Experimental Challenge of Chinook Salmon (Oncorhynchus tshawytscha) With Tenacibaculum maritimum and Tenacibaculum dicentrarchi Fulfils Koch's Postulates.

The bacterial skin disease tenacibaculosis, caused by Tenacibaculum species, affects numerous economically important marine fish, including salmonids. This study reports the ability of three Tenacibaculum maritimum strains, belonging to different molecular O-AGC types, and a single Tenacibaculum dicentrarchi strain to induce tenacibaculosis in farmed Chinook salmon (Oncorhynchus tshawytscha, Walbaum 1792) in Aotearoa New Zealand. Naïve Chinook salmon were exposed to T. maritimum (2 × 108 cells/mL) and T. dicentrarchi (2 × 107 cells/mL) by immersion using natural seawater. Clinical signs of ten

Comparative genomic assessment of members of genus Tenacibaculum: an exploratory study.

Tenacibaculosis is an ulcerative skin disorder that affects finfish. It is caused by members of the genus Tenacibaculum, resulting in eccentric behavioural changes, including anorexia, lethargy, and abnormal swimming patterns that often result in mortality. Currently, species suspected of causing fish mortality include T. ovolyticum, T. gallaicum, T. discolor, T. finnmarkense, T. mesophilum, T. soleae, T. dicentrarchi, and T. maritimum. However, pathogenic members and the mechanisms involved in disease causation, progression, and transmission are limited due to the inadequate sequencing effort

New Zealand rickettsia-like organism and Tenacibaculum maritimum vaccine efficacy study.

A cohort of Chinook salmon juveniles was vaccinated, with an autogenous bivalent vaccine against New Zealand RLOs (NZ-RLO1) and Tenacibaculum maritimum. A proportion of the cohort was not vaccinated to act as controls. At smoltification, the fish were challenged with NZ-RLO1, NZ-RLO2, or T. maritimum. We found that challenge with T. maritimum by immersion in (7.5 × 105  cfu/mL of water) did not yield any pathology. Challenge with RLOs produced clinical signs that were more or less severe depending on the challenge route, dose or vaccination status. Survival was significantly higher for vaccina

New salmonid hosts for Tenacibaculum species: Expansion of tenacibaculosis in Chilean aquaculture.

The success and sustainability of Chilean aquaculture largely depends on the control of endemic and emerging pathogens, including several species of the genus Tenacibaculum. Tenacibaculum dicentrarchi and "Tenacibaculum finnmarkense" have been detected and confirmed in Chilean Atlantic salmon (Salmo salar). However, no outbreaks of tenacibaculosis in rainbow trout (Oncorhynchus mykiss) or coho salmon (Oncorhynchus kisutch) have been reported, either in Chile or globally. The aims of this study were to determine whether the mortalities recorded for rainbow trout and coho salmon from five marine

Genotyping of Tenacibaculum maritimum isolates from farmed Atlantic salmon in Western Canada.

Mouthrot infections (bacterial stomatitis) have a significant impact on the Atlantic salmon aquaculture industry in Western Canada due to economic losses and fish welfare. Bacteria isolated from lesions in the field have been identified as Tenacibaculum maritimum. Mouthrot is different to classical tenacibaculosis, which is most commonly associated with ulcerative lesions, frayed fins and tail rot. The marine fish pathogen T. maritimum is found worldwide; however, in Western Canada, the knowledge of the genetic profile of T. maritimum is limited. This study looked at increasing this knowledge

Pathology of experimentally induced mouthrot caused by Tenacibaculum maritimum in Atlantic salmon smolts.

Mouthrot, caused by Tenacibaculum maritimum is a significant disease of farmed Atlantic salmon, Salmo salar on the West Coast of North America. Smolts recently transferred into saltwater are the most susceptible and affected fish die with little internal or external clinical signs other than the characteristic small (usually < 5 mm) yellow plaques that are present inside the mouth. The mechanism by which these smolts die is unknown. This study investigated the microscopic pathology (histology and scanning electron microscopy) of bath infected smolts with Western Canadian T. maritimum isolates

Outer membrane vesicles (OMVs) from Tenacibaculum maritimum as a potential vaccine against fish tenacibaculosis.

Outer membrane vesicles (OMVs) have been gained increasing attention in vaccinology due to their ability to induce strong protective humoral and cell-mediated immunity. The Gram-negative bacterium Tenacibaculum maritimum, the causative agent of marine tenacibaculosis, poses a significant challenge to the global aquaculture industry due to its difficult prophylaxis. In previous studies, we demonstrated that OMV production is a key virulence mechanism in T. maritimum. Building on this, the present study aimed to evaluate the efficacy of a natural, encapsulated multi-antigen vaccine made from adj

Discovery and Biosynthesis of Antimicrobial Phenethylamine Alkaloids from the Marine Flavobacterium Tenacibaculum discolor sv11.

The bacterial genus Tenacibaculum has been associated with various ecological roles in marine environments. Members of this genus can act, for example, as pathogens, predators, or episymbionts. However, natural products produced by these bacteria are still unknown. In the present work, we investigated a Tenacibaculum strain for the production of antimicrobial metabolites. Six new phenethylamine (PEA)-containing alkaloids, discolins A and B (1 and 2), dispyridine (3), dispyrrolopyridine A and B (4 and 5), and dispyrrole (6), were isolated from media produced by the predatory bacterium Tenacibac

Characterization of the Immune Response of Atlantic Salmon to Tenacibaculum dicentrarchi Using In Vivo and In Vitro Approaches.

Tenacibaculosis, primarily caused by Tenacibaculum dicentrarchi in Chile, is now the second leading infectious cause of mortality in Chilean Atlantic salmon (Salmo salar) farming, yet the host immune response remains uncharacterized. Here, we combined in vitro and in vivo approaches to characterize the immunopathological interaction between Atlantic salmon and a Chilean T. dicentrarchi isolate. Primary head kidney macrophages stimulated with live bacteria showed early and sustained upregulation of tnfα, il1β, and il8 (4-24 h), followed by late induction of inos at 24 h, consistent with a class

Tenacibaculum salmonis sp. nov., isolated from Atlantic salmon (Salmo salar L.) fish farmed in Chile.

Strain P3-BQ1T is a Gram-negative, aerobic, non-spore-forming bacterium with gliding motility and filamentous cells. It was isolated in 2018 from the gills of a diseased Atlantic salmon (Salmo salar) during an outbreak of tenacibaculosis in a Chilean fish farm. Phylogenetic analysis based on 16S rRNA gene sequencing confirmed that strain P3-BQ1T belongs to the genus Tenacibaculum and is most closely related to Tenacibaculum dicentrarchi 35/09T (98.53%), 'Tenacibaculum pacificus' 18-2881-A (98.46%), Tenacibaculum aestuariivivum JDTF-79T (98.46%), Tenacibaculum finnmarkense TNO010 (97.85%) and T

Immune Response of Atlantic Salmon (Salmo salar) Experimentally Infected by Immersion Challenge With Tenacibaculum dicentrarchi.

Tenacibaculosis, caused by Tenacibaculum dicentrarchi, is an emerging bacterial disease affecting salmonid aquaculture worldwide; however, host-pathogen interactions and the immune response of Atlantic salmon (Salmo salar) remain poorly understood. This study evaluated the expression of 20 immune-related genes in Atlantic salmon experimentally infected by immersion with T. dicentrarchi and in uninfected controls under identical conditions. The 10-day challenge trial included sampling every two days (skin/muscle, gills, spleen and head kidney) for gene expression analyses. Koch's postulates wer

Tenacibactins K-M, cytotoxic siderophores from a coral-associated gliding bacterium of the genus Tenacibaculum.

HPLC/DAD-based chemical investigation of a coral-associated gliding bacterium of the genus Tenacibaculum yielded three desferrioxamine-class siderophores, designated tenacibactins K (1), L (2), and M (3). Their chemical structures, comprising repeated cadaverine-succinic acid motifs terminated by a hydroxamic acid functionality, were elucidated by NMR and negative MS/MS experiments. Compounds 1-3 were inactive against bacteria and a yeast but displayed cytotoxicity against 3Y1 rat embryonic fibroblasts and P388 murine leukemia cells at GI50 in submicromolar to micromolar ranges. Isolation, identification, virulence potential and genomic features of Tenacibaculum piscium isolates recovered from Chilean salmonids.

Tenacibaculum piscium, a gram-negative bacterium isolated from the skin ulcers of sea-farmed fish, has only been described in Norway. In the present study, we examined 16 Chilean Tenacibaculum isolates recovered from different organs in moribund and dead Atlantic salmon (Salmo salar), Rainbow trout (Oncorhynchus mykiss) and Coho salmon (Oncorhynchus kisutch) cultured at different fish farms between 2014 and 2018. The present study applied biochemical, phenotypic, fatty acid and whole-genome sequence-based analyses to confirm the taxonomic status of the Chilean isolates. The obtained results ar

Tenacibaculum maritimum can boost inflammation in Dicentrarchus labrax upon peritoneal injection but cannot trigger tenacibaculosis disease.

INTRODUCTION: Despite being a bacterial pathogen with devastating consequences, Tenacibaculum maritimum's pathogenesis is not fully understood. The aim of the present study was to elucidate if different inoculation routes (intraperitoneal - i.p - injection and bath challenge - known to induce mortality) can induce tenacibaculosis (i.e., using the same T. maritimum inoculum), as well as evaluate the short-term immune response of European sea bass (D. labrax). Additionally, the host response against i.p. injection of extracellular products (ECPs) was also studied. METHODS: Fish were i.p. challe

Identification, Characterisation, and Pathogenic Potential of Chilean Isolates of Tenacibaculum finnmarkense Recovered From Coinfection in Farmed Salmonids.

The Chilean salmon farming industry faces major challenges from bacterial diseases, notably tenacibaculosis, which is primarily caused by Tenacibaculum maritimum and T. dicentrarchi. However, T. finnmarkense was recently identified as an emerging pathogen. This bacterium was previously described only in Norway, where two genomovars (finnmarkense and ulcerans) were reported. The present study focuses on the phenotypic and genetic characterisation of isolates recovered from salmonids in Chile to elucidate intraspecific diversity, identify the dominant genomovar, and evaluate pathogenic potential

Dietary 1.3-1.6 yeast β-glucans enhance immune response and disease resilience in European seabass challenged with Tenacibaculum maritimum.

INTRODUCTION: Nutritional strategies have emerged as promising tools to modulate immune responses in marine fish, particularly at mucosal surfaces involved in host-pathogen interactions. Dietary immunomodulators such as yeast-derived β-glucans (BG) have gained attention for their capacity to enhance innate immune defenses, however their role in coordinating these mechanisms during bacterial infections remains incompletely understood. METHODS: This study evaluated the effects of two dietary inclusion levels (0.06 and 0.12%) of purified yeast β-1,3/1,6-glucans on the immune responses of Europea

Efficacy of a Vaccine for Atlantic Salmon (Salmo salar) Using a Tenacibaculum dicentrarchi Strain Cultured Under Iron-Limited Conditions.

Tenacibaculosis, caused by T. dicentrarchi, results in skin lesions, ulceration, yellow plaques on teeth, and haemorrhaging in the operculum, peduncle and pectoral fins. It is the second leading cause of mortality in farmed Atlantic salmon (Salmo salar) in Chile. Currently, no vaccine is available to prevent the disease. In silico and in vivo studies have shown that iron plays a key role in T. dicentrarchi infection. We hypothesised that culturing the TdCh05 vaccine strain under iron-limiting conditions (i.e., with non-assimilable iron chelator 2.2'-dipyridyl [DIP]) would enhance Atlantic salm
