TNF Receptor Associated Factor 2 (TRAF2) Signaling in Cancer.

Tumor necrosis factor (TNF) receptor associated factor-2 (TRAF2) has been originally identified as a protein interacting with TNF receptor 2 (TNFR2) but also binds to several other receptors of the TNF receptor superfamily (TNFRSF). TRAF2, often in concert with other members of the TRAF protein family, is involved in the activation of the classical NFκB pathway and the stimulation of various mitogen-activated protein (MAP) kinase cascades by TNFRSF receptors (TNFRs), but is also required to inhibit the alternative NFκB pathway. TRAF2 has also been implicated in endoplasmic reticulum (ER) stres

The deubiquitinase OTUD6B regulates cardiac hypertrophy by stabilizing TNF receptor associated factor 2.

AIMS: Pathological cardiac hypertrophy serves as the primary pathological driver of heart failure, significantly impacting patient prognosis. Deubiquitinating enzymes, which play pivotal roles in protein homeostasis, are emerging as key regulators in cardiac hypertrophy and dysfunction. This study sought to elucidate the contribution of a specific deubiquitinase, Ovarian tumor (OTU) Domain-Containing Deubiquitinase 6B‌ (OTUD6B), to cardiac hypertrophy and explore its functional mechanisms. MATERIALS AND METHODS: A mouse model of pressure overload cardiac hypertrophy was established through tr

The Deubiquitinase OTUD5 regulates cardiac hypertrophy by stabilizing TNF receptor associated factor 2.

BACKGROUND: Pathological cardiac hypertrophy acts as a major pathological contributor to heart failure, profoundly influencing patient outcomes. Deubiquitinating enzymes, which are critical for maintaining protein homeostasis, are increasingly recognized as essential regulators in cardiac hypertrophy and dysfunction. This study aimed to investigate the role of a specific deubiquitinase, Ovarian tumor domain-containing protein 5 (OTUD5), in cardiac hypertrophy and elucidate its functional mechanisms. METHODS: A pressure overload-induced cardiac hypertrophy model was created in mice using trans

Rotavirus NSP1 inhibits interferon induced non-canonical NFκB activation by interacting with TNF receptor associated factor 2.

TNF receptor associated factor 2 (TRAF2) plays a very important role in cellular innate immune as well as inflammatory responses. Previous studies have reported TRAF2 mediated regulation of TNF and Interferon (IFN) induced canonical and non-canonical activation of NFκB. In this study, we show that rotavirus NSP1 targets TRAF2 to regulate IFN induced non-canonical NFκB activation. Here we found that rotavirus Non-Structural Protein-1 (NSP1) interacts with TRAF2 and degrades it in a proteasome dependent manner. C-terminal part of NSP1 was sufficient for interacting with TRAF2 but it alone could

Susceptibility to Burkholderia pseudomallei is associated with host immune responses involving tumor necrosis factor receptor-1 (TNFR1) and TNF receptor-2 (TNFR2).

Melioidosis is caused by the facultative intracellular bacterium, Burkholderia pseudomallei. Using C57BL/6 mice, we investigated the role of macrophages, TNF-alpha, TNF receptor-1 (TNFR1) and TNF receptor-2 (TNFR2) in host defense against B. pseudomallei using an experimental model of melioidosis. This study has demonstrated that in vivo depletion of macrophages renders C57BL/6 mice highly susceptible to intranasal infection with B. pseudomallei, with significant mortality occurring within 5 days of infection. Using knockout mice, we have also shown that TNF-alpha and both TNFR1 and TNFR2 are

TNF receptor-associated factor 6 sensitizes prostate cancer cells to CD8+ T cell-mediated ferroptosis via Lipocalin 2 degradation.

Prostate cancer remains a significant therapeutic challenge in male urologic oncology. Characterized by an immunosuppressive microenvironment, it is commonly described as an immunologically "cold" tumor. This study elucidates the role of TNF receptor-associated factor 6 (TRAF6) within the immune landscape of prostate cancer. We observed marked downregulation of TRAF6 in both patient-derived tissues and cultured prostate cancer cell lines. Functional assays demonstrated that TRAF6 upregulation robustly suppresses cancer cell proliferation and migration. Mechanistically, we discovered that TRAF6

TNF receptor-associated factor (TRAF) domain and Bcl-2 cooperate to induce small B cell lymphoma/chronic lymphocytic leukemia in transgenic mice.

Transgenic mice overexpressing in B lymphocytes either Bcl-2 or a TNF receptor-associated factor (TRAF)2 mutant lacking the N-terminal RING and zinc finger domains located at the N terminus of the molecule (TRAF2DN), which mimics TRAF1, developed lymphadenopathy and splenomegaly due to polyclonal B cell expansion. Remarkably, TRAF2DN/Bcl-2 double-transgenic mice contained B cell populations similar to those observed in TRAF2DN mice. However, over time, they developed severe splenomegaly and lymphadenopathy, and most animals also developed leukemia, pleural effusion, and, in some cases, ascites

CD40-TRAF2/3/5 Signaling Promotes Cardiac Repair by Mediating Macrophage Efferocytosis After Myocardial Infarction.

BACKGROUND: After myocardial infarction (MI), macrophage-mediated clearance of dead cells, a process known as efferocytosis, represents a pivotal role in tissue remodeling. Efficient efferocytosis contributes to rescuing neighboring viable cardiomyocytes, drives the phenotypic transition of reparative macrophages, and facilitates the resolution of inflammation. In this study, we explored the roles of CD40 and the signals transduced by its 2 downstream adaptor-protein binding sites (TRAF2/3/5 and TRAF6) in the cardiac macrophage efferocytosis after MI. METHODS: Systemic, myeloid- and macrophag

The TRAF2 and TRAF6 expression in myomas and myometrium of women in reproduction and perimenopausal age.

Uterine myomas represent one of the most common female diseases. Uterine myomas or fibromas are benign, hormone-responding tumours of, respectively, smooth muscles and fibroblasts and their aetiology induces a significant interest. In myomas the presence of aromatase was detected and, in addition, oestrogen was found to be synthesized in myoma cells. The studies were performed on myoma patients of generative age and those in peri-menopausal age. Expression of TRAF2 and TRAF6 proteins was examined using immunohistochemistry and Western blot approach in small and large uterine myomas isolated fr

Functional relevance of soluble TNF-alpha, transmembrane TNF-alpha and TNF-signal transduction in gastrointestinal diseases with special reference to inflammatory bowel diseases.

As a result of extensive clinical and basic research, the pivotal role of tumour necrosis factor (TNF) in the pathogenesis of chronic inflammatory diseases such as inflammatory bowel disease (IBD) has now generally been acknowledged. This has led to promising clinically effective anti-TNF-strategies. Of note, there is more and more evidence that TNF seems to play a key role in other gastrointestinal diseases including Helicobacter pylori infection, pancreatitis, viral hepatitis and toxic liver damage, too. The action of TNF at the cellular level is mediated by two cell surface receptors, TNF-R

An Internally Translated MAVS Variant Exposes Its Amino-terminal TRAF-Binding Motifs to Deregulate Interferon Induction.

Activation of pattern recognition receptors and proper regulation of downstream signaling are crucial for host innate immune response. Upon infection, the NF-κB and interferon regulatory factors (IRF) are often simultaneously activated to defeat invading pathogens. Mechanisms concerning differential activation of NF-κB and IRF are not well understood. Here we report that a MAVS variant inhibits interferon (IFN) induction, while enabling NF-κB activation. Employing herpesviral proteins that selectively activate NF-κB signaling, we discovered that a MAVS variant of ~50 kDa, thus designated MAVS5

CD40-mediated tumor necrosis factor receptor-associated factor 3 signaling upregulates IL-4-induced germline Cepsilon transcription in a human B cell line.

Induction of germline C epsilon transcription in B cells by IL-4, which is a critical initiating step for IgE class switching, is enhanced by CD40 engagement. Although signaling by CD40 is initiated by the binding of tumor necrosis factor receptor-associated factor (TRAF) family members to its cytoplasmic domain, whether those TRAF family proteins mediate enhancement of germline Cepsilon transcription is not evident. We report here that CD40-induced TRAF3-dependent activation of mitogen-activated protein kinase (MAPK)/extracellular signal-regulated kinase (ERK) kinase 1 (MEK1) is involved in t

[Utility of next-generation sequencing for diagnosing non-secretory multiple myeloma].

A 62-year-old man was referred to our hospital for leukocytosis with an increase (25.5%) of plasma cell-like cells in the peripheral blood. M-protein was not detected, and no deviation of the κ/λ ratio of free light chain was observed. Bone marrow biopsy showed increased plasma cell-like cell counts; however, immunohistochemical staining was positive only for CD138, and negative for both κ and λ chains. Non-secretory multiple myeloma (MM) was diagnosed based on morphological findings, although differential diagnosis from lymphoplasmacytic lymphoma was difficult. After remission induction treat

Global quantitative proteomic analysis of human glioma cells profiled host protein expression in response to enterovirus type 71 infection.

Enterovirus 71 (EV71) is one of the leading causes of hand, foot and mouth disease with neurological complications in some cases. To study the pathogenesis of EV71 infection, large-scale analyses of EV71 infected cells have been performed. However, most of these studies employed rhabdomyosarcoma (RD) cells or used transcriptomic strategy. Here, we performed SILAC-based quantitative proteomic analysis of EV71-infected U251 cells, a human glioma cell line. A total of 3125 host proteins were quantified, in which 451 were differentially regulated as a result of EV71 infection at 8 or 20 hpi or bot

UBD participates in neutrophilic asthma by promoting the activation of IL-17 signaling.

Neutrophilic asthma is a persistent and severe inflammatory lung disease characterized by neutrophil activation and the mechanisms of which are not completely elucidated. Ubiquitin D (UBD) is a ubiquitin-like modifier participating in infections, immune responses, and tumorigenesis, while whether UBD involves in neutrophilic asthma needs further study. In this study, we initially found that UBD expression was significantly elevated and interleukin 17 (IL-17) signaling was enriched in the endobronchial biopsies of severe asthma along with neutrophils increasing by bioinformatics analysis. We fu

TRAF2/3 deficient B cells resist DNA damage-induced apoptosis via NF-κB2/XIAP/cIAP2 axis and IAP antagonist sensitizes mutant lymphomas to chemotherapeutic drugs.

Deletion of TRAF2 or TRAF3 in B cells prolongs their survival. However, it remains unknown whether deletion of such factors affects B cells' ability to tolerate DNA damage, which can be induced by chemotherapeutics and cause apoptosis. Genetic alterations of TRAF2 or TRAF3 are observed in subsets of human B-cell lymphomas and B cell-specific deletion of TRAF3 led to lymphoma development in aged mice. However, it remains unknown whether double deficiency of TRAF2 and TRAF3 accelerates B-cell lymphomagenesis. Here, we showed that B cell-specific TRAF2/3 double deficient (B-TRAF2/3-DKO) B cells w

The Role of Apoptotic Genes and Protein-Protein Interactions in Triple-negative Breast Cancer.

BACKGROUND/AIM: Compared to other breast cancer types, triple-negative breast cancer (TNBC) has historically had few treatment alternatives. Therefore, exploring and pinpointing potentially implicated genes could be used for treating and managing TNBC. By doing this, we will provide essential data to comprehend how the genes are involved in the apoptotic pathways of the cancer cells to identify potential therapeutic targets. Analysis of a single genetic alteration may not reveal the pathogenicity driving TNBC due to the high genomic complexity and heterogeneity of TNBC. Therefore, searching th

Cutting Edge: The RNA-Binding Protein Ewing Sarcoma Is a Novel Modulator of Lymphotoxin β Receptor Signaling.

Lymphotoxin β receptor (LTβR) signaling is crucial for lymphoid tissue organogenesis and immune homeostasis. To identify novel regulatory mechanisms for signaling, we implemented a two-step screen that uses coexpression analysis of human fibroblasts undergoing LTβR stimulation and affinity-purification mass spectrometry for the LTβR signaling protein TNFR-associated factor 3 (TRAF3). We identify Ewing sarcoma (EWS) protein as a novel LTβR signaling component that associates with TRAF3 but not with TNFR-associated factor 2 (TRAF2). The EWS:TRAF3 complex forms under unligated conditions that are
