Papillary thyroid cancer (PTC) is a heterogeneous malignancy in which current prognostic markers often fail to distinguish between indolent and aggressive disease. The need for reliable biomarkers is especially urgent in cases of angioinvasion, where improved risk stratification could guide decisions such as extended surgery and early radioiodine therapy. This study evaluates thioredoxin (TRX), a redox-regulating protein, and tetraspanin 30 (CD63), a mediator of tumor-endothelial interactions, as potential non-invasive biomarkers of angioinvasive PTC. We analyzed serum samples from 90 patients

Small membranous extracellular vesicles (EV) incorporate proteins and nucleic acids from the parent cell. Proteins exposed on EV surface are dictated by cellular origin and biogenesis pathway. To better understand the EV origin and function, it is important to develop methods that reveal surface protein composition of heterogeneous EV populations in culture supernatants and in biofluids. Tetraspanins CD9, CD63, and CD81 are common and abundant EV markers. However, their relative enrichment (profile) on EVs of specific cellular origins is not fully elucidated. We introduce LuminEV, a novel vers

BACKGROUND: Exosomes, as key mediators of intercellular communication, are increasingly implicated in the pathogenesis of various diseases. Multiomics investigations have uncovered functional roles of low-molecular-weight biomolecules in exosomes. However, conventional methods for isolating exosomes from biological matrices are time-consuming and laborious, limiting the sample preparation for multiomics analysis.
RESULTS: We proposed an innovative approach termed as multi-exosomics, which integrates a rapid isolation protocol and multiomics for simultaneous analysis of exosomal metabolites an

Small extracellular vesicles (sEVs) play pivotal roles in modulating the pathological processes of various diseases and have emerged as promising biomarkers for disease diagnosis, including acute spinal cord injury and cancers. This is attributed to their ability to transport multiple proteins that reflect the molecular signatures of their parent cells. The evaluation of surface proteins presents a robust strategy for identifying a comprehensive set of biomarkers. In this study, we developed a high-throughput device capable of characterizing surface proteins on intact sEVs. Our approach employ

Exosomes are secreted vesicles of ~30 to 150 nm diameter that play important roles in human health and disease. To better understand how cells release these vesicles, we examined the biogenesis of the most highly enriched human exosome marker proteins, the exosomal tetraspanins CD81, CD9, and CD63. We show here that endocytosis inhibits their vesicular secretion and, in the case of CD9 and CD81, triggers their destruction. Furthermore, we show that syntenin, a previously described exosome biogenesis factor, drives the vesicular secretion of CD63 by blocking CD63 endocytosis and that other endo

BACKGROUND: Exosomes (EXOs) are promising biomarker sources for liquid biopsy. Due to their low abundance in biofluids and the presence of non-target vesicle proteins during the isolation process, traditional methods such as ultracentrifugation (UC) are time-consuming, yield low purity, and can cause damage to EXOs. Achieving rapid, high-purity isolation remains a significant challenge. The magnetic beads (MBs) method is the most effective way to enhance EXO purity, but existing MBs face bottlenecks, including poor suspension stability, limited surface functionalization, slow magnetic response

Extracellular vesicles (EVs) are natural, cell-derived nanoparticles released into biofluids, such as plasma, and hold great potential as a new class of biomarkers. However, the utility of analyzing EVs in clinical samples has been hampered by a lack of suitable EV isolation methods that can be performed reproducibly in a scalable manner. The current method of choice for isolating EVs, size exclusion chromatography (SEC), is performed manually one column at a time, and thus does not have the throughput for isolating EVs from clinical samples. In this work, we adapt SEC to a plate-based format

The important role of extracellular vesicles, which are considered key mediators of intercellular communication under physiological and pathological conditions, in various cellular processes, including those crucial for mammalian reproduction, has been increasingly studied. Tetraspanins, including CD63, are widely used as markers of extracellular vesicles, but they may also play a role in their biogenesis, cargo selection, cell targeting, and uptake. This study aimed to map the journey of the extracellular vesicle protein tetraspanin CD63 from the testis through the epididymis into mature bull

Extracellular vesicles (EVs) are lipid bilayer-bound nanoparticles secreted by nearly all cells, with notable applications in intercellular communication and therapeutic delivery. However, conventional EV isolation techniques, such as ultracentrifugation and size exclusion chromatography, often face challenges like high cost, specialized equipment requirements, and low yield. In this study, we present a modified aqueous two-phase system (ATPS) as an alternative method for isolating EVs from raw bovine milk. This approach is scalable, cost-effective, and requires minimal specialized equipment,

To prepare antibodies that specifically recognize the conserved domain in the large extracellular loop of the CD63 protein, we expressed anti-CD63 single-chain variable fragment (scFv) antibody in Pichia pastoris in a secreted form. The purified expression product was found to bind specifically with CD63 protein and recognize CD63 on the surface of SK-MEL-28 cells. The variable region of the anti-CD63 monoclonal antibody in an anti-CD63-positive cell line was sequenced. The anti-CD63 scFv consisted of a variable heavy chain and a variable light chain linked by a flexible peptide was then desig

Hermansky-Pudlak syndrome (HPS) is an autosomal recessive disorder characterized by clinical features including oculocutaneous albinism (OCA) and bleeding diathesis due to platelet storage pool deficiency. In this article, we report a 12-year-old boy with recurrent epistaxis who was referred to the Iranian Blood Transfusion Organization (IBTO) reference coagulation laboratory for platelet function analysis. Based on the laboratory diagnostic tests in this study and the patient's clinical presentation, the probability of HPS type 2 is more likely.

Despite the adenoids are regularly removed in patients with mucopolysaccharidoses (MPS), the underlying tissue and cellular pathologies remain understudied. We characterized an (immuno)histopathologic and ultrastructural phenotype dominated by lysosomal storage changes in a specific subset of adenotonsillar paracortical cells in 8 MPS patients (3 MPS I, 3 MPS II, and 2 MPS IIIA). These abnormal cells were effectively detected by an antibody targeting the lysosomal membrane tetraspanin CD63. Important, CD63+ storage vacuoles in these cells lacked the monocytes/macrophages lysosomal marker CD68.

Small extracellular vesicles and nanoparticles (sEVPs) are cell-secreted entities with potential as diagnostic biomarkers and therapeutic vehicles. However, significant intrinsic sEVP heterogeneity impedes analysis and understanding of their composition and functions. We employ multidimensional fluorescent labelling on sEVPs, leveraging the robustness of a newly developed membrane probe-conjugated oligoelectrolytes (COEs), and conduct total internal reflection fluorescence (TIRF) microscopy on sEVP arrays. These arrays comprise single sEVPs anchored to a soft material functionalized surface wi

The considerable abundance and remarkable stability of sEVs provide substantial benefits for diagnosing Alzheimer's disease. Therefore, precise tracking subtypes of small extracellular vesicles (sEVs) is crucial for screening novel diagnostic biomarkers and developing therapeutic technologies. We propose a three-target recognition-mediated proximity ligation assay for the precise identification of sEV subtypes utilizing three specifically designed probes: one for the exosomal surface protein CD63 recognition, one for fixing the biolipid layer, and the third for the identification of distinctiv

Large dense-core vesicles (LDCVs) are characterized as a class of lysosome-related organelles (LROs), which undergo regulated release and play important roles in development, metabolism and homeostasis. The Muted protein is a subunit of the biogenesis of lysosome-related organelles complex-1 (BLOC-1), which functions in the biogenesis of lysosomes and LROs. CD63 is a membrane component of lysosomes and LROs. Whether and how CD63 is sorted into LDCVs is largely unknown. In this study, we aim to identify the localization of CD63 in chromaffin cells by colocalization, living cell imaging and cell

Burkholderia pseudomallei is the causative agent of melioidosis, a disease with high morbidity that is endemic in South East Asia and northern Australia. An unusual feature of the bacterium is its ability to induce multinucleated giant cell formation (MNGC), which appears to be related to bacterial pathogenicity. The mechanism of MNGC formation is not fully understood, but host cell factors as well as known bacterial virulence determinants are likely to contribute. Since members of the tetraspanin family of membrane proteins are involved in various types of cell:cell fusion, their role in MNGC

BACKGROUND: Autophagy is an intracellular degradation process involving the lysosomal breakdown of unnecessary or abnormal cellular components. Targets of autophagy include pathogens, altered organelles, and protein aggregates, which are sequestered within double-membrane-limited vesicles known as autophagosomes. A variety of lysosomal membrane proteins play essential roles in numerous cellular processes, including lysosomal acidification, metabolite transport, and interaction with other membrane systems. Cluster of differentiation 63 (CD63), a member of the transmembrane 4 superfamily, is pre

Salivary exosomes are extracellular vesicles (EVs) with abundant CD63 immunoreactivity on their surface. Based on their size and protein composition, these exosomes can be categorized into two classes of exosomes I (mean diameter of 83.5 nm) and II (mean diameter of 40.5 nm). We have attempted to review the features of these exosomes, including origin, composition, separation methods, and their application in medicine. Not only the composition of salivary exosomes is invaluable in term of diagnosis, but can also afford an understanding in roles of the contents and components of these exosomes
