Production of Tiger Puffer Takifugu rubripes Offspring from Triploid Grass Puffer Takifugu niphobles Parents.

The tiger puffer Takifugu rubripes is one of the most popular aquacultural fish; however, there are two major obstacles to selective breeding. First, they have a long generation time of 2 or 3 years until maturation. Second, the parental tiger puffer has a body size (2-5 kg) much larger than average market size (0.6-1.0 kg). The grass puffer Takifugu niphobles is closely related to the tiger puffer and matures in half the time. Furthermore, grass puffer can be reared in small areas since their maturation weight is about 1/150 that of mature tiger puffer. Therefore, to overcome the obstacles of

Multi-Omics Reveals Tetrodotoxin Transport and Accumulation Mechanisms in Takifugu bimaculatus.

The potent toxicity of tetrodotoxin (TTX) has long constrained sustainable growth in pufferfish aquaculture. Takifugu bimaculatus, an economically important species farmed along the coast of Fujian, China, remains poorly understood regarding how it transports and accumulates this potent neurotoxin. To address this gap, we combined transcriptomic and proteomic analyses to characterize the molecular responses of T. bimaculatus to TTX exposure. After oral administration, TTX primarily accumulated in the liver, ovaries, and skin. Multi-omics profiling revealed 163 differentially expressed genes (D

Comparative transcriptome analysis identified genes involved in ovarian development in Takifugu rubripes.

Ovarian development is a complex process involving multiple genes, but the molecular mechanisms underlying this process in Takifugu rubripes remain poorly understood. This study aimed to identify genes associated with ovarian development in T. rubripes and to investigate the regulatory mechanisms of oocyte maturation. Transcriptome data were compared across four different developmental stages (stage II to V) to identify differentially expressed genes (DEGs) and perform GO and KEGG enrichment analysis. The expression patterns of randomly selected genes were then validated by qPCR. The results y

A Novel Angiotensin-I-Converting Enzyme (ACE) Inhibitory Peptide from Takifugu flavidus.

Alcalase, neutral protease, and pepsin were used to hydrolyze the skin of Takifugu flavidus. The T. flavidus hydrolysates (TFHs) with the maximum degree of hydrolysis (DH) and angiotensin-I-converting enzyme (ACE)-inhibitory activity were selected and then ultra-filtered to obtain fractions with components of different molecular weights (MWs) (<1, 1-3, 3-10, 10-50, and >50 kDa). The components with MWs < 1 kDa showed the strongest ACE-inhibitory activity with a half-maximal inhibitory concentration (IC50) of 0.58 mg/mL. The spinal sympathetic preganglionic cell column in the puffer fish, Takifugu niphobles.

Little is known about the spinal sympathetic organization in teleosts. We examined the location of the sympathetic preganglionic neurons with horseradish peroxidase (HRP) labeling. After HRP application to the sympathetic trunk or celiac ganglion, labeled neurons were found just dorsal - dorsolateral to the central canal. They form a cell column (central autonomic nucleus) at the level of the posterior rootlet of the first spinal nerve to the third spinal nerve. HRP application to the sympathetic trunk produced labeling in almost the entire central autonomic nucleus, but HRP application to the

Comparative transcriptome analysis identified genes involved in testicular development in Takifugu rubripes.

To identify candidate genes and pathways involved in testicular development in Takifugu rubripes, a comparative transcription analysis was conducted across the various developmental stages of the testis (stages II to V). A total of 9520 differentially expressed genes (DEGs) were identified among the different stages, and they were significantly clustered into six clusters (P < 0.05). One thousand four hundred eleven DEGs such as gndf, wnt1, and cyp17b1 were found to be decreased from stage II to V. In contrast, 994 DEGs such as fn1, ift81, and cdc25a were found to be increased from stage II to

Occurrence of Natural Interspecific Hybrids in the Pufferfish, Genus Takifugu, and Their Tetrodotoxin-accumulating Tissues.

Pufferfish in the genus Takifugu accumulate the potent neurotoxin tetrodotoxin (TTX) in specific tissues that vary by species; nevertheless, certain species are consumed as a luxury food in Japan. Therefore, the edible species and tissues of Takifugu pufferfish are regulated in Japan. In recent years, unidentifiable individuals with atypical morphological characteristics caught in waters around Japan have been found to be natural interspecific hybrids among Takifugu species. Four cases of natural hybridization have been reported; in all cases, hybrids had external characteristics intermediate

Effect of Ichthyophthirius multifiliis infection on host immunity and microbiota shifts of Takifugu fasciatus.

Ichthyophthirius multifiliis (Ich) is a major pathogen responsible for high mortality rates in Takifugu fasciatus aquaculture. Despite its significance, the local and systemic immune responses of pufferfish to Ich infection remain poorly understood. Additionally, the interaction between ectoparasite infection and gut microbiota has not been thoroughly investigated. To address these gaps, we established an Ich infection model in pufferfish through bath administration. RT-PCR confirmed that Ich invades the skin, gill, and fin at 10 days post-infection (dpi) using 5000 theronts per fish. RT-qPCR

Muscle Characteristics and Transcriptomic Analysis of Diploid and Triploid Tiger Pufferfish (Takifugu rubripes).

Sexual maturity in tiger pufferfish (Takifugu rubripes) consumes substantial metabolic energy, constraining somatic growth and limiting meat yield. Artificial triploid induction (sterility) may redirect energy toward somatic growth. Cold-shock induced triploids were compared with diploid controls using muscle and liver tissues, and the phenotype, histology, nutritional composition, hormone levels, as well as transcriptome profiles were assessed. After 5 months, triploids attained significantly greater body length and body weight, with larger muscle fiber diameter but lower fiber density. The t

Tetrodotoxin functions as a stress relieving substance in juvenile tiger puffer Takifugu rubripes.

We tested whether tetrodotoxin (TTX) functions as a stress relieving substance in puffer fish. We orally administered TTX to the juveniles of hatchery-reared non-toxic tiger puffer Takifugu rubripes and measured the effects of TTX on brain corticotropin-releasing hormone (CRH) mRNA expression and plasma cortisol levels in comparison with effects in non-toxic juveniles. Firstly, the reciprocal connections of CRH and adrenocorticotropic hormone (ACTH) were confirmed by dual-label immunohistochemistry. CRH-immunoreactive (ir) cell bodies were detected in the hypothalamus and CRH-ir fibers were ob

Identification and transcriptional analysis of eight C1q genes in Takifugu obscurus during bacterial infection.

As an initiating molecule of the classical complement pathway, C1q performs diverse biological functions. In this study, eight C1q genes were identified in Takifugu obscurus and designated as ToC1q1-ToC1q8. Bioinformatics analysis revealed that the proteins encoded by these cDNAs consist of 197-261 amino acids, with varying molecular weights and isoelectric points. SMART domain analysis indicated that all ToC1q proteins contain a signal peptide and a C1q domain, one of which also contains a collagen-like domain. Sequence alignment and phylogenetic analysis showed low sequence similarity among

Profile of gene expression changes during estrodiol-17β-induced feminization in the Takifugu rubripes brain.

BACKGROUND: As the critical tissue of the central nervous system, the brain has been found to be involved in gonad development. Previous studies have suggested that gonadal fate may be affected by the brain. Identifying brain-specific molecular changes that occur during estrodiol-17β (E2) -induced feminization is crucial to our understanding of the molecular control of sex differentiation by the brains of fish. RESULTS: In this study, the differential transcriptomic responses of the Takifugu rubripes larvae brain were compared after E2 treatment for 55 days. Our results showed that 514 genes

Screening of immunogenic antigens from scuticociliate in Takifugu rubripes, and the development and evaluation of subunit vaccines.

In recent years, scuticociliatosis has emerged as a major parasitic disease in the aquaculture of Takifugu rubripes, inflicting significant economic losses on the pufferfish farming industry. The conventional approach of using chemical drugs to prevent and control parasitic diseases poses substantial risks to the environment and food safety. Consequently, the utilization of vaccines for the prevention and treatment of aquatic animal diseases is currently regarded as the mainstream direction for future aquatic animal disease prevention and control.
