[Taurodeoxycholic acid in the treatment of primary biliary cirrhosis. A controlled study in comparison to ursodeoxycholic acid].

Thirty patients with primary biliary cirrhosis were enrolled; 25 completed the study period (6 months). Of these, 12 were randomised to TUDCA treatment and 13 to UDCA. Dosage range 12-15 mg/kg daily for both groups which were comparable at to age, sex, duration and stage of the disease, as well as for all liver laboratory tests. Altogether, the results obtained were satisfactory in both groups. What was surprising was that clinical experience failed to confirm the greater efficacy of TUDCA compared to UDCA. On the contrary, UDCA appeared to be more effective than TUDCA in improving liver funct

The microbiota-derived bile acid taurodeoxycholic acid improves hepatic cholesterol levels in mice with cancer cachexia.

Alterations in bile acid profile and pathways contribute to hepatic inflammation in cancer cachexia, a syndrome worsening the prognosis of cancer patients. As the gut microbiota impinges on host metabolism through bile acids, the current study aimed to explore the functional contribution of gut microbial dysbiosis to bile acid dysmetabolism and associated disorders in cancer cachexia. Using three mouse models of cancer cachexia (the C26, MC38 and HCT116 models), we evidenced a reduction in the hepatic levels of several secondary bile acids, mainly taurodeoxycholic (TDCA). This reduction in hep

Recombinant Bile Salt Hydrolase Enhances the Inhibition Efficiency of Taurodeoxycholic Acid against Clostridium perfringens Virulence.

Clostridium perfringens is the main pathogen of chicken necrotic enteritis (NE) causing huge economic losses in the poultry industry. Although dietary secondary bile acid deoxycholic acid (DCA) reduced chicken NE, the accumulation of conjugated tauro-DCA (TDCA) raised concerns regarding DCA efficacy. In this study, we aimed to deconjugate TDCA by bile salt hydrolase (BSH) to increase DCA efficacy against the NE pathogen C. perfringens. Assays were conducted to evaluate the inhibition of C. perfringens growth, hydrogen sulfide (H2S) production, and virulence gene expression by TDCA and DCA. BSH

Taurodeoxycholic acid-YAP1 upregulates OTX1 in promoting gallbladder cancer malignancy through IFITM3-dependent AKT activation.

Orthodenticle homeobox (OTX1) is reported to be involved in numerous cancers, but the expression level and molecular function of OTX1 in gallbladder cancer (GBC) remain unknown. Here, we found the elevated level of OTX1 associated with poor prognosis in human gallbladder cancer. In vitro and in vivo studies of human gallbladder cancer cell lines demonstrated that overexpression of OTX1 promoted cell proliferation, whereas the downregulation inhibited it. Additionally, we found a tight correlation between the serum level of taurodeoxycholic acid (TDCA) and OTX1 expression. TDCA-induced activati

Taurodeoxycholic, taurocholic, and glycocholic acids promote hepatic gluconeogenesis via TGR5 in dairy cows.

BACKGROUND: Ruminants and monogastric animals exhibit significant differences in gluconeogenic efficiency. In dairy cows, hepatic gluconeogenesis serves as the primary source of glucose. Metabolites modulate gluconeogenesis efficiency through allosteric regulation, redox state, and signal transduction pathways. However, the liver-enriched metabolites that regulate hepatic gluconeogenesis in dairy cows and their specific regulatory mechanisms remain incompletely characterized. RESULTS: Six Holstein dairy cows and six Duroc × (Landrace × Yorkshire) (DLY) crossbred pigs served as research subjec

Taurodeoxycholic acid alleviates intestinal inflammation by modulating gut microbiota and TGR5-NF-kappaB axis in DSS-induced colitis.

Taurodeoxycholic acid (TDCA), a metabolite of cholesterol, has been shown to be able to regulate various inflammatory responses and improve intestinal inflammation. However, its potential to alleviate colitis through modulating the gut microbiota-bile acid axis remains unclear. Mice were simultaneously administered dextran sulfate sodium (DSS) and TDCA via drinking water to estimate the effect of TDCA on colitis. TDCA-treated mice showed markedly relieved DSS-induced colitis. 16S rDNA sequencing revealed TDCA selectively remodeled the gut microbiota, notably decreasing the abundances of Desulf

Multi-omics reveals that gut microbiota-derived taurodeoxycholic acid mediates the protective effect of Lactobacillus rhamnosus GG against LPS-induced liver injury.

Gut microbiota-derived lipopolysaccharide (LPS) is a critical mediator in the pathogenesis of nonalcoholic fatty liver disease and metabolic syndrome. The liver plays a crucial role in mediating immune responses and detoxifying endotoxins through bile secretion. However, the precise role of bile acid metabolism in LPS-induced liver injury and the underlying regulatory mechanisms remain poorly understood. RNA sequencing of liver and ileum tissues, combined with targeted metabolomic profiling of liver and cecal chyme and full-length 16S rRNA sequencing of cecal microbiota, revealed that LPS disr

Taurodeoxycholic acid impairs the function and expression of organic anion transporter 2 through farnesoid X receptor activation during acute kidney injury to chronic kidney injury transition.

Acute kidney injury (AKI) is a worldwide health issue with risk of progressing to chronic kidney disease (CKD). The kidney harbors various transporters that may be involved in certain disease processes, in addition to mediating drug transport. Kidney injury may compromise the effectiveness and safety of medications by disturbing functions of these transporters. However, so far, there is a poor understanding of the changes in transporters and their mechanisms during AKI-to-CKD transition. In the present study, the expression of organic anion transporter 2 (OAT2) was found to be gradually decrea

Enterobacteria modulate intestinal bile acid transport and homeostasis through apical sodium-dependent bile acid transporter (SLC10A2) expression.

In our study, ampicillin (AMP)-mediated decrease of enterobacteria caused increases in hepatic bile acid concentration through (at least in part) elevation of bile acid synthesis in C57BL/6N mice. We investigated the involvement of enterobacteria on intestinal bile acid absorption in AMP-treated mice in the present study. Fecal enterobacterial levels and fecal bile acid excretion rates were markedly decreased in mice treated with AMP (100 mg/kg) for 3 days, whereas bile acid concentrations in portal blood were significantly increased compared with those in mice treated with a vehicle. Effect of acid, pepsin, and bile acid on the stenotic progression of traumatized subglottis.

BACKGROUND AND AIMS: Gastroesophageal reflux disease is related to various laryngeal sequelae. However, there is a lack of established reflux animal models covering longer observation periods. We evaluated the effects of acid, pepsin, and bile acid on healing of the traumatized subglottis in a simulated reflux model. METHODS: In the first experiment, 48 rabbits were inflicted with a posterior subglottic injury using a diode laser, and there were 10 unwounded controls. After catheter insertion under transoral endoscopic guidance, animals were randomly assigned to one of the four groups: acid r

Effects of sphincter of Oddi bypass on bile acid metabolism in fed and fasted intact and cholecystectomized dogs.

The effects of cholecystectomy and sphincter of Oddi bypass on bile acid (BA) metabolism in dogs have been studied. Cholecystectomy and sphincter bypass decreased the BA pool half-life and increased the percent of taurodeoxycholic acid in the pool. A 48-hour fast had no effect on total BA pool size of intact and intact sphincter-of-Oddi-bypassed dogs but caused a marked decrease in cholecystectomized dogs. It was concluded that while the sphincter of Oddi is unnecessary to maintain bile acid pool size in fasting dogs, the gallbladder is. Alimentation is necessary to maintain pool size in chole

Impairment of rat oesophageal muscle contractility associated with experimental non-erosive oesophageal mucosal damage.

NEW FINDINGS: What is the central question of this study? Is the responsiveness of isolated segments of the rat oesophagus to contractile or relaxant stimuli susceptible to acute luminal exposure of the oesophagus to an acid solution that contains pepsin and bile salt? What is the main finding and its importance? The study reveals that luminal acidity is an important factor that disrupts barrier function in the oesophagus to allow the diffusion of noxious agents, such as bile acid, that alter the contractile status of the oesophageal body, even in the absence of inflammation. ABSTRACT: We inv

Stachyose Attenuates Hepatic Cholesterol Deposition in Nonalcoholic Fatty Liver Mice via Gut Microbiota-Driven Bile Acid Profile Remodeling.

Nonalcoholic fatty liver disease (NAFLD), driven by liver lipid accumulation, presents a global health challenge. Bile acid metabolism critically regulates cholesterol homeostasis, but whether prebiotic stachyose can modulate it for NAFLD intervention is unknown. In NAFLD mice induced by a high-fat-high-sucrose diet, stachyose (200 mg/kg bw, 20 weeks) expanded the bile acid pool, elevating the primary bile acids (such as cholic acid and chenodeoxycholic acids) in both serum and liver while reducing secondary bile acid levels. Mechanistically, stachyose upregulated the expression of bile acid s

A small intestinal bile acid modulates the gut microbiome to improve host metabolic phenotypes following bariatric surgery.

Bariatric surgical procedures such as sleeve gastrectomy (SG) provide effective type 2 diabetes (T2D) remission in human patients. Previous work demonstrated that gastrointestinal levels of the bacterial metabolite lithocholic acid (LCA) are decreased after SG in mice and humans. Here, we show that LCA worsens glucose tolerance and impairs whole-body metabolism. We also show that taurodeoxycholic acid (TDCA), which is the only bile acid whose concentration increases in the murine small intestine post-SG, suppresses the bacterial bile acid-inducible (bai) operon and production of LCA both in vi

Cloning and analysis of bile salt hydrolase genes from Lactobacillus plantarum CGMCC No. 8198.

Genes coding for bile salt hydrolase of Lactobacillus plantarum CGMCC 8198, a novel probiotic strain isolated from silage, were identified, analyzed and cloned. L. plantarum strongly resisted the inhibitory effects of bile salts and also decreased serum cholesterol levels by 20% in mice with hypercholesterolemia. Using RT-PCR analysis, bsh2, bsh3 and bsh4 were upregulated by bile salts in a dose-dependent manner. All three bsh genes had high similarity with those of other Lactobacillus strains. All three recombinant BSHs had high activities for the hydrolysis of glycodeoxycholic acids and taur
