Synaptogyrins regulate Ca2+-dependent exocytosis in PC12 cells.

Synaptogyrins constitute a family of synaptic vesicle proteins of unknown function. With the full-length structure of a new brain synaptogyrin isoform, we now show that the synaptogyrin family in vertebrates includes two neuronal and one ubiquitous isoform. All of these synaptogyrins are composed of a short conserved N-terminal cytoplasmic sequence, four homologous transmembrane regions, and a variable cytoplasmic C-terminal tail that is tyrosine-phosphorylated. The localization, abundance, and conservation of synaptogyrins suggest a function in exocytosis. To test this, we employed a secretio

Synaptogyrin-3 Prevents Cocaine Addiction and Dopamine Deficits.

Synaptogyrin-3, a functionally obscure synaptic vesicle protein, interacts with vesicular monoamine and dopamine transporters, bringing together dopamine release and reuptake sites. Synaptogyrin-3 was reduced by chronic cocaine exposure in both humans and rats, and synaptogyrin-3 levels inversely correlated with motivation to take cocaine in rats. Synaptogyrin-3 overexpression in dopamine neurons reduced cocaine self-administration, decreased anxiety-like behavior, and enhanced cognitive flexibility. Overexpression also enhanced nucleus accumbens dopamine signaling and prevented cocaine-induce

Transient expression of synaptogyrin in the ganglionic eminence of the human fetal brain.

The ganglionic eminence (GE) representing a conspicuous bulb-like elevation of the telencephalic proliferative zone has recently been shown to be involved in the establishment of cortical connections. This study demonstrates the presence of synaptogyrin-immunoreactivity in a large number of cell bodies of the human GE between 12 and 20 weeks of gestation. From the 20th week onwards synaptogyrin expression sharply declines. No immunoreactive structures are detectable in the 23rd week or later. As the GE persists nearly throughout the entire fetal period these results show that its neurochemical

Cellugyrin (Synaptogyrin-2) Regulates Macrophage Phagocytosis of Aggregatibacter actinomycetemcomitans (Aa).

Grade C molar-incisor pattern periodontitis (C-MIP) is a rapidly progressive form of periodontal disease affecting young individuals that is often linked to a highly virulent genotype of Aggregatibacter actinomycetemcomitans (Aa). Although Aa is present in the healthy oral microbiome, its transition into subgingival tissue correlates with the conversion from healthy to diseased status within the periodontal pocket. These changes may be due to immune evasion strategies attributed to Aa exotoxins. We previously demonstrated that a host cell protein, cellugyrin, plays a critical role in exotoxin

In vitro investigation of host Synaptogyrin-2 role in BVDV-1 replication.

Bovine viral diarrhea virus type 1 (BVDV-1) is an RNA virus known for lifelong persistent infection in affected animals and substantial economic impact in cattle. While current approaches of infection control rely on biosecurity and immunization, recent advances in genome editing provide opportunities for improving genetic resistance of livestock to viral diseases. Any improvements in our understanding of the role of host genes in viral replication could lead to new gene-editing applications, supporting future efforts to control BVDV-1 worldwide. Recent evidence demonstrates that Synaptogyrin-

Synaptogyrin-3 plays a critical role in addiction-related dopamine dysfunction and behavioral maladaptations.

Cocaine use disorder is marked by deficits in dopamine signaling; however, the molecular mechanisms driving these deficits remain unclear. The synaptic vesicle protein Synaptogyrin-3 (Syngr3) has recently garnered attention for its association with disorders involving dopamine dysfunction and impaired cognition. Here, we found low Syngr3 expression in the ventral tegmental area (VTA) of men who died of cocaine overdose and in male rats that had chronically self-administered cocaine. Syngr3 was confirmed to be in dopamine neurons, and its expression was correlated with dopamine markers in both

Exploring the Relationship Between Lone Star Virus and Synaptogyrin-2 Using Novel Viral and Host Models of Infections.

BACKGROUND: There is limited understanding of the replication and transmission of bandaviruses and the influence of host genotype in successful infection. An in vitro Bandavirus model, such as Lone Star virus (LSV, Bandavirus amblyommae), capable of propagating in standard cell lines, could provide some of this critical information. In this study, we sequenced the genome of LSV and profiled its relationship with a key host viral-interacting protein, Synaptogyrin-2 (SYNGR2), known to influence the replication of another Bandavirus, Bandavirus dabieense.Materials and Methods: The genome of the L

Analysis in vivo using a new method, ARGO (Analysis of Red-Green Offset), reveals complexity and cell-type specificity in presynaptic turnover of synaptic vesicle protein Synaptogyrin/SNG-1.

In long-lived cells such as neurons, proteostasis involves the regulated degradation and replacement of proteins to ensure their quality and appropriate abundance. Synaptic vesicle (SV) protein turnover in neurons is important for controlling the SV pool size and sustaining appropriate levels of neurotransmission; yet, it is incompletely understood, partly due to limited tools for quantifying protein turnover in vivo. We present ARGO (Analysis of Red-Green Offset), a fully genetically encoded ratiometric fluorescence imaging method that visualizes and quantifies protein turnover with subcellul

Early synaptic pathology is associated with small tau aggregates in Alzheimer's disease.

Alzheimer's disease (AD) is phenotypically characterised by progressive memory loss, which has been linked to tau aggregation and synaptic dysfunction. Here we characterised the nanoscopic tau aggregates in individual synaptosomes from AD cases and controls, measuring their number and size using SynPull with direct stochastic optical reconstruction microscopy (dSTORM). A total of 7888 synaptosomes from pre-frontal cortex samples were studied, showing the presence of AT8-positive tau aggregates in a small fraction of synaptosomes (~ 3%) from control brains, reaching ~ 20% by Braak stage 6. Thes

Advancing medicine for Alzheimer's disease: A plasma neural exosome platform.

Enrichment of neurally derived extracellular vesicles of several cell-types from plasma for protein quantification longitudinally in living patients with Alzheimer's disease has permitted the development of a tentative temporal framework of initiating events, progression mechanisms, and amplification processes. Interactions of beta-amyloid peptides with an elevated level of their normal prion protein dendritic receptor and of phospho-tau species with their synaptogyrin-3 synaptic vesicle receptor replace excessive production and accumulation of neuropathic proteins as the major initiating even

Unveiling the significance of synaptic proteins in parkinson's pathogenesis: A review.

Parkinson's disease (PD) is a progressive neurodegenerative disorder that leads to death of dopaminergic neurons and deficiency of dopamine. It is the second most common neurodegenerative disorder worldwide after Alzheimer's disease (AD). It is mostly prevalent in elderly people above age 60. Clinical manifestations of PD include motor symptoms like tremor, akinesia, rigidity and gait imbalance; whereas non-motor symptoms include impaired olfaction and GI dysfunction. Α-synuclein is the major pathological hallmark of PD pathology. It aggregates and leads to formation of fibrils and Lewy bodies

Exploring the Potential of Exosomal Biomarkers in Mild Traumatic Brain Injury and Post-Concussion Syndrome: A Systematic Review.

BACKGROUND: Alongside their long-term effects, post-concussion syndrome (PCS) and mild traumatic brain injuries (mTBI) are significant public health concerns. Currently, there is a lack of reliable biomarkers for diagnosing and monitoring mTBI and PCS. Exosomes are small extracellular vesicles secreted by cells that have recently emerged as a potential source of biomarkers for mTBI and PCS due to their ability to cross the blood-brain barrier and reflect the pathophysiology of brain injury. In this study, we aimed to investigate the role of salivary exosomal biomarkers in mTBI and PCS. METHOD

Chronic Social Isolation Stress during Peri-Adolescence Alters Presynaptic Dopamine Terminal Dynamics via Augmentation in Accumbal Dopamine Availability.

Chronic peri-adolescent stress in humans increases risk to develop a substance use disorder during adulthood. Rats reared in social isolation during peri-adolescence (aSI; 1 rat/cage) period show greater ethanol and cocaine intake compared to group housed (aGH; 4 rats/cage) rats. In addition, aSI rats have a heightened dopamine response in the nucleus accumbens (NAc) to rewarding and aversive stimuli. Furthermore, single pulse electrical stimulation in slices containing NAc core elicits greater dopamine release in aSI rats. Here, we further investigated dopamine release kinetics and machinery

Co-immunoprecipitation with Tau Isoform-specific Antibodies Reveals Distinct Protein Interactions and Highlights a Putative Role for 2N Tau in Disease.

Alternative splicing generates multiple isoforms of the microtubule-associated protein Tau, but little is known about their specific function. In the adult mouse brain, three Tau isoforms are expressed that contain either 0, 1, or 2 N-terminal inserts (0N, 1N, and 2N). We generated Tau isoform-specific antibodies and performed co-immunoprecipitations followed by tandem mass tag multiplexed quantitative mass spectrometry. We identified novel Tau-interacting proteins of which one-half comprised membrane-bound proteins, localized to the plasma membrane, mitochondria, and other organelles. Tau was

SYNGR3 Accelerates α-Synuclein Aggregation and Neurodegeneration in Parkinson's Disease.

BACKGROUND AND AIMS: The aggregation of α-synuclein (α-syn) is a central event in Parkinson's disease (PD) pathogenesis. However, the cellular factors that initiate and accelerate the process are not fully understood. Synaptogyrin-3 (SYNGR3) is a synaptic vesicle protein whose role in α-syn pathology remains unexplored. This study investigated whether SYNGR3 is a key factor triggering the pathological process of PD. METHODS: This study investigated the expression of SYNGR3 in the brains of transgenic A53T α-syn mutant mouse line M83 (TgA53T) PD model mice using Western blot. The direct intera

SYNGR4 and PLEKHB1 deregulation in motor neurons of amyotrophic lateral sclerosis models: potential contributions to pathobiology.

Amyotrophic lateral sclerosis is the most common adult-onset neurodegenerative disease affecting motor neurons. Its defining feature is progressive loss of motor neuron function in the cortex, brainstem, and spinal cord, leading to paralysis and death. Despite major advances in identifying genes that can cause disease when mutated and model the disease in animals and cellular models, it still remains unclear why motor symptoms suddenly appear after a long pre-symptomatic phase of apparently normal function. One hypothesis is that age-related deregulation of specific proteins within key cell ty

SYNGR1 is associated with schizophrenia and bipolar disorder in southern India.

Chromosome 22q11-13 is one of the most consistent linkage regions for schizophrenia (SCZ) and bipolar disorder (BPAD). The SYNGR1 gene, which is associated with presynaptic vesicles in neuronal cells, is located on 22q13.1. We have previously identified a novel nonsense mutation in the SYNGR1 gene in a SCZ pedigree. In the present study, a detailed analysis of this gene was performed in a case-control cohort (198 BPAD, 193 SCZ and 107 controls from southern India) to test for association with SCZ and BPAD. Sequence analysis of all exonic and flanking intronic regions of the SYNGR1 gene in 198

Synaptic gene dysregulation within hippocampal CA1 pyramidal neurons in mild cognitive impairment.

Clinical neuropathologic studies suggest that the selective vulnerability of hippocampal CA1 pyramidal projection neurons plays a key role in the onset of cognitive impairment during the early phases of Alzheimer's disease (AD). Disruption of this neuronal population likely affects hippocampal pre- and postsynaptic efficacy underlying episodic memory circuits. Therefore, identifying perturbations in the expression of synaptic gene products within CA1 neurons prior to frank AD is crucial for the development of disease modifying therapies. Here we used custom-designed microarrays to examine prog

An integrated multi-level analysis reveals learning-memory deficits and synaptic dysfunction in the rat model exposure to austere environment.

Austere environment existing in tank, submarine and vessel has many risk factors including high temperature and humidity, confinement, noise, hypoxia, and high level of carbon dioxide, which may cause depression and cognitive impairment. However, the underlying mechanism is not fully understood yet. We attempt to investigate the effects of austere environment (AE) on emotion and cognitive function in a rodent model. After 21 days of AE stress, the rats exhibit depressive-like behavior and cognitive impairment. Compared with control group, the glucose metabolic level of the hippocampus is signi
