Review of Australian species of Theileria, with special reference to Theileria buffeli of cattle.

This review describes 4 species of parasites in Australia generally regarded as Theileria. The physiological and epidemiological characteristics of the species that occurs in cattle are discussed. Confusion still surrounds the relationship of this species with benign species of Theileria in other countries. Arguments are presented to support the proposal that the bovine species in Australia is correctly designated Theileria buffeli and that this name should apply to all the benign species of Theileria of cattle in Eurasia. The other 3 species considered in the review are proposed species from

Theileria orientalis: a review.

Theileria orientalis (also known historically as T. sergenti and T. buffeli) is responsible for benign or non-transforming theileriosis, and exerts its major effect through erythrocyte destruction. The life cycle of T. orientalis is essentially similar to that of other Theileria species, except that the schizonts do not induce transformation and fatal lymphoproliferation. The pathogenesis of anaemia as a result of infection is not clearly established and may be multifaceted. Clinical signs of weakness, reluctance to walk and abortion are early but non-specific indications of disease, particula

Simultaneous detection of Theileria annulata and Theileria orientalis infections using recombinase polymerase amplification.

Theileriosis is a disease of domesticated animals in tropical and subtropical countries and causes significant reductions in livestock productivity. The arid region of Punjab in Pakistan is notorious for the presence of the vector tick (Acari: Ixodidae) and tick-borne diseases, such as theileriosis and babesiosis. The distribution of Theileria annulata and T. orientalis in the Chakwal district of Punjab was determined by developing a multiplex recombinase polymerase amplification (RPA) assay as a scientific basis for formulating control strategies for bovine theileriosis. Specificity was evalu

Prevalence and genetic characterization of Theileria annulata and Theileria orientalis in Post-abortion cattle.

Tropical theileriosis is a significant health risk for cattle especially in tropical and subtropical regions where it contributes to high mortality rates and economic losses. Tick-borne protozoan parasites belonging to Theileria genera are responsible for this infection. Theileriosis has been associated with abortion and stillbirth in cattle, primarily as an indirect consequence of maternal illness; however, this association has not been investigated in Pakistan. Therefore, the present study aimed to determine the molecular prevalence and distribution of Theileria species in cattle with a rece

Proliferation of Theileria annulata and Theileria parva macroschizont-infected bovine cells in scid mice.

Theileria annulata and Theileria parva macroschizont-infected bovine cells formed tumours at the inoculation site when injected subcutaneously into C.B.-17 scid mice. T. annulata tumours showed more vigorous growth than T. parva tumours. The tumours did not regress and infected cells spread to other tissues. Intraperitoneal injection of high doses of T. annulata-infected cells resulted in the development of ascites: the infected cells colonized abdominal organs, in particular mesenteric tissue. Low doses of cells did not establish when administered by this route. Evidence for a role for macrop

Investigation of Theileria spp. in Livestock in Northern Algeria and Identification of a Novel Haplotype of Theileria orientalis.

PURPOSE: Theileria species are tick-borne, intracellular apicomplexan protozoan parasites that pose a significant threat to livestock health and productivity. Theileria orientalis, the etiological agent of oriental theileriosis, is a benign Theileria species with worldwide distribution, including Algeria, where investigations on its prevalence and genetic diversity in domestic ruminants are limited. This study aimed to investigate the molecular and phylogenetic profiles of Theileria spp. in Algeria. METHODS: A total of 149 blood samples were collected from cattle, sheep, and goats in Bouira a

Theileria ovis discovered in China.

A polymerase chain reaction (PCR) using 989/990 primers was conducted to identify a newly isolated Theileria sp. in Xinjiang Province of China. The target DNA fragments of the complete 18S rRNA gene were cloned and sequenced. The phylogenetic relationship of newly isolated Theileria spp. was inferred based on the 18S rRNA gene. The results showed that the new Theileria sp. belonged to the cluster of Theileria ovis. Moreover, the findings were confirmed by T. ovis species-specific PCR. An expected 520 bp fragment of T. ovis DNA was obtained from 25 out of 320 (8%) field blood samples, and blood

Detection of naturally occuring mixed infection of Theileria lestoquardi and Theileria annulata in buffalo, in Pakistan.

Theileriosis caused by Theileria lestoquardi (malignant ovine theileriosis) in sheep and Theileria annulata (topical theileriosis) in cattle, is a tick-borne hemoprotozoal disease that causes major economic losses in animal production caused by a tick-transmitted haemoprotozoan Theileria species, infecting various domestic and wild animals. It is a threat to the buffalo population in Pakistan due to high mortality rates and economic losses. Extensive mixed farming in Pakistan poses threats due to mixed infections and increased mortality rates. The current case report describes the mixed infect

Babesia, Theileria, Plasmodium and Hemoglobin.

The Propagation of Plasmodium spp. and Babesia/Theileria spp. vertebrate blood stages relies on the mediated acquisition of nutrients available within the host's red blood cell (RBC). The cellular processes of uptake, trafficking and metabolic processing of host RBC proteins are thus crucial for the intraerythrocytic development of these parasites. In contrast to malarial Plasmodia, the molecular mechanisms of uptake and processing of the major RBC cytoplasmic protein hemoglobin remain widely unexplored in intraerythrocytic Babesia/Theileria species. In the paper, we thus provide an updated co

First molecular detection of Theileria capreoli and Theileria cervi in Haemaphysalis longicornis from New Zealand.

The tick Haemaphysalis longicornis Neumann, 1901 is a vector of important medical and veterinary pathogens. In New Zealand, it primarily affects livestock, with impacts on animal health and economic productivity, and to date it has been reported only as a vector of Theileria orientalis, which can cause theileriosis in cattle. This exploratory study aimed to investigate the presence of piroplasmids in convenience samples of H. longicornis collected from farmed livestock (cattle, sheep, deer, and horses) across different regions of New Zealand's North Island. A total of 457 ticks were analysed f

[Molecular diagnostics of Babesia and Theileria].

Protozoa Babesia and Theileria are a group of parasites transmitted by ticks and invading mammals' blood cells. Infections by these protozoa cause heavy losses among farm animals. Since human babesiosis was described forty years ago, the number of reported cases has been increasing. Conventional methods of parasitological diagnostics are unreliable in these diseases, especially in stages with low parasitemia. Methods of nucleic acids detection have been developing fast in recent years and are more and more specific and sensitive. Polymerase Chain Reaction (PCR) enables detection of DNA of Babe

Characterization of species and strains of Theileria.

A variety of methods is now available for characterizing species and strains of Theileria. For many practical purposes involving field control of theileriosis, characterization on a broad basis may be sufficient, but in other areas much more precise characterization is required. Such precision can be usefully exploited only when cloned parasite populations are involved, and methods to improve parasite characterization and parasite cloning should be developed concurrently. The current methods of immunization against theileriosis involve the use of live parasite populations which are generally p

Discovery of a new Theileria sp. closely related to Theileria annulata in cattle from Sri Lanka.

Theileria annulata is a haemoprotozoan parasite that causes a cancer-like illness known as tropical theileriosis in cattle. In the course of analyzing the genetic diversity of T. annulata in Sri Lanka, we observed that merozoite-piroplasm surface antigen (tams1) and surface protein (tasp)-like gene sequences obtained from bovine blood DNA samples, which were PCR-positive for T. annulata, were conserved but shared low identity with T. annulata GenBank sequences. Moreover, the 18S rRNA sequences from the Sri Lankan samples contained ten unique single-nucleotide polymorphisms compared with all kn

Molecular characterization of Theileria species and Theileria parva genetic diversity associated with bovine theileriosis outbreaks in Zimbabwe.

Several Theileria species infect cattle and wildlife in eastern and southern Africa. Among them, Theileria (T.) parva is the most economically important, causing severe disease and substantial livestock losses in Zimbabwe. Recent shifts in spatio-temporal patterns of theileriosis outbreaks highlight the need for updated information on circulating Theileria species and strains to better understand the infection process and to guide control strategies. Between November 2020 and May 2021, 147 whole blood, 145 dried blood spots, 87 fecal, and 22 organ samples were collected from 158 cattle in Bind

Molecular detection and characterization of Anaplasma ovis, Theileria ovis, and Theileria lestoquardi in sheep and goats in Luxor, Egypt.

BACKGROUND: Tick-borne diseases cause economically significant losses to animal production globally, and anaplasmosis and theileriosis are associated with the greatest losses. However, the spread of the relevant pathogens in flocks of domesticated animals in southern Egypt is little understood. Accordingly, in this study, we aimed to determine the prevalences of Anaplasma ovis, Theileria ovis, and Theileria lestoquardi in southern Egyptian sheep and goats through blood tests, and to make a molecular characterization of the A. ovis detected in sheep targeting a specific gene. RESULTS: We colle

A perspective on Theileria equi infections in donkeys.

The donkey population has remained unchanged in the last two decades despite a decrease in the overall population of equids, emphasizing the usefulness of the donkey as a draught and pack animal. Piroplasmosis in donkeys, caused by Theileria equi and Babesia caballi, has been recognized as a serious problem of major economic importance as the affected animals manifest decreased working capacity, loss of appetite, etc. In tropical countries, T. equi infections are more wide-spread and pathogenic than those caused by B. caballi. Donkeys usually remain asymptomatic carriers with positive antibody
