Schwann cell precursors generate sympathoadrenal system during zebrafish development.

The autonomic portion of the peripheral nervous system orchestrates tissue homeostasis through direct innervation of internal organs, and via release of adrenalin and noradrenalin into the blood flow. The developmental mechanisms behind the formation of autonomic neurons and chromaffin cells are not fully understood. Using genetic tracing, we discovered that a significant proportion of sympathetic neurons in zebrafish originates from Schwann cell precursors (SCPs) during a defined period of embryonic development. Moreover, SCPs give rise to the main portion of the chromaffin cells, as well as

WNT signaling, the development of the sympathoadrenal-paraganglionic system and neuroblastoma.

Neuroblastoma (NB) is a tumor of the sympathoadrenal system arising in children under 15 years of age. In Germany, NB accounts for 7% of childhood cancer cases, but 11% of cancer deaths. It originates from highly migratory progenitor cells that leave the dorsal neural tube and contribute neurons and glial cells to sympathetic ganglia, and chromaffin and supportive cells to the adrenal medulla and paraganglia. Clinically, histologically and molecularly, NBs present as extremely heterogeneous, ranging from very good to very poor prognosis. The etiology of NB still remains unclear and needs to be

Abnormal sympathoadrenal development and systemic hypotension in PHD3-/- mice.

Cell culture studies have implicated the oxygen-sensitive hypoxia-inducible factor (HIF) prolyl hydroxylase PHD3 in the regulation of neuronal apoptosis. To better understand this function in vivo, we have created PHD3(-/-) mice and analyzed the neuronal phenotype. Reduced apoptosis in superior cervical ganglion (SCG) neurons cultured from PHD3(-/-) mice is associated with an increase in the number of cells in the SCG, as well as in the adrenal medulla and carotid body. Genetic analysis by intercrossing PHD3(-/-) mice with HIF-1a(+/-) and HIF-2a(+/-) mice demonstrated an interaction with HIF-2

Pressor effect of nalbuphine in hemorrhagic shock is dependent on the sympathoadrenal system.

Nalbuphine, an analgesic with opiate agonist and antagonist properties, increases heart rate, mean arterial pressure (MAP), and cardiac function following a moderate hemorrhage in the rat. The interaction between opiate receptors and the sympathoadrenal system was examined in control, beta receptor-blocked, and adrenal-demedullated animals. MAP and heart rate were measured continuously in chronically instrumented conscious animals. Hemorrhage decreased MAP in the control group by 50%, and nalbuphine (1 mg/kg) returned this to prehemorrhage values within 5 min. MAP remained significantly greate

Influences of thyroid status and sympathoadrenal system on extrarenal potassium disposal.

Effects of hyper- and hypothyroidism on the ability of rats to transfer acute intravenous loads of potassium from the extracellular to the intracellular milieu (extrarenal potassium disposal, ERPD) were studied. We also examined the effects of the sympathoadrenal system on ERPD, as well as the manner in which it interacts with thyroid status. Experiments were performed in thyroidectomized (hypothyroid), sham-operated (euthyroid), or 3,5,3'-triiodo-L-thyronine-treated (thyrotoxic) rats. In anesthetized, acutely nephrectomized animals given a constant infusion of KCl over a 90-min period, ERPD w

Abnormal regulation of the sympathoadrenal system in deoxycorticosterone acetate-salt hypertensive rats.

With the use of circulating norepinephrine (NE) and epinephrine (E) levels, the sympathoadrenal activity as well as its local modulation by adrenoceptors were studied in normotensive (NT) and DOCA-salt hypertensive (HT) rats. In anesthetized hypertensive rats, plasma NE levels were higher, whereas in conscious animals both NE and E levels were found to be increased, suggesting an increased basal sympathoadrenal tone in these animals. The finding of a close correlation between blood pressure levels and NE levels suggests that the elevation of blood pressure may be linked to sympathetic system a

Role of the central nervous system in hemodynamic and sympathoadrenal responses to cocaine in rats.

These studies were undertaken to examine the contribution of central nervous system mechanisms to the cardiovascular and sympathoadrenal effects of cocaine. Changes in systolic and diastolic blood pressure, heart rate and plasma catecholamine concentrations were determined in response to cocaine injected i.a. or i.c.v. in conscious unrestrained rats. Systemically administered cocaine produced brisk, transient dose-related increases in systolic and diastolic pressure at doses of 0.05 to 5 mg/kg i.a. Plasma catecholamine concentrations increased in a dose-related manner, reaching peak levels at

[The function of the cholinergic and sympathoadrenal systems in miners with long-term exposure to manganese oxides].

Long-term exposure to manganese oxides (MO) exceeding MPL leads at first to adaptation strain such as an increase in the activity of acetyl chlonesterase and correspondingly a decline in acetylcholine; greater excretion of catecholamin precursors with subsequent depletion of adaptive mechanisms. Under exposure to manganese concentration of 5.9 to 16.3 mg/m3, the adaptation exhaustion reactions in the sympathoadrenal system (SAS) and the cholinergic system (ChS) develop 7 to 9 years earlier than with 0.4-2.8 mg/m3 of MO. Moreover, ChS responds sooner than SAS to manganese exposure--about ten ye

[Reaction of the sympathoadrenal system in young children with congenital heart defects during cardiopulmonary bypass surgery].

Reactions of the sympathoadrenal system were studied during surgical interventions in children aged 1.5-3 years with congenital heart disease operated under conditions of artificial circulation and hypothermia. The state of the sympathoadrenal system was different in uneventful and complicated course of the operation, which may indicate a different efficiency of the neurovegetative blocking and antistress protection from surgical trauma.

Slow but Steady-The Responsiveness of Sympathoadrenal System to a Hypoglycemic Challenge in Ketogenic Diet-Fed Rats.

The sympathoadrenal counterregulatory response to hypoglycemia is critical for individuals with type 1 diabetes due to impaired ability to produce glucagon. Ketogenic diets (KD) are an increasingly popular diabetes management tool; however, the effects of KD on the sympathoadrenal response are largely unknown. Here, we determined the effects of KD-induced ketosis on the sympathoadrenal response to a single insulin-induced hypoglycemic challenge. We investigated how a 3 week KD feeding regimen affected the main components of the sympathoadrenal counterregulatory response: adrenal sympathetic ne

[The effect of neurohumoral factors of the sympathoadrenal system on the results of cataract extraction with intraocular lens implantation in diabetic patients].

Effect of the sympathoadrenal system (SAS) on the results of cataract extraction with IOL implantation was studied by analyzing the stress index in 64 patients with diabetes mellitus and 20 reference patients. SAS reaction to surgical stress was abnormal in 34 (53.1%) diabetics. The level of SAS activity, postoperative complications, and kinetics of regenerative repair processes in the cornea were related. In 15 (23.%) diabetics with a low stress index the postoperative period was characterized by frequent inflammations, longer repair of normal corneal homeostasis, and coarse cicatrization. In

Cocaine: cardiovascular effects in relation to inhibition of peripheral neuronal monoamine uptake and central stimulation of the sympathoadrenal system.

Cocaine (0.03-5.6 mg/kg i.v.) produced a dose-dependent and prolonged increase in mean arterial blood pressure and heart rate in conscious rats. The 0.3 and 1 mg/kg doses of cocaine potentiated the pressor response to exogenous norepinephrine (0.2 microgram/kg), whereas lower (0.03 and 0.1 mg/kg) and higher (3 and 5.6 mg/kg) doses were ineffective. Desipramine (0.03-1 mg/kg), a prototype norepinephrine uptake blocker, did not alter blood pressure or heart rate. Nisoxetine (0.01-1 mg/kg), another norepinephrine selective uptake blocker, produced a small and brief (< 5 min) pressor response, but

Dopamine: an important neurohormone of the sympathoadrenal system. Significance of increased peripheral dopamine release for the human stress response and hypertension.

In human plasma, the concentration of free dopamine (DA) is approximately equal to that of epinephrine (E). Like norepinephrine (NE) and E its concentration may increase during physical effort and stress (Fig. 1). Most of the stress-related increase in plasma DA is derived from peripheral noradrenergic nerve terminals and the adrenal medulla. Because virtually all of the DA is rapidly conjugated upon release, it is necessary to measure plasma total (free + conjugated) DA. When dietary sources are controlled, the total DA concentration can be used as an indicator of the intensity of the sympath

Role of catecholamine-induced activation of vagal afferent pathways in regulation of sympathoadrenal system activity: negative feedback loop of stress response.

Stress-induced activation of the hypothalamic-pituitary-adrenal axis and sympathoadrenal system is precisely regulated by well-documented negative feedback mechanisms. These include direct negative feedback effect of glucocorticoids on brain structures regulating the hypothalamic-pituitary-adrenal axis activity. However, since the blood-brain-barrier is impermeable to circulating catecholamines, the role of circulating epinephrine and norepinephrine in feedback regulation of the sympathoadrenal system activity is unclear. Here we show that vagal innervation of the adrenal medulla combined with

The Carotid Body as Part of a Unified Sympathoadrenal System of Neural Crest Derivatives: Insights from Two Centuries of Research.

The carotid body-a chemoreceptive derivative of the neural crest located at the bifurcation of the carotid artery-has been studied for over 282 years. The history of research into this small but vital organ is full of unexpected turns and offers many valuable lessons. Initially considered part of a unified system of paraganglia performing the endocrine function, the carotid body was later reclassified and recognized as a chemosensory organ. This article highlights the key controversies encountered by past researchers. These contradictions though largely forgotten, remain unresolved. The aim of
