The ferredoxin:sulphite reductase gene from Synechococcus PCC7942.

The structural gene of the ferredoxin:sulphite reductase (EC 1.8.7.1) from the cyanobacterium Synechococcus PCC7942 (formerly 'Anacystis nidulans') was cloned and sequenced. The gene termed 'sir' was detected by heterologous Southern hybridisation with the structural gene cysI from Escherichia coli encoding the iron-sulphur haemoprotein of the NADPH:sulphite reductase. The open reading frame is comprised of 1875 bp encoding for a polypeptide of M(r) 70.028. The deduced amino acid sequence is 35.6% identical with the enterobacterial iron-sulphur haemoprotein. This putative fd-dependent sulphite

Biotechnological Applications of Cyanobacteria: Synechocystis and Synechococcus Strains.

Cyanobacteria as phototrophic microorganisms bear great potential for biotechnological application and a truly sustainable bioeconomy. Besides production of biomass and natural compounds, CO2-based production of diverse value-added compounds with engineered strains enjoys ever-growing interest. Representatives of the genera Synechocystis and Synechococcus are the most used cyanobacterial model organisms for this purpose, with studies ranging from basic research to their utilization as cell factories. For both genera, genetic tools become more and more established, being, however, still far les

The synthesis of elemental selenium particles by Synechococcus leopoliensis.

Exposure of Synechococcus leopoliensis to selenite in the light resulted in orange-colored granules associated with the cells. No such particles were made in dark grown cells or when selenite was replaced by selenate. Light and scanning electron microscopy revealed that the particles formed inside the cells. Furthermore, these were easily extracted and shown to be composed of selenium as determined by energy-dispersive X-ray spectroscopy. During selenium particle synthesis there was a concurrent loss of organic pigments in the cyanobacteria. Cells also become heavier as they produced and accum

Pan-proteome regulation of marine Synechococcus nutrient stress plasticity.

Variable phytoplankton resource demands can buffer productivity to climate-driven declines in nutrient supply, but the extent of this buffering depends on limits to cellular elemental plasticity. Elemental plasticity is driven by a combination of genomic adaptation and physiological regulation, but their mechanistic links remain unresolved. Here, we combine continuous culture experiments with proteomics and elemental analyses to determine how genomic diversity shapes nutrient stress plasticity in six strains of the globally distributed cyanobacterium Synechococcus. By maintaining fixed growth

Dissolved organic phosphorus bond-class utilization by Synechococcus.

Dissolved organic phosphorus (DOP) contains compounds with phosphoester, phosphoanhydride, and phosphorus-carbon bonds. While DOP holds significant nutritional value for marine microorganisms, the bioavailability of each bond-class to the widespread cyanobacterium Synechococcus remains largely unknown. This study evaluates bond-class specific DOP utilization by Synechococcus strains from open and coastal oceans. Both strains exhibited comparable growth rates when provided phosphate, a phosphoanhydride [3-polyphosphate and 45-polyphosphate], or a DOP compound with both phosphoanhydride and phos

From phenomenon to mechanism in the circadian clock of Synechococcus elongatus.

Creative and collaborative research in the early 21st century revealed the mechanistic secrets of the cyanobacterial circadian clock, a nanomachine whose function can be studied in vitro. The structural dynamics of the Synechococcus elongatus circadian oscillator, comprising KaiA, KaiB, and KaiC, establish a near 24 h cycle of activity of the global transcription regulator RpaA by rhythmically changing the interaction platform of kinases that control RpaA phosphorylation.

Role of polyphosphate in thermophilic Synechococcus sp. from microbial mats.

Synechococcus OS-B', a thermophilic unicellular cyanobacterium, recently isolated from the microbial mats in Octopus Spring (Yellowstone National Park), induces a suite of genes, including phosphatases and transporters, in response to phosphorus (P) starvation. Here we describe two different approaches to examine the ability of Synechococcus OS-B' to synthesize and break down polyphosphate (poly P), a key storage compound in many prokaryotes. First, we developed a transformation protocol to create mutants in the polyphosphate kinase (ppk), the major enzyme responsible for the synthesis of poly

Employing Nanotechnology to Enhance Ethanol Production in Two Synechococcus elongatus Strains.

Cyanobacteria have emerged as promising photosynthetic cell factories. However, low product yields remain a bottleneck to industrial exploitation. Here, we use bacterial microcompartment technology to improve product titres using ethanol biosynthesis as the model pathway. Two strategies compared to unmodified enzyme production were tested: first, targeting enzymes to nanofilaments by fusion with encapsulation peptides (EPs) which form non-covalent interactions with the filaments. Second, the enzyme-EP fusions were used in the absence of nanofilaments, as EPs tend to form soluble aggregates. Bo

Characterization of zinc stress response in Cyanobacterium Synechococcus sp. IU 625.

The ability of cyanobacteria to survive many environmental stress factors is a testament to their resilience in nature. Of these environmental stress factors, overexposure to zinc is important to study since excessive zinc intake can be a severe hazard. Zinc toxicity in freshwater has been demonstrated to affects organisms such as invertebrates, algae and cyanobacteria. Cyanobacteria which possess increased resistance to zinc have been isolated. It is therefore important to elucidate the mechanism of survival and response to determine what factors allow their survival; as well as any remediati

Utilization of lignocellulosic hydrolysates for photomixotrophic chemical production in Synechococcus elongatus PCC 7942.

To meet the need for environmentally friendly commodity chemicals, feedstocks for biological chemical production must be diversified. Lignocellulosic biomass are an carbon source with the potential for effective use in a large scale and cost-effective production systems. Although the use of lignocellulosic biomass lysates for heterotrophic chemical production has been advancing, there are challenges to overcome. Here we aim to investigate the obligate photoautotroph cyanobacterium Synechococcus elongatus PCC 7942 as a chassis organism for lignocellulosic chemical production. When modified to i

[Characteristics of different growth stages of the blue-green alga Anacystis nidulans (Synechococcus)].

The rate of growth of the blue-green alga Anacystis nidulans is highest during the first 5--8 days but the number of the cells increases up to 30 days when the stationary phase takes place (30--60 days). The content of chlorophyll is almost constant during the first 20 days of growth while the content of phycocyanin increases; then the content of the both pigments decreases. The rate of O2 evolution and the rate of electron transport to ferricyanide decrease exponentionally during the first 10 days of growth. The rate of endogenous respiration and the rate of O2 absorption, induced by methylvi

Safety of photosynthetic Synechococcus elongatus for in vivo cyanobacteria-mammalian symbiotic therapeutics.

The cyanobacterium Synechococcus elongatus (SE) has been shown to rescue ischaemic heart muscle after myocardial infarction by photosynthetic oxygen production. Here, we investigated SE toxicity and hypothesized that systemic SE exposure does not elicit a significant immune response in rats. Wistar rats intravenously received SE (n = 12), sterile saline (n = 12) or E. coli lipopolysaccharide (LPS, n = 4), and a subset (8 SE, 8 saline) received a repeat injection 4 weeks later. At baseline, 4 h, 24 h, 48 h, 8 days and 4 weeks after injection, clinical assessments, blood cultures, blood counts,

Functional elements of the strong psbAI promoter of Synechococcus elongatus PCC 7942.

The psbAI gene of the cyanobacterium Synechococcus elongatus PCC 7942 is one of three psbA genes that encode a critical photosystem II reaction center protein, D1. Regulation of the gene family in response to changes in the light environment is complex, occurs at transcriptional and posttranscriptional levels, and results in an interchange of two different forms of D1 in the membrane. Expression of psbAI is downregulated under high-intensity light (high light) in contrast to induction of the other two family members. We show that, in addition to a known accelerated degradation of the psbAI mes

Nucleoid compaction influences carboxysome localization and dynamics in Synechococcus elongatus PCC 7942.

UNLABELLED: The bacterial nucleoid is not just a genetic repository-it serves as a dynamic scaffold for spatially organizing key cellular components. ParA-family ATPases exploit this nucleoid matrix to position a wide range of cargos, yet how nucleoid compaction influences these positioning reactions remains poorly understood. We previously characterized the maintenance of carboxysome distribution (Mcd) system in the cyanobacterium Synechococcus elongatus PCC 7942, where the ParA-like ATPase McdA binds the nucleoid and interacts with its partner protein, McdB, to generate dynamic gradients tha

Efficient utilization of xylose requires CO2 fixation in Synechococcus elongatus PCC 7942.

Cyanobacteria show great promise as autotrophic hosts for the renewable biosynthesis of useful chemicals from CO2 and light. While they can efficiently fix CO2, cyanobacteria are generally outperformed by heterotrophic production hosts in terms of productivity and titer. Photomixotrophy, or co-utilization of sugars and CO2 as carbon feedstocks, has been implemented in cyanobacteria to greatly improve productivity and titers of several chemical products. We introduced xylose photomixotrophy to a 2,3-butanediol producing strain of Synechococcus elongatus PCC 7942 and characterized the effect of

Rubisco packaging and stoichiometric composition of the native β-carboxysome in Synechococcus elongatus PCC7942.

Carboxysomes are anabolic bacterial microcompartments that play an essential role in CO2 fixation in cyanobacteria. This self-assembling proteinaceous organelle uses a polyhedral shell constructed by hundreds of shell protein paralogs to encapsulate the key CO2-fixing enzymes Rubisco and carbonic anhydrase. Deciphering the precise arrangement and structural organization of Rubisco enzymes within carboxysomes is crucial for understanding carboxysome formation and overall functionality. Here, we employed cryoelectron tomography and subtomogram averaging to delineate the 3D packaging of Rubiscos

Engineering and characterization of copper and gold sensors in Escherichia coli and Synechococcus sp. PCC 7002.

The anthropogenic release of toxic metals into the environment poses danger to the health of both humans and the local ecosystem. Biosensors for the detection of metals have been developed to improve our ability to monitor these environmental contaminants, yet most of these sensors use heterotrophic bacterial hosts, which require a fixed carbon source and do not typically grow in natural waterways. In this study, we constructed and characterized metal sensors for development of a photoautotrophic biosensor using Synechococcus sp. PCC 7002. We characterized gold and copper sensors based on modi
