Identification of Thalictrum squarrosum as an alternate host for Puccinia triticina and pathogen analysis of Thalictrum squarrosum rust.

Puccinia triticina (Pt) is a heteroecious fungus needing two different plants as primary and alternate hosts throughout its life cycle. Thalictrum spp. were first identified as alternate hosts of Pt in 1921, and over 100 species have been identified. However, within China, only T. petaloideum L., T. minus L., T. minus var. hypoleucum and T. baicalense have been reported as alternate hosts of Pt. During the six-year (2015-2018, 2023-2024) field surveys in Zhangbei County (41.26°N, 115.14°E), Zhangjiakou City, Hebei Province, our research team found rust disease on T. squarrosum. This persistent

[Anti-tumor effect of hernandezine and other components extracted from Thalictrum glandulosissimum].

The total alkaloids of T. glandulosissimum and its main component hernandezine were found to be effective for treatment of mice bearing P388 leukemia, S180 ascites and C26 colon cancer. Although hernandezine inhibited the growth of mouse L1210 cells and human oral cancer KB cells in vitro markedly, its inhibitory effect on normal hemopoietic progenitor cells (CFU-GM) in mice was relatively low. Preliminary results showed that hernandezine blocked cell-cycle transfer from G1 to S phase, and its cytocidal action might be cell cycle specific. In addition, two other components, thalidezine and iso

Identification of chemical components in Thalictrum glandulosissimum using UPLC-Q-TOF-MS/MS and pharmacokinetic analysis of multiple alkaloids in normal and ulcerative colitis rats.

Thalictrum glandulosissimum (TG) is traditionally used for treating ulcerative colitis (UC). Given the continuously increasing global incidence of UC and the limitations of current therapeutic options, there is an urgent need for safer treatments. However, most existing studies have focused on single component. Studies on the multi-component profile of TG's active alkaloids, particularly systematic pharmacokinetic investigations under pathological conditions, remain insufficient. Without a systematic understanding of which components are absorbed into the bloodstream and how their disposition

Understanding the Enzyme (S)-Norcoclaurine Synthase Promiscuity to Aldehydes and Ketones.

The (S)-norcoclaurine synthase from Thalictrum flavum (TfNCS) stereoselectively catalyzes the Pictet-Spengler reaction between dopamine and 4-hydroxyphenylacetaldehyde to give (S)-norcoclaurine. TfNCS can catalyze the Pictet-Spengler reaction with various aldehydes and ketones, leading to diverse tetrahydroisoquinolines. This substrate promiscuity positions TfNCS as a highly promising enzyme for synthesizing fine chemicals. Understanding carbonyl-containing substrates' structural and electronic signatures that influence TfNCS activity can help expand its applications in the synthesis of differ

A pharmacological study of two bisbenzylisoquinoline alkaloids, thalistyline and obamegine.

Thalistyline, a monoquaternary bisbenzylisoquinoline alkaloid isolated from Thalictrum sp. inhibited respiration in anesthetized dogs. Thalistyline is about one-fourth as potent as d-tubocurarine in blocking neuromuscular transmission in the rat hemidiaphragm preparation. The pharmacological mechanism of action of the alkaloid is similar to that of d-tubocurarine. Obamegine did not exhibit curare-like activity. On the isolated rabbit aorta, contractions induced by an alpha-adrenoreceptor agonist, phenylephrine, were antagonized by both alkaloids. Increasing concentrations of thalistyline produ

Role of the phloem in the biochemistry and ecophysiology of benzylisoquinoline alkaloid metabolism.

Benzylisoquinoline alkaloids (BIAs) are a diverse group of biologically active specialized metabolites produced mainly in four plant families. BIA metabolism is likely of monophyletic origin and involves multiple enzymes yielding structurally diverse compounds. Several BIAs possess defensive properties against pathogenic microorganisms and herbivores. Opium poppy (Papaver somniferum: Papaveraceae) has emerged as a model system to investigate the cellular localization of BIA biosynthesis. Although alkaloids accumulate in the laticifer cytoplasm (latex) of opium poppy, corresponding biosynthetic

Cepharanthine analogs mining and genomes of Stephania accelerate anti-coronavirus drug discovery.

Cepharanthine is a secondary metabolite isolated from Stephania. It has been reported that it has anti-conronaviruses activities including severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2). Here, we assemble three Stephania genomes (S. japonica, S. yunnanensis, and S. cepharantha), propose the cepharanthine biosynthetic pathway, and assess the antiviral potential of compounds involved in the pathway. Among the three genomes, S. japonica has a near telomere-to-telomere assembly with one remaining gap, and S. cepharantha and S. yunnanensis have chromosome-level assemblies. Following b

Hernandezine alleviates inflammatory bowel disease by enhancing gut barrier integrity via the AMPK/NRF2 pathway and regulating immune-metabolic balance.

Inflammatory bowel disease (IBD), comprising Crohn's disease and ulcerative colitis, is characterized by chronic inflammation and impaired intestinal barrier function. Current therapeutic approaches primarily aim to suppress the overly active immune response using aminosalicylates and immunosuppressive agents, which are frequently associated with significant adverse effects. In this study, we evaluated the therapeutic potential and underlying mechanisms of Hernandezine (HER), a bioactive compound extracted from Thalictrum, using a dextran sulfate sodium (DSS)-induced murine colitis model. Cyanogenesis in Aralia spinosa (Araliaceae).

A systematic survey of Aralia spinosa (Araliaceae), covering an entire growing season and including aboveground organs at various developmental stages, revealed that only about half of all samples collected showed cyanogenesis. Cyanogenesis was detected in inflorescences and leaves but is apparently restricted to certain harvest times or developmental stages. The structurally unusual triglochinin, characterized by a hex-2-enedioic acid partial structure, was the only cyanogenic glycoside detected. This is the first description of triglochinin in this species and in the family of Araliaceae. Tr
