Three decades of analyzing Wolbachia bacterial endosymbionts in arthropods: Trends and gaps.

Wolbachia are the most pervasive bacterial endosymbionts yet described, infecting half of all arthropod species. These bacteria trigger outstanding phenotypes in their hosts including cytoplasmic incompatibility (CI), which has also been used as a mechanism to control pest species. Here, we analyzed peer-reviewed articles published in the 30 years from 1995 to 2024. Our results show that most studies continue to use traditional methods such as PCR and Sanger sequencing, approaches that remain sufficient and appropriate in many cases, particularly when working with well-characterized strains or

Wolbachia-induced proteostasis remodeling in ovaries supports cytoplasmic incompatibility rescue in Drosophila.

The endosymbiont Wolbachia can modulate host reproduction to promote its own spread through host populations. Cytoplasmic incompatibility (CI) is a prevalent reproductive strategy induced by Wolbachia in insects, wherein embryos from infected males and uninfected females cannot develop normally; however, infected females rescue this embryonic lethality, thereby favoring its maternal transmission. The ovarian mechanisms that facilitate Wolbachia transmission and enable CI rescue remain largely unclear. Here, we integrated ovarian proteomics and ubiquitin-proteomics, and compared these datasets

The symbiotic Wolbachia in Anopheles and its role in reducing the transmission of Plasmodium: updates and prospects.

Reducing malaria transmission and the prospects for vector control include multi-pronged strategies, such as interrupting the parasite cycle in both vectors and mosquitoes. Effective vector control remains essential to prevent malaria transmission. This is all the more important as problems such as resistance to insecticides and the lack of a highly effective malaria vaccine remain. New generation vector control measures and optimised products are essential to address the public health needs for malaria eradication. Strategies to reduce malaria transmission include the use of insecticide-treat

Effects of different X-ray irradiation dosages on Wolbachia trans-infected and uninfected Aedes aegypti mosquitoes.

BACKGROUND: Aedes aegypti is a primary vector of several arboviral diseases, including dengue, chikungunya, Zika, and yellow fever. Eliminating Ae. aegypti can disrupt virus transmission cycles, thereby reducing the incidence of these diseases. Releasing sterile Ae. aegypti males has been implemented to suppress their natural populations, using three approaches, i.e., ionizing radiation-induced sterilization (Sterile Insect Technique, SIT), Wolbachia-induced cytoplasmic incompatibility (Incompatible Insect Technique, IIT), and a combination of both approaches (SIT/IIT). METHODOLOGY/PRINCIPAL

Analysis of the genomes of a male-killing Spiroplasma and its co-infecting Rickettsia reveals a case of concerted genome expansion.

Maternally inherited symbionts are central to arthropod biology, functioning both as mutualistic partners and as reproductive parasites. Genomic analyses provide critical insight into these interactions. Here, we sequenced, assembled and examined the genomes of Spiroplasma and Rickettsia co-infecting the lacewing Mallada desjardinsi, with the aim of elucidating the male-killing phenotype of Spiroplasma and predicting potential phenotypes for Rickettsia. In Spiroplasma, we identified a set of candidate effector genes. However, Spaid-like genes, where present, lacked the functional domains previ

Establishment and characterization of a Wolbachia (wAlbB)-infected Aedes aegypti line (Tw-Kao) for dengue control.

Global trade and climate change are driving the geographic expansion of dengue vectors, contributing to the global spread of dengue. Conventional vector control measures have proven insufficient to prevent substantial disease burdens, highlighting the need for innovative and sustainable strategies. The release of Wolbachia-infected mosquitoes offers a promising alternative for dengue suppression. Here, we developed a locally derived Ae. aegypti line carrying the wAlbB strain (wAlbB-Tw-Kao) and systematically evaluated its fitness, viral interference, and potential for vector population control

Tsetse-Wolbachia symbiosis: comes of age and has great potential for pest and disease control.

Tsetse flies (Diptera: Glossinidae) are the sole vectors of African trypanosomes, the causative agent of sleeping sickness in human and nagana in animals. Like most eukaryotic organisms, Glossina species have established symbiotic associations with bacteria. Three main symbiotic bacteria have been found in tsetse flies: Wigglesworthia glossinidia, an obligate symbiotic bacterium, the secondary endosymbiont Sodalis glossinidius and the reproductive symbiont Wolbachia pipientis. In the present review, we discuss recent studies on the detection and characterization of Wolbachia infections in Glos

Mathematical modelling of Wolbachia replacement in Aedes aegypti for dengue control: a scoping review.

Wolbachia replacement in dengue virus vectors is a promising tool for controlling transmission, and models can explore its potential in novel and complex scenarios. We analyse how research questions in Wolbachia replacement modelling have developed over time and in response to empirical findings. A scoping review was conducted of Wolbachia replacement models. For each study, we extracted research questions, key findings and modelling methodology and thematically categorized research questions. From 726 search results, 115 studies met inclusion criteria. Four themes were identified: identifying

Symbiosis Comes of age at the 10th Biennial Meeting of Wolbachia Researchers.

Wolbachia pipientis is an alpha-proteobacterial, obligate intracellular microbe and arguably the most successful infection on our planet, colonizing 40-60% of insect species. Wolbachia are also present in most, but not all, filarial nematodes where they are obligate mutualists and are the targets for anti-filarial drug discovery. Although Wolbachia are related to important human pathogens they do not infect mammals, but instead are well known for their reproductive manipulations of insect populations, inducing the following phenotypes: male-killing, feminization, parthenogenesis induction, or

Wolbachia variability and host effects on crossing type in Culex mosquitoes.

Wolbachia is a common maternally inherited bacterial symbiont able to induce crossing sterilities known as cytoplasmic incompatibility (CI) in insects. Wolbachia-modified sperm are unable to complete fertilization of uninfected ova, but a rescue function allows infected eggs to develop normally. By providing a reproductive advantage to infected females, Wolbachia can rapidly invade uninfected populations, and this could provide a mechanism for driving transgenes through pest populations. CI can also occur between Wolbachia-infected populations and is usually associated with the presence of dif

Impulsive Release Strategies for Wolbachia-Infected Mosquitoes Under Temperature-Induced Infection Loss.

The release of Wolbachia-infected mosquitoes is a promising strategy for controlling Aedes aegypti populations, although high-temperature exposure may temporarily reduce infection levels and compromise long-term persistence. We propose a population-dynamics model based on impulsive differential equations that incorporates cytoplasmic incompatibility, periodic mosquito releases, and temperature-driven infection loss. Analytical results establish fundamental qualitative properties of the continuous and impulsive models, derive conditions for the existence and stability of periodic solutions, and

Optimal control approach to developing sex-structured release strategies for dengue prevention.

Dengue remains a public health challenge due to the persistence of Aedes aegypti populations and the limited effectiveness of medical and vector control interventions. The release of Wolbachia-infected mosquitoes has been used in multiple regions; yet, release protocols are often heuristically structured, with limited optimization. In particular, the impacts of mosquito sex structure and strain-specific effects remain unclear. This study develops a sex-structured dynamical model that links within-vector Wolbachia dynamics, such as cytoplasmic incompatibility and vertical transmission, with bet

Wolbachia-induced cytoplasmic incompatibility triggers intergenerational dysregulation of the small RNA regulatory network in offspring.

The intracellular symbiont Wolbachia, which is widespread among insects, may induce cytoplasmic incompatibility (CI) between hosts with different infection statuses. Increasing evidence indicates that symbiotic bacteria can influence host reproduction, metabolism, and other biological processes by modulating non-coding small RNAs. However, it is still unclear how Wolbachia-induced CI affects the offspring reproduction. In this study, using Drosophila melanogaster as a model system, small RNA and transcriptome sequencing were conducted on the reproductive systems of the offspring resulting from

Wolbachia-mediated reproductive manipulation in rice planthoppers.

Rice planthoppers, including brown (Nilaparvata lugens), small brown (Laodelphax striatellus), and white-backed (Sogatella furcifera) planthoppers, are major agricultural pests in China and severely affect rice production and food security. The endosymbiotic bacterium Wolbachia is commonly found in these insects, where it regulates reproduction through mechanisms such as cytoplasmic incompatibility (CI) and increased fertility. In this review, we discuss the strain-specific effects of Wolbachia: wLug (in N. lugens, < 50% infection) increases fecundity without CI; wStri (in L. striatellus, 99%

Male development time influences the strength of Wolbachia-induced cytoplasmic incompatibility expression in Drosophila melanogaster.

Cytoplasmic incompatibility (CI) is the most widespread reproductive modification induced in insects by the maternally inherited intracellular bacteria, Wolbachia. Expression of CI in Drosophila melanogaster is quite variable. Published papers typically show that CI expression is weak and often varies between different Drosophila lines and different labs reporting the results. The basis for this variability is not well understood but is often considered to be due to unspecified host genotype interactions with Wolbachia. Here, we show that male development time can greatly influence CI expressi

On the Spread of Microbes That Manipulate Reproduction in Marine Invertebrates.

AbstractBacterial symbionts are functionally integral to animal reproduction and development, some of which have evolved additional mechanisms to override these host programs. One habitat that is increasingly recognized to contain phylogenetically related lineages of reproductive manipulators is the ocean. The reproduction of marine invertebrates often occurs by free spawning instead of by the physical contact of copulation in terrestrial systems. We developed an integrated model to understand whether and when microbes that manipulate host reproduction by cytoplasmic incompatibility, feminizat

Spiroplasma in Glossina fuscipes fuscipes: a potential manipulator of male reproduction.

Glossina fuscipes fuscipes (Gff) is a riverine species of the Glossina genus (tsetse flies). It is one of the principal vector species responsible for transmitting human African trypanosomiasis across a broad swath of sub-Saharan Africa. Consequently, it remains a primary target for vector-control interventions in endemic regions. As with other Glossina species, G. fuscipes houses a highly streamlined microbiota, composed of maternally transmitted endosymbionts mandatory for survival, which also significantly impacts the vectorial capacity. Among them, the obligate Wigglesworthia provides esse

Wolbachia induce cytoplasmic incompatibility and affect mate preference in Habrobracon hebetor to increase the chance of its transmission to the next generation.

Wolbachia are common intracellular bacteria that are generally found in arthropods, including a high proportion of insects and also some nematodes. This intracellular symbiont can affect sex ratio with a variety of reproductive anomalies in the host, including cytoplasmic incompatibility (CI) in haplodiploids. In this study, we questioned if the parasitoid wasp, Habrobracon hebetor (Hym.: Braconidae), an important biological control agent of many lepidopteran larvae, is infected with Wolbachia. To test this, DNA was extracted from adult insects and subjected to PCR using specific primers to Wo

Genomic stability and differential expression of type V cif genes in Wolbachia with variable cytoplasmic incompatibility phenotypes in spider mites.

The two-spotted spider mite, Tetranychus urticae, is a globally significant agricultural pest frequently infected by the endosymbiont Wolbachia. Wolbachia induces cytoplasmic incompatibility (CI) with varying intensity across geographic populations. Despite the importance of CI, its molecular mechanisms in T. urticae remain poorly understood. Here, we demonstrate that CI differences are driven by both Wolbachia (wTurt) strain variations and host nuclear background. The wTurt genome harbors two pairs of cif genes (cifA1-B1 and cifA2-B2), both phylogenetically classified as Type V. Genomic analy
