Small membranous extracellular vesicles (EV) incorporate proteins and nucleic acids from the parent cell. Proteins exposed on EV surface are dictated by cellular origin and biogenesis pathway. To better understand the EV origin and function, it is important to develop methods that reveal surface protein composition of heterogeneous EV populations in culture supernatants and in biofluids. Tetraspanins CD9, CD63, and CD81 are common and abundant EV markers. However, their relative enrichment (profile) on EVs of specific cellular origins is not fully elucidated. We introduce LuminEV, a novel vers

Exosomes are secreted vesicles of ~30 to 150 nm diameter that play important roles in human health and disease. To better understand how cells release these vesicles, we examined the biogenesis of the most highly enriched human exosome marker proteins, the exosomal tetraspanins CD81, CD9, and CD63. We show here that endocytosis inhibits their vesicular secretion and, in the case of CD9 and CD81, triggers their destruction. Furthermore, we show that syntenin, a previously described exosome biogenesis factor, drives the vesicular secretion of CD63 by blocking CD63 endocytosis and that other endo

Extracellular vesicles (EVs) are natural, cell-derived nanoparticles released into biofluids, such as plasma, and hold great potential as a new class of biomarkers. However, the utility of analyzing EVs in clinical samples has been hampered by a lack of suitable EV isolation methods that can be performed reproducibly in a scalable manner. The current method of choice for isolating EVs, size exclusion chromatography (SEC), is performed manually one column at a time, and thus does not have the throughput for isolating EVs from clinical samples. In this work, we adapt SEC to a plate-based format

BACKGROUND: Retinal degenerative diseases (RDD) cause irreversible vision loss due to photoreceptor (PR) loss. Stem cell-derived PR precursors hold promise for retinal repair, but genetic labeling limits clinical translation. Surface marker-based sorting offers a safer alternative.
METHODS: We analyzed single-cell RNA sequencing (scRNA-seq) datasets of human retinal organoids to identify PR-specific surface markers and validated them by immunofluorescence and flow cytometry. Cells with a CD9⁻CD81mid (negative for CD9 and intermediate levels of CD81) phenotype were isolated via fluorescence-ac

This study introduces a highly sensitive fluorescence array that leverages immunosorbent reversible addition-fragmentation chain transfer (immuno-RAFT) signal amplification for detecting CD81 protein, a key biomarker used to diagnose preeclampsia. By integrating the operational efficiency of traditional enzyme-linked immunosorbent assay (ELISA) in 96-well plates with the robust signal amplification capabilities of RAFT technology, this method enables rapid and quantitative detection of low-concentration CD81 protein. This advancement provides substantial theoretical and technical support for t

Hepatitis C virus (HCV) is the second viral agent that causes the majority of chronic hepatic infections worldwide, following Hepatitis B virus (HBV) infection. HCV infection comprises several steps, from the attachment to the receptors to the delivery of the viral genetic material and replication inside the cells. Tetraspanin CD81 is a key entry factor for HCV as it accompanies the virus during attachment and internalization through clathrin-mediated endocytosis. HCV-CD81 binding takes place through the viral glycoprotein E2. We performed full-atom molecular dynamics simulations reproducing t

Tetraspanin family proteins, CD9, CD81 and CD82 are expressed in the oligodendrocytes and Schwann cells. We investigated autoantibodies to tetraspanin proteins in patients with demyelinating diseases. Sera were collected from 119 multiple sclerosis patients, 19 neuromyelitis optica, 42 acute inflammatory demyelinating polyneuropathy, 23 chronic inflammatory demyelinating polyneuropathy and 13 acute motor axonal neuropathy as well as 55 healthy controls. Few multiple sclerosis and acute inflammatory demyelinating polyneuropathy patients had autoantibodies that were weakly reactive to CD9 or CD8

Burkholderia pseudomallei is the causative agent of melioidosis, a disease with high morbidity that is endemic in South East Asia and northern Australia. An unusual feature of the bacterium is its ability to induce multinucleated giant cell formation (MNGC), which appears to be related to bacterial pathogenicity. The mechanism of MNGC formation is not fully understood, but host cell factors as well as known bacterial virulence determinants are likely to contribute. Since members of the tetraspanin family of membrane proteins are involved in various types of cell:cell fusion, their role in MNGC

Tetraspanins are four-span transmembrane proteins of host cells that facilitate infections by many pathogens. Burkholderia pseudomallei is an intracellular bacterium and the causative agent of melioidosis, a severe disease in tropical regions. This study investigated the role of tetraspanins in B. pseudomallei infection. We used flow cytometry to determine tetraspanins CD9, CD63, and CD81 expression on A549 and J774A.1 cells. Their roles in B. pseudomallei infection were investigated in vitro using monoclonal antibodies (MAbs) and recombinant large extracellular loop (EC2) proteins to pretreat

Extracellular vesicles (EVs) are nanoscale particles secreted by cells that carry diverse biomolecules reflecting their cell of origin. Single-EV imaging approaches have enabled precise characterization of heterogeneous EV populations; however, their broader application is limited by low-throughput workflows and cumbersome EV isolation procedures. Here, we introduce a streamlined, high-throughput imaging platform capable of analyzing protein expression of individual intact EVs directly from unprocessed biological samples at the single-vesicle level. Our approach employs a functionalized glass

Extracellular vesicles (EVs) facilitate intercellular communication and have emerged as valuable diagnostic and prognostic biomarkers, therapeutic tools, and delivery systems. Due to their heterogeneity, precise profiling of EVs requires sub-EV resolution beyond standard EV biomarkers. Additionally, there is a growing need to trace the biogenesis of individual EVs. While super-resolution microscopy enables EV profiling at the nanoscale with biochemical specificity, the lack of standardized workflows for EV purification, immobilization, and labeling continues to limit reproducibility and biolog

Hepatitis C virus (HCV) infection is a major public health concern, and almost two million people are infected per year globally. This is occurred by the diverse spectrum of viral genotypes, which are directly associated with chronic liver disease (fibrosis, and cirrhosis). Indeed, the viral genome encodes three principal proteins as sequentially core, E1, and E2. Both E1 and E2 proteins play a crucial role in the attachment of the host system, but E2 plays a more fundamental role in attachment. The researchers have found the "E2-CD81 complex" at the entry site, and therefore, CD81 is the key

Submembrane cortical actin (CA) plays a large role in regulating the dynamic organization of cell surface receptors, which in turn regulates receptor signaling. Many receptors have short intracellular domains and no known link to actin. Here, we identified the β1-integrin subunit and several tetraspanins (CD9, CD81, and CD151) as part of the hitherto unknown molecular link between the receptor CD36 and CA. Our data indicate that CD36 in vascular endothelial cells interacts with these proteins, with stronger interactions near the cell edge. Compromising these interactions via the point mutation

Human CD81 and CD9 are members of the tetraspanin family of proteins characterized by a canonical structure of four transmembrane domains and two extracellular loop domains. Tetraspanins are known as molecular facilitators, which assemble and organize cell surface receptors and partner molecules forming clusters known as tetraspanin-enriched microdomains. They have been implicated to play various biological roles including an involvement in infections with microbial pathogens. Here, we demonstrate an important role of CD81 for the invasion of epithelial cells by Salmonella enterica. We show th

Gastric infection by Helicobacter pylori (H. pylori) is associated with extra-gastric pathologies. H. pylori can promote these pathologies by generating extracellular vesicles (Hp-EVs), which cross biological barriers and access other tissues through the circulation. However, the presence of Hp-EVs in the human circulation has not been demonstrated. We isolated and characterized EVs from serum of non-infected and infected patients, and used immunoblots (IB) to identify markers from host cells EVs (h-EVs) and Hp-EVs. We also assessed the predictive capacity of IB with an anti-Urease subunit B (

The tuberous sclerosis complex (Tsc) proteins regulate the conserved mTORC1 growth regulation pathway. We identified that loss of the Tsc2 gene in mouse inner medullary collecting duct (mIMCD) cells induced a greater than two-fold increase in extracellular vesicle (EV) production compared to the same cells having an intact Tsc axis. We optimized EV isolation using a well-established size exclusion chromatography method to produce high purity EVs. Electron microscopy confirmed the purity and spherical shape of EVs. Both tunable resistive pulse sensing (TRPS) and dynamic light scattering (DLS) d

The tetraspanin CD81 is one of the main entry receptors for Hepatitis C virus, which is a major causative agent to develop liver cirrhosis and hepatocellular carcinoma (HCC). Here, we identify CD81 as one of few surface proteins that are downregulated in HCV expressing hepatoma cells, discovering a functional role of CD81 beyond mediating HCV entry. CD81 was downregulated at the mRNA level in hepatoma cells that replicate HCV. Kinetics of HCV expression were increased in CD81-knockout cells and accompanied by enhanced cellular growth. Furthermore, loss of CD81 compensated for inhibition of pro

BACKGROUND: The global outbreak of COVID-19, and the limited availability of clinical treatments, forced researchers around the world to search for the pathogenesis and potential treatments. Understanding the pathogenesis of SARS-CoV-2 is crucial to respond better to the current coronavirus disease 2019 (COVID-19) pandemic.
METHODS: We collected sputum samples from 20 COVID-19 patients and healthy controls. Transmission electron microscopy was used to observe the morphology of SARS-CoV-2. Extracellular vesicles (EVs) were isolated from sputum and the supernatant of VeroE6 cells, and were char

The Parkinson's disease-associated protein, alpha-synuclein (α-syn; SNCA) is suspected of promoting melanoma progression. We recently knocked out SNCA in the human cutaneous melanoma cell line SK-MEL-28 to try to deduce the role of α-syn in melanoma progression. Compared to control cells, the SK-MEL-28 SNCA-knockout (KO) cells have significantly inhibited growth, invasion, and migration, and the levels of the neural adhesion protein L1CAM and the transferrin receptor (TFR1) are significantly reduced. In this study, using transmission electron microscopy and immunofluorescence we show that SK-M
