Derivatives of Tenuazonic Acid as Potential New Multi-Target Anti-Alzheimer's Disease Agents.

Alzheimer's disease (AD) is generally recognized as a multifactorial neurodegenerative pathology with an increasing impact on society. Tenuazonic acid (TA) is a natural compound that was recently identified as a potential multitarget ligand with anti-cholinesterase, anti-amyloidogenic and antioxidant activities. Using its structure as a chemical scaffold, we synthesized and evaluated new derivatives (1-5), including tenuazonic-donepezil (TA-DNP) hybrids (4 and 5) due to the clinical importance of the anti-AD drug donepezil. These novel compounds all achieved activity in the micromolar range to

Opposing immunomodulatory effects of the Alternaria mycotoxin tenuazonic acid in immune and intestinal epithelial cells.

Tenuazonic acid (TeA) is one of the most frequently detected Alternaria mycotoxins in contaminated food. Despite its frequent occurrence, its immunomodulatory effects remain insufficiently characterized. Therefore, the present study investigated the impact of TeA on inflammatory signaling and cytokine regulation in monocytes and intestinal epithelial cell (IEC) models. NF-κB activity was assessed using a reporter gene assay in THP1-Lucia™ monocytes, while cytokine mRNA expression and protein secretion were quantified in Caco-2 and HCEC-1CT cells by qRT-PCR and ELISA, respectively. In THP-1 mon

Rational design and herbicidal activity of tenuazonic acid derivatives based on photosystem II D1 protein.

BACKGROUND: Natural products are an important source for the development of herbicides. Tenuazonic acid (TeA) is a natural phytotoxin produced by phytopathogenic fungi that strongly inhibits photosystem II (PS II) by preventing electron flow from the primary quinone acceptor (QA) of PS II to the secondary quinone acceptor (QB), thereby directly disrupting the key chain of plant photosynthesis. RESULTS: A series of TeA derivatives were rationally designed and synthesized based on the PS II D1 protein, and their herbicidal activities were evaluated. Compound C11 achieved > 90% control of Abutil

Integrated histone and proteome analyses reveal convergent and distinct hepatotoxic mechanisms of tenuazonic acid and deoxynivalenol.

Mycotoxins are widespread dietary contaminants whose health impacts are expected to intensify under climate change. Although their mechanisms of toxicity remain incompletely understood, epigenetic dysregulation has been increasingly implicated. Here, mass spectrometry-based multi-omics was used to profile histone post-translational modifications and proteome dynamics in HepG2 cells exposed to seven mycotoxin conditions. Time-resolved analyses identified tenuazonic acid as the dominant cellular disruptor, inducing alterations in H3K27 and H1 variants, and revealing a previously unrecognized oxi

The mechanism behind tenuazonic acid-mediated inhibition of plant plasma membrane H+-ATPase and plant growth.

The increasing prevalence of herbicide-resistant weeds has led to a search for new herbicides that target plant growth processes differing from those targeted by current herbicides. In recent years, some studies have explored the use of natural compounds from microorganisms as potential new herbicides. We previously demonstrated that tenuazonic acid (TeA) from the phytopathogenic fungus Stemphylium loti inhibits the plant plasma membrane (PM) H+-ATPase, representing a new target for herbicides. In this study, we further investigated the mechanism by which TeA inhibits PM H+-ATPase and the effe

Alternaria toxin-induced resistance in rose plants against rose aphid (Macrosiphum rosivorum): effect of tenuazonic acid.

Many different types of toxins are produced by the fungus, Alternaria alternata (Fr.) Keissler. Little is known, however, regarding the influence of these toxins on insects. In this study, we investigated the toxin-induced inhibitory effects of the toxin produced by A. alternata on the rose aphid, Macrosiphum rosivorum, when the toxin was applied to leaves of the rose, Rosa chinensis. The results demonstrated that the purified crude toxin was non-harmful to rose plants and rose aphids, but had an intensive inhibitory effect on the multiplication of aphids. The inhibitory index against rose aph

Structural basis of a CDR3-embedded binding mechanism in a nanobody for sensitivity enhancement toward tenuazonic acid.

Nanobodies (Nbs) have shown great potential for use in immunoassays targeting small-molecule contaminants in food safety monitoring. However, a limited understanding of their recognition mechanisms has hindered the development of high-performance Nbs and the improvement of assay performance. Herein, a previously developed nanobody (Nb) 3F9 against tenuazonic acid (TeA) was selected as a model to resolve its X-ray crystal structure. Notably, Nb3F9 adopts a typical immunoglobulin fold, with TeA deeply inserted into the complementary-determining region 3 (CDR3) and buried in a binding pocket form

Preventive role of cinnamaldehyde against tenuazonic acid- and Freund's adjuvant-induced histopathological and biochemical alterations in the mouse model.

INTRODUCTION: This study was designed to assess the protective role of cinnamaldehyde (Cin) against the synergistic effect of tenuazonic acid (TeA) and Freund's adjuvant on different organs of Swiss albino mice. METHODS: TeA was administered singly and in combination with Freund's adjuvant intra-peritoneally. The mice were divided into control (vehicle treated), mycotoxicosis-induced (MI) groups, and treatment groups. The route of administration of TeA was intra-peritoneal. The treatment group (FAICT) received Cin orally as a protective agent against TeA-induced mycotoxicosis. The effects on

Elucidating the occurrence patterns of Alternaria toxin tenuazonic acid in individual tomatoes through MALDI-MSI and LC-MS/MS.

Tomato (Solanum lycopersicum) is susceptible to Alternaria fungi, accumulating Alternaria toxins including tenuazonic acid (TeA). Previous studies have primarily focused on overall contamination levels; however, this study employs MALDI-MSI and LC-MS/MS techniques to provide the first detailed analysis of the spatial distribution and average levels of TeA in individual tomatoes. Among 77 lesioned processing tomato samples, the exceedance rate of TeA was notably high. MALDI-MSI reveals TeA's spatial distribution, while LC-MS/MS offers quantitative data but is affected by sample processing. Spec

Development and validation of a green spectrofluorimetric method using a rhodamine 6G probe and FCCCD-optimized dispersive liquid-liquid microextraction for tenuazonic acid surveillance in dried figs.

Tenuazonic acid (TeA) is a major Alternaria mycotoxin frequently contaminating dried figs, yet its routine surveillance remains constrained by reliance on costly chromatographic instrumentation. To address this analytical gap, a novel "turn-off" spectrofluorimetric method was developed for TeA determination in dried fig samples using rhodamine 6G as a cationic xanthene fluorescent probe. Given the complexity of the sugar-rich matrix, dispersive liquid-liquid microextraction was first optimized through a face-centered central composite design coupled with Derringer's desirability function. Liquid chromatographic analysis system for cyclopiazonic acid in peanuts.

A liquid chromatographic procedure has been developed for analysis of cyclopiazonic acid in peanuts. The chromatographic system specifies a C18 or C8 column loaded with 4-dodecyldiethylenetriamine and an aqueous mobile phase containing 4-dodecyldiethylenetriamine, zinc acetate, ammonium acetate, 2-propanol, and acetonitrile. Cyclopiazonic acid was extracted from peanuts with methanol-chloroform (20 + 80), partitioned into aqueous sodium bicarbonate, acidified, and back-extracted into dichloromethane. The limit of detection (280 nm UV) was approximately 4 ng and detector response was linear to

Multiple mycotoxin exposure assessment through human biomonitoring in an esophageal cancer case-control study in the Arsi-Bale districts of Oromia region of Ethiopia.

BACKGROUND: Esophageal cancer (EC) is a malignancy with a poor prognosis and a five-year survival rate of less than 20%. It is the ninth most frequent cancer globally and the sixth leading cause of cancer-related deaths. The incidence of EC has been found to vary significantly by geography, indicating the importance of environmental and lifestyle factors along with genetic factors in the onset of the disease. In this work, we investigated mycotoxin exposure in a case-control study from the Arsi-Bale districts of Oromia regional state in Ethiopia, where there is a high incidence of EC while alc

AvmM catalyses macrocyclization through dehydration/Michael-type addition in alchivemycin A biosynthesis.

Macrocyclization is an important process that affords morphed scaffold in biosynthesis of bioactive natural products. Nature has adapted diverse biosynthetic strategies to form macrocycles. In this work, we report the identification and characterization of a small enzyme AvmM that can catalyze the construction of a 16-membered macrocyclic ring in the biosynthesis of alchivemycin A (1). We show through in vivo gene deletion, in vitro biochemical assay and isotope labelling experiments that AvmM catalyzes tandem dehydration and Michael-type addition to generate the core scaffold of 1. Emerging Alternaria Toxins Drive LX-2 Cells Transdifferentiation into Myofibroblasts for Liver Fibrosis and CotA Detoxification.

Alternariol (AOH), alternariol monomethyl ether (AME), and tenuazonic acid (TeA) are three major emerging Alternaria toxins in food. In this lncRNA-mRNA omics-based study, we first decrypt the blueprint: how emerging Alternaria toxins induce hepatic stellate cell LX-2 transdifferentiation for liver fibrosis. AOH and AME caused the fibrotic marker α-smooth muscle actin, extracellular matrix collagen expression, and cell contraction, while TeA had no significant effect. Mechanistically, AOH, AME, and AAT (their combination) activated the NF-κB pathway and induced ferroptosis and AMPK/AKT/m-TOR-r

Mycotoxins and plant alkaloids in plant-based protein foods and meat alternatives: Occurrence, exposure and risk assessment.

Plant-based protein-rich foods and meat alternatives are increasingly used as substitutes or supplements to animal-based products. Despite their potential nutritional and environmental benefits, data on their contamination with natural toxins remain limited, particularly for plant alkaloids. In this study, 57 mycotoxins, 16 tropane alkaloids and 32 pyrrolizidine alkaloids were analysed in 101 plant-based protein samples, including protein isolates/concentrates, extruded products, veggie burgers and other meat alternatives, using U-HPLC-HRMS/MS. Twenty-five mycotoxins and eight alkaloids were q

DFT study of the Lewis acid mediated synthesis of 3-acyltetramic acids.

The synthesis of 3-acyltetramic acids by C-acylation of pyrrolidine-2,4-diones was studied by density functional theory (DFT). DFT was applied to the mycotoxin tenuazonic acid (TeA), an important representative of these bioactive natural compounds. Lewis acid mediated C-acylation in combination with previous pH-neutral domino N-acylation-Wittig cyclization can be used for the efficient preparation of 3-acyltetramic acids. Nevertheless, quite harsh conditions are still required to carry out this synthetic step, leading to unwanted isomerization of stereogenic centers in some cases. In the prese

Multi-Mycotoxin Quantification in Soy Protein Powders, Soymilk, Soybeans (Glycine max) and Other Legumes Using a New Targeted Differential Ion Mobility LCMS Method.

Foods contaminated with fungal mycotoxins pose a serious risk to human health. Soy and other legumes are a vital source of alternative protein, yet the rates of mycotoxin contamination are not understood. We therefore developed a sensitive stable-isotope-dilution-differential-ion-mobility-spectrometry-LC-MS method for the quantification of 43 mycotoxins in soylike matrixes. Extraction by QuEChERS-d-SPE-PSA reduced matrix effects, while Differential-Ion-Mobility-Spectrometry (DMS) decreased background-noise and enabled the selective quantification of acetyl-deoxynivalenol (ACDON) isomers.
