Reproduction of Mesobiotus: Comparison of Morphology and Behavior in the Family Macrobiotidae (Tardigrada: Eutardigrada).

The genus Mesobiotus was separated from the genus Macrobiotus in 2016 and the name referred to its phylogenetic position among the family Macrobiotidae; however, knowledge of the reproductive behavior of this genus is limited compared to those of Paramacrobiotus and Macrobiotus. This study comprehensively provides the reproductive traits, including the gamete morphologies and behavioral observations, of Mesobiotus. The morphology of its spermatozoon showed a length that was intermediary among those of Paramacrobiotus and Macrobiotus species. The sequence of mating behavior was generally conser

Candidate Cyanide Resistance Genes in Eutardigrade (Tardigrada) Genomes and KCN Resistance of Hypsibius exemplaris.

Several eutardigrade species can withstand high doses of potassium cyanide (KCN), a potent inhibitor of mitochondrial respiration, suggesting defense mechanisms that remain poorly understood. We surveyed the available tardigrade genomes in search for 11 protein families associated with cyanide detoxification and resistance in other organisms. We identified 28 sequences putatively encoding mitochondrial alternative oxidase (AOX), as well as proteins from the nitrilase superfamily and rhodanese superfamily in three eutardigrades, but found no evidence for other canonical cyanide-detoxifying enzy

Tardigrada: An Emerging Animal Model to Study the Endoplasmic Reticulum Stress Response to Environmental Extremes.

Tardigrada (also known as "water bears") are hydrophilous microinvertebrates with a bilaterally symmetrical body and four pairs of legs usually terminating with claws. Water bears are quite complex animals and range from 50 to 1200 μm in length. Their body is divided into a head segment and four trunk segments, each bearing a pair of legs. They inhabit almost all terrestrial and aquatic environments, from the ocean depths to highest mountains ranges. However, one of their best known and unusual features is their capability for cryptobiosis. In this state tardigrades are able to survive extreme

"Let's Dry up and Survive Together": Is Anhydrobiosis in Two Paramacrobiotus Species (Tardigrada) Associated with a Specific Microbiome Community?

This study reports, for the first time, changes in the microbiome community associated with anhydrobiosis in two tardigrade species of the genus Paramacrobiotus. To identify bacteria linked to the anhydrobiosis phenomenon and to track microbiome changes under anhydrobiotic stress, next-generation sequencing of bacterial 16S rRNA genes was conducted. Microbiome profiling was performed across various developmental and physiological stages of tardigrades, including: eggs; active adult specimens (both before and after 7, and 120 days of anhydrobiosis, referred to as short- and long-term anhydrobio

CAT-Posterior Mean Site Frequencies Improves Phylogenetic Modeling Under Maximum Likelihood and Resolves Tardigrada as the Sister of Arthropoda Plus Onychophora.

Tardigrada, the water bears, are microscopic animals with walking appendages that are members of Ecdysozoa, the clade of molting animals that also includes Nematoda (round worms), Nematomorpha (horsehair worms), Priapulida (penis worms), Kinorhyncha (mud dragons), Loricifera (loricated animals), Arthropoda (insects, spiders, centipedes, crustaceans, and their allies), and Onychophora (velvet worms). The phylogenetic relationships within Ecdysozoa are still unclear, with analyses of molecular and morphological data yielding incongruent results. Accounting for across-site compositional heterogen

A transcriptome approach to ecdysozoan phylogeny.

The monophyly of Ecdysozoa, which comprise molting phyla, has received strong support from several lines of evidence. However, the internal relationships of Ecdysozoa are still contended. We generated expressed sequence tags from a priapulid (penis worm), a kinorhynch (mud dragon), a tardigrade (water bear) and five chelicerate taxa by 454 transcriptome sequencing. A multigene alignment was assembled from 63 taxa, which comprised after matrix optimization 24,249 amino acid positions with high data density (2.6% gaps, 19.1% missing data). Phylogenetic analyses employing various models support t

Pigment-dispersing factor neuropeptides act as multifunctional hormones and modulators in tardigrades.

Pigment-dispersing factors (PDFs) are neuropeptides that play key roles in controlling the circadian rhythms in various insects, whereas their function remains elusive in other protostomes including tardigrades (water bears). Here we show that the three PDFs of the tardigrade Hypsibius exemplaris are co-localized in two pairs of inner lobe cells in the brain, whereas only one PDF occurs in four additional cerebral and two extracerebral cells. The axons of the inner lobe cells pass through the contralateral brain hemisphere, descend to the ventral nerve cord and terminate in two pairs of potent

Multi-Species Tardigrade RNA Isolation Protocol.

Tardigrades are microscopic animals that are known for their exceptional tolerance to environmental stress. These animals can survive desiccation, extreme pressure, low temperatures, and high levels of ionizing radiation. However, the molecular mechanisms underlying these extremotolerant capabilities remain incompletely understood. The molecular approaches used to investigate these mechanisms, which include RNA sequencing, RNA interference (RNAi), and heterologous expression of tardigrade genes in systems such as bacteria, require the isolation of high-quality messenger RNA (mRNA) from limited

Neural markers reveal a one-segmented head in tardigrades (water bears).

BACKGROUND: While recent neuroanatomical and gene expression studies have clarified the alignment of cephalic segments in arthropods and onychophorans, the identity of head segments in tardigrades remains controversial. In particular, it is unclear whether the tardigrade head and its enclosed brain comprises one, or several segments, or a non-segmental structure. To clarify this, we applied a variety of histochemical and immunocytochemical markers to specimens of the tardigrade Macrobiotus cf. harmsworthi and the onychophoran Euperipatoides rowelli. METHODOLOGY/PRINCIPAL FINDINGS: Our immunol

Patterning on Living Tardigrades.

Micro/nanofabrication techniques have revolutionized modern photonics and electronics. However, conventional methods remain incompatible with living organisms due to inherent constraints including nonconformal coating, radiation damage, and toxic solvent requirements. Here, we present ice lithography for direct fabrication of micro/nanoscale patterns on the surfaces of tardigrades in their cryptobiotic state. Remarkably, upon rehydration the tardigrades revive, retaining the patterns on their surfaces. By precisely controlling parameters such as ice thickness, beam energy, and substrate proper

Verification of Hypsibius exemplaris Gąsiorek et al., 2018 (Eutardigrada; Hypsibiidae) application in anhydrobiosis research.

Anhydrobiosis is considered to be an adaptation of important applicative implications because it enables resistance to the lack of water. The phenomenon is still not well understood at molecular level. Thus, a good model invertebrate species for the research is required. The best known anhydrobiotic invertebrates are tardigrades (Tardigrada), considered to be toughest animals in the world. Hypsibius. exemplaris is one of the best studied tardigrade species, with its name "exemplaris" referring to the widespread use of the species as a laboratory model for various types of research. However, av

The biomedical potential of tardigrade proteins: A review.

Tardigrades are ubiquitous microinvertebrates exhibiting extreme tolerance to various environmental stressors like low and high temperatures, lack of water, or high radiation. Although exact pathways behind the tardigrade extremotolerance are yet to be elucidated, some molecules involved have been identified. Their evidenced properties may lead to novel opportunities in biomedical and pharmacological development. This review aims to present the general characteristics of tardigrade intrinsically disordered proteins (TDPs: Dsup, CAHS, SAHS, MAHS) and late embryogenesis-abundant proteins (LEA) a

Modified meiosis in the tardigrade Hypsibius exemplaris maintains heterozygosity across the genome.

In asexual reproduction, meiosis must be bypassed or altered to maintain ploidy from mother to daughter without fertilization. Most of the ways meiosis can be modified to this end are expected to reduce heterozygosity within individuals; however, many asexual species are highly heterozygous. Asexual reproduction is especially common among species of microscopic, desiccation-tolerant animals such as rotifers, nematodes, and tardigrades, but the cellular and genetic mechanisms underlying asexual reproduction have not been definitively documented in any species of tardigrade. Here, we show that t

A Non-Invasive, Label-Free Method for Examining Tardigrade Anatomy Using Holotomography.

BACKGROUND/OBJECTIVES: Holotomography is an advanced imaging technique that enables high-resolution, three-dimensional visualization of microscopic specimens without the need for fixation or staining. Here we aim to apply holotomography technology to image live Hypsibius exemplaris in their native state, avoiding invasive sample preparation procedures and phototoxic effects associated with other imaging modalities. METHODS: We use a low concentration of 7% ethanol for tardigrade sedation and sample preparation. Holotomographic images were obtained and reconstructed using the Tomocube HT-X1 sy

Genome of the tardigrade Cucumibius annulatus: functional annotation and specific proteins.

Tardigrades are microscopic invertebrates renowned for their capacity to survive extreme environmental conditions, yet stress tolerance strategies vary substantially across the phylum. Here we present the first genome assembly and annotation for the superfamily Isohypsibioidea - the most basal group within the order Parachela - represented by freshwater species Cucumibius annulatus. The draft genome (73 Mb, N50 = 96.4 kb, BUSCO 87.1%) was assembled from short-read data. Orthogroup-based comparison against three reference tardigrades and two ecdysozoan outgroups (Caenorhabditis elegans, Drosoph

Ultrastructure of Oogenesis in Actinarctus doryphorus (Heterotardigrada, Arthrotardigrada, Tanarctidae).

Ultrastructural studies of oogenesis in Heterotardigrada are limited. Here, we examined the ovarian organization of Actinarctus doryphorus using transmission electron microscopy and three-dimensional reconstruction. Four female specimens at different developmental stages, including a juvenile, were analyzed. The ovary is a single sac-like structure dorsally above the digestive tract, partially supported by longitudinal dorsal muscles. Early germ cells form anterior clusters interconnected by cytoplasmic bridges. Previtellogenic oocytes contain numerous mitochondria clustered around a prominent

Nematode-derived drosomycin-type antifungal peptides provide evidence for plant-to-ecdysozoan horizontal transfer of a disease resistance gene.

Drosomycin-type antifungal peptides (DTAFPs) are key innate immunity components of Drosophila and plants and confer resistance to fungal infection. Here we report the discovery of a multigene family of DTAFPs, comprising of 15 members (termed cremycin-1 to crymycin-15), in the fruit nematode Caenorhabditis remanei. Cremycins share highly similar amino-acid sequences and identical precursor organization to drosomycins. Of the 15 cremycin genes, 10 are found to be transcriptionally active and 6 are upregulated after fungal challenge. Synthetic cremycin-5 is active on filamentous fungi and a seri

A Versatile Technique for Measuring the Stiffness of Small Organisms Based on Microscopic Observation of Cantilever Strain.

Small organisms such as water fleas and nematodes change their body stiffness in response to external stimuli and environmental alterations. Atomic force microscopy (AFM) is commonly used for stiffness measurement. However, the compression of the object by the cantilever is performed in the same direction as the observation, and this makes it difficult to observe the deformation of the object. Furthermore, the placement of the cantilever relative to the object is spatially quite limited and the driving distance is also very short: these limitations complicate measurements in organisms of vario

Fast autooxidation of a bis-histidyl-ligated globin from the anhydrobiotic tardigrade, Ramazzottius varieornatus, by molecular oxygen.

Tardigrades, a phylum of meiofaunal organisms, exhibit extraordinary tolerance to various environmental conditions, including extreme temperatures (-273 to 151°C) and exposure to ionizing radiation. Proteins from anhydrobiotic tardigrades with homology to known proteins from other organisms are new potential targets for structural genomics. Recently, we reported spectroscopic and structural characterization of a hexacoordinated haemoglobin (Kumaglobin [Kgb]) found in an anhydrobiotic tardigrade. In the absence of its exogenous ligand, Kgb displays hexacoordination with distal and proximal hist

Tardigrades as a potential model organism in space research.

Exposure of living organisms to open space requires a high level of tolerance to desiccation, cold, and radiation. Among animals, only anhydrobiotic species can fulfill these requirements. The invertebrate phylum Tardigrada includes many anhydrobiotic species, which are adapted to survive in very dry or cold environmental conditions. As a likely by-product of the adaptations for desiccation and freezing, tardigrades also show a very high tolerance to a number of other, unnatural conditions, including exposure to ionizing radiation. This makes tardigrades an interesting candidate for experiment
