Elemental sulfur reduction to H2S by Tetrahymena thermophila.

Eukaryotic nucleocytoplasm is believed to be descended from ancient Archaea that respired on elemental sulfur. If so, a vestige of sulfur reduction might persist in modern eukaryotic cells. That was tested in Tetrahymena thermophila, chosen as a model organism. When oxygenated, the cells consumed H2S rapidly, but when made anoxic they produced H2S mostly by amino acid catabolism. That could be inhibited by adding aminooxyacetic acid, and then H2S production from elemental sulfur became more evident. Anoxic cell lysates produced H2S when provided with sulfur and NADH, but not with either substr

Calcium regulates the regeneration of cilia in Tetrahymena thermophila.

A study of calcium metabolism in Tetrahymena during the regeneration of cilia evidenced that the process is inhibited by nifedipine and trifluoperazine. This suggests that calcium ions play an important regulatory role in this process. This was confirmed by studies on calcium uptake and efflux which showed that there was a net increase in calcium uptake prior to the reinitiation of motility. The increase coincided with a period of sensitivity to the calcium antagonist TMB-8 and with an increase in the intracellular level of cGMP. The process was also inhibited by neomycin and stimulated by pho

Organization and pairing of meiotic chromosomes in the ciliate Tetrahymena thermophila.

During meiotic prophase in the ciliate Tetrahymena thermophila micronuclei dramatically elongate and form thread-like crescents. The arrangement of the chromosomes within the crescent as well as the timing of chromosome pairing and recombination with respect to the elongation process have been subjects of ongoing debate. Here, we addressed these issues by means of fluorescence in situ hybridization, labeling of individual chromosomes by BrdU (BrdU-painting) and by immunostaining of the recombination protein, Rad51. BrdU-painting indicated that chromosomes are arranged as parallel bundles withi

Biochemical evidence for a P2Y-like receptor in Tetrahymena thermophila.

Extracellular nucleotides are ubiquitous signaling molecules. ATP signals through two receptor types: the ionotropic P2X receptors, and the metabotropic P2Y receptors. ATP acts as a chemorepellent in Tetrahymena thermophila, where it causes a distinct avoidance response. The intracellular mechanisms by which ATP causes avoidance in this organism, however, are unknown. In this study, we use in vivo pharmacological assays along with enzyme immuno-assays to obtain information about the ATP chemorepellent pathway and its associated second messenger systems. Our data show strong similarities betwee

Characterization and properties of cholesterol desaturases from the ciliate Tetrahymena thermophila.

Live Tetrahymena thermophila transforms exogenous cholesterol into 7,22-bis, dehydrocholesterol (DHC) by desaturation at positions C7(8) and C22(23) of the cholesterol moiety. In this first report on expression, isolation, characterization, and reconstitution of Tetrahymena's cholesterol desaturases in cell-free extracts, we describe conditions for increasing the expression of both desaturases based on the addition of specific sterols to the culture medium. Reactions performed in vitro, with isolated microsomes, yield only the mono-unsaturated derivatives, 7-DHC and/or 22-DHC. However, selecti

MicroRNAs in Tetrahymena thermophila: An epigenetic regulatory mechanism in the response to cadmium stress.

Among the epigenetic mechanisms based on non-coding RNA are microRNAs (miRNAs) that are involved in the post-transcriptional regulation of mRNAs. In many organisms, the expression of genes involved in the cellular response to biotic or abiotic stress depends on the regulation, generally inhibitory, performed by miRNAs. For the first time in the eukaryotic microorganism (ciliate-model) Tetrahymena thermophila, miRNAs involved in the post-transcriptional regulation of transcripts linked to the response to cadmium have been isolated and analyzed. Forty de novo miRNAs (we named tte-miRNAs) have be

Organic matter excreted by the protozoan Tetrahymena thermophila and its effects on the bioaccumulation of nanoparticles.

Although organic matter (exudate) excreted by aquatic organisms is an important component of dissolved organic matter (DOM) in the natural environment, its potential effects on the bioaccumulation of nanoparticles (NPs) remain unclear. In the present study, we examined the effects of the exudates from the protozoan Tetrahymena thermophila on the bioaccumulation (including uptake and cell surface adsorption) of iron oxide (Fe2O3, polyacrylate coated) and silica (SiO2) NPs in T. thermophila. The exudates were mostly (93.6 %, in carbon) composed of < 1-kDa molecules (e.g., lipids). When the exuda

Effects of six pyrimidine analogs on the growth of Tetrahymena thermophila and their implications in pyrimidine metabolism.

Tetrahymena are ciliated protists that have been used to study the effects of toxic chemicals, including anticancer drugs. In this study, we tested the inhibitory effects of six pyrimidine analogs (5-fluorouracil, floxuridine, 5'-deoxy-5-fluorouridine, 5-fluorouridine, gemcitabine, and cytarabine) on wild-type CU428 and conditional mutant NP1 Tetrahymena thermophila at room temperature and the restrictive temperature (37°C) where NP1 does not form the oral apparatus. We found that phagocytosis was not required for pyrimidine analog entry and that all tested pyrimidine analogs inhibited growth

Recombinant Haemagglutinin Derived From the Ciliated Protozoan Tetrahymena thermophila Is Protective Against Influenza Infection.

Current influenza vaccines manufactured using eggs have considerable limitations, both in terms of scale up production and the potential impact passaging through eggs can have on the antigenicity of the vaccine virus strains. Alternative methods of manufacture are required, particularly in the context of an emerging pandemic strain. Here we explore the production of recombinant influenza haemagglutinin using the ciliated protozoan Tetrahymena thermophila. For the first time we were able to produce haemagglutinin from both seasonal influenza A and B strains. This ciliate derived material was im

Preparation and biological effect of nucleotide-capped CdSe/ZnS quantum dots on Tetrahymena thermophila.

In this paper, we described the preparation and characterization of different types of modified CdSe/ZnS quantum dots (QDs) and explored the biological effects of QDs with different surface modifications on the whole growth of unicellular protozoan Tetrahymena thermophila BF(5) using a thermal activity monitor air isothermal microcalorimeter. Our results demonstrated that adenosine 5'-monophosphate (AMP) showed stronger interaction with QDs than other types of nucleotide. AMP-QDs could stimulate the growth of T. thermophila while mercaptoacetic acid-capped CdSe/ZnS quantum dots inhibited it. I

Development of a Tetrahymena thermophila-Based Vaccine Expressing Miamiensis avidus Ciliary Proteins to Combat Scuticociliatosis.

Miamiensis avidus is a parasitic pathogen responsible for scuticociliatosis, a lethal infection affecting marine fish worldwide, including Japanese flounder. Immunisation with formalin-killed M. avidus has shown promise in inducing protective immunity, positioning it as a potential vaccine candidate against scuticociliatosis. However, challenges such as the high cost of producing sufficient cells and inconsistent quality due to the lack of cryopreservation methods hinder its development. In this study, we expressed M. avidus ciliary proteins in Tetrahymena, a culturable ciliate, and used these

Comparative transcriptome analysis uncovers roles of hydrogen sulfide for alleviating cadmium toxicity in Tetrahymena thermophila.

BACKGROUND: Cadmium (Cd) is a nonessential heavy metal with potentially deleterious effects on different organisms. The organisms have evolved sophisticated defense system to alleviate heavy metal toxicity. Hydrogen sulfide (H2S) effectively alleviates heavy metal toxicity in plants and reduces oxidative stress in mammals. However, the function of H2S for alleviating heavy metal toxicity in aquatic organisms remains less clear. Tetrahymena thermophila is an important model organism to evaluate toxic contaminants in an aquatic environment. In this study, the molecular roles of exogenously H2S a

Glutathione Relevance in the Response of Ciliate Tetrahymena thermophila Against Metals/Metalloids and Other Environmental Stressors.

The tripeptide glutathione (GSH) is the primary and most abundant non-protein thiol compound present in all aerobic organisms. The main function of this molecule is cellular antioxidant defense. For the first time in ciliates, using the eukaryotic microorganism Tetrahymena thermophila as a model organism, several parameters (cell mortality, molecules containing -SH groups, and total glutathione) have been evaluated under the toxic effects of various chemical stressors (including several metals/metalloids), either in combination with or without the inhibitory effects of BSO (a GSH biosynthesis

Big1 is a cell-cycle regulator linking cell size to basal body number in Tetrahymena thermophila.

Cell-size control is necessary for the optimum function and fitness of cells. In many eukaryotes, cell size is controlled through the initiation of cell division once a size threshold is achieved.1 Cells sense their size through the relationship of cell-size-dependent and independent factors, though the mechanistic details are known for few eukaryotes.2,3 An additional dimension of cell-size control is the subcellular localization and control of cell-cycle regulators. The cell cortex is a known site of cell-cycle regulator accumulation that maintains an established cell size based on surface a

The H1 phosphorylation state regulates expression of CDC2 and other genes in response to starvation in Tetrahymena thermophila.

In Tetrahymena thermophila, highly phosphorylated histone H1 of growing cells becomes partially dephosphorylated when cells are starved in preparation for conjugation. To determine the effects of H1 phosphorylation on gene expression, PCR-based subtractive hybridization was used to clone cDNAs that were differentially expressed during starvation in two otherwise-isogenic strains differing only in their H1s. H1 in A5 mutant cells lacked phosphorylation, and H1 in E5 cells mimicked constitutive H1 phosphorylation. Sequences enriched in A5 cells included genes encoding proteases. Sequences enrich

Tetrahymena thermophila granule lattice protein 3 improves solubility of sexual stage malaria antigens expressed in Escherichia coli.

The requirement for low cost manufacturing makes bacterial cells a logical platform for the production of recombinant subunit vaccines for malaria. However, protein solubility has been a major stumbling block with prokaryotic expression systems. Notable examples include the transmission blocking vaccine candidates, Pfs25 and Pfs48/45, which are almost entirely insoluble when expressed as recombinant proteins in Escherichia coli. Various solubility tags have been used with limited success in improving solubility, although recent studies with granule lattice protein 1 (Grl1p) from the ciliated p

Protection of Grouper Against Cryptocaryon irritans by Immunization With Tetrahymena thermophila and Protective Cross-Reactive Antigen Identification.

Vaccination is an effective method to prevent Cryptocaryon irritans infection. Although some vaccines have been developed, large-scale production of these vaccines is costly. Development of a heterogenous vaccine generated by low-cost antigens is an alternative method. In the present study, grouper immunized with Tetrahymena thermophila, a free-living ciliate that easily grows in inexpensive culture media at high density, showed protective immunity against C. irritans infection. Higher immobilization against C. irritans theronts was detected in T. thermophila-immunized grouper serum, which sug

Characterization of Cystathionine β-Synthase TtCbs1 and Cysteine Synthase TtCsa1 Involved in Cysteine Biosynthesis in Tetrahymena thermophila.

Cysteine is implicated in important biological processes. It is synthesized through two different pathways. Cystathionine β-synthase and cystathionine γ-lyase participate in the reverse transsulfuration pathway, while serine acetyltransferase and cysteine synthase function in the de novo pathway. Two evolutionarily related pyridoxal 5'-phosphate-dependent enzymes, cystathionine β-synthase TtCBS1 (TTHERM_00558300) and cysteine synthase TtCSA1 (TTHERM_00239430), were identified from a freshwater protozoan Tetrahymena thermophila. TtCbs1 contained the N-terminal heme binding domain, catalytic dom

Photocatalytic degradation of methylene blue by CdS quantum dots biosynthesized by cysteine synthetase TtCsa1 from Tetrahymena thermophila.

H2S is a crucial endogenous gaseous signaling molecule involved in various metabolic pathways and is associated with the biological response to heavy metal stress. The biomineralization of CdS serves as a critical mechanism for responding to cadmium stress and removing Cd2+ in different organisms. CdS is also a widely utilized semiconductor material for the photocatalytic degradation of dyes. In this study, we found that cysteine synthetase 1 (TtCsa1) is involved in CdS formation under cadmium stress in Tetrahymena thermophila. TtCsa1 also catalyzed CdS formation in vitro, and the synthesized

Cystathionine β-synthase TtCbs1 from Tetrahymena thermophila catalyzes the synthesis of CdS quantum dots for methyl orange decolorization.

UNLABELLED: Cystathionine β-synthase (CBS) is a pivotal enzyme in the transsulfuration metabolic pathway, catalyzing the synthesis of H2S and cystathionine. These metabolites mediate stress responses and maintain cellular redox homeostasis. In Tetrahymena thermophila, TtCBS1 expression was upregulated under cadmium stress, with cysteine further enhancing its transcription. The recombinant His-TtCbs1 protein was expressed in Escherichia coli and purified by affinity chromatography. The His-TtCbs1 catalyzed the synthesis of monodisperse cadmium sulfide quantum dots (CdS QDs) in cysteine and Cd2+
