OBJECTIVE: To determine whether tertiary lymphoid structures (TLSs), which reflect organized immune cell aggregates present in non-lymphoid tissues, are consistent features of endometriosis lesions.
DESIGN: Detailed histopathological analysis of endometrial and lesion tissue from patients with endometriosis and controls was performed. Multiplex immunofluorescence on select samples was then conducted to identify canonical cell populations present within TLSs: CD3+ and CD8+ T-cells, CD79a+ B-cells, CD208+ dendritic cells, CD21+ follicular dendritic cells, and PNAd+ high endothelial venules.

Tertiary lymphoid structures (TLSs) are ectopic lymphoid tissues that form in response to chronic inflammation, such as in autoimmune diseases or cancer. Their presence has been increasingly recognized as a significant factor in determining patient prognosis and response to cancer treatments. The formation and development of TLSs are intricately linked to inflammatory cytokines and chemokines, which can be induced by the innate immune system. The innate immune system serves as the body's first line of defense against pathogens by producing cytokines and chemokines upon the detection of foreign

OBJECTIVE: This study aimed to investigate the presence of tertiary lymphoid structures (TLSs) and tumor-infiltrating B cells within the germinal centers of TLSs in the tumor microenvironment of thyroid cancer, utilizing a morphological approach.
MATERIALS AND METHODS: Histological samples from patients with papillary thyroid carcinoma (PTC) (n = 112) stained with hematoxylin and eosin were examined. The presence of lymphoid neogenesis in PTC was determined based on morphological features and classified according to TLS location and maturation status. Immunofluorescence staining was performed

Tertiary lymphoid structures (TLSs) are transient ectopic lymphoid-like structures potentiating the adaptive anti-tumor immunotherapy efficiency. They have been observed across diverse malignancies since they were initially found to correlate with favorable prognosis in non-small cell lung cancer. Importantly, TLSs could systematically enhance immune responses efficiency by optimizing the immune ecology of the tumor microenvironmen (TME). They have attracted considerable interest as predictive biomarkers for anti-tumor treatment responses, including immune checkpoint blockade (ICB) and potenti

Desmoplastic melanomas (DM) have unique and challenging clinical presentations and histomorphology. A characteristic feature is the presence of scattered lymphoid aggregates. However, the nature of these aggregates is not defined. We hypothesized that they may be tertiary lymphoid structures (TLS), and may be associated with programmed death ligand 1 (PD-L1) expression. We searched our tissue database for 'pure' DMs and for scars as control tissues, collected clinical information, and reviewed H&E histology. We performed multispectral imaging after staining for CD8, CD20, PNAd, FoxP3, CD83, an

Tertiary lymphoid structures (TLSs) are immune cell aggregates that emerge in nonlymphoid tissues during various disease states, including chronic inflammation, autoimmunity, and cancer. TLSs are structurally and functionally analogous to secondary lymphoid organs, and exhibit a maturation continuum (progressing from initial aggregation to mature structures with germinal centers). TLS formation is synergistically regulated by local chemokine networks (e.g. CXCL13, CCL19, and CCL21), lymphotoxin signaling axes, stromal cells, metabolic reprogramming, and the microbiome. This review comprehensiv

Pilonidal sinus disease (PSD) is characterized by recurrent purulent abscesses or sinus tracts in the sacrococcygeal region. Although its pathophysiology remains unclear, mixed inflammatory infiltrates are seen on histopathology. We aimed to understand the spatial organization of immune cells using histologic and immunofluorescence techniques. We examined skin samples from 10 patients with chronic PSD and 5 site-matched healthy controls. H&E staining revealed dense immune infiltrates in PSD samples, including lymphoid and plasma cell aggregates. Immunofluorescence staining for T cells, B cells

The central nervous system (CNS) constitutes a tightly regulated milieu, where immune responses are strictly controlled to prevent neurological damage. This poses considerable challenges to the therapeutic management of CNS pathologies, such as autoimmune disorders and cancer. Tertiary lymphoid structures (TLS) are ectopic, lymph node-like structures containing B- and T-cells, often associated with chronic inflammation or cancer, which have been shown to be detrimental in autoimmunity but beneficial in cancer. In-depth studies of TLS induction in CNS disorders, as well as their precise role in

Lymphoid organ development occurs in ontogeny in the absence of antigenic stimulation; inflammation occurs after some immunological insult. These subjects had originally been considered as separate fields of research. Inflammation is due to inflammatory mediators such as lymphotoxin (LT) and its close relative, tumor necrosis factor (TNF). The two fields converged with the realization that LT is also crucial for secondary lymphoid organ (SLO) development and that it induces chronic lymphoid infiltrates, called tertiary lymphoid organs (TLOs) or tertiary lymphoid structures (TLSs) that are rema

Tertiary lymphoid structures (TLSs) frequently occur at sites of chronic inflammation. A more advanced stage of multiple sclerosis (MS) has been associated with certain TLSs. However, tumor-associated TLSs have been shown to correlate with a greater treatment response rate and a better prognosis in glioma mouse models. In this review, we evaluate the clinical significances of TLSs in prognosis and treatment response, as well as the status of TLS-directed therapies targeting alternative biochemical pathways in various central nervous system (CNS) disorders. Potential molecular mechanisms underl

Tertiary lymphoid structures (TLSs) are lymphoid cell clusters that form in non-lymphoid tissues in response to chronic inflammation and function as sites for localized, antigen-specific immune responses, potentially enhancing anti-tumor immunity. This review examines TLS presence, composition, and clinical significance across genitourinary (GU) cancers to evaluate their potential as prognostic and therapeutic targets. In prostate cancer, TLSs are infrequently found due to a typically immunologically inactive tumor microenvironment (TME), but when present, they correlate with improved outcomes

Tertiary lymphoid structures (TLS) are essential immune components within the tumor microenvironment. The cellular composition, heterogeneity, and maturity of TLS are closely associated with tumor progression, response to therapy, and patient prognosis. Although numerous studies have highlighted the antitumor potential of TLS, their role can vary-either inhibiting or promoting tumor growth-depending on the specific microenvironment or disease stage. Hepatocellular carcinoma (HCC), the most common and aggressive primary liver cancer, frequently resists immune checkpoint therapy. In this context

Ovarian cancer remains resistant to immunotherapy in most patients, highlighting the need to make these tumors more immunogenic. A recent study unveils how chemotherapy is able to induce cancer cell stress in metastatic ovarian carcinomas, inducing the formation of tertiary lymphoid structures that create a "hotter" tumor microenvironment. See related article by Lanickova et al., p. 164.

Chronic kidney disease is a global health problem with high morbidity and mortality rates. Acute kidney injury substantially increases the risk of chronic kidney disease progression, particularly in the elderly, partly because of prolonged inflammation that exacerbates kidney fibrosis and dysfunction. Tertiary lymphoid structures (TLSs) are ectopic lymphoid aggregates that develop in non-lymphoid organs during chronic inflammation, such as autoimmune diseases, cancers, and age-related inflammation. Age-dependent TLS formation is observed in various organs, such as the kidneys, bladder, lacrima

Treatment with immune checkpoint blockade (ICB) has revolutionized cancer therapy. Until now, predictive biomarkers1-10 and strategies to augment clinical response have largely focused on the T cell compartment. However, other immune subsets may also contribute to anti-tumour immunity11-15, although these have been less well-studied in ICB treatment16. A previously conducted neoadjuvant ICB trial in patients with melanoma showed via targeted expression profiling17 that B cell signatures were enriched in the tumours of patients who respond to treatment versus non-responding patients. Tertiary lymphoid structures (TLS) are ectopic immune aggregates that form in response to chronic inflammation, and their presence across many solid tumors has been associated with improved survival and enhanced responses to immunotherapy. Although current models primarily characterize TLS through the lens of immune aggregation and organization, their formation also depends on reciprocal interactions with the non-immune tumor microenvironment. Fibroblasts, vasculature, extracellular matrix architecture, and tumor metabolism collectively influence TLS development, suggesting that these structur

Mitochondria activate immunity, but the role in cancer is unknown. Here, we report that transgenic expression of Parkin, a regulator of mitochondrial fitness, induces inflammation and interferon (IFN) gene signatures, suppresses prostate cancer formation and generates CD8+ and CD20+ intraprostatic immune aggregates. These had hallmarks of mature tertiary lymphoid structures (TLS), expressing markers of B cell maturation (CXCL13, CCL21), germinal center formation (BCL6, GL7), mature dendritic cells (MHC-II/CD208) and high endothelial venules (LYVE-1). Parkin TLS showed high Ig gene expression,

Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), is a major public health burden responsible for over a million deaths each year. A deeper understanding of the mechanisms that balance protective immunity and immunopathology is essential for developing more effective therapeutics. This review focuses on the dynamic interplay between CD4+ T cells and B cells in the lung, with an emphasis on their interactions in tertiary lymphoid structures (TLS). TLS are immune cell aggregates that arise in inflamed, nonlymphoid tissues, range from loosely to highly organized clusters, and serve a

Checkpoint blockade therapies that reactivate tumour-associated T cells can induce durable tumour control and result in the long-term survival of patients with advanced cancers1. Current predictive biomarkers for therapy response include high levels of intratumour immunological activity, a high tumour mutational burden and specific characteristics of the gut microbiota2,3. Although the role of T cells in antitumour responses has thoroughly been studied, other immune cells remain insufficiently explored. Here we use clinical samples of metastatic melanomas to investigate the role of B cells in
