the oral cavity, snout and hooves of pigs. This infection is clinically indistinguishable from trade-restrictions-related diseases such as foot-and-mouth disease. Other clinical manifestations include diarrhoea, anorexia, lethargy, neurological signs and mortality in piglets during their first week of age. Before this study, Chile was considered free of vesicular diseases of swine, including SVV
