Initial identification of genomic islands in Taylorella equigenitalis and Taylorella asinigenitalis and their distribution in isolates from around the world.

Taylorella equigenitalis, the causative agent of contagious equine metritis (CEM), is endemic in several countries throughout the world and impacts the global movement of horses. It presents with symptoms ranging from non-clinical to endometritis, vaginitis, and acute infertility in females, while males remain asymptomatic. Studies of the disease have focused primarily on understanding pathology, transmission dynamics, diagnostics, and phenotypic and genotypic characteristics. The goal of this study was to establish a more comprehensive global phylogenetic structure and generate reference-qual

Development of a real time PCR for the detection of Taylorella equigenitalis directly from genital swabs and discrimination from Taylorella asinigenitalis.

A discriminatory real time PCR for the detection of Taylorella equigenitalis, the causative agent of contagious equine metritis (CEM), and the related species T. asinigenitalis was developed for the direct examination of genital swabs. The 112bp amplicons produced from the two species were discriminated from each other using TaqMan probes labelled with different fluorophores. The TaqMan PCR was shown to be specific for the 16S ribosomal DNA of the two species of taylorella and did not cross-hybridise with the 16S ribosomal DNA of other bacteria tested. Direct amplification from genital swabs w

Prevalence and persistence of Taylorella asinigenitalis in male donkeys.

This study was undertaken to investigate the prevalence of Taylorella asinigenitalis in a subset of the donkey population of Michigan and in other equids on farms on which the organism was identified. Other aims were to further characterize the carrier state in terms of persistence and preferred sites of colonization of T. asinigenitalis in the male donkey as well as determine the genotype of any isolates of the organism. Initial testing of 43 donkeys and 1 mule turned up 4 (9.3%) donkeys culture positive for T. asinigenitalis. The 4 culture-positive donkeys resided on 2 farms accommodating a

Production and characterization of monoclonal antibodies against Taylorella equigenitalis.

Monoclonal antibodies were produced against Taylorella equigenitalis using two reference strains. Out of the 79 hybridoma clones shown to express antibodies to T equigenitalis by indirect immunofluorescence assay, 16 were selected for monoclonal antibody production and characterization. These clones recognized different field strains of T equigenitalis isolated in France. They showed no cross-reaction with bacterial strains with previously reported antigenic cross-reactivity, nor did they react with other bacteria commonly found in genital flora. The epitopes recognized by eight of the monoclo

Taylorella equigenitalis infections in Poland - results of current diagnostic investigations.

INTRODUCTION: Contagious equine metritis (CEM) is a cosmopolitan infectious, venereal disease of equids caused by the bacterium Taylorella equigenitalis. Its frequently asymptomatic course leads to its prevalence sometimes being underestimated, and knowledge of the spread of infections with its causative agent is insufficient. The aim of this study is to summarise and present data on the incidence of T. equigenitalis infections in horses in Poland. MATERIAL AND METHODS: In the years 2018-2023, routine laboratory tests of horses for CEM were carried out. Between 52 and 99 horses were examined

Preservation of viable Taylorella equigenitalis in different commercially available transport systems.

The cultural diagnosis of the causal agent of contagious equine metritis (Taylorella equigenitalis) using transport swabs is challenging. Swabs must be placed in Amies charcoal medium, refrigerated during transport, and plated out at the laboratory no later than 48 h after sampling. In this study, the viability of T. equigenitalis strain CIP 79.7T in 11 commercial swab transport systems was initially compared at 1 day and 2 days of storage at ambient (20 ± 3 °C) or refrigerated (5 ± 3 °C) temperature. The four best swab transport systems, systems B, E, F (used as the reference) and K, were the

Growth kinetics of Taylorella equigenitalis and its survival in straw-based bedding.

Taylorella equigenitalis, the causative agent of contagious equine metritis, is a fastidious bacterium with limited tolerance to extra-host conditions; however, quantitative data on its growth dynamics and survival outside the host remain scarce. This study evaluated the growth of T. equigenitalis under aerobic and microaerophilic conditions in liquid medium and its survival in wet and dry straw-based bedding under controlled laboratory conditions.Growth was assessed at 20 °C and 37 °C using real-time optical density monitoring and viable cell counts. No proliferation was observed at 20 °C, wh

Differences between Taylorella equigenitalis strains in their invasion of and replication in cultured cells.

The ability of Taylorella equigenitalis, the causative agent of contagious equine metritis, to invade and replicate in equine derm cells was studied. The kinetics of invasion and replication were determined for four T. equigenitalis strains. On the basis of these experiments, a simpler assay in which the invasive as well as the replicative properties of a particular strain could be determined was developed. This assay was used to characterize 32 strains, which had previously been typed by field inversion gel electrophoresis of genomic restriction fragments. The invasiveness of T. equigenitalis

Acute Endometritis due to Taylorella equigenitalis Transmission by Insemination of Cryopreserved Stallion Semen.

Taylorella equigenitalis can be transmitted during artificial insemination. This report describes clinical T. equigenitalis transmission by cryopreserved stallion semen. T. equigenitalis isolates from a mare's vaginal discharge and semen from the same batch of the cryopreserved semen used for the insemination gave identical API ZYM, antibiotic susceptibility, and multilocus sequence typing results (ST-46); furthermore, the multilocus sequence typing lineage ST-46 is known to circulate in the country of semen collection. These results support the need for strict contagious equine metritis scree

Quantitative assessment of the risks of reducing the routine swabbing requirements for the detection of Taylorella equigenitalis.

The transmission of contagious equine metritis (CEM) on stud farms in Britain, Ireland and other European countries is prevented by following the recommendations in the Horserace Betting Levy Board's Code of Practice on CEM. A quantitative risk assessment was undertaken to estimate the likely impact of removing the recommendation, from the 2002 code, to culture endometrial or cervical swabs microaerophilically for the presence of Taylorella equigenitalis, the causative organism. The scientific literature was reviewed for evidence about the anatomical distribution of T. equigenitalis at differe

Identification of anatomic features of the equine clitoris as potential growth sites for Taylorella equigenitalis.

A median clitoral sinus, as a space canalized from epithelial cells, was distinguishable developmentally in equine fetuses from 33-mm crown-rump length (CRL) to 500-mm CRL (including a mule of 21-mm CRL). In saggital sections of the clitoris of a 480-mm CRL fetus, indentations under the transverse frenular fold were identified as lateral sinuses of the clitoris. Unlike the median sinus, they were shallow; it therefore could not be anatomically substantiated that the lateral sinuses were of sufficient depth to support the growth of the partial anaerobe Taylorella equigenitalis, the organism of

Prevalence, genomic diversity and emerging antimicrobial resistance of Taylorella asinigenitalis in Grand Noir du Berry breeding donkeys.

Taylorella asinigenitalis is a fastidious Gram-negative bacterium of the equine genital tract, found more commonly in donkeys than horses. Although considered non-pathogenic, it is closely related to Taylorella equigenitalis, the agent of contagious equine metritis, and has been linked to experimental and natural cases of endometritis. The SAFRAN project (French acronym for "Sauvegarde de la fertilité de l'âne", meaning "Preservation of donkey fertility"), launched in 2024 by the French National Research Institute for Agriculture, Food and Environment (INRAE) with support from the Centre-Val d

Evaluation of a newly developed real-time PCR for the detection of Taylorella equigenitalis and discrimination from T. asinigenitalis.

A 'culture-LightCycler PCR' assay has been developed for the detection of Taylorella equigenitalis, the causative agent of contagious equine metritis (CEM) in horses. The primers and hybridisation probes were derived from the 16S rDNA sequence. Their specificity was determined in two closely related organisms and six commensal bacteria of the genital tract. The assay was specific for T. equigenitalis and discriminates T. asinigenitalis isolates. It also avoids misidentifications of morphologically and phenotypically similar organisms. The sensitivity was evaluated in comparison to a standard b

Possible misidentification of Bacteroides sp., probably B. ureolyticus as Taylorella equigenitalis: implications for the laboratory diagnosis of CEM.

A wild-type isolate with similar morphological and phenotypic properties to Taylorella equigenitalis, the causative bacterial agent of contagious equine metritis (CEM), was referred for molecular identification by PCR amplification of the 16S rRNA gene. A species-specific PCR failed to yield a product compatible with that of T. equigenitalis. The direct sequencing of the universal 16S rRNA PCR amplicon suggested the presence of a Bacteroides sp., probably Bacteroides ureolyticus, with no consequent effects on the movement and transportation of the animal. Adoption of such a molecular means of
